Effects of ChangbuDodamTang on the Polycystic Ovaries Induced by Estradiol Valerate in Rats

Purpose : This study was designed to investigate Effects of ChangbuDodam-Tang(CDT) on the Polycystic Ovaries induced by Estradiol Valerate(EV) in Rats. Materials and Methods : A type of polycystic ovary resembling some aspects of human polycystic ovarian syndrome can be induced in the rat with a single injection of long-acting EV. Among several theories behind the polycystic ovary, the involvement of sympathetic nervous system draws much attention. This study was conducted to prove the possible hypothesis of neurogenic participation in the form of nerve growth factor(NGF) in the pathology of ovarian dysfunction. CDT used in the present study is known to relieve the abnormal symptoms of polycystic ovary. The morphological changes of ovaries were also evaluated. The administration of CDT was done every other day for 60 days. Results : The results from CDT treatment were compared to those from oil-treated control group and EV-injected group. The nerve growth factor was analyzed in the central nervous system and ovaries by immunocytochemistry. The main findings of the present study were 1) polycystic ovary was fully developed in the rat with a single intramuscular injection of estradiol valerate, 2) polycystic ovary resulted in the expression of NGF-immunoreactive material in the ovaries and the brain tissues, and 3) CDT administration significantly decreased the elevated NGF staining in the ovaries without affecting the brain tissues. Conclusion : This study shows that ChangbuDodamTang have beneficial effects Polycystic Ovaries.
A Study on the inhibitory effect of Gaejibokryungwhan on the growth of Human Uterine Myomal Cells

This study was designed to examine the inhibitory effect of Gaejibokryungwhan on the growth of human uterine myomal cells. Human uterine myomal cells were collected from a consented of a patient(age of 39) with myoma of uterus. The cells were induced to grow by being cultured in a Dulbecco's modified eagle medium (DMEM) culture solution containing 10
fetal bovine serum (FBS). For the direct inhibitory effects of Gaejibokryungwhan, the uterine myomal cells and monolayered uterine myomal cells were cultured in the DMEM containing different concentration of Gaejibokryungwhan solution. Afterwards, I executed the analysis of the effect of Gaejibokryungwhan solution on cell growth inhibition using molecular biological method through MAP kinase activity and a expression of bcl-2 in uterine myomal cells. The growth of the human uterine myomal cells were increased in a culture solution containing with FBS whereas cell growth inhibition was observed in cell cultures treated with Gaejibokryungwhan solution. Especially 0.5
Gaejibokryungwhan solution was decreased in a culture solution after the formation of monolayer. MAP kinase activity in uterine myomal cells cultured with Gaejibokryungwhan solution appeared to decrease in a concentration proportional fashion in human uterine myomal cells. The expression of bcl-2 for Gaejibokryungwhan solution appeared to increase in a concentration proportional fashion in human uterine myomal cells. This study showed that Gaejibokryungwhan took effects on the inhibition of human uterine myomal cells growth and decreased of MAP kinase activity, and increased the expression of bcl-2. These results suggest that Gaejibokryungwhan contributes to the chemical inhibitor and treatment of human uterine myoma.
nfluence of Gammakdaejo-Tang on brain Fos-liks immunoreacivity in response to immobilization stress in ovariectomized rats

Objective : During the Postmenopausal state, which is over one third of a women's life, the issue of quality of life is very important. Various kinds of Climacteric Syndrome are found to be problematic and psychoneurotic problems are the most common symptoms especially in Korean women. Postmenopausal depression is similar to Jangjo-jeung(臟燥症) and Gammakdaejo-Tang (GD) is used clinically as a model in Oriental medicine. Thus this experiment is made to elucidate whether GD has an action in relieving stress or anxiety in postmenopausal state. Method : Female Sprague-Dawley(230-260g) rats were used. Temperature was maintained at
Water and food were not limited. The environment was manipulated to simulate 12 hours of day and 12 hours of night. Pentobarbital sodium(50mg/kg) was injected into the intrapentoneal to anesthetize, A 1 cm cut on the abdomen of the rat was made and the ovaries were removed. 12 days after the ovariectomy, the female rats were administered with GD (50 mg or 100 mg daily) in drinking water for 10 days. Rats were subjected to 2 hours of immobilization stress and were perfused 1 hour after immobilization stress. FLI was visualized by immunohistochemistry. We measured the Fos-like immunoreactivity (FLI) of female ovariectomized rats in response to immobilization stress in neural circuits related to stress response. Result : Immobilization stress induced a significant increase in FLI in all areas (paraventricular nucleus, bed nucleus of the stria terminalis, medial prefrontal cortex, lateral septum, central nucleus of amygdala, basolateral nucleus of amygdala, medial nucleus of amygdala), that were significant, compared to control-unstressed animals, at the area of paraventricular nucleus, lateral septum, and medial nucleus of amygdala. Immobilization stress-induced increase of FLI was blocked significantly by pretreatment of GD 50 in LS, and GD100 in paraventricular nucleus, medial prefrontal cortex, lateral septum, and central nucleus of amygdala. No effect was observed in medial nucleus of amygdala. Conclusion : These results show that GD pretreatment decreases immobilization stress induced increase of FLI in brain areas related to stress in ovariectomized rats. Taken together, these results suggest that GD has an action in relieving anxiety and HPA axis activation in response to stress in the postmenopausal state.
Effects of Carthami Semen on the Hormones and Cytokines Associated with Bone Metabolism in Ovariectomized Rats

