Bioethanol Production from Rice straw by Irpex consors

This study was initiated to evaluate ethanol production by a Korean isolate of white rot fungus Irpex consors. It was found that the fungus could produce ethanol by converting glucose, mannose, xylose, and cellobiose under semi-aerobic condition with yields of 0.23, 0.19, 0.21, and 0.17 g ethanol per g sugars, respectively. Furthermore, the strain produced ethanol by simultaneous saccharification and fermentation of rice straw treated with steam pressured boiling water, 3% NaOH, and 3%
with maximum yields of 0.12, 0.15, and 0.19 g ethanol per g rice straw, respectively. These results suggested that I. consors could produce ethanol from the components of cellulose and hemicellulose including glucose, mannose, xylose, cellobiose as well as rice straw treated with steam pressured boiling water, dilute sodium hydroxide, and dilute sulfuric acid. This is the first report that I. consors mycelia produce ethanol from various sugars and lignocellulosic substance including rice straw.
Characteristics and breeding of a new brown variety `Geumhyang` with short cultivation period in Flammulina velutipes

`Geumhyang`, a new variety with short cultivation period and yellowish ivory color in Flammulina velutipes, was bred by crossing between monokaryons isolated from `Garlmoe` and wild strain `CBMFV-33`. In bottle cultivation, the period of mycelial growth, primordia1 formation, and growth of fruit body was required 23 days, 7 days, and 17 days, respectively. Total culvivation period was 47 days, showing 7 days shorter than that of `Garlmoe`. Pileus diameter was greater than that of `Garlmoe`, and pileus color showed yellowish ivory. Fruit body production per 850 ml bottle was 160 g, which was 8% higher than that of `Garlmoe`. The taste, derived from `Garlmoe` and wild strain, was excellent because of better chewing feeling than white ones. `Geumhyang` with differentiated color and taste, has an advantage to target a niche market of white ones.
Optimum cultivation conditions for mass production of antagonistic bacterium Pseudomonas azotoformans HC5 effective in antagonistic of brown blotch disease caused by Pseudomonas tolaasii

This study was conducted to investigate optimum conditions for mass production of antagonistic microbes Pseudomonas azotoformans HC5. P. azotoformans HC5 is a potent biological control agent to control brown blotch disease caused by Pseudomonas tolaasii. This markedly showed the antagonistic activity against P. tolaasii, the most destructive pathogen of cultivated mushrooms. To define the optimum conditions for the mass production of the P. azotoformans HC5, we have investigated optimum culture conditions and effects of various nutrient source on the bacterial growth. The optimum initial pH and temperature were determined as pH 6.0 and
, respectively. The optimal concentration of medium elements for the growth of pathogen inhibitor bacterium was determined as follows: 0.6% adonitol, 1.5% yeast extract, 0.8%
, and 0.2% asparagine.

`Heukhyang`, a new variety with dark brown color, short cultivaltion period, and good taste in Flammulina velutipes, was bred by crossing between monokaryons isolated from `Garlmoe` and wild strain `CBMFV-10`. In bottle cultivation, the period of mycelial growth, primordia1 formation, and growth of fruit body was required 23 days, 6 days, and 17 days, respectively. Total culvivation period was 46 days, showing 8 days shorter than that of `Garlmoe`. Pileus diameter and thickness were greater than those of `Garlmoe`, and pileus color showed dark brown. Fruit body production per 850 ml bottle was 135 g, which was 10% lower than that of `Garlmoe`. However, the overall acceptance was most excellent in sensory evaluation, and `Heukhyang` has an advantage to target a niche market of white ones due to differentiated color and taste.
Effect of dietary supplementation of fermented spent mushroom substrates from Pleurotus eryngii on Hanwoo Steers

This study was carried out to investigate the feeding effects of dietary supplementation of fermented spent mushroom substrates (F-SMS) from Pleurotus eryngii with Bacillus subtilis CS21 and Saccharomyces cerevisiae on Hanwoo steers. The cellulase and xylanase producing bacteria, designated CS21, was isolated from freshly spent mushroom substrates from Pleurotus eryngii and used as probiotics to fermented spent mushroom substrates. Twenty Hanwoo steers were allocated into two feeding groups and assigned equally to two dietary treatments; Control (TMR) and TMR including 30% F-SMS (30% F-SMS TMR). Total gain and feed intake was significantly greater in the 30% F-SMS TMR than control (p<0.05), but carcass grades were not influenced by the experimental diets. Based on this study, fermented spent mushroom substrates from Pleurotus eryngii with B. subtilis CS21 and Saccharomyces cerevisiae is able to use as an ingredient feed in TMR for Hanwoo steers.
Growth characters and harvest time for the artificial cultivation of Mycoleptodonoides aitchisonii

Mycoleptodonoides aitchisonii has been used a culinary material and traditional medicine for a long time in worldwide and recently the researches to find biological effects have been increased such as dopamine activation, preventive effect against phytopathogens, inhibitive effect against erythrocyte coagulation, anti-oxidative effect, anti-tumorigenic effect, etc. However it is hard to cultivate Mycoleptodonoides aitchisonii it is impossible to be mass-produced and in order to solve the problem in this study we found the appropriate cultivation period and the harvest point for it. For life cycle (from primordium formation to harvest) the morphology, weight, and quantity of polysaccharide of Mycoleptodonoides aitchisonii were measured using with 5 bodies per a day and it could be divided for 4 stages; primordium formation, growth, needle maturation, and aging. And then from the results the Zeide nonlinear growth curve could be gotten. At 13th day after cultivation there is the relation between the change of media weight and body weight and at 14th day after cultivation the rate of polysaccharide in the body was 11 %. However in the case of O2 insufficient supply the malformation of them was observed.
Organic acid and quality change in Flammulina velutipes fruit body by various storage temperature treatments and packaging films application

