Effects of Hominis placenta on mice sperm motility and in vitro fertilization

Objective: This study was conducted to investigate the effects of Hominis placenta (紫河車) on the reproductive competence in male mice. Materials and Method: We administered the decoction of Hominis placenta to 8-week-old and 18-week-old male mice once a day for 8 days. And we observed the count, the motility and the morphology of epididymal sperm from the male mice and the result of in vitro fertilization and embryonic development of oocytes. We examined the pregnancy rate and the delivery rate from the mice. Results: It was effective to be treated with Hominis placenta on the increase of the count and the motility of epididymal sperm in 8-week old mice but not effective on the change of the morphology of that. It was effective to be treated with Hominis placenta on the increase of the motility of epididymal sperm in 18-week old mice but not effective on the increase of the count and the change of the morphology of that. There was no signigicant effectiveness in the rate of in vitro fertilization and embryonic development of oocytes of the Hominis placenta treated group and that of control group in 8-week and 18-week old mice. There was no signigicant effectiveness in the rate of pregnancy and delivery of in vitro cultured blastocyst from mice of the Hominis placenta treated group and that of control group in 8-week and 18-week old mice. Conclusion: This study shows that Hominis placenta has no significant effects on the morphology of epididymal sperm from the male mice and in vitro fertilization and embryonic development of oocytes but has significant effects on the count and motility. We can suggest that Hominis placenta has an effect on the improvement of reproductive competence in male mice.
Hangreunghwan water extract induced apoptosis in Human Cervical carcinoma HeLa cells.

To address the ability of Hangreunghwan(HRH) to induce cell death, we investigated the effect of HRH on cell viability. Twenty-four hours later, loss of viability occurred following HRH exposure in a dose-dependent manner. The treatment of HRH, a commonly used herb formulation in Korea, Japan and China, caused a decrease in cell viability. HRH also resulted in apoptotic morphology a brightly blue-fluorescent condensed nuclei by Hoechst 33258-staining, and reduction of cell volume. Our results show that 2mg/ml HRH induces mitochondria membrane potential collapse. Immunoblotting data also shows that the expression of Bcl-2, antiapoptotic protein, decrease by the addition of HRH. This GFP-Bax overexpression system shows that an important pro-apoptotic Bcl-2-family protein, Bax is translocated to mitochondria by the addition of 2 mg/ml HRH. Interestingly, the treatment of BAPTA-AM, calcium chelator, shows that calcium is involved in HRH-induced apoptosis including caspase-3 activation in HeLa cells. BAPTA-AM reverses HRH-induced inhibition of XIAP expression. In addition, the calcium chelator inhibits the increase of Bax expression which is induced by HRH. However, the treatment of BAPTA-AM does not have any effect on HRH-induced Bax translocation. In conclusion, calcium is involved in both HRH-increased Bax expression and HRH-regulated XIAP expression, resulting in apoptosis in HeLa human cervical carcinoma cells.
Effect of Jeoje and Tongyu-tang except Cheonsangap on lactation in postpartum rat

Objective: This study was conducted to investigate the effect of Jeoje(저蹄) and Tongyu-tang except Chunsangap on lactation in postpartum rat. Material and Method: We used 18 weeks pregnant Spraque Dawley rats and administered the decotions of Jeoje and Tongyu-tang except Cheonsangap to rats (treated group) and normal saline to control group once in a day for 4days and 8days. Then we observed the changes of the body weight changes of postpartum rats and pup rats, level of serum prolactin,
-casein m-RNA, WAP m-RNA and mammary gland tissue of postpartum rats. Result: We found that the body weight changes of postpartum rats and pup rats and the level of serum prolactin in Jeoje treated group and Tongyu-tang except Chunsangap treated group showed no significant difference. In the level of -casein and WAP gene, Jeoje treated group and Tongyu-tang except Chunsangap treated group increased than control group. Mammary gland tissue of Jeoje treated group and Tongyu-tang except Chunsangap treated group showed significantly increased angiogenesis of blood vessle ompared with the control group. Conclusion: This study shows that Jeoje and Tongyu-tang except Chunsangap have an effect on lactation in postpartum rat. But the mechanism of it may be other way than the hormon pathway.
Effects of Gamikyogamhwan on the Ovulation and Ovary in Rats

