Characterization of γ-Aminobutyric acid(GABA) produced by a lactic acid bacterium from button mushroom bed

-Aminobutyric acid(GABA) is a four carbon non-protein amino acid that has several well-known physiological functions, such as a postsynaptic inhibitory neurotransmitter in the brain and induction of hypotensive and tranquilizer effects. A lactic acid bacterium was isolated from button mushroom bed, which is showing high GABA productivity by TLC or HPLC analysis. The strain was identified as Lactobacillus hilgardii by analysis of 16S rDNA gene sequence. When the maximum production of GABA by L. hilgardii was investigated with various concentration of monosodium glutamate, the yield of GABA reached to be 53.65 mM at 1% mono sodium glutamate (MSG) in flask cultivation. A Glutamate decarboxylase (GAD) enzyme, which was known to convert MSG to GABA, was purified from a cell-free extract of L. hilgardii and the molecular weights of purified GAD was estimated to 60,000 by SDS-PAGE. The optimum pH and temperature of GAD were at pH4.6 and at
, respectively. The GAD activity was increased by the addition of sulfate ions such as ammonium sulfate, sodium sulfate and magnesium sulfate, indicating that the increase of hydrophobic interaction causes the increase of GAD activity.
Production of lactic acid by Lactobacillus paracasei isolated from button mushroom bed

A galactose fermentation bacterium producing lactose from red seaweed, which was known well to compromise the galactose as main reducing sugar, was isolated from button mushroom bed in Buyeo-Gun, Chungchugnamdo province. The lactic acid bacteria MONGB-2 was identified as Lactobacillus paracasei subsp. tolerans by analysis of 16S rRNA gene sequence. When the production of lactic acid and acetic acid by L. paracasei MONGB-2 was investigated by HPLC analysis with various carbohydrates, the strain MONGB-2 efficiently convert the glucose and galactose to lactic acid with the yield of 18.86 g/L and 18.23 g/L, respectively and the ratio of lactic acid to total organic acids was 1.0 and 0.91 g/g for both substrates. However, in the case of acetic acid fermentation, other carbohydrates besides galactose and red seaweed hydrolysate could not be totally utilized as carbon sources for acetic acid production by the strain. The lactic acid production from glucose and galactose in the fermentation time courses was gradually enhanced upto 60 h fermentation and the maximal concentration reached to be 16-18 g/L from both substrates after 48 h of fermentation. The initial concentration of glucose and galactose were completely consumed within 36 h of fermentation, of which the growth of cell also was maximum level. In addition, the bioconversion of lactic acid from the red seaweed hydrolysate by L. paracasei MONGB-2 appeared to be about 20% levels of the initial substrates concentration and this results were entirely lower than those of galactose and glucose showed about 60% of conversion. The apparent results showed that L. paracasei MONGB-2 could produce the lactic acid with glucose as well as galactose by the homofermentation through EMP pathway.

Polyporus umbellatus (Syn. Grifola umbellata) is a sclerotium forming mushroom belongs to family Polyporaceae of Polyphorales, Basidiomycota. The sclerotia of P. umbellatus have long been used for traditional medicines in China, Korea and Japan. This study was initiated to obtain the basic data for artificial sclerotial production of P. umbellatus. Here, we investigated the favorable conditions for mycelial growth of P. umbellatus and its symbiotic fungus Armillaria mellea. We also evaluate the favorable carbon and nitrogen sources for sclerotial formation in dual culture between P. umbellatus and A. mellea. Molecular identification of fungus gnats from shiitake mushroom in Korea

Fungus gnats are usually found in mushroom farm and have recently become important pest because they can cause severe damage and reduce the production on shiitake mushroom. Usually shiitake mushrooms are cultivated on both oak bed logs and in the artificial sawdust beds in greenhouses. Using yellow sticky trap, the dipteran species in shiitake mushroom farm were collected from May to September in Kyonggi-do and Chungcheong-do in 2013. To identify the main species of fungus gnat on the shiitake farm in Korea, the collected samples were determined the sequence of cytochrome c oxidase subunit I (COI) by DNA barcoding. The phylogeny based on maximum likelihood analyses from COI sequence showed that Bradysia difformis and B. alpicola were main species of shiitake bed log and Scatopsidae sp. and B. difformis were dominant species of sawdust beds.
Composition analysis of raw material constituting the medium for mushroom cultivation

