The Experimental Study on Antithrombotic activities of Saegeumsan(瑞金散)

This study was performed to evaluate antithrombotic activities of Saegeumsan (瑞金散, SGS) which has effects of activating blood, removing thrombus. This study is designed to measure the effect which was given to blood flow rate through the regular volume of glass tube after the blood was diluted five times with ACD solution. Antithrombotic effect was calculated as a percentage of the experimental animal figure protected from the paralysis of hind legs or death of the mouse that is caused from the administration of platelet aggregation regent. We standardized the time when the experimental animals were incapable of functioning the hind legs more than 20 minutes or maintained trembling. Being classified one group of eight mice, each of them was divided into Normal, Control, and SGS. The normal group supplied a saline solution and the control group brought the dextran extravasated blood after an hour of administering the saline solution. Also, SGS was dissolved in 2㎖ saline solution and then we dosed it to the experimental mice with Oral Zonde one day before the experiment. After that, the mice were abstained from food. And then we gave a measured amount of it before an hour. Finally, it gave rise to dextran extravasated blood in the same way as the Control group. The results were obtained as follows, SGS significantly inhibited platelet aggregation induced by ADP and epinephrine when analyzed by the Sigmoid Emax model in WinNonlin. EC50 values of SGS were 4.61 mg/ml and 12.41 mg/ml for ADP and epinephrine respectively.SGS showed fibrinolytic activity insignificantly as compared with the control group. SGS increased blood flow rate significantly as compared with the control group in vitro. SGS inhibited pulmonary embolism induced by collagen and epinephrine(inhibitive rate is 37.5 %). SGS increased number of platelet and fibrinogen amount significantly, and shortened PT and APTT as compared with the control group in thrombus model induced by dextran.According to, SGS is effective antithrombotic activity from experimental result.
Kung-kyung-IlHo-jeon on Induced Apoptosis in Human Cervical Careinoma HeLa Cells

To address the ability of Kung-Kyung-Ilho-Jeon(KK) to induce cell death, we investigated the effect of KK on cell viability. Forty-eight hours later, loss of viability occurred following KK exposure in a dose-dependent manner. The treatment of KK, a commonly used herb formulation in Korea and China, caused a decrease in cell viability. KK also resulted in apoptotic morphology a brightly blue-fluorescent condensed nuclei by Hoechst 33258-staining, and reduction of cell volume. Our results show that KK induces caspase-3 and -9 activation in a time-dependent manner. In addtion, the translocation of cytochrome c release into cytoplasm has been observed under the presence of 5 ㎎/㎖ KK. The subsequent loss of mitochondria membrane potential is collapsed by the addition of KK. Our immunoblotting data show that PARP, a well known caspase-3 and -6 substrate, is cleaved by KK. We show that a pro-apoptotic protein, Bax is increased in the presence of KK but that the amount of Bcl-2 is not changed. We suggest that Bax, a critical protein which can regulate channel of mitochondria to release cytochrome c, is a key protein in KK-induced apoptosis of Hela human cervical carcinoma cells
Herba Portulacae induced Apoptosis in Human CervicalCarcinoma HeLa Cells

To address the ability of Herba Portulacae(HP) to induce cell death, we investigated the effect of HP on cell viability. Twenty-four hours later, loss of viability occurred following HP exposure in a dose-dependent manner. The treatment of HP, a commonly used herb formulation in Korea, Japan and China, caused a decrease in cell viability. HP also resulted in apoptotic morphology a brightly blue-fluorescent condensed nuclei by Hoechst 33258-staining, and reduction of cell volume. Our results show that 2mg/ml HP induces mitochondria membrane potential collapse. Immunoblotting data also shows that the expression of Bcl-2, antiaoptotic protein, decrease by the addition of HP. This GFP-Bax overexpression system shows that an important pro-apoptotic Bcl-2-family protein, Bax is translocated to mitochondria by the addition of 2mg/ml HP. Inerestingly, MAPK inhibitor study shows that p38 MAPK inhibitor, SB203580 inhibits HP-induced cell death and caspase-3 activation in HP-treated HeLa cells. Furthermore, HP transiently but significantly induces p38 activation. But P38 MAPK inhibitor does not have any effect on the translocation of Bax. Considering these results, HP induces apoptosis via p38 MAPK activation. But the pathway does not involve the translocation of Bax.