This study was carried out to characterize the chemical components of Flammulina velutipes during storage by different packaging film application for developing export winter mushroom to south east asia area. Changes of weight and growth rate from Flammulina velutipes were the lowest in testing packaging film at

. The changes of colors was measured by Hunter`s color value, L value and yellowness (+b) were slightly decreased during storage. But the L value and yellowness (+b) were not affected by packaging films. Six organic acids were detected such as oxalic acid, lactic acid, malic acid, citric acid, acetic acid and succinic acid. The content of total organic acids were increased by storage period.
Growth properties of Neolentinus lepideus according to the light environment

We found about cultivation light environment conditions of Neolentinus lepideus. For the high quality of N. lepideus, lights-out was useful. In the lights-out, the diameter of pileus of N. lepideus was smaller, and the length and the thickness of stipes were longer and larger than in that of lighting. At the LED effect, the orange and red LEDs prolonged the cultivation period in N. lepideus. The yields of N. lepideus under the white, blue and green LEDs were higher, and the number of available stipes of N. lepideus under the white and blue LEDs was higher than that of other LEDs. The diameter of pileus of N. lepideus under the white, blue and green LEDs was higher, and the length and the thickness of stipes under the orange and red LEDs was higher than those of other LEDs.
Characteristics and breeding of a new oyster mushroom (Pleurotus ostreatus) variety `Chunhwashim` by using cytoplasmic hybrid

Hybrid strains was selected by crossing between dikaryotic strain ASI 0628 (Dagul) bred from ASI 2596 (Suhan No.3) and ASI 2782(Black pileus mutant) and monokaryotic strain ASI 2344-84. The strain 84 that shown the best cultural characteristics was selected to be a new cytoplasmic hybrid variety and named as `Chunhwashim`. The `Chunhwashim` shown incompatibility among them forming the confrontation growth line against parental strains ASI 0628 (Dagul) and ASI 2344 (chunchu 2ho). Fruiting body produced about
per bottle. And also the individual generation of fruit body is multiple than chunchu2ho as 40.9. The `Chunhwashim` was cytoplasmic hybrid included hybrid DNA bands of parental strains ASI 0628 (Dagul) and ASI 2344 (chunchu 2ho) by Random Amplified Polymorphic DNA (RAPD) primer, and mitochondria DNA band of monokaryotic ASI 2344 (chunchu 2ho) using mitochondria microsatellite DNA marker. This new cytoplasmic hybrid variety `Chunhwashim` of oyster mushroom is characterized by multiple of individual generation and deeply grey color of pileus.
Breeding of a new oyster mushroom cultivar `Daejang 3ho`

`Daejang 3ho` is a new cultivar of oyster mushroom for high productivity than that of Chunchu 2ho. This cultivar was bred by Di-mon mating between monokaryotic strains isolated from `Daejang 1ho` and dikaryotic strain `Jangan 5ho`. The color of pileus and the shape of pileus of new cultivar were dark gray and deeply funnel shape, respectively. The length and thickness of stipe were longer and thicker than those of Chunchu 2ho. It took 4~5 days to formation of primordia, that was a similar to Chunchu 2ho. The optical temperature of fruit body was
. Yield of `Daejang 3ho` was 13.9% higher than that of Chunchu 2ho. Analysis of RAPD by URP-primer of #03, #08, #10 and #11 showed that different band pattern those of other strains. There were no significant differences between the texture properties of `Daejang 3ho` and Chunchu 2ho.
The formula and the utilizing method for adjusting moisture contents of the substrates on the mushroom bottle cultivation

This study was done the calculation formula for calculating the below item. That is the moisture content of the substrates raw materials, water content of the mixed substrates; the amount of media, dry weight, amount of liquid, bulk density in the bottle; dried materials volume, water volume, mixing ratio in a substrates mixing materials; water requirement amount, replenishment amount for water content control of the substrates and the total amount of substrates. And each was prepared in an Excel file for automatically calculated. The calculation of the Excel file will be able to take advantage for the mushroom growers; to make raw material costs, per bottle nutrient content and moisture content maintained throughout the year and new media composition. At this time we expect, while minimizing the quantity or deterioration in the daily ups and downs continued growing courses throughout the year contribute to stabilizing production.
Damages and Developmental Characteristics of Fungus Gnat, Lycoriella ingenua (Diptera: Sciaridae) in Button Mushroom Cultivation

Mushroom sciarid fly Lycoriella ingenua (Dufour), syn. L. mali (Fitch) is one of the most common fly pests affecting the mushroom cultivation in Korea. This study was carried out to investigate the development at different temperatures and damages of L. ingenua in A. bisporus mushroom cultivation. Rearing of mushroom flies were carried out on mycelial culture in Petri dishes. The development of L. ingenua from egg to adult at the temperature of 16, 20, 24,
were 35.2, 25.8, 23.5, and 22.2 days, respectively. Adult flies invade mushroom farms and oviposit in freshly spawned compost. Damages are mainly caused by the larvae of 1-7 mm, which feed on growing mycelium and also developing stipes and caps of mushrooms. Adult flies spoil the appearance of the fruiting bodies, spread various fungal and bacterial disease, and transmit mites. The damages by mushroom flies increased without pest control causing yield loss of up to 27%.