The contents of raw materials which are components of mixed substrate for mushroom cultivation were analyzed to optimize the composition. The pure protein(amino acid) level of soybean meal was the highest, 44.02% followed by those of soybean curd residue(31.5%) and cotton seeds meal(30.6%). The non protein nitrogen(NPN) contents in crude protein of main nitrogen materials were 2.4% for soybean meal and 5.6% for dried soybean curd residue, while those of wheat bran and rice bran used as the carbon source were relatively higher, 17.6% compared to that of nitrogen supplying media. Crude protein content per price was 6.0 for rapeseed meal, indicating that it is high crude protein content per price. Nitrogen-free extract(NFE) considering as an ingredient for mycelial growth were high in alphacorn(72.9%) and wheat bran B(57.2%). Acid detergent fiber(ADF) was high in corncob, 51.88%, its use for cultivation of brown rot fungi including Lentinus lepideus should pay attention because the fungi lack complete lignin degradation activity.
Heating and cooling properties in cultivation room and cultural characteristics of oyster mushroom according to type of air exchange

This study was carried out to find out suitable input method for air exchange about bottle cultivation of oyster mushroom. There was no difference of average temperature in cultivation room, but T1(direct introduction of outside air) was higher than T2(heat exchanger) and T3(air buffer) in the standard deviation. The ratio of cooling operating was the highest in August and in the descending order, T1 54%, T3 43%, T2 33%. At ratio of energy reduction, T2 and T3 were higher than T1. The operating ratio of heater was highest in January and in the descending order, T1 53%, T3 37%, T2 30%. At ratio of energy reduction(%), T2 and T3 were higher than T1 similar to result of cooling operating. Therefore there were largest in August about Cooling Degree-Hour and in January about Heating Degree-Hour. And fruitbody yields quality was excellent in T2 and T3 than T1. The suitable effective type of air exchange in oyster mushroom cultivation are heat exchange and air buffer system.
Yields of oyster mushroom(Pleurotus ostreatus) on addition rate of Juglans mandshurica sawdust

This study was carried out to investigated optimum mixing ratio of Korean natural Juglans mandshurica using as functional plants. Total nitrogen and carbon source of J. mandshurica was 0.21% and 46.0%, respectively and C/N ratio was 219. Total nitrogen source and pH of substrate mixed with J. mandshurica was 2.3~2.9 and 5.0, respectively. The contents of CaO, MgO and
at J. mandshurica media were higher at harvest media than inoculation media. Mycelial growth was the fastest at J. mandshurica 20%, and slower by increase of J. mandshurica substrate. Yields of fruiting body show the highest to 163 g/850 mL of medium which are addition 10% of J. mandshurica sawdust, and diameter and thick of pileus were the highest, too. The L value of pileus and stipes were increased by increase of J. mandshurica substrate, but there was no significant difference in the a-value and the b-value. The contents of

Effects of cultural characteristics of Lentinula edodes according to LED wavelength with sawdust substrate cultivation

This study was carried out to elucidate suitable wavelength of light during development of fruit body in Lentinula edodes. The four colors of LED(Light Emitting Diode), blue, green, red and yellow, were irradiated for formation of fruit-body. The effect of color of LED at all growth stage, the lightness of cap showed darker in blue and green LED irradiation than that of red and yellow LED. The longer stipe were resulted in longer wavelength. And the activity of anti-oxidant did not showed big differences according to LED wavelength. We obtained higher commercial yields and lower ratio of abnormal fruit body in green LED than those in control(fluorescent lamp).
Biological control of mushroom flies using the predatory mite Hypoaspis aculeifer in a shiitake cultivation

