The Effect of JokyungSan on the Ovarian Functions and Differential Gene Expression of Caspase-3, MAPK and MPG in Female Mice

Purpose : These experiments were undertaken to evaluate the effect of adminis tration of JokyungSan on ovarian functions and differential gene expressions related cell viabilities caspase-3, MAPK and MPG in female mice. Methods : We administered the JokyungSan to 6-week-old female ICR mice for 4, 8, or 12 days. The female mice were injected PMSG and hCG for ovarian hyperstimulation. We chose the caspase-3 for cell apoptosis, MAPK and MPG genes for cell viability and DNA repair. To compare the differences, we set a control group treated with plain water at the same volume by the same way. Results : In case of 4-day administration of JokyungSan, the mean number of total ovulated oocytes and the number of morphologically normal oocytes increased significantly compared to a control group. The administration of JokyungSan, were beneficial effect of embryonic development in preimplantation period and play a role of prevention of cell apoptosis and DNA damages and also increased cell proliferation resulted in ovarian functions. Conclusion : From our results suggested that the medication of JokyungSan has beneficial effect on reproductive functions of female mice via prevention of cell apoptosis and DNA damaging and promotion of cell proliferation.
Induced apoptosis in human Uterine Leiomyoma Cells by treatment with Chiljehyangbu-hwan

Purpose : Uterine leiomyoma (fibroids) are benign smooth muscle tumors originating from the myometrium. These benign neoplasms of monoclonal origin are typically diagnosed during the reproductive years, occurring only after puberty and tending to regress after menopause. In the present study we used Chiljehyangbu-hwan to determine its growth inhibitory effect and apoptosis in human uterine leiomyoma cells. Methods : Primary cultured human uterine leiomyoma cells were treated with Chiljehyangbu-hwan. Cell viability analysis was analyzed by MTS assay and FACS was performed to ascertain the effects Chiljehyangbu-hwan. DNA fragmentation analysis and casapase-3 activity test were done. Expression of apoptosis related proteins were evaluated by Western blot analysis. Results : Cell viability was significantly influenced by Chiljehyangbu-hwan treatment in a dose-dependent manner in leiomyoma cells compare to normal myometrial cells. FACS showed that Chiljehyangbu-hwan induced Sub G1 arrest. DNA fragmentation assay was carried out and apoptosis was detected. Activation of caspase-3, down-regulation of Bcl-2, with concomitant increased expression in Bid and Bax were observed. Chiljehyangbu-hwan treatment of uterine leiomyoma cells resulted in a concentration-dependent cell death induced via the mitochondrial pathway.
Effects of Astragalus membranaceus on Surgically Induced Endometriosis in Rats

Purpose : Astragalus membranaceus is a herb which has effectiveness of energy increase, protection against pathogenic and organ immune response control. Endometriosis has been thought to be related with immune response in uterus. Therefore in this study, the effects of Astragalus membranaceus on endometriosis were investigated. Methods : The endometrial tissue was autografted to rat's small intestine. Rats with surgically induced endometriosis was administerd with Astragalus membranaceus extracts for 40 days orally or rectally. We observed the size of ectopic uterine implants at the mesenteric with the naked eye and examined the concentrations of estradiol, progesterone,
and IL-2, IL-4, IL-6 and IL-10 in serum. Also, we inquired out the efficiency between oral and rectal administration groups. Results : The size of ectopic uterine implants of oral and rectal treated groups were much smaller than those of control groups. The concentration of estradiol, progesterone.
and IL-2 were significantly decreased in oral administration experimental group compared with the control group. The concentration of progesterone were significantly decreased in rectal administration experimental group compared with the control group. The efficiency of oral administration group was better than that of rectal administration group in the serum concentration of estradiol,
and IL-2. Conclusion : Our data suggest that Astragalus membranaceus reduce the size of ectopic uterine implants at the mesenteric wall and inhibit the growth of ectopic uterine implants. Judging from the above results, it can be suggested that Astragalus membranaceus should be a useful agent for inhibiting the proliferation of uterine endometrial tissue. May be, oral administration of Astragalus membranaceus is more effective than rectal administration for inhibiting the proliferation of endometriosis.
Short term effect of Allii tuberosi semen on muscular fatigue and muscular strength

Purpose: This study was conducted to investigate the muscle anti-fatigue effects of Allii tuberosi semen after short term administration. Methods: 4-6 weeks old ICR mice were used in this study and we administered the water soluble extracts of Allii tuberosi semen in the concentration of 1, 10 and 100mg/0.3ml to each 5 mice (ATS group) and same volume of normal saline was administered to 5 mice (control group) once a day for 30 or 60 days. After the administration we performed the swimming exercise test and the grip strength test. And we measured the concentration of glucose and the activity of lactate dehydrogenase in plasma regarded as biochemical indicator related with the muscular fatigue.. Results: In swimming exercise test. 10 and 100mg/0.3ml ATS groups showed increased time compared with relative control group after 30 days administration. But after 60 days adminstration all ATS groups showed increased results. In grip strength test, 100mg/0.3ml ATS group increased grip strength after 30days administration and 10mg/0.3ml ATS group showed increased result after 60days administration. All the results of plasma glucose and lactate dehydrogenase didn't showed statistically significant difference after 30 and 60 days administration. Conclusion: From this study we could find the anti-fatigue effect of Allii tuberosi semen in the muscle strength related test.