Vasodilatory Activities of the Water Extracts of Seventeen Medicinal Remedy

The pharmacological effects of medicinal remedies traditionally used in Asian countries for improving the blood circulation were examined on isolated rat thoracic aorta strips in organ baths. Each experimental medicine was consecutively extracted under reflux with water. Of 17 plants, Curcuma longa (CL) having the strongest acute relaxant activity in endothelium-intact arteries, Mucunae caulis (MC), Cirsium pendulum (CP), Rumex longiflius (RL), Paeonia suffruticosa (PS), Curcuma zedoaria (CZ), Scirpus maritimus (SM), Siphonostegia chinensis (SC), Leonurs sibiricus (LS) and Typha orientalis (TO) were showing dose-dependent relaxant activity. Long-term relaxant effects were showed in Curcuma aromatia (CA), MC, CP, RL, PS, Potulacae grandiflorae (PG), CZ, Panax notoginseng (PN), Achyranthes japonica (AJ), CL, SC, Lycoppus lucidus (LL) and Corydalis turtschaninovii (CT). In endothelium-injury test using carbachol, CL, SC, MC, RL and PS which are having the acute vasorelaxing activity and CA and CT which are not showing vasorelaxaing activity were damaged to endothelium. As a result of this study, the possibility that a part of medicinal remedy may contribute to the beneficial effects in blood circulation was proposed, but inter-individual variation has been observed. Also, further studies on the vasorelaxant effects of these remedies are still required.
Anti-inflammatory Effects of Hongdeung-Tang on Mice with LPS Injected in Vagina

Purpose : There are various oriental medicine therapy of treating Pelvic Inflammatory Disease(PID) in clinics. We made the PID mice model by injection Lipopolysaccharide (LPS) in vagina, and investigate anti-inflammatory effects of Hongdeung-Tang among oral medication, retention enema therapy and herbal-acupuncture treatment. Method : The ICR(20-30g) mice(♀) were used. To examine the occurrence of inflammation, LPS in different concentration was injected into the vagina of the mice, and White Blood Cell(WBC) in blood was counted. To examine anti-inflammatory effects, 6 mice were assigned to each of the normal group, the control group and the sample group. Hongdeung-Tang was medicated in oral and rectal with 1.0g/kg low dose and 3.0g/kg high dose and by herbal acupucture for 5 days, 2 days before LPS injection. After 3days from LPS injection, blood was collected from retro-orbital plexus, and WBC, Interleukin-6(IL-6), Tumor Necrosis Factor-
) in the blood was counted. Result : After LPS injection with each dose, WBC count showed significant increase depending on LPS concentration from 100
/kg, and it was maximized at 3 or 4 days after LPS injection. the Hongdeung-Tang treatment groups, The number of WBC was decreased significantly only in low dose and high dose oral medication, and IL-6 concentration showedsignificant decrease in oral and rectal medication as well as in herbal acupuncture treatment. TNF-
concentration was decreased significantly in oral and rectal medication of low dose and high dose. herbal-acupuncture treatment group datas showed reductive tendency. Conclusion : Based on above results, we made the PID mice model by injection LPS in vagina , and demonstrated anti-inflammatory effect of Hongdeung-Tang of oral medication, retention enema therapy and herbal-acupuncture treatment.
Effects of Kamikwibitang on Serum Levels of Corticosterone and Histamine and Immune Response after Immobilization Stress in Mice

