In vitro antioxidant, anti-hyperglycemic, anti-cholinesterase, and inhibition of nitric oxide production activities of methanol and hot water extracts of Russula rosacea mushroom

Russula rosacea, a mycorrhizal fungus, has been used for edible and medicinal purposes. This study was conducted to evaluate the in vitro antioxidant, anti-hyperglycemic, anti-cholinesterase, and nitric oxide inhibitory effects of the fruiting bodies from R. rosacea extracted with methanol, and hot water. The 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activities of the methanol and hot water extracts (2.0 mg/ml) of R. rosacea were comparable with BHT, the positive control. The chelating effects of the mushroom and hot water extracts were significantly higher than that of BHT. The reducing power of methanol and hot water extract (6 mg/ml) were significantly lower than that of BHT. Seven phenolic compounds were detected from acetonitrile and hydrochloric acid solvent extract of the mushroom. alpha-amylase and alpha-glucosidase inhibitory activities of methanol and hot water extracts were lower than that of acarbose, the positive control. The acetylcholinesterase and butyrylcholinesterase inhibitory effects were moderate compared with galanthamine, the standard drug. Nitric oxide (NO) production in lipopolysaccharide (LPS) induced RAW 264.7 cells were inhibited significantly by the mushroom extracts in a concentration dependent manner. Therefore, we demonstrated that fruiting bodies of R. rosacea possess in vitro antioxidant, anti-hyperglycemic, anti-cholinesterase, and NO production inhibitory activities. The experimental results suggest that the fruiting bodies of R. rosacea are good natural antioxidant, anti-hyperglycemic, anti-cholinesterase, and anti-inflammatory sources.
The difference of occurring pattern of Hericium erinaceus by pinheading induction methods

In recent years, Hericium erinaceus became famous for comsumer because it contains some materials to assist in human health. This study was carried out to find the difference of occurring pattern of H. erinaceus by pinheading induction methods. Harvest period of T1, T2 treatments were longer than T3~T6. All treatments fresh weights were not significantly different, but T3, T6 treatments's dry weight in 2nd flush were lower than others. T5 treatment's 2nd flush's regeneration rate was the highest than others and looks best quality. Therefore, it is suggested that pinheading induction method would be changed not T1, T2 treatment which were used some farm as Pluerotus ostreatus cultivation, but to scratch surface method which is T5 treatment for cultivation of high quality H. erinaceus.
Effect of spent mushroom substrates on Phythopthora Blight disease and growth promotion of pepper

Water extracts from spent mushroom substrate (SMSE)of edible mushrooms, Pleurotus eryngii, Hericium erinaceus and Lentinula edodes promoted growth of pepper seedling. Mycellial growth rate of Phythopthora capsici and Fusarium oxysporum was dramatically inhibited by 100% and 70% on PDA added with SMSE of H. erinaceus. SMSEs from H. erinaceus, P. eryngii, and L. edodes effectively reduced the disease severity of Phytophthora blight of pepper caused by Phytophthora capsici to 75%, 10% and 35%, respectively. These results suggested that SMSE from the mushrooms have dual effects that suppress phythopthora blight disease and promote plant growth of pepper.
Cultural characteristics of Pholiota adiposa according to substrates composition of sawdust medium by bottle cultivation

This study was carried out to develop optimum sawdust medium for bottle cultivation of Pholiota adiposa. Five kinds of sawdust media were tested. Period of primordial formation was shorter 1-5 days in T3 and in T4 than in other treatments. Total cultivation period was 3-5 days longer in treatments including oak sawdust than in T5, treatment of poplar+rice bran(8:2). Stipe diameter, stipe length, and number of valid stipe were the biggest in T3. Yield of fruiting body was increased 33% in T3 and 12% in T4, respectively compared to T1 as control. Therefore, T3, treatment of oak sawdust+beet pulf+cottonseed meal(5:3:2), would be appropriate for the commercial production of Pholiota adiposa by bottle cultivation.