A study on effects of Ligustri Lucidi Fructus, Ecliptae Herba and Yijihwan on antioxidant activity and blood pressure

Purpose: Ligustri Lucidi Fructus and Ecliptae Herba has long been used for clinical therapy associated especially with menopausal symptoms in Korea. To provide a scientific rationale for such use, we have investigated the antioxidant and vasorelaxant effects of Ligustri Lucidi Fructus, Ecliptae Herba and its mixture. Methods: The antioxidant activity of the extracts from Ligustri Lucidi Fructus, Ecliptae Herba and its mixture were evaluated and compared with that of BHA (butylated hydroxyanisole), BHT (butylated hydroxytoluene), vitamin C and vitamin E, using the
(DPPH) radical scavenging method. Results: Antioxidant activity of all extracts using the DPPH radical scavenging method decreased in the order vitamin C>BHA>vitamin E>Ligustri Lucidi Fructus>Ligustri Lucidi Fructus:Ecliptae Herba(1:1)>Ecliptae Herba>BHT. The vasorelaxant effects of extracts were investigated on the vasomotor tone of the rat thoracic aorta in an organ bath. All of the extracts elicited along-term relaxing response in the endothelium-intact as well as endothelium-denuded rat aorta contracted with norepinephrine. This relaxant effect was abolished by Precontraction with 72 mM KCI. Thus, it is suggested that the mechanism of vasorelaxant effect of extracts not involve voltage-operated
channel blocking but receptor-mediated route. Conclusion: These antioxidant and vasorelaxant effecs of the extracts may contribute to the beneficial effects in postmenopausal women.
A study on the short-term effects of Ginseng Radix Alba extract on muscle fatigue and blood components of the rats.

Purpose: This study was conducted to investigate the short-term effects of Ginseng Radix Alba extract solution on muscle fatigue and concentration of blood components especially in glucose and lactate dehydrogenase of the rats. Methods: The 4~6-week-old ICR rats are used in the study, and we administered the extract solution of Ginseng Radix Alba concentration of 1, 10, 100mg/0.3ml to each rats and water to another rats(control group) once a day for each 30 and 60 days. After 30 and 60 days. we measured the persistent time of swimming exercise test and the results of grip strength test. And we also examined the plasma concentration of glucose, and lactate dehydrogenase. Results: In swimming exercise test, the swimming time of the 1, 10, 100mg/0.3ml groups for 30 days and the 100mg/0.3ml group for 60 days significantly increased than control group. and we found that as the concentration increased, the swimming time was also increased for 30 and 60 days experiment. And in grip strength test, the grip strength of the 1, 10, 100mg/0.3ml groups for 30 days and the 10mg/0.3ml group for 60 days significantly increased than control group. and we found that as the concentration increased, the grip strength was also increased for 30 days experiment. Plasma concentration of lactate dehydrogenase was significantly increased in the 10mg/0.3ml group for 60 days than control group. And there was no significant differences between the control and the treatment group in the plasma concentration of glucose. Conclusion: This study show that Ginseng Radix Alba can increase the muscle strength and antifatigue effect.
Effects of Pinelliae Tuber on the Surgical Induction of Endometriosis in the Rat

Purpose: Endometriosis has been thought to be related with blood stasis in uterus. Pinelliae Tuber is a herb which has effectiveness of dispelling phlegm which can be transformed into blood stasis, so is used in gynecology blood stasis-desease. Therefore in the present study, the effects of Pinelliae Tuber on endometriosis were investigated. Also retention enema therapy acts on the limited part directly, so is used frequently in gynecology clinic. This study was to examine the comparative effects on the surgical induction of endometriosis in the rat between orally and rectally administered with Pinelliae Tuber extract. Results: There was no difference in the size of ectopic uterine implants of experimental group compared with control group. The concentration of IL-10 level was significantly decreased in orally administered experimental group and the concentration of progesterone, IL-6 level was significantly decreased in rectally administered experimental group. There was no significant difference between orally and rectally administerd with Pinelliae Tuber. Conclusion: Judging from the above results, it can be suggested that Pinelliae Tuber should be not a useful agent for inhibiting the proliferation of uterine endometrial tissue. There is no difference between orally and rectally administerd with Pinelliae Tuber extract.
Antimicrobial effects of the interior warming herbs on vaginal microbe

