The Effect of Houttuynia cordata thunberg(魚腥草) on the inhibition of growth of leiomyomas and apoptosis

Purpose: Uterine leiomyomas (fibroids) are common estrogen-dependent uterine tumors. Houttuynia cordata thunberg has cancer-preventing properties and often used in Chinese medicine. In the present study we used Houttuynia cordata thunberg to determine its effect on cell proliferation and apoptosis in human uterine leiomyoma cells. Methods: Primary cultured human uterine leiomyoma cells were treated with Houttuynia cordata thunberg. Cell viability analysis was analyzed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTS) assay and Flow cytometry was performed to ascertain the effects Houttuynia cordata thunberg. Expression of cell cycle related proteins and apoptosis related proteins were evaluated by Western blot analysis. Results: Cell viability was significantly influenced by Houttuynia cordata thunberg treatment in a dose-dependent manner in leiomyoma cells compare to normal myometrial cells. Flow cytometric analysis showed that Houttuynia cordata thunberg induced Sub G1 arrest. DNA fragmentation assay was carried out and apoptosis was detected. Activation of caspase-3, down-regulation of Bcl-2, with concomitant increase in p21 was observed. Houttuynia cordata thunberg treatment of uterine leiomyoma cellsresulted in a concentration-dependent cell death induced via the caspase dependent mechanism. Conclusion: These results suggest that Houttuynia cordata thunberg treatment in uterine leiomyoma cells leads to growth inhibition and induced apoptosis. These results suggest that Houttuynia cordata thunberg will be a promising agent for use in therapeutics agents against human uterine endometrial cancer.
The Effect of Guibitang(歸脾湯) on the Ovarian Functions and Differential Gene Expression of Caspase-3, MAPK and MPG in Female Mice

Purpose: To evaluate the effect of administration of Guibitang on ovarian functions and differential gene expressions related cell viabilities caspase-3, MAPK and MPG in female mice. Methods: We administered the Guibitang to 6-week-old female ICR mice for 4, 8, or 12 days. Then, the female mice were injected PMSG and hCG for ovarian hyperstimulation. The mice divided into 3 different groups for each experiment. To compare the differences, we set a control group treated with plain water at the same volume by the same way. Results: administration of Guibitang, the mean number of total ovulated oocytes and the number of morphologically normal oocytes increased significantly compared to control group. And the rates of blastocyst formation from 2-cell stages alsa increased significantly compared to control group. Capase-3 gene expression which is known to maker gene for cell apoptosis were significantly lower than that of control group. And MAPK and MPG gene expressions for cell viability and DNA repair were same that of control group. Conclusion: From our results suggested that the medication of Guibitang has beneficial effect on reproductive functions of female mice via prevention of cell apoptosis and DNA damaging and promotion of cell proliferation.
Anti-proliferative Effect of Ulmi Pumilae Cortex Extracts on MCF-7 cells

Purpose: This study was conducted to investigate the anti-proliferative effects of Ulmi Pumilae Cortex Extracts(UPCE) on MCF-7(human, breast, adenocaecinoma) and NIH3T3 (human, murine, fibroblast). Methods: MCF-7 cells and NIH3T3 cells were cultured and seeded in cell culture plates, respectively. UPC was extracted with hot water and then further fractionated it into five types: hexane, chloroform, ethyl acetate, butanol, and water soluable fractions. These five different fractions from UPCE were tested for their anti-proliferative effects on MCF-7 cells and NIH3T3 cells by MMT assay. Results: Among the five solvent-fractions of UPCE, n-hexane fraction and ethyl acetate fraction showed a strong anti-proliferative effects on MCF-7 cells but they displayed significant cytotoxicity on NIH3T3 cells, too. On the other hand, chloroform fraction showed a marked anti-proliferative effects on MCF-7 cells and low cytotoxicity on NIH3T3 cells. Conclusion: Chloloform fraction from UPCE showed selective anti-cancer activities on human breast cancer cell MCF-7 relatively to the other fractions.
The Experimental Study on Anti-thrombotic and Anti-inflammatory Effect of NeungaSoJeokTang(NSJT)