Gamikyogamhwan(GMKGH : 加味交感丸) is used in female infertility, caused by ovulation disorder. An attempt was made to evaluate the influences of GMKGH on the serum concentrations of FSH, LH, and estradiol(
), the histological changes of ovary and the immunohistochemic-al staining for $AT_2$receptor in ovary of rats. The results of the study were as follows : 1. In blood FSH level, experimental group as compared with control group showed no efficacy. 2. In blood LH level, experimental group as compared with control group showed no efficacy. 3. Blood E2 level significantly increased in experimental group as compared with control group. 4. In histological observations of ovary, ovulation significantly increased in experimental group as compared control group. 5. In observations of immunohistochemical staining for
receptor in ovary, there is no difference between control group and experimental group. According to these results, it can be concluded that GMKGH influences ovary to increase the ovulation of rats.
Effects of Epimedii koreanum on the Motility of Rat Sperm

Objective : This study was conducted to investigate the effects of Epimedii koreanum (淫羊藿) on the motility of rat sperm which is important factor of male fertility. Materials and Method : We chose the 10 of 2-month-old Sprague Dawley rats, and administered the extracts powder of Epimedii koreanum to 5 rats (EK group) and normal saline to another 5 rats (control group) once in a day for 28 days. We observed the change of the body weights before and after administration of Epimedii koreanum extracts and normal saline. After 28 days we isolated their gonad surgically and then we observed the weight of testis, epididymis, vascular gland, and prostate. We looked rats testis tissue through a microscope. Also we examined the total, normal and motile sperm count in testis, and the activity of testicular catalase and peroxidase. Results : We found that the count of total, normal, motile sperm in testis of Epimedii koreanum group showed the significant statistical difference comparing with the control group. The angiogenesis in seminiferous tubule was increased and the increasement of the number of spermatogonia, primary and secondary spermatocyte was observed in Epimedii koreanum group through a microscope. The activity of testicular catalase and peroxidase also showed statistically significant difference in the Epimedii koreanum group. The body weight and the weight of testis, epididymis, vascular gland, prostate shows no significant difference between two groups. Conclusion : This study shows that Epimedii koreanum can effect the count and motility of sperm, the important factor in male fertility and also promote the activity of antioxidants, atalase and peroxidase, which is the important factor in spermatogenesis.

Purpose : This study was done to probe the direction and to research the usefulness of aquaacupunture therapy in OB&GY by surveying scientific tendency in journal of oriental OB&GY, the journal of korean acupunture & moxibustion society and the journal of korean institute of herbal-acupunture. Method : 33 treatises was surveyed in the journal of oriental OB&GY from Vol 1. No1, 1987 to Vol 16 1. No 3 2003 and the journal of korean acupunture& moxibustion society and the journal of korean institute of herbal-acupunture Results : The results were as follows; About treatises types experimental treatises was 27, and clinical treatises was 6. About diseases osteoporosis was 10, 瘀血 was 9, infertility was 4, abnormal uterine bleeding was 3, obeisity, urinary incontinence was 1. Aqua-acupunture therapy in OB&GY were effective Conclusion : So we would like to report that aquaacupunture therapy in OB&GY in the oriental medicine can be applied to 瘀血, infertility, osteoporosis, abnormal uterine bleeding etc.
The Study on Recurrent Miscarriage in PubMed

Objective : To research the trend of the study related to recurrent miscarriage and to the hereafter direction for the study on recurrent miscarriage Methods : We referred a PubMed site by using searching word of "recurrent miscarriage"(Limits: 1 Year, only items with abstracts, Human) Results : 62 journals with 110 papers were searched. Conclusion : 1. We have searched 110 papers in 62 journals on recurrent miscarriage. 2. The publication type of study was as follows: 31 papers of review articles, 6 papers of clinical trials, 13 papers of case reports, 3 papers of meta-analysis, a paper of evaluation study and a paper of multicenter study. 3. The most researched subjects on recurrent miscarriage were 67 papers of Immunologic factors. 4. Immunologic therapy using heparin, aspirin or intravenous immunoglubulin and cervical cerclage for incompetent cervix were major treatments on recurrent miscarriage. 5. Antiphospholipid syndrome was a main subject of immunologic factors on recurrent miscarriage.