The major species of fungus gnats which caused the severe damage in shiitake farm were identified as a Bradysia difformis, B. alpicola, and Camtomyia cortocalis on oak log beds cultivation. The B. difformis occurred early in the middle of March while B. alpicola and C. cortocalis appeared since the beginning of May. The occurrence rate for adults of B. difformis showed highly at the end of July (11.9~1,774.2 in dong-myeon and 0.4~2,583.3 in pungse-myeon) in 2012 and mid-June (10.7~4,650 in dong-myeon and 36.8~4740 in pungse-myeon) in 2013. The counting numbers on the traps for B. alpicola reached highest peak in the middle of June (2.1~63.2 in dong-myeon and 1.0~21.7 in pungse-myeon) and the end of May (0.8~163.7 in dong-myeon and 0.5~280.5 in pungse-myeon) in 2012 and 2013, respectively. The number of C. cortocalis showed high record in the middle of May in 2012 (0.6~4.7) and in the middle of June (2.1~17.3) in 2013 in dong-myeon whereas showed the peaks in the middle of May (0.6~4.7) in 2012 and in the late of May (1.3~17.6) in 2013 in pungse-myeon. The fruiting bodies of shiitake mushroom by fungus gnats were severely damaged from mid-June to late-July and the damage rate were 0.625.5% (2012) and 0.7~30.5% (2013) in dong-myeon and 1.5%~21.6% (2012) and 1.9~36.8%(2012) in pungse-myeon. To investigate the control effect for fungus gnats by Hypoaspis aculeifer, H. aculeifer (30 mixutre of nymph and adult per
) were treated to oak log beds shiitake cultivation for six times (May 2 and 28, June 25, July 10 and 25 and August 28). The occurrence rate of adults and damage rate of fruiting bodies of 3 major species reduced 79.3% (adult numbers) and 74.8% (fruiting bodies) in dong-myeon and 64.1% (adult numbers) and 65.5% (fruiting bodies) in pungse-myeon, respectively, compared to non-treatment. Accordingly, H. aculeifer effectively controlled the fungus gnats on shiitake mushroom and can be used as good control agent.
Development of the log cultivation techniques of the maitake Grifola frondosa

This study was conducted to develop the log cultivation techniques of the maitake Grifola frondosa. We used two kinds of Grifola frondosa strains for this study. Fully incubated logs were buried into soil directly and were buried with soil inside of container box in 70% shaded vinyl house. In the vinyl house Yipsaeilho cultivar buried by the soil was delayed about 15days in harvest date compared with Dabak cultivar. Color of fruit body of Yipsaeilho cultivar was dark brown and that of Dabak cultivar was grayish brown. Yields of Yipsaeilho cultivar was 15-16
, more than that of Dabak cultivar, 11-13
. In the container box, Yipsaeilho cultivar buried by the soil was also delayed about 5days in harvest date compared with Dabak cultivar. Color of fruit body was similar to the result of vinyl house soil reclamation. But yields difference was not shown between two cultivars, 1.2-1.4 kg/box.
Characteristics of a new cultivar 'Hwaseong 5ho' in Pleurotus ostreatus

"Hwaseong 5ho" was developed by the method of Di-mon mating between monokaryotic strains derived from "Hwaseong 1ho" and dikaryotic strain "PSC109". The color of pileus was dark grayish brown, the shape of pileus was convex or infundibuliform. The length of stipe was longer and the thickness of stipe was some thinner than Suhan 1ho. Material properties of stipe of "Hwaseong 5ho" was higher in strength, hardness, chewingness and brittleness than Suhan 1ho, but similar in elasticity and cohesion. RAPD using URP-primer showed not the same between two strains. Days of primordia formation period were 22-27 days after spawning, that was a little later than Suhan 1ho. In the trial using culture box containing composted cotton waste, yield index of 'Hwaseong 5ho' was 16.6% higher than Suhan 1ho. The farm field trial were showed stable productivity in each different growing conditions.
Characteristics of a new variety 『Gonji10ho』 in Neolentinus lepideus