Purpose: This study was performed to evaluate anti-thrombotic and anti-inflammatory effects of NeungaSoJeokTang water extract (NSJT). Methods: In the study of anti-inflammatory effects, NSJT was investigated using cultured cells and murine models. As for the parameters of inflammation, levels of several inflammatory cytokines and chemical mediators which are known to be related to inflammation were determined in mouse lung fibroblast cells(mLFC) and RAW 264.7 cells. Results: Prior to the experiment, we evaluated sGOT, sGPT, BUN and creatine after the treatment. As a result, NSJT was innoxious on liver and kidney. In experiment of anti-thrombotic effect, NSJT inhibited the platelet aggregation induced by ADP and epinephrine, and inhibited pulmonary embolism induced by collagen and epinephrine. NSJT did not affect significantly the blood flow rate both in vitro and in vivo. NSJT increased platelet number and fibrinogen amount, and NSJT shortened PT and APTT in thrombus model induced by dextran. In experiment of anti-inflammatory effect, NSJT inhibited
, COX-2 and NOS-II mRNA expression in a concentration-dependent manner in RAW 264.7 cell line, and inhibited significantly NO production at 50, 100
, and also inhibited ROS production in a concentration-dependent manner. NSJT inhibited

production significantly in serum of acute inflammation-induced Balb/c mice, and decreased

production in spleen tissue, but increased

production in liver tissue. NSJT increased survival rate at the 3th day in ICR mice with lethal endotoxemia induced by LPS. Conclusion: These results suggest that NSJT can be used for treating diverse female diseases caused by thrombosis and inflammation such as pelvic pain, pelvic inflammatory disease as well as vulvar pain due to vulvitis, vulvar vestibulitis and so on.
Effects of the oral administration of Epedra Sinica Extract on suppression of body weight gains and the DNA chip expression of obese rats.

Purpose: This study is to examine anti-obesity effect and cytotoxicity of the long-term oral administration of Ephedra Sinica(Ma-hwang, ES) Methods: Using diet-induced obesity C57BL/6 mouse model, anti-obesity effect and DNA chip expression and cytotoxicity of the long-term oral administration of this herbal extract were investigated. Results: The herbal extract treated groups were arrested in weight increment only when they were lodged together. Such effects were abolished when they kept individually. ES fed mice behaved very rudely and violently. On the basis of histological studies of liver tissues and also in vitro cytotoxicity tests of the liver and kidney cell lines, no significant toxicity was found by 14 weeks of ES treatments. However, we found significant changes in gene expression profile in ES treated group by micro-array analysis. In case of ES group, up-regulated genes were 113 and down-regulated were 120. Some of lipid metabolism related genes also significantly changed in treatment groups. Conclusion: ES had effects of increasing the basal metabolic rate by stimulating the sympathetic nervous systems.
Long Term Effect of Allii tuberosi semen on Muscle Fatigue and Muscular Strength

Purpose: This study was conducted to investigate the muscle anti-fatigue effects of Allii tuberosi semen after long term administration. Materials and Methods: 4-6 weeks old ICR mice were used in this study and we administered the water soluble extracts of Allii tuberosi semen in the concentration of 1, 10 and 100mg/0.3ml to each 5 mice (ATS group) and same volume of normal saline was administered to 5 mice (control group) once a day for 90 or 120 days. After the administration we performed the swimming exercise test and the grip strength test. And we measured the concentration of glucose and the activity of lactate dehydrogenase in plasma regarded as biochemical indicator related with the muscular fatigue. Results: In swimming exercise test, all ATS groups showed increased time compared with relative control group after 90 days administration. But after 120 days adminstration 10 and 100mg/0.3ml ATS groups showed increased results. In grip strength test, all ATS group increased grip strength after 90 days administration and 10mg/0.3ml ATS group showed increased result after 120 days administration. All the results of plasma glucose didn't showed statistically significant difference after 90 and 120 days administration. In the plasma concentration of activity level of lactate dehydrogenase, 1mg/0.3ml and 10mg/0.3ml ATS groups showed decreased concentration compared with relative control group after 90 days administration. But after 120 days adminstration 1mg/0.3ml ATS groups showed decreased results. Conclusion: From this study we could find the anti-fatigue effect of Allii tuberosi semen in the muscle strength related test.
The Effect of Palmultang(八物湯) on the Ovarian Functions and Differential Gene Expression of Caspase-3, MAPK and MPG in Female Mice

Purpose : These experiments were undertaken to evaluate the effect of administration of Palmultang on ovarian functions and differential gene expressions related cell viabilities caspase-3, MAPK and MPG in female mice. Materials and Methods : We administered the Palmultang to 6-week-old female ICR mice for 4, 8, or 12 days. The female mice were injected PMSG and hCG for ovarian hyperstimulation. And then recovered ovaries were minced and extracted mRNA and analyzed cell viability related gene expression. We chose the caspase-3 for cell apoptosis, MAPK and MPG genes for cell viability and DNA repair. To compare the differences, we set a control group treated with plain water at the same volume by the same way. Results : In case of administration of Palmultang, the mean number of total ovulated oocytes and the number of morphologically normal oocytes increased significantly compared to a control group. We were also examined the embryonic developmental competence in vitro. The administration of Palmultang in a concentration with 10 and 100 mg/ml were beneficial effect of embryonic development in preimplantation period. The administration of Palmultang play a role of prevention of cell apoptosis and DNA damages and also increased cell proliferation resulted in ovarian functions. Conclusion : From our results suggested that the medication of Palmultang has beneficial effect on reproductive functions of female mice via prevention of cell apoptosis and DNA damaging and promotion of cell proliferation.