The purpose of this research was to investigate the effects of Kamikwibitang water extract (KKT) on immobilization stress in C57BL/6J mice.KKT decreased the serum level of histamine and corticosterone increased by immobilization stress. In addition, KKT decreased the cell viability of thymocytes and enhanced the cell viability of splenocytes decreased by immobilization stress. Also, KKT decreased the viability of thymocytes and splenocytes in vitro. KKT decreased DNA fragmentation of splenocytes increased by immobilization stress. KKT decreased the population of thymic CD4+CD8- cells increased by immobilization stress, and did not affect the population of B220+ cells and the population of Thy1+ cells changed by immobilization stress and enhanced the population of splenic CD4-CD8+ cells increased by immobilization stress. KKT enhanced the production of γ-interferon and did not affect the production of interleukin-2 and interleukin-4 decreased by immobilization stress. Also, KKT decreased the phagocytic activity and the level of nitric oxide decreased by immobilization stress.These results indicate that KKT may be useful for the prevention and treatment of stress via suppression of serum histamine and corticosterone level and enhancement of specific-immune response.
Effects of Mokhyangsaenghwa-tang and Mokhyangsaenghwa-tang plus Cervi Pantotrichum Cornu on Postpartum Recovery and Lactation

Objective:This study was conducted to investigate the effect of Mokhyangsaenghwa -tang (MS) and Mokhyangsaenghwa-tang plus Cervi Pantotrichum Cornu (MS- C) on postpartum recovery and lactation. Materials and Methods: We used 18-week pregnant Spraque Dawley rats and administered the decoctions of MS and MS-C to rats once a day for 4 days or 8 days. Then we observed changes in the body weight of pup rats and complete blood cell count, liver function test, renal function test, mammary gland tissue, level of serum prolactin, β-casein and WAP of postpartum rats. Result: A significant increase in body weight was observed in MS-C treated pup rats compared with in MS treated group. The levels of WBC and platelet from MS group and MS-C group were decreased compared with the control group. The levels of RBC, hemoglobin and hematocrit from MS group and MS-C group showed statistically significant increases compared with the control group. The levels of protein, albumin from MS group and MS-C group were increased compared with the control group. The levels of BUN, creatinine from MS group and MS-C group did not show statistically significant changes compared with the control group. The mammary gland tissues from MS group and MS-C group showed increased angiogenesis. The levels of serum prolactin from MS group and MS-C group were increased compared with the control group. The expression of
-casein and WAP genes from postpartum rats treated with MS and MS-C was increased. Conclusion: This study shows that MS and MS-C improved postpartum recovery and lactation in rats.
Effects of Eucommiae Cortex (杜庶) on the Bone Histomorphometryand Metabolism in Ovariectomized Rats

Objective: This study was undertaken to investigate the effects of Eucommiae Cortex on the osteoporosis in ovariectomized rats. Materials and Methods: We used Sprague-Dawley female rats in 8-week-old. They were divided into three groups. Sample group was ovariectomized and administered with 10 ㎎/100 g/day Eucommiae Cortex extract solution for 10 weeks. Control group was ovariectomized and sham group was conducted by sham's operation. And control group and sham group were administered with normal saline as the same way. We measured rats's body and uterus weight and also measured the serum levels of Ca,phosphorus and ALP. We stained the specimens of rat's tibial bones with Goldner's modified Masson's Trichrome and then examined bone histomorphometry with Bioquant computer program of image analysis system. We measured the thickness of osteoid and callus as static parameters and measured bone volume and mineral apposition rate as dynamic parameters. We observed the expressions of RANKL and OPG mRNA of the tibial bone by RT-PCR. Results: The body weight was significantly (p<0.05) increased in control and sample groups compared with sham group, respectively. The uterus weight was significantly (p<0.05) decreased in control and sample group compared with sham group. In the change of Ca, phosphorus and ALP there were no significant changes among three groups. There were no significant changes of trabecula cortical and osteoid bones' thickness and volume. But trabecula mineral apposition rate (MAR) was significantly (p<0.05) increased in sham and sample group compared with control group. In the expression of RANKL mRNA, sample group was decreased compared with control group, and in that of OPG mRNA, sample group was increased compared with control group. Conclusion: This study shows that Eucommiae Cortex has the beneficial effects on bone histomorphometry and metabolism in the ovariectomized rats. We suggest that Eucommiae Cortex be useful for the treatment of osteoporosis.
Dose Dependent Effects of Epimedii Herba on theReproductive Function in the Mice