Since button mushroom (Agaricus bisporus) cultivation is performed in closed environment, the understanding of indoor environment becomes essential for the quality and quantitative production of the greenhouse-grown mushroom. To generate information on indoor environmental factors affecting on fruiting body quality, we investigated temperature, humidity, and bacterial concentration and species in a greenhouse located in Buyeo, Chungnam Province. Fibrinolytic, thrombin inhibitory, anti-oxidative and anti-inflammatory activities of Pleurotus ferulea

Our study investigated the fibrinolytic, thrombin inhibitory, anti-oxidative and anti-inflammatory activities of the water extract and solvent fractions isolated from Pleurotus ferulea. Fibrinolytic activity was investigated using the fibrin plate method. Thrombin inhibitory activity was used to analyze thrombin inhibitor assay. The DPPH assay was used to estimate anti-oxidative activity. Inhibition of NO production was measured for anti-inflammatory activity in LPS-activated murine RAW 264.7 macrophage cells. An MTS assay was used to evaluate the effects of the water extract and solvent fractions isolated from Pleurotus ferulea on cell viability. Our results showed the fibrinolytic activity to be strong in the ethyl acetate fraction at 1.33 plasmin units. The ethyl acetate fraction also showed high thrombin inhibitory activity at 94.45%. The anti-oxidative activity of the water extract was 37.01% and the anti-inflammatory activity of the chloroform fraction was 98.13%. These findings suggest that Pleurotus ferulea's extract and fractions could be applicable in the development of functional foods for the treatment and prevention of cardiovascular diseases.
Genetic relationship analysis of Pleurotus pulmonarius strains using RAPD

Genetic relationship analysis by PCR reactions with 25 primers could produce relevant classifications of the 21 P. pulmonarius (Fr) Quel. isolates collected from 2005. They were classified by two groups with similarity coefficient 0.37. All of the P. pulmonarius cultivars belonged to group 2. New cultivar 'Hwasan' presented high similarity coefficient to parental strains, and that of 'Hosan' and 'Kangsan' cultivars was very high owing to shared crossing parents, and that of two strains from China was high, as well.
Selection of parental strain on the sawdust cultivation and mycelial growth and cultural characteristics of Lentinula edodes hybrid strains

In order to breed new strains of oak mushroom, Lentinula edodes, on the sawdust cultivation, we collected 10 from Korea, Taiwan and China respectively and examined somatic incompatibility, morphological features of fruiting body and productivity. Four strains(SANJO701HO, FMRI2534, FMRI2337 and FMRI2613) shown remarkable results, were confirmed with parental strains. 80 monokaryotic strains derived from the selected 4 parental strains, were selected and 117 hybrid strains were made by mono-mono mating. Aslo, Physiological characteristics were investigated. Average of the mycelial growth of hybrid strains of mating combination SANJO701HO-FMRI2337 was approximately 10% lower than other mating combinations. Overall, This study was founded to develop new varieties of L. edodes. The productivity of new 117 strains on sawdust cultivation needs continued research.

Glucan has been shown to have a significant role in the activation of the immune system, including increased activity of macrophages and so on. Sparassis latifolia (formerly S. crispa) is an edible mushroom abundant in dietary fiber and widely known to contain high levels of
-glucan. In the present study, fermentation broths containing soluble
-glucan were prepared by fermentation with mushrooms with four Lactobacillus species (L. plantarum subsp. Plantarum, L. acidophilus, L. helveticus, and L. delbrueckii subsp. Bulgaricus). Biodiesel production using lipase producing bacteria isolated from button mushroom bed

A lipase producing bacterium was isolated from button mushroom bed, which showing high clear zone on agar media containing Tributyrin as the substrate. The strain was identified as Burkholderia cepacia by analysis of 16S rDNA gene sequence. Crude lipase (CL) was partially purified from 70% ammonium sulfate precipitation using the culture filtrate of B. cepacia. Immobilized lipases were prepared by cross-linking method with CL from B. cepacia and Novozyme lipase (NL) onto silanized Silica-gel as support. Residual activitiy of the immobilized CL (ICL) and immobilized NL (INL) was maintained upto 61% and 72%, respectively. Biodiesel (Fatty acid methyl ester, FAME) was recovered by transesterification and methanolysis of Canola oil using NaOH, CL and ICL as the catalysts to compare the composition of fatty acids and the yield of FAME. Total FAME content was NaOH
, in which the highest levels of FAME was observed to 50% oleic acid (C18:1) and 22% stearic acid (C18:0). In addition, the unsaturated FAME (C18:1, C18:2) decreased, while saturated FAME (C16:0, C18:0) increased according to increasing the reaction times with both CL and ICL, supporting CL possess both transesterification and interesterification activity. When reusability of ICL and INL was estimated by using the continuous reaction of 4 cycles, the activity of ICL and INL was respectively maintained 66% and 79% until the fourth reaction.
Biodegradation of triphenyl methane dyes by white rot fungus, Trametes versicolor