The evaluation of usefulness for far-infrared radiating under inner-wear on dysmenorrhea

Purpose: We intended to observe the usefulness of far-infrared radiation functional under inner-wear for dysmenorrhea. Methods: We introduced far-infrared radiation functional under inner-wear to women in childbearing-age by internet portal site. Then, we preliminarily examined dysmenorrhea severity by VAS(visual analogue scale) questionnaire to women interested in far-infrared radiation functional under inner-wear. We selected women scored 5 and above of 10 measurement VAS score. Finally, 121 women were the subject of study and used far-infrared radiation functional under inner-wear for one menstrual cycle. Before and after use of far-infrared radiation functional under inner-wear, we conducted a questionnaire survey of dysmenorrhea severity by VAS. After that, we compared VAS score before and after use. Also, we studied correlation between frequency of far-infrared radiation functional under inner-wear use and
(VAS score before use minus VAS score after use). For statistics, we used Paired samples test and Spearman's rho correlations, SPSS 13.0 for windows. Results: Before and after use of far-infrared radiation functional under inner-wear, VAS score means were different. Frequency of far-infrared radiation functional under inner-wear use was correlated to
. Statistically they showed significant result (p<0.05>. Conclusion: The results showed that dysmenorrhea severity by VAS decreased after far-infrared radiation functional under inner-wear use. As frequency of use increased,
increased. So we can consider far-infrared radiation functional under inner-wear effects dysmenorrhea severity.
The Research on the Characteristics of MMPI Profile and Sasang Constitutional on Dysmenorrhea Patients

Purpose: The purpose of this study was to investigate Minnesota Multiphasic Personality Inventory on clinical scales and personality scales of dysmenorrhea patients. Methods : 29 dysmenorrhea patients were tested by MMPI and QSCC II. It is analyzed by one-way ANOVA, Crosstabulation, t-test, and by Duncan method in case of significant difference. Results and Conclusion : 1. The MMPI scores of dysmenorrhea patients were normal range that was elevated in Hs, Hy clinical scales, 1-3 profile type which is profile of conversional neurosis. 2. All of the profiles can be classified three types of profile. Group 1(44.8%) showed a normal profile. Group 2(20.7%) showed a normal profile but elevated in Mf, Ma scales, suggesting mainly extroverted and psychopathic personality. Group 3(34.5%) showed 1-3 profile type which is profile of conversional neurosis. 3. MDQ score and age of dysmenorrhea patients are related to personality in MMPI profile of dysmenorrhea patients.
A Study on the Relation of Dysmenorrhea of some patients and Body Composition Analysis

Purpose: Dysmenorrhea is a women's common disease. The aim of this study is to investigate the relation between body composition and VAS, VRS, MVRS of dysmenorrhea. Methods: The subject were 17 women who took body composition(body mass index, percent body fat, waist-hip ratio) analysis(Inbody 720) and answered the questionnaire have been calculated VAS, VRS and MVRS in Kangnam Korean Hospital, Kyunghee Univ. from Apr. 6, 2007 to Mar. 25, 2007. Results: There was statistically significant correlation between BMI and dysmenorrhea VRS, MVRS. There was statistically significant correlation between BMI and dysmenorrhea VRS, MVRS. There was statistically significant correlation between percent body fat and dysmenorrhea VRS, MVRS. There was statistically significant difference of dysmenorrhea VRS, MVRS between BMI underweight group and BMI normal or overweight group. There was statistically significant difference of dysmenorrhea VAS between waist-hip ratio normal group and waist-hip ratio abnormal group. Conclusion: Body composition analysis(body mass index, percent body fat, waist-hip ratio) is useful to predict pain grade of dysmenorrhea.
Analysis on the Heart Rate Variability of Dysmenorrhoea Patients