Objective: This study was undertaken to evaluate the dose dependent effects of Epimedii Herba extract solution on the spermatogenic abilities such as concentration, motility and morphological normality of sperm from the testis, and the activities of spermhyaluronidase and antioxidants.Materials and Method: We choose the 2-month-old mice, and administered the extract powder of Epimedii Herba in the different concentration once in a day for 60 days. The control group was administerde to normal water in the isolated testis tissue. Also we observed changes of isolated testis at the before and after administration of Epimedii Herba extracts in the mice. And we compared to the testicular tissue especially seminiferous tubules between control and treated group by histochemical methods.Results: The significant dose dependent differences were observed in the concentration of total sperm, the motility and normality of spermatozoa of the Epimedii Herba extract administered groups compared than that of control group, respectively. In the histological analysis of the testicular lobes were observed in the Epimedii Herbaextract administered groups than control group, respectively. Also, the activity hyluronidase was significantly increased in the Epimedii Herba extract administered groups than that of the control group. In case of antioxidant activity analysis, the activity of peroxidase and catalase were significantly increased in the Epimedii Herba extract administered groups than that of control group, respectively.Conclusion: This study shows that Epimedii Herba can effect the count and motility of sperm, the important ractor in male fertility and also promote the activity of antioxidants, catalase and peroxidase, which is the important factor in spermatogenesis.
Clinical Study on the Efficacy and Safety of Chiljehyangbuhwan in the Treatment of Dysmenorrhea

Objective:The purpose of this study is to identify the clinical effects of Chiljehyangbuhwan in the treatment of dysmenorrhea patients.Methods : We studied fifty patients who visited Kyung Hee University Oriental Medical Center from July, 2004 to August, 2004. Women with organic disease such as ut. myoma, ov. cyst and pelvic inflammatory disease were excluded from this study. We treated them with Chiljehyangbuhwan for one menstrual cycle. The severity of dysmenorrhea were measured by MVRS (Multidimensional Verbal Rating scale), VRS (Verbal Rating scale) and VAS (Visual Analog Scale).Results : Chiljehyangbuhwan significantly decreased the severity of dysmenorrhea. Furthermore Chiljehyangbuhwan had an effect on dysmenorrhea for next menstrual cycle without taking. Chiljehyangbuhwan did not show hepatic and renal virulence. Conclusion : This study shows that Chiljehyangbuhwan has remarkable effects on dysmenorrhea patients and that effects continue to next menstrual cycle without taking medicine.
The effect of the stem of Spatholobus suberectus Dunn on the proliferation and gene expression related apoptosis in human cervical cancer cells

Objective : Spatholobus Suberectus Dunn stems, Chinese vine plants, have been used for the relief of menstrual disorders and rheumatic arthralgia. In this study, we investigated the antitumor effect of Spatholobus Suberectus Dunn on cervical cancer in vitro.Methods : HeLA cervical cancer cell lines were used as targets. We examined the effect of water extract from Spatholobus Suberectus Dunn on cell proliferation, cell cycle regulation and cell cycle-regulating gene expression. Further, we investigated the apoptotic effects of Spatholobus Suberectus Dunn on cervical cancer cell lines. Results : Spatholobus Suberectus Dunn significantly inhibited the proliferation of cervical cancer cell lines in a dose-dependent and time dependent manner. Fluorescence activated cell sorter (FACS) analysis indicated that Spatholobus Suberectus Dunn induced G1 cell cycle arrest. Spatholobus Suberectus Dunn enhanced the expression of p21waf1 and p27kip1 with cell cycle arrest. Further, Spatholobus Suberectus Dunn stimulated apoptosis via caspase3 pathway.Conclusions : These findings suggest that Spatholobus Suberectus Dunn is a candidate agent for the treatment of cervical cancer. p21waf1 and p27kip1 may play an important role in Spatholobus Suberectus Dunn-induced cell cycle arrest and cell growth inhibition.
The effect of -sitosterol proliferation and apoptosis in human uterine leiomyoma cells