Purpose: This study was conducted to investigate the antimicrobial effects of the interior warming herbs on vaginal microbes. Methods: Staphylococcus aureus, Methicillin-resistant Staphylococcus aureus, Candida albicans and Gardnerella vaginalis were used for vaginitis-induced microbes. Lactobacillus gasseri, Streptococcus spp. and Escherichia coli HB101 were used for normal vaginal florae. And herbs for warming the interior (Zingiberis Rhizoma, Alpiniae Officinari Rhizoma, Aconiti Tuber, Anethi Fructus, Evodiae Fructus, Cinnamomi Cortex Spissus, Caryophylli Flos, Aconiti Tube, Zanthoxyli Pericarpium, Piperis Longi Fructus, Piperis Nigri Fructus) were used in this study. Antimicrobial activities were tested by the change of optical densities (OD) and colony test in vitro. Results: In the results of vaginitis-induced microbes, Anethi Fructus was decreased the OD values on MRSA and Gardnerella vaginalis and Aconiti Tuber was decreased on MRSA. There were no viable MRSA and Gardnerella vaginalis colony forming against Evodiae Fructus, Staphylococcus aureus and MRSA colony forming against Piperis Longi Fructus, Staphylococcus aureus colony forming against Piperis Nigri Fructus and MRSA colony forming against Zanthoxyli Pericarpium. In the results of normal vaginal florae, Zingiberis Rhizoma was decreased the OD values on Streptococcus spp. and all normal vaginal florae were showed viable colony forming against all experimental herbs. Conclusion: According to these results, we can suggest that some kinds of interior warming herbs have antimicrobial effects on vaginal microbes. So there might be needed to make furthermore studies to seek the herbs which have selective antimicrobial effect on pathologic vaginal microbes.
Cell differentiation and Anti-oxydative effect of Dioscoreae Rhizoma on HeLa Cell

Purpose: This study examined the Cell differentiation and the anti-oxidative effect of Dioscoreae Rhizoma on HeLa cells. We are interested in whether Dioscoreae Rhizoma is capable of causing apoptosis processes on HeLa cell, and whether cotreatment of NCS with Dioscoreae Rhizoma reduces cell viability. Methods: We used aqueous extract to treat HeLa cell with different concentrations treated with a water or a MeOH extract of Dioscoreae Rhizoma (0, x10, x20, x40, x80). The MTT (3, (4, 5-dimethyl-thiazol) 2, 5-diphenyl-tetraxolium bromide) reduction assay was employed to quantify the differences in cell activity and viability. Cells were stained with DAPI and visualized by fluorescent Microscope. The caspase-3, Bcl-2, PARP, p53 expression level was monitored using western-blotting techniques. The patterns of the changes in expression were scanned and analyzed. Results: The survival rate of cells treated with Dioscoreae Rhizoma extracts increased by 20% at specific concentration. The other side Dioscoreae Rhizoma extracts induced apoptotic features including chromatin condensation and fragmentation. And Dioscoreae Rhizoma extracts increased the expression of caspase-3, p53 and the cleavage of PARP protein. However, co-treatment with Dioscoreae Rhizoma with NCS attenuated the activations of p53 and PARP protein that were mediated by NCS treatment alone. This is indicated Dioscoreae batatas extracts attenuated cytotoxicity induced by oxidative agents including NCS. Conclusion: Our results suggest Dioscoreae Rhizoma extracts induce cell differentiation or apoptosis connected with concentration. Further elucidation of concentration of Dioscoreae Rhizoma awaits many other biochemical investigative studies.
Effect of Morindae officinalis Radix extract on muscle fatigue and blood component

Purpose: This study was conducted to investigate the muscle anti-fatigue effects of Morindae officinalis Radix liquid extract. Methods: 4-6 weeks old ICR rats are used in the study, and we administered the extracts powder of Morinda officinalis concentration of 1, 10, 100mg/0.3 ml to each to rats(MO group) once a day for each 30 and 60 days. After 30 and 60 days, we examined the plasma concentraion of lactate dehydrogenase, glucose. And we measured the persistent time of swimming exercise test and conducted grip strength test Results: We found that there are no significantly differences between the control and the treatment group in the plasma concentration of activity level of lactate dehydrogenase. And we found that in 30days group, level of glucose concentration is significant. In swimming exercise test, as administration time and concetration growed up, swimming time is increased. And in grip strength test, as concentraion is growed up, grip strength is increased. Conclusion: This study show that Morinda officinalis is effective against muscle fatigue.
The effect of Kami-Saenghwatang and Kami-Bohertang to postpartum weight retention