Purpose : This study was aimed to find out the acupuncture therapy on distocia in oriental medicine Methods : We surveyed the 20 oriental medical books that have articles Distocia Results 1. Acupuncture points used for distocia in ancient literature are Samu mgyo(SP6), Hapkok(LI4), Chiu m,(BL67), Kollyun(B60), T aech ung(Liv3), Ko gwol(CV14), Kich ung(S30), Chungbong(Liv4) 2. Acupuncture points used for distocia in modern literature are Samu mgyo(SP6), Hapkok(LI4), Chiu m(B67), Chok-Samni(S36), Palnyo(B31~B34), Toku m, Kwanwon (CV4), T aech ung(Liv3), Nae-gwan(P6), Kihae(CV6), Kyo njo ng(SI9), T ae-gye (K30), Puryu(K7) 3. Samu mgyo(SP6), Hapkok(LI4) have been significantly together used corelatedly in acupuncture therapy on distocia 4. The Current Perception Thresholds in Patients with cold hypersensitivity on hands

Cold hypersensitivity is the condition feeling unusual sensation of cold at temperature when others don't feel so. The diagnosis of this was based on clinical symptoms and nowadays for which DITI is used. Current perception threshold (CPT) was measured using Neurometer
CPT/C device (Neurotron Inc., Baltimore, Malyland, USA ; neurometer) to evaluate relationship between cold hypersensitivity and peripheral sensory nerve fiber. Neurometer is a quantitative sensory test equipment for analyzing the functions of A-beta, A-delta and C-fibers at three frequencies (5-Hz, 250-Hz and 2000-Hz). 37 patients with cold hypersensitivity or numbness on hands were selected as subjects who visited the women medical center in Kangnam Korean Hospital, KyungHee University. We measured Visual Analogue Scale(VAS) about coldness and numbness and CPT on hands. The coldness VAS scores had statistically significant negative relationship with CPTs at 5-Hz and a little negative relationship with CPTs at 2000-Hz and 250-Hz, but no significance in statistics. However no difference could be found correlation between numbness VAS scores and CPTs. Patients with cold hypersensitivity have an unusual sensitivity in peripheral sensory nerve and a low thresholds of C-fibers, which convey pain and cold sensation.
Effects of Dalsaeng-san on Duration of Lavor in Primiparas

This clinical study investigated the effect of Dalsaengsan on duration of delivery time for primiparas. Dalsaengsan was administerd to 41 primiparas who were expected to undergo vaginal delivery from 2 weeks before estimated delivery date(EDD). The mean time of sample group was significantly shortened compared to that of each control group. The mean age of primiparae was 27.8. The more the age of women in labor was, the longer the delivery time was. Inspite of that result, mean delivery time of the sample group over 30 was significantly shorter than that of the control groups. The mean medication period was 10.8 days. There was no correlation between medication periods and the delivery time. The mean gestational weeks was 39.8 and no correlation was founded between gestational weeks and the delivery time. The average weight of neonates was 3.33kg and the mean time of <3.5kg group was significantly shortened compared to >3.5kg group. So far, no abnormal condition has been detected in both the neonates and the primiparas.
A study of Taping Therapy In Labor and Delivery

Purpose : The aim of this study is to investigate the effects of taping therapy from the view of the primipara's having easy laboring(Sunsan). In this study, both total time and mode of labor were observed. Method : The 58 primipara who have visited to deliver on due in department of ObGy in Bundang CHA Hospital from October, 2002 to September, 2003 divided to two groups, experimental(N=30) and control(N=28). The members in the experimental group have taken the (subcutaneous) taping therapy on Acupoints in the accelation phrase in labor process I. The members in the control group was selected by random sampling within the same period. Then we estimated total necessary time from the activation phase to the full stage and followed up the pattern of labor, NSVD and C-sec. The general data were collected that were age, height, weight, past history etc, and the data reflecting the rate of the labor that Bishop score, NST, Apagar score, breakage of the amnion in labor. Results : The duration of labor of the experimental group was 227.2
33.1(min) and that was 240
35.7(min) in control group, then there was no significant difference in two groups, irrespective of the slight reduction of time. In respect of mode of delivery, there were 19 childbirth on NSVD(Sunsan) of 30 in the experimental group comparing to the 16 childbirth on C-sec(Nansan) of 28 in the control group. Also there was no significant difference in two groups. Conclusion : According to the above result, the taping therapy was not effective more than control group to help primipara to give easy birth.
Clinical Research about the Correlation between Defecation Type and Cold Hypersensitivity of Lower Abdomen, Hand and Foot