-sitosterol on the proliferation of human uterine leiomyoma cells and the expression of gene related the mechanism of cell apoptosis.Methods : We counted the number of death cells treated with indicated time of the
-sitosterol and investigated cell death rate by cell count assay. Furthermore, flow cytometry analysis and DNA fragmentation assay were used to dissect between necrosis and apoptosis. and then we observed the differential gene expression by western blot analysis.Results :1)The inhibitory effect on the growth of uterine leiomyoma cell treated with the
M was increased in a time dependent.2)The result of flow cytometry analysis, subG1 phase arrest related cell apoptosis was investigated 16.97% in uterine leiomyoma cell treated with the
M and showed the fashion of proportional time dependent.3)The gene expression of p27, p21 related cell cycle was increased according to increasing time interval but cyclin E-CDK2 complex was decreased expression.4)The character of apoptosis, DNA fragmentation was significantly observed on the time dependent.5)The expression of pro-caspase 3 and PARP were decreased dependent on treatment with time dependent.Conclusion : This study showed that the
-sitosterol have the inhibitory effect on the proliferation of human uterine leiomyoma cell and the effect was related with apoptosis.
Systemic Review :The Study on Ovarian hyperstimulation syndrome(OHSS)

Objective : To know about ovarian hyperstimulation syndrome pathophysiology, risk factors and clinical features and to research the trend of the study related to OHSS. Methods : We referred a PubMed site by using searching word of "ovarian hyperstimulation syndrome"(Limits: 1 Year, only items with abstracts, Human). Results : 28 journals with 49 papers were searched. Conclusion 1. The study of OHSS subjects on pathophysiology, prevention and medical treatment. 2.As OHSS is an exaggerated response to ovulation induction therapy, it's emphasized that aspect of prevention OHSS. 3.Preventing OHSS are the following. The first is to give a GnRH agonist or antagonist in substitute for hCG. The second is to screen out prevalence of thrombophilia. The third is to monitor E2 levels. The forth is to aspirate of Mediculous follicle. The fifth is cryopreservation of all embryos. The sixth is that administration of albumin for treatment of OHSS. But, it's not useful to administration of albumin for prevention of OHSS. 4. There's no therapy of OHSS. But, there's only symptomatic treatment of OHSS.
The clinical study on one case of cervical sprain patient due to traffic accident in pregnancy

Purpose : With the development of society, traffic accident(TA) rate is increasing accordingly. And the number of women in pregnancy who are damaged by TA is increasing as more women are involved in economical and social activities. TA victim in pregnancy cannot get proper treatment(Tx) in western medicine. Oriental medicine Tx is noted as alternative in treating TA victims recently. The main purpose of this clinical study is to make a survey of effective way of the oriental gynecological Tx about TA victim in pregnancy. Methods : The patient in this case, 28-year-old, was admitted for 14days(27th/May/ 2004 - 9th/June/2004) to cure severe neck pain and limitaion of cervical ROM(range of motion) occured from TA on the 4th week of pregnancy. This patient was treated with the acupunture, herbal medicine and physical treatment. Result and Conclusion : The symptoms of patient in this case are remarkable alleviated. And we think that the TA victims in pregnancy can be managed effectively by oriental gynecological Tx due to consider preservation of fetus and pregnant woman. After this, further study, approach and case will be needed about oriental gynecological Tx of TA victims in pregnancy.
Analysis on Infertility Patients in Dept. of Oriental Gynecology of Conmaul Oriental Hospital