This study was aimed at proving the effects of Carthami Semen on the hormones and cytokines associated with bone metabolism in ovariectomized rats. Twenty-four female Sprague-Dawley rats were divided into sham operated (normal) group, ovariectomized(control) group, ovariectomized and treated with extract of Carthami Semen(treated) group. Each group was monitored the changes of body weight at 0,3,5,7weeks after treatment, and changes of femur weight, femur/body weight, thickness of compact bone, area of cancellous bone, serum estrogen, serum calcitonin, serum parathonnone, serum tumor necrosis factor-

) at 7weeks after treatment. The results can be summerized as follows ; 1. The treated group showed no change in body weight compared with the control group. 2. The treated group revealed significant increases in femur weight and femur/body weight compared with the control group. 3. The treated group showed much thicker compact bone of body of femur than the control group. 4. The area of cancellous bone of distal epiphysis of femur in treated group was increased significantly compared with the control group. 5. The level of serum estrogen showed no change in the treated group compared with the control group. 6. The level of serum calcitonin showed no change in the treated group compared with the control group. 7. The level of serum parathormone was decreased in treated group significantly compared with the control group. 8. The level of serum TNF-
was decreased in the treated group significantly compared with control group. 9. The level of serum IL -1
showed no change in the treated group compared with the control group. These results indicate that Carthami Semen inhibits bone resorption in ovariectomized rats by it's lowering effects on serum parathormone and TNF-
The effect of hominis pacenta on reproductive performance in aged mice

Objective : This study was designed to investigate the effect of hominis Placenta on reproductive performance in aged mice Materials & Method : The decoction of hominis placenta was administered to 18 week old mice, once a day for 4 days or 8 days. And the number of ovulated oocytes and the rate of morphologically normal oocytes, and the in vitro developmental rate of the 2-cell stage oocytes up to the stage of blastocyst were observed Result : The 18 week old mice before medication showed the significant increase in their body weigt and ovary weight after the superovulation induction comparing with the 4 week old mice while there were significant decreases in the total number of ovulated ova and the rate of the morphologically normal ova, the number of embryos recovered and the in vitro develomental rate up to the stage of blastocyst. After medication, there was the statistically significant increase (p〈0.05) of the total number of ovulated ova and the rate of morphologically normal ova in the experiment group comparing with the control group. There was no significant difference (p〈0.01) of the in vitro developmental rate of embryos recovered between the experiment group and the control group. There was no significant difference between the group of the hominis placenta medication for 4 days and the one for 8 days. Conclusion : This study shows that hominis placenta has significant effects on the increase of the total number of ovulated ova and the rate of morphologically normal ova in aged mice and augments ovarian functions.
The Effect of Cistanches Deserticola Y. C. Ma. On the Reproductive Performance in Mice