'Gonji10ho', a new variety of Neolentinus lepideus, was bred by mating two monokaryotic strains isolated from 'GMNL66037' and 'GMNL66014' in Mushroom Research Institute, Gyonggi-Do ARES in 2012. The optimum temperature for the mycelial growth of 'Gonji10ho' was
on PDA medium and that for the primordia formation and the growth of fruiting body of 'Gonji10ho' was
. In bottle cultivation of 'Gonji10ho', the period of spawn running was around 35 days at
and the period from scratching of inoculum to harvesting was 27 days. 'Gonji10ho' had thicker pileus and stipe than control variety('Solhyang'). The number of available stipe was fewer than that of control variety. During the cultivation of 'Conji10ho', the occurrence rate of Trichoderma spp. was lower than that in control variety. In bottle cultivation, the yield of fruiting bodies of 'Gonji10ho', which was 38% higher than that of control variety, was 69.6 g/900 ml bottle, and in bag cultivation, that of 'Gonji10ho', which was 10% higher than that of control variety, was 114.2 g/1 kg bag. In physical characteristics, the strongness and brittleness of the pileus of 'Gonji10ho' were higher than those of control variety. When considered in the period available for sale, the shelf life of 'Gonji10ho', which was 5 days longer than that of control variety, was 34 days.
Antioxidant and tyrosinase inhibitory activity of Hypsizygus marmoreus (brown cultivar) methanol extracts

The objective of this study was to evaluate antioxidant effect and tyrosinase inhibitory activity of methanol extracts from Hypsizygus marmoreus. The Hypsizygus marmoreus was divided into two parts (pileus and stipe) and extracted with methanol. Total polyphenolics and flavonoids in the methanol extracts were measured by spectrophotometric methods and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities have been determined for antioxidant activities. The total polyphenolics and flavonoids contents of methanol extract of the pileus were higher than methanol extract of the stipes. The total polyphenolics contents in methanol extracts of the pileus and stipes were 8.7 ug/mg and 5.6 ug/mg, respectively. The total flavonoids contents in methanol extracts of the pileus and stipes were 2.8 ug/mg and 1.4 ug/mg, respectively. The tyrosinase inhibitory activity was proportional to concentration of methanol extract. The tyrosinase inhibitory activity of the methanol extract (200 mg/ml) of pileus (66.9%) and stipe (57.97%) was lower than those of positive control 2% arbutin. The DPPH radical scavenging activity of the methanol extract (20 mg/ml) of pileus and stipes was 52.55% and 30.35%, respectively. Moreover, the effects of methanol extarcts on cell proliferation of B16BL6 mouse melanoma cells were investigated using WST-1 assay (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulphonate) and B16BL6 mouse melanoma cells treated with methanol extract of 200-2,000 ug/ml were higher proliferation rate than those of 0.04% adenosine.
Analysis of nutritional contents and physiological activities of Neolentinus lepideus

This study was conducted to investigate the physiological activities and nutrition contents of Gonji No.10 and Solhyang, the strains of Neolentinus lepideus. In nutritional contents of two strains of N. lepideus, the content of free sugars and sugar alcohols and total polyphenol were more in Gonji No.10 than Solhyang. The total contents of amino acids and
-glucan in Solhyang are more than those in Gonji No.10. Solhyang also showed higher DPPH radical scavenging activity than Gonji No.10. In particular, hot water extract of Solhyang has higher inhibitory activity of
-amyloglucosidase while Gonji No.10 has higher anti-inflammatiory effect than Solhyang. But, they have similar effects on nitrite scavenging activity.

Dictyophora indusiata is an edible mushroom belongs to Family Phallaceae of Phallales, Basidiomycota. The purpose of this study was to investigate the antioxidant and anti-inflammatory activities of methanol and hot water extracts prepared from fruiting bodies of Dictyophora indusiata. Besides measuring of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, a reducing power and a chelating activity on ferrous ions were also measured to evaluate the antioxidant activity for those extracts. To measure the anti-inflammatory activities for the extracts, nitric oxide(NO) production from lipopolysaccharide(LPS) treated RAW 264.7 macrophage cells and carrageenan-induced acute hind paw edema of rats were investigated. The results showed that the extracts have excellent DPPH scavenging and chelating activity on the ferrous ions compared with positive control. The nitric oxide(NO) production in LPS-stimulated RAW 264.7 macrophage cells were decreased as we increased the concentration of the mushroom extracts. Significant reduction of paw edema of rats were observed at 2~6 h after treatment of methanol and hot-water extracts with 50 mg/kg concentration to the rats which are induced acute hind paw edema by carrageenan administration. Therefore, the experimental results suggested that methanol and hot-water extracts of Dictyophora indusiata fruiting bodies might be used for natural sources of antioxidant and anti-inflammatory agents.