Purpose : To investigate clinical-epidemologic aspect of infertility patients in Conmaul Oriental Hospital. Methods : From 2003, 5 to 2004, 4, a total 1223(female 943, male 280) patients was recruited, and their medical records were reviewed retrospectively. Results : The mean age of female was 31.83

3.03 in male. The most common occupation of female was profession, administration, management.The primary infertility was 52.9% and the secondary infertility 47.1%. The most common past history in female was laparotomy. The mean duration of infertility was 3.32

2.63 years in female and male. 83.6% of secondary infertility women experienced abortion. The incidence of patients with both combinded infertility factors of female and male was the most highest, and the most common single factor was ovulatory. 92.4% of female patients took sterility tests, 71.0% of male patients took semen analysis. 50.8% female took ovulation induction, IUI, IVF before coming. 8.91% of female and 72.14% of male were accompanied by their spouse. 61.2% of female took sterility test with their spouse. Female's duration of treatment was longer than man's. The most reliable source of choices was encouragement of family members, acquaintance. Conclusion : In this study, we presented staus concerend with infertility and the characteristics of patients went to oriental hospital. Furthermore, the study about oriental treatments and the results of that treatments is required.
A Case Report of 1 Patient Complaining ofHyperemesis Gravidarium

Hyperemesis gravidarum is a severe and intractable form of nausea and vomiting in pregnancy. It is a diagnosis of exclusion and may result in weight loss; nutritional deficiencies; and abnormalities in fluids, electrolyte levels, and acid-base balance. The peak incidence is at 8-12 weeks of pregnancy, and symptoms usually resolve by week 16. Interestingly, nausea and vomiting of pregnancy is generally associated with a lower rate of miscarriage. The cause of severe nausea and vomiting in pregnancy has not been identified. Extreme nausea and vomiting may be related to elevated levels of estrogens or human chorionic gonadotropin. Once the diagnosis is made, treatment consists of mainly supportive care. Until vomiting is controlled, these patients should be maintained as NPO. We treated 1 patient who had hyperemesis gravidarum and visited Kyung-Won Incheon Oriental Hospital by Herbal medication, acupuncture, moxa and nega therapy and got good result from them.
A Study on the Measured Method ofMenstrual Quantity

Purpose : This study carried out to develop the questionnaires that can measure objectively menstrual quantity and determine quantity index. Method : First, we made questionnaires to measure menstrual quantity by six question. Then we investigated 1309 women's questionnaires who has the menses. Conclusion : 1.the questionnaires that can measure objectively menstrual quantity and determine quantity index was high a correlation coefficient therefore those were used Method of Menstrual Quantity(here after MMQ) calculation. Those were added and averaged. 2.We examined correlation of calculated MMQ and women's questionnaires who has the menses therefore we got 0.30～0.47 correlation. 3.MMQ 1～3(hypomenorrhea) was 39 persons 3.19%, MMQ 4～6(normal menorrhea) was 1050 persons 85.85% and MMQ 7～10(hypermenorrhea) was 134 persons 10.96%. MMQ 4 tended to be little and MMQ 6 tended to be much. 4.MMQ confidence was high as 82% comparatively so we thought MMQ was appropriate as the measured method of menstrual quantity.

This study was designed to evaluate the effects of oriental medicine therapy on a chronic pelvic inflammatory disease. The patient was admitted to the OBGY department of Dong-Shin University in Kwang-Ju, on September 15, 2004 and remained until October 20, 2004. She was treated with herbal medicine, herbal retention enema, herbal acupuncture etc. After treatment, improvement was seen in pelycalgia, low abdomen pain, dysuria, leukorrhea. The study suggests that oriental medicine therapy is significantly effective in the treatment of chronic pelvic inflammatory disease .