Objective : To investigate the effect of Cistanches Deserticola Y. C. Ma. (肉총蓉) on ovulation and in vitro development competence from pronuclear stage oocytes into blastocyst stage embryos in mice Materials & Method : The decoction of Cislanches Deserticola Y. C. Ma. was administered to 4-week-old and 18-week-old mice, once a day for 4 days or 8 days. The total number of ovulated oocytes, the rate of morphologically normal oocytes, and the in vitro developmental competence from pronuclear stage oocytes into blastocyst stage embryos were observed. Result : There was no significant difference between treated and untreated groups in the comparison of the body and ovarian weight after Cistanches Deserticola Y. C. Ma. administration for 4 and 8 days. The total number of released ova showed significant increase(p〈0.05) after 4-day administration in 4-weck-old (treated) group. After 8-day administration, the total number of released ova and the normal ova rate showed significant decrease (p〈0.05) in 4-week-old (treated) group. The total number of released ova showed significant increase (p〈0.01) after. 4-and 8-day administration and the normal ova rate showed significant increase (p〈0.0001) after 8-day administration in 18-week-old (treated) mice In the comparison of the developmental competence from pronuclear stage oocytes into blastocyst stage embryos after 4- and 8-day administration, 4-week-old (treated) group showed significant increase (p〈0.01, 0.01) and 18-week-old (treated) group showed significant increase (p〈0.01, 0.0001). Conclusion : This study shows that Cistanches Deserticola Y. C. Ma. has significant effects on the increase of the total number of ovulated ova and the rate of morphologically normal ova, and the in vitro developmental competence in 4- and 18-week-old mice.
A Literal Study on retention enema therapy for PID(focused on Chinese medical magazines)

I came to a few conclusions as follows through investigation of 21 kinds Chinese medical magazmes on retention enema therapy for Pelvic Inflammation Disease(PID). 1. PID is a genital inflammatory disease, of which symptoms are lower abdominal pain, lumbago, much leucorrhoea, fever and painful menstruation. 2. The method of retention enema therapy is using
herbal extract 100ml, injecting this decoction into anus by the depth 12-20cm and keeping for 30 minutes in anus. 3. The herbs of enema therapy for PID are Patriniae Radix(敗醬草), Paeoniae Radix Rubra(赤芍藥), and Violae Herba(紫花地丁), which have effects of Clearing away heat and toxic meterial(淸熱解毒) and Drainage of pus and swelling(消腫排膿). 4. The treatment rate of retention enema therapy for PID is 91.6
. It means this therapy is better than antibiotic treatment and oral medication from the viwepoint of effective rate. And there is not a significant difference of treatment rate between only using enema therapy and using enema and oral medication together.
The Relationship between peripheral blood flow and cerebral blood suply on D.I.T.I. and TCD

Purpose : There are many patients with cold hypersensitivity who worry about having cerebro-vascular disease. There has been no studies about it. So we examined rebtionship between peripheral blood flow and cerebral blood supply. Method : We observed the 80 patients who visited KyungHee Univ., Kangnam Hospital of Oriental Medicine from December, 1, 2001 to March, 31, 2002. To rule out an bias, we excluded the patients with obesity, skin diseases, CVA and HNP of L-spine and seniors. We measured body temperature by D.I.T.I. and middle cerebral artery flow velocity by TCD. We used SPSS program (version 8.0) for statisties. Result : Average age of the patients was

Research of relatioship on cold hypersensitivity for the patients in OB&GY of Dong Eui Medical center

Cold hypersensitivity means a condition feeling cold at a temperature when one should not so. Many patients of Oriental Gynecology are conscious of cold hypersensitivity. Purpose : We aimed to study cosciousness of cold hypersensitivity of Oriental Gynecology patients and cosciousness classified by disease Method : For 5 months from January to May in 2001, we investigated 235 patients who answerd by a questionnnaire at Dong Eui Medical center. Result : 1. 94.47
of patients are conscious of cold hypersensitivity. 2. In the distribution according to the part, there are hole body, hands and feet, lower abdomen, external genitals, lumbus and knees, the back. 3. In the distribution according to the age, there are many cold hypersensitivity at the group of more than 28 4. Cold hypersensitivity of hole body, lumbus and knees, external genitals are increased according to the age but, hands and feet, the back, lower abdomen are not increased. 5. The rate of conscious of cold hypersensitivity classified by disease there are many morbid leukorrhea, postpartum general aching, natural abortion but, there is a few myoma. 6. The patients who take with morbid leukorrhea are conscious of cold hypersensitivity in external genitals severely. 7. The patients who are extracted at the uterus are conscious of cold hypersensitivity similary with other patients. According in the fact mentioned above, many patients of Oriental Gynecology are conscious of cold hypersensitivity, therefore management of cold hypersensitivity is important at the treat of Oriental Gynecology disease.
A Study of Pelvic Congestion Syndrome(PCS) causing Chronic Pelvic Pain