Purpose: The purpose of this study is to analysis on the Heart Rate Variability(HRV) of some dysmenorrhoea patients. Methods : We studied 30 patients visiting Kyung Hee University East-West Neo Medical Center from 7th Febrary 2007 to 22th Febrary 2007. Women with organic disease were excluded from this study. The severity of dysmenorrhoea were measured by Multidimensional Verbal Rating scale(MVRS), Verbal Rating scale (VRS) and Visual Analog Scale(VAS). And HRV was recorded. Results : According to comparison of Means of HRV values among the groups of None, Mild and Moderate, the points of almost all values were kept by order of Moderate group-Mild group-None group. Among these, the statistically significant things are PR, SDNN, Ln (TP), Ln (VLF), Ln (LF). According to correlation analysis between dysmenorrhoea and HRV, specially, there were many correlations in VRS2. Among these, the statistically significant things are LF norm, HF norm, LF/HF ratio(p<0.05L). Conclusion : The results suggest that HRV is related to dysmenorrhoea. We may analyze that dysmenorrhoea is related to, specially, increased activities of sympathetic nerve. We need more study for settling this.
A Study on Characters of Heart Rate Variability in Postpartum Disease Patients

Purpose: During postpartum period, many women complain multiple pain, sweating, cold hypersensitivity, abdominal pain, hot flush and so on. We called the postpartum disease as San Hu Pung. The San Hu Pung could related with dysautonomia. So we studied the character of the postpartum disease patients as heart rate variability(H.R.V) Methods: The subjects were 63 women who visited Kangnam Kyunghee hospital for postpartum management from February 2006 to June 2007. From history taking, 42 symptomatic women was divided at the postpartum disease group, 21 asymptomatic women was divided at the control group. All women was measured HRV, we studied the difference of HRV between two group. Results: Compared with the control group, SDNN of the disease group was significant low. TP, LF and HF of the disease group were significantly lower than the control group. Conclusion: The difference of HRV between the disease group and the control group was significant. The results of HRV on the disease group could mean that San hu pung is related dysautonomia. The more studies to diagnose San hu pung would be needed.
Analysis for 23 studies of patients that used Herbal Medicine during Pregnancy

Purpose: This study was designed to establish safety and efficacy of using herbal medicine during pregnancy. Methods : We searched through 23 sources, which had been previously published frome 1988 to 2007, concerned patients that used herbal medicine during pregnancy. We searched diseases for that used herbal medicine, kinds of herbal medicine taken, the period of herbal medicine taken, the duration of herbal medicine taken and results of herbal medicine taken. Results: 1. Whaltae(habitual abortion) was the most of diseases that used herbal medicine during pregnancy. Second disease was hyperemesis gravidum. Third disease was Taegibulan (fetal restlessness). 2. The most of Herb medicines of used for Whaltae was Kyoesamultang and Anjeonecheontang. The most of Herb medicines of used for hyperemesis gravidum was Bosaengtang. The most of Herb medicines of used for Taegibulan was Kyoesamultang. 3. The time of herbal medicine taken mostly was the first period of pregnancy. The rate was 78%. 4. The duration of herbal medicine taken mostly was
at mean. 5. The rate of normal delivery was 85% and herbal medication during pregnancy does not cause any deformity to the newborn and of patients used herbal medicine. 6. The patients of taken herbal medicines were recovered from their symptoms and prevented miscarriage. The rate of recovery was 87%. Conclusion: The results of this study show that herbal medication during pregnancy is effective to cure disease and maintain the pregnancy. It is thought that have to actively try to treat diseases and symptoms on pregnancy by using herbal medicine.
A study on the minimal acupuncture using a modified guide tube for Korean female participants familiar with acupuncture therapy

Purpose: To find objective validity of minimal acupuncture using a modified guide tube as a method to verify the effect of acupuncture. Methods: Subjects of this study were participants of a randomized clinical trial which was practiced to verify the effect of acupuncture on postmenopausal hot flash. There were 2 arms of treatment, one's for active acupuncture group(AG) with manipulation and De qi, the other's for minimal acupuncture group(MG) using a modified guide tube which was designed to give minimal stimulation to the patient. After 8 weeks' treatment followed by 4 weeks' observation, participants were asked to fill in self-report forms on their education, previous experiences of acupuncture, original idea on the efficacy of acupuncture and recognition of blinding. Results: 19 participants of 26 in AG and 19 of 26 in MG completed the self-report form. There was no difference between AG and MG in their education(p=0.5976, Fisher exact test) and previous experiences of acupuncture(p=0.9999, Fisher exact test). In their original ideas on the efficacy of acupuncture, most of AG and MG respondents have been thought that acupuncture is very effective or quite effective, and there was no difference between 2 groups(p=0.5065). 16 of 19 in AG and 14 of 19 in MG believed that they underwent more effective way, there was no statistical difference between 2 groups(p=0.6928, Fisher exact test). Participants believed in undergoing more effective way not because they recognized treatment method, but they were satisfied with the effectiveness of treatment retrospectively. Conclusion: As the result of this study, these authors suggest that minimal acupuncture using a modified guide tube is acceptable as a method to verify the effect of acupuncture in acupuncture-familiar Korean culture. Also further studies on the stimulation-specific effect of minimal acupuncture are demanded.
A Clinical Trial to Verity the Quality of Life Improvement Efficacy of Dangguijakyak-san and Gyejibongnyeong-hwan Granulation in Postmenopausal Women.