White rot fungi produce lignin-degrading enzymes such as laccase, manganese peroxidase and lignin peroxidase. These extracellular oxidases efficiently degrade recalcitrant synthetic dyestuffs with diverse chemical structures. Here, we examined the activities of lignin-degrading enzymes in Trametes versicolor using triphenyl methane dyes, crystal violet (CV) and malachite green (MG). Both dyes were decolorized by T. versicolor in solid and liquid culture conditions. T. versicolor decolorized MG more quickly than CV in both conditions. Among three ligninolytic enzymes, laccase was most abundantly found in the decolorization processes of CV and MG. However, higher activity of laccase was needed to degrade CV than MG. The much less activity of MnP was also detected. But the increase of MnP activity was well corresponded to the decolorization efficiency of CV, suggesting the involvement of MnP in CV degrading process. However, its role in the degradation process of MG is supposed to be subsidiary to laccase.
Physiology characteristics of genus Hypocrea isolated at oyster mushrooms of the shelf cultivation

Hypocrea disease is the most severe disease of oyster mushroom cultivation in Korea. Physiological and ecological studies were performed on the pathogens (Hypocrea spp.) to obtain basic information for developing the integrated disease management system. Fourteen isolates of Hypocrea were collected from oyster mushroom house in five areas. Pathogenic fungi causing disease of oyster mushroom were identified as Hypocrea sp. based on morphological characteristics and pathogenicity. Two isolates (H-1, H-12) showed the fastest growth at
but four isolates (H-8, H-9, H-13, H-14) showed slower growth than those of other isolates at
. Stroma with ascocarps and ascospore were produced on PDA under fluorescent light. The five isolates produced stroma with ascocarps and ascospores. Formation of fruiting body of strains H-14 of Hypocrea were the best out of all the strains on the potato dextrose agar (PDA). Also, fruiting bodies and ascospores were completely produced under fluorescent light. The growth of the isolates was correlated with total carbon content. The stroma of the isolates was formed mainly in histidine and asparagine treatment and especially in histidine-70 and asparagine-100 treatment. In the test of pathogenicity, after and before spawning showed very fast incidence of disease.
Characteristics of new mid-high temperature adaptable oyster mushroom variety 『Heuktari』 for bottle culture

The 'Heuktari', a new mid-high temperature adaptable variety of oyster mushroom for the bottle culture, was bred by mating with monokaryons isolated from 'P11056' and 'MT07156'. The optimum temperature for the mycelial growth was
on PDA medium and that for the primordia formation and the growth of fruiting body of 'Heuktari' was
on sawdust substrate. In case of bottle cultivation, the period of mycelial growth was required about 30 days. In addition, the period of primordia formation and growth of fruiting body was 4 days and 5 days, respectively. In the characteristics of fruiting body, shape and color of pilei were round type and dark grayish brown, stipe color was white color and stipe shape was short and thick. The yield of fruiting bodies was 180 g/900 ml bottle which was 15% higher than that of Suhan-1ho. The gumminess and brittleness of stipe tissue were 110% and 140% stronger than those of Suhan-1ho, respectively.
Development of strain-specific SCAR marker for selection of Pleurotus eryngii strains with higher β-glucan

In this study, SCAR marker that differentiates Pleurotus eryngii strains with higher
-glucan from control strain was developed. Genomic DNAs of 9 control strains of Pleurotus eryngii and 9 Pleurotus eryngii strains with higher
-glucan were analyzed by bulked segregant analysis (BSA) using randomly amplified polymorphic DNA (RAPD). One-hundred twenty RAPD primers were screened on bulked DNA samples and a unique DNA fragment with the size of 91 bp was yielded by OP-R03 primer from the Pleurotus eryngii strains with higher
-glucan. A sequence characterized amplified region (SCAR) marker, designated as OP-R03-1-F and OP-R03-1-R, was designed on the basis of the determined sequence. The PCR analysis with the OP-R03-1 primer showed that this SCAR marker can clearly distinguish the Pleurotus eryngii strains with higher
-glucan from the control strains.