In pelvic congestion syndrome mental tension or sexual tension brings the congestion in pelvic vein also pelvic pain is the main symptom. So the pelvic congestion syndrome is an important causing factor to women's chronic pelvic pain. The blood stasis is a major factor of PCS in Traditional Korean Medicine(T.K.M) ctiology. Due to increase in women social workers having a lot of stress and wide open sexual contact, the woman could cause part infection and this result in blood stasis in pelvic area. Because of all these reasons the many patients are making complaints about theses diseases and diagnosis opportunity has been increased since wide use of non-invasive diagnostic method been used to find pelvic symptoms. So theses diseases are now one of the very often seen disorders in clinical field of T.K.M Ob&Gy. For this reason the author explains in this thesis that clinical treatment as T.K.M combining with modern diagnosis for PCS causing chronic pelvic pain, T.K.M herbal treatment, acupuncture, and heating therapy could treat blood stasis in pelvic area and timid status. The mind-body combined treatment is very effective on PCS through T.K.M. Individually variated pelvic circulation where treat local symptom and a whole body symptom to combine with herbal therapy and local acupuncture therapy has control the pain and beneficially to the pathologic factors. Also in case of deficiency of vital energy or yang-energy occurred, one should to treat heat therapy such as TDP and intravaginal heatingl therapy(herb-com) of pelvic area is very helpful to the symptoms.
A Study of Traditional Korean Medicine Treatment in endometriosis

Endometriosis is an important causing factor to pelvic pain. In Traditional Korean Mcdicine(T.K.M), its etiology is blood stasis. Due to increase in women social workers having a lot of stress and wide open sexual contact, the woman could cause part infection and this result in blood stasis in pelvic area. Because of all these reasons the many patients are making complaints about this disease and diagnosis opportunity has been increased since wide use of non-invasive diagnostic method been used to find pelvic pathologies. So this disease is now one of the very often seen disorders in clinical field of T.K.M Ob&Gy. For this reason the author explains in this thesis that clinical treatment as T.K.M combining with modern diagnosis for endometriosis. T.K.M herbal treatment, acupuncture, and Reserved-enema therapy could treat blood stasis in pelvic area and timid status, it has bright future. If there are patients willing to have pregnancy, one should look out ovulation cycle and T.K.M Treatment is very effective.
The Clinical Analysis of the Symptoms and Characteristics on 50 Cases of Postmenopause syndromes

The clinical analysis was done to characterize postmenopausal symptoms by use of the one of worldwide woman's health assesment scales, Kupperman's Index which consisted of qustionnaires about onset, severity and some other characteristics of the postmenopausal symptoms. The patients were chosen from the outpatients at woman's health clinic of Kangnam Kyung Hee Korean hospital from March to May 2001. According to Kupperman's Index analysis. The age ditribution of them was from 42 to 58, of which mean age was 49.9, 31 of them was postmenopause wemen, theire mean age was 50.8. The average time from recognition of symptoms from menopause was 11.2 months. 38
of them time was found to visit cilnic within 3 months after menopause, although 12
of them was examined after 6 monthes of menopause. It was suggested that we could get more accurate and practical information about postmenopause symptoms by analyzing clinical cases exactly, which would be very fundamental for more advanced skills to cure them.
Study on living style of obesity patient

Obesity is, by definition, the condition which is excess fat stored in the body. If there are more calories than the basic nutritional needs in physical activity and growth, the excess is converted to fat and stored. largely obesity comes from psychological and socioeconomic factors like overeating, lack of exercise, heredity, unbalanced caloric intake, excessive drinking, marriage, prohibitioh of smoking. Nowadays a lot of treatment of obesity have been tried such as diet, exercise, surgical operations, medicines, acupuncture-Tx, herbal medication and so on. But a person could regain his/her weight after losing it unless there is a fundamental change of his/her bad living style. Therefore, in obesity therapy it is essential for a doctor carefully observe patient's living style and train them to get over their bad habits.
The clinical study of a patient hospitalized due to Abnormal Uterine Bleeding for 5 consecutive months

The 25-year-old female patient who has been suffering from uterine bleeding for five months underwent oriental medical treatment after she failed to see a remedial value from treatment at a obstetrics and gynecology hospital. The authors report that she stopped bleeding at the uterus after she received herb medicine, acupuncture and moxa treatments at the same time in accordance with 寒流([sailiu]-stanch blood), 澄源([chengyuan]-correct origin) and 複舊([fujiu]-restore), which are principles of treatment of 崩漏([benglou]-metrorrhagia).