Purpose: This master‘s thesis to evaluate the grade of life after medication a Dangguijakyak-san(pasabu) and Gyejibongnyeong-hwan(cheongwal) with in postmenopausal women get lowering quality of life caused by vasomotor symptoms with hot flush. Methods: A subject who signing on the clinical trial written consent by self-will is registered this clinical trial after decided suitable by selection and exception standard, after take a medical experiment and checkup according to clinical trial plan. Registered subject should valuated by settled schedule after take the testing medicine 1,2 during thirty-day. In this period, a subject allocated at relatively better suited experimental group by oriental doctor after consideration of general efficacy and nature of a medicine at second visiting. Results: The result of comparison in the remedial value with sixteen patients who ended the experiment is like next list. 1. There's no regarded difference of comparison in general conditions between two patient groups. 2. There's no regarded difference during observation period before take medicine. 3. By period of measurement, there's something regarded differences most of patients of two parts after taking medicine. 4. There's no regarded difference at alteration phase of each standard according to kind of medicine. 5. There's no abnormal views reflected at allergy, Laboratory and Physical Examination during experiment. Conclusion: This experiment evidence a Dangguijakyak-san and Gyejibongnyeong-hwan can help for improvement generally life quality of postmenopausal women and certify safety of herbs.
4 Cases Report of Hyeolheopungyeol type(血虛風熱型) Dermatoses Gravidarum treated with sopungsan-gami(消風散加味)

Purpose: The purpose of this study was to report the clinical application of traditional korean medicine(TKM) treatment to hyeolheopungyeol type of dermatoses gravidarum. Methods: We experienced four patients who had been suffering from itching, plaque, erythema during pregnancy. They had already been treated with antihistamine or ointment, but they were not improved. We diagnosed their case as hyeolheopungyeol type of dermatoses gravidarum, and treated with sopungsan-gaml. Results: After TKM treatment, itching, plaque and erythema were alleviated or resolved. And they gave birth to a healthy baby. Conclusion: The results indicate that TKM, sopungsan-gami can be effective for the management of hyeolheopungyeol type of dermatoses gravidarum.
A Case Report of Postpartum Disease Induced by Inadequate Postpartum Care

Purpose: In postpartum period, women are very weak and liable to various diseases. So postpartum care is very important. But excessive heating and sweating cause postpartum diseases. Methods: The patient in this case, 37 years-old female was in Kyunghee University East-West neo medical center for 8days(27th/Oct/2006 - 3rd/Nov/2006). Her chief complains when she was admitted in the hospital were sweating, chilling, and mild dizziness. These symptoms were caused by excessive heating and sweating. We thought that her condition was deficiency of Qi. After giving her the therapies with herb medicine, acupuncture and moxibustion, her symptoms got almost disappeared and her condition got better. Results: After the oriental medical treatment, the clinical symptom of postpartum disease was improved. Conclusion: This case study shows that excessive heating and sweating cause postpartum diseases and the oriental medical therapy is effective in treating postpartum disease. And the guideline of postpartum care is needed.
Clinical study for the one case who contracts of Arthritis after parturition

Purpose: The purpose of this study is to report the effect of TKM to Rhumatitis Arthritis after parturition and show the importance of recuperation after parturition. Methods: The patient was a 33-year-old woman who complains arthralgia, edema and sense of coldness after her third parturition. The patient was treated by Herb medicine, Acupuncture treatment, Moxa treatment and Physical Treatment. And the progress of symptoms was evaluated by cheking the change of VAS, and inspecting the DITI. Results: The Pain and edema have declined. After discharge, she got definite diagnosis as Rhumatitis Arthritis. But We already estimated it from the results of serum test. So we had treated the patience with TKM and we could explain the prognosis. We regulated the pain and edema successfully at the acute phase. And the patient could take care of herself, she didn't play her disease down as a simple symptoms of parturition. Conclusion: TKM is expected to have positive effects on Rheumatic arthritis with quick onset after parturition.