Purpose: The purpose of this study was to investigate the relationships between the herbal medicines used for women in puerperium on postpartum care, Kami-Saenghwatang(SHT) and Kami-Bohertang(BHT) and postpartum body composition change. We also examined the influence of gestational age, the number of parity and the route of delivery for gestational weight gain and postpartum weight loss. Methods: 239 women followed by postpartum care center. Variety factor s were conformed and then checked body composition analysis. 8 weeks later, we followed up body composition of 36 women among the former checked. and additionally asked the body weight of another 42 former checked women by telephone call. . The number of parities had corrrelation with gestational weight gain. Age, number of parities and route of delive were not significantly correlated with postpartum weight loss during 8 weeks after delivery, and also it was not correlated with body water mass, body fat mass, body mass index and body weight on postpartum 8 weeks whether they treated with SHT and BHT. conclusion: It was not correlated with body water mass, body fat mass, body mass index and body weight on 8 weeks postpartum whether they treated with SHT and BHT.
A study on Skin Resistance Variability(SRV) of Women with Abnormal Menstrual cycle

Purpose: Recent studies show that Skin Resistance Variability(SRV) could be related to the functions of Hypothalamus-Pituitary-Ovary(H-P-O) axis. 7-Zone-Diagnostic System is oriental medicine measuring system using SRV. To know the differences of SRV of normal and abnormal menstrual cycle groups, we research the SRV of the two groups. Methods: We measured SRV of two groups that were consist of 126 women who took CP-6000A test in Sangji Oriental Medical Hospital from Mar. 2004 to Feb. 2006. After detection of SRV, we performed correlation analysis by SPSS 12.0. Results: The SRV was measured twice in 7 areas. By the results of 1st trial on Factor AA of the SRV, the patient group's results were lower than normal group's results in all 7 areas. And there were remarkable differences in 1, 2, 3, 4, 5, 7 areas between two groups. By the results of 2nd trial on Factor AA of the SRV, the patient group's results were lower than normal group's results in all 7 areas similar to 1st trial results. And there were remarkable differences in 2, 3 areas between two groups. Conclusion: Comparing normal group with patient group, these results may reflect the relations of abnormal menstrual cycle and H-P-O axis functions. Because patient's results were lower than normal's in all 7 areas. And remarkable different results in 2, 3 areas were repeated by twice trial all. Further study will be needed.
Evaluation of Health Information on Internet Sites Dealing with Oriental Obstetrics & Gynecology about Menstrual Pain

Purpose: The purpose of this study was to analyze current status of the web-sites dealing with oriental obstetrics & gynecology in the internet, and to evaluate their quality and confidence of health information. Methods: It was selected 54 local sites by searching for such key words as menstrual pain. The evaluation tool consisted of six major classification (Content, Authorship, Purpose, Functionality, Feedback and Privacy). Results: In the evaluation of web-sites, the lowest and the highest scores were 7 and 19 out of 26, and the average score was 13.94. The scores of Purpose, Content, Feedback were higher than the average, but the scores of Authorship, Functionality, Privacy were lower than the average. Conclusion: There is a need to develop a high-quality information about latest, confidence, authorship of documents, and to prevent a exposure of privacy.
A case report of Cervical incompetence

Purpose: The purpose of this study is to report the effect of oriental treatments on the cervical incompetence. Methods: Treatment of incompetent cervix has traditionally been surgical correction of presumed physical deficit in tissue strength with an encircling or cerclage suture, placed electively between 12 and 15 weeks. Despite the prolonged controversy about the role of cerclage, a randomized trial of cerclage versus bed rest or no therapy in women with atypical history of incompetent cervix has not been concluded. We treated a 32-year-old female who had been suffering from cervical incompetence with Herbal medicine(Gamidangkwisan). Results: After oriental medical treatment, the clinical symptom of Cervical incompetence was improved and she was delivered in 42 weeks. Conclusion: This case study shows that the oriental medical therapy is effective in treating cervical incompetence. If we make more studies about this disease, it will bring the high satisfaction of patient and the improvement of treatment rate.
A Case Report of Chronic Pelvic Inflammatory Disease

Purpose: The purpose of this study is to report the effect of Korean Medicine therapy to chronic pelvic inflammatory disease. Methods: A 34 years old woman, who suffered from chronic pelvic inflammatory disease was admitted to the OBGY department of Se-Myung University, Je-Cheon. In the beginning of treatment, she had pain and cold in the lower abdomen, dysuria, displeasure of sexual intercourse, etc. She received Korean Medicine therapies such as herbal medicine, moxibustion, acupuncture and got good result from them. Results: As a result, symptoms are remarkably alleviated. Conclusion: The present study suggests that Korean Medicine therapies have significant effect on chronic pelvic inflammatory disease.