Cold hypersensitivity is the condition with unusual cold sensitivity at temperature when others don't feel so. It is not disease in itself, but is known as having relationship with infertility, dysmenorrhea, anemia and endocrine disorder. The symptoms of cold hypersensitivity appear on limbs and abdomen especially, and may affect bowel movement such as constipation or diarrhea. We made a research of 86 healthy young girls who took medical examination and examined subjective bowel habit. The patients were divided into three group by defecation type, constipation(42), diarrhea(14) and normal group(30). Temperature differences
measured by DITI on upper and lower abdomen of each group had not statistically significance. Otherwise
between upper arm and palm and between upper leg and foot were statistically signigicant. The severity of cold hypersensitivity on hands was in order constipation, normal and diarrhea group, and same as feet. The diarrhea patients had more severe cold hypersensitivity as compared with constipation patients. Correlation between
on abdomen and hands or feet didn't exist.
on hands and feet, however, had positive relationship. This research showed cold hypersensitivity could be related with diarrhea.
A Clinical Study on the Efficacy of Electrostimulation on AcupunctureLoci in the Treatment of Dysmenorrhia

Purpose : The purpose of this study is to identify the clinical effects of electrostimulation on acupuncture loci in the treatment of dysmenorrhia. Method : This study is carried out to the 20 volunteers who suffering from menorrhalgia. The volunteers was selected from January, 2004 to March, 2004 through Bundang CHA Oriental Hospital. We invesigated general characteristics, menstrual and dysmenorrheic charateristics through Case Research Form(CRF) and camparison of before and after electrostimulation on acupuncture loci by using Visual Analogue Scale(VAS) for sample patients. We divided to two groups, primary(N=12) and secondary(N=8) and compared the effects of the treatment. And we invesigated the change of VAS score as time goes on. Results : 1. The VAS score on before and after treatment were 89 12.52 and 7 9.23. The VAS score of after electrostimulation on acupuncture loci was decreased significantly compared with that of before(P〈0.01). 2. The VAS score on primary group and secondary group were 88.33 10.3 and 72.5 11.34 after treatment. The VAS score of primary group after electrostimulation on acupuncture loci was decreased significantly compared with that of secondary group(P〈0.0071). 3. The VAS score decreased above 50 points around 15 minutes after electrostimulation on acupuncture loci. Conclusion 1. The electrostimulation on acupuncture loci will be efficacious against dysmenorrhea. 2. Primary group was more effective than secondary group in electrostimulation on acupuncture loci. 3. It seemed that electrostimulation on acupuncture loci should be over 15 minutes.
Clinical Study about The Effects of Gamisamultang on Lactogenesis and Prolactin in Early Puerperal Period

Purpose : To determine the efficiency of Gamisamultang on lactogenesis and serum prolactin level in the early puerperal period. Methods : Participants were enrolled at the Postpartum care center of Conmaul Oriental Hospital. Participants were randomized into a sample group and a control group. 136 women participated in the study, 29 women discontinued participation in the study. In total there were 52 in the sample group, 55 in the control group. Both groups were administrated Gamisaenghwatang for 5 days, then sample group had Gamisamultang and control group had Boheotang, respectively, for 5 more days. Participants' daily milk volume and lactation times were recorded on a provided table. Serum prolactin levels were measured in the 6th and 11th day of the trial. Results : Gamisamultang was discovered to raise both the milk volume(68.93
) and serum prolactin level(27.80
) of the early puerperal period compared with Boheotang. Milik volume was significantly increased(P<0.05), while the serum prolactin level was not. Conclusion : The present findings indicate that Gamisamultang is useful for increasing lactogenesis in the early puerperal period and may have a positive effect on breast-feeding practice.
A study on clinical managements of traffic accident victimsduring pregnancy

Purpose : With the development of society and economy, traffic accident rate is increasing accordingly. And as the number of women who are involved in social activities and driving is going up, more women in pregnancy get hurt and damaged by traffic accident. It is true that these traffic accident victims during pregnancy don't get proper treatment in western medicine due to the difficulty of instant diagnosis and positive treatment. this is where approach from the perspective of oriental OB & GY is definitely needed. So the purpose of this study is to report clinical managements of traffic accident victims during pregnancy. Methods : The patient in this case, 26-year-old, was hospitalized in order to cure neck pain and low back pain occured from traffic accident on the 13th week of pregnancy. This patient was treated with the acupunture and herbal medicine. And we investigate the clinical managements of oriental OB & GY for traffic accident victims during pregnancy. Result and Conclusion : We think that we have to take three things into account ; maintenance of pregnancy(保胎), relief(安神) and pain control(止痛) in order to manage traffic accident victims during pregnancy. And studies on concrete methods of clinical managements should be continued.
The Clinical Study of twenty cases of menorrhalgia patients withadministration of Soeumin Kwakhyangjeonggisan-gamibang