Phellinus xeranticus is an medicinal mushroom belongs to Family Hymenochaetaceae of Polyporales, Basidiomycota. The purpose of this study was to investigate the antioxidant, anti-inflammatory and anti-acetylcholinesterase activities of methanol and hot water extracts prepared from fruiting bodies of Phellinus xeranticus. Besides measuring of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, a reducing power and a chelating activity on ferrous ions were also measured to evaluate the antioxidant activity of the extracts. The nitric oxide (NO) production in LPS-induced RAW 264.7 macrophage cells were decreased as the concentration of the mushroom extracts increased. Significant reduction of paw edema of rats were observed at 2~6 h after treatment of methanol and hot-water extracts with 50 mg/kg concentration to the rats which are induced acute hind paw edema by carrageenan administration. The anti-acetylcholinesterase activity of the methanol extract of the mushroom showed 83.34% inhibition on AcHE which is lower than that of positive control galanthamine. The experimental results suggested that methanol and hot-water extracts of Phellinus xeranticus fruiting bodies might be used for good sources of antioxidant, anti-inflammatory and anti-acetylcholinesterase agents.
Quality standard of Pleurotus ostreatus in a market and changes of mushroom quality during storage

This study was carried out to improve standardization of agricultural products and investigate quality changes during storage at different treatments. The standardization does much to improve merchantable quality, distribution efficiency and fair dealings by shipping of the standard agricultural products. Therefore, modification of these standards is required to fit farmhouse situations. Average pileus diameter and thickness of A grades was 38.9 mm and 4.5 mm at shelf culture and 22.2 mm and 3.9 mm at bottle culture. Average stipe length of A grades was 49.6 mm at shelf culture and 66.7 mm at bottle culture, and stipes thick was the highest in A grades of shelf culture. The diameter of pileus of Pleurotus ostreatus stored for 14 days at
showed no significant difference. but the thickness of pileus decreased rapidly after 7 days. L-value and hardness of stipes and pileus were higher at shelf culture than bottle culture.

Recently, Pleurotus eryngii, King oyster mushroom is the main export item in South Korea mushroom industry. For appropriate transportation to Europe and USA markets, it is necessary to elucidate suitable conditions of storage and treatment of post-harvest during long time shipping. The cultivation of Poria cocos on tree-stumps and logs, was first described at the beginning of the 13th century. Poria cocos has been used for medicinal purposes to treat physical and mental recuperation, promote diuresis, forgetfulness and physical weakening. The artificial cultivation techniques of P. cocos was reported in China in 1957. are reported. The basic study on morphological characteristics and artificial cultivation method of Poria cocos with pine tree log buried under ground were initiated by Rural Development Administration. To widen the utilization of harvested sclerotia of Poria cocos as new food stuffs, powder of the sclerotia were used for producing new soft drinks and supplemented to wheat flour to make functional bread. Last 30 years, the supply of P. cocos in Korea through cultivation could not meet the domestic demand, Korea collected P. cocos in the mountainous areas and also import the sclerotia from China. Since the domestic demand of the sclerotia in China increased, the price of imported sclerotia of P. cocos also increased. Recently, high-quality and high-yielding cultivation methods were developed in Gyeongbuk Agricultural Research and Extension Services (GARES). The new cultivation methods of P. cocos developed by GARES was to change traditional cultivation method from under ground to above ground cultivation house, and shortened cultivation period by half, and improved the quality of the sclerotia. Therefore, it is suggested that advanced new technology for production of sclerotia of P. cocos from under ground and above ground must be developed for fullfil the domestic demand of the sclerotia.