Purpose : Female is liable to disease of psycho-somatic and neurosis because female is more emotional than male, generally. For that reason, I think, it is significant things to correlate the chronic disaeses of Gynocology and the Sasang Constitutional Medicine with psycho-somatic and constitutional character. In order to know the effect of Sasang Constitutional Medical therapy of menorrhalgia, this clinical study was conducted. Method : I invited applications for the patients from April 2002 to May 2002 for this study. I studied twenty patients with interior disease of Soeumin(少陰人 裏證) from twenty-one to fouryt-one years old. We diagnosed Sasang constitution of patients by Questionnaire of Sasang Constitution classification (1) and QSCC(2)and(1) (Questionnaire for Sasang Constitution classification(2)and (1)) Soeumin Kwakhyangjeonggisan-gamibang was administered twice a day for three months. The change of pain and symptoms was measured by the methods of VAS(Visual analog scale), VRS(Verbal Rating scale). after menstration monthly, three times. After three months, we checked up their pain and symptoms by telephone. Results : 1. The portion of menorrhalgia is in the lower abdominen(19 patients, 85

The change of pain during menstration duration according to VRS improved all significantly. 9. The change of symptoms which has in menstration according to VRS improved in hypermenorrhea, brest pain, headache, general pain, dizziness, indigestion, the coldness, stress. It has significant. As mentioned above, the treatment of Sasang Constitution Medical therapy of menorrhalgia patients with interior disease of Soeumin(少陰人 裏證) is effective.
Clinical study on a case of postpartum disease causedby artificial abortion

Purpose : The purpose of this study is to report the effect of oriental treatments for the postpartum disease caused by artificial abortion before marriage. Methods : The patient in this case, 30-years-old female, was admitted at Se-Myung University oriental medical hospital for 15 days(24th/February/2004-9th/March/2004) due to postpartum disease caused by artificial abortion before marriage. This patient was treated with oriental treatments such as herb medication, acupuncture and moxibustion therapy, and so forth. Results : After lasting oriental treatments, the patient was improved in all complaints of the postpartum disease caused by artificial abortion before marriage. Conclusion : The postpartum disease caused by artificial abortion before marriage would be treated more carefully in oriental medicine considering women's difiiculties.
A Clinical study of acute climacteric symptoms measured byKupperman's index

Purpose : The climacteric disturbance has biological, social and psychologycal character. It's very difficult that treatment's effect of climacteric symptoms by Traditional Korean Medicine(TKM) is appeared objectively. We want to propose that the climacteric symptom be measured objectively and diagnosed definitely by climacteric index. Treatment by TKM of the climacteric symptom can be valued by climacteric index. Method : We measured each climacteric symptom by Kupperman's index and summed up each climacteric index. Then, We compared climacteric disturbance before treatment with one after treatment by Kupperman's index. Result : We treated her who had haven acute climacteric symptoms with herb medication, accupucture, and moxibustion during short term admission. During the admission, her climateric symtoms were improved. The sum of patient's climacteric index was rapidly decreased(from 48 to 20). Conculsion : The treatment by TKM which improves climacteric symptoms is very effective. It would be valid to measure by Kupperman's index for treatment's effect of climacteric symptoms.
Pregnancy in Premature Ovarian Failure afterOriental Medicine Treatment. Two Cases Report

Premature ovarian failure (POF) is a condition causing amenorrhea, hypoestrogenism and elevated gonadotropins before the age of 40 years. There is no ensure way to treat this disease in western medicine. Oriental medicine doctors have made reference about diagnosis and treatment of POF long ago, but also have not an efficacious remedy. So, case report about pregnancy case report in premature ovarian failure is rare in all oriental-western medicine. Two patients taken herbal medicine with each diagnosis, also herbal-acupuncture treatment, moxibustion. In result, they recovered the menstrual cycle and became pregnant & delivery.
A Clinical Study on Postmenstrual Colic Treated by Gami-sibjeon-tang

We treated two patients who had dysmenorrhea. One had an endometriosis, the other had an adenomyosis uteri. Severe pain was after post menstruation as well as menstruation period. After treatment, relief of postmenstrual colic was less than menstrual pain. Then we thought the postmenstrual colic was concern with inflammation. Gami-sibjeon-tang was used when the inflammation didn't healed because of deficiency of Ki and blood. We administerd Gami-sibjeon-tang and their postmenstrual colic was relieved.
