Hepatoprotective Effect of Forest frog's oviduct oil on Acetaminophen-induced Liver Injury in Mice.

Objectives : The purpose of this study is that the protective effects of habmayou on acetaminophen (AP)-induced hepatotoxicity were investigated in mice. Methods : Before administering AP mice supplied with only water were left alone for 18 hours. after concentration and dissolution in poly ethylglycol AP 400mg per 1kg of mouse weight, we injected AP titrated density with a physiological saline solution into the abdominal cavity of mouse to induce hepatotoxicity. we researched mortality rate and the shape of liver tissue of mouse. Results : Treatment with habmayou (250 mg/kg, p.o.) 0.5 h after AP administration significantly prevented an increase in plasma alanine aminotransferase and aspartate aminotransferase activities and AP-induced hepatic necrosis, and also reduced AP-induced mortality from 46% to 0%. In addition, oral treatment with habmayou significantly prevented AP-induced depletion of glutathione (GSH) contents. However, habmayou treatment, by itself, did not affect hepatic GSH contents. Conclusion : These results show that the hepatoprotective effects of habmayou against AP overdose may be due to its ability to block the bioactivation of AP by regeneration of hepatonecrotic cells.
Effects of Hwangryunhaedok-tang on DNA Damage, Antioxidant Enzymes Expression and Acetylcholinesterase Activity

Objectives : In Alzheimer's disease(AD), free radical oxidative stress caused by amyloid beta-peptide may lead to DNA damage, neuronal dysfunction, neurotoxicity and cell death, Hwangryunhaedok-tang(HHT) is traditionally used for the treatment of pyrogenetic diseases. To develop a new anti-AD drug from natural herb, HHT was selected and extracted in this study. Methods : The antioxidant activities of HHT water extract powder were examined by hydroxyl radical-induced DNA strand nicking assay, and antioxidative enzymes expression assay in H4IIE cell. In addition, HHT was examined for the inhibitory effect on the acetylcholinesterase(AChE) using by Ellman's coupled assay. Results: The HHT exhibit DNA protective effect in the hydroxyl radical-induced DNA Strand nicking assay, mRNA expression of superoxide dismutase and glutathione peroxidase were recovered at a normal level by HHT treatment in H4IIE cell. Furthermore, water extract of HHT showed inhibitory effect on AChE activity. Conclusion : These results suggest that HHT may be effective in delaying and preventing AD progression related to the free radical-induced DNA damage and AChE activity.
A Herbological study on the plants of Selaginellales, Equisetales, Psilotales in Korea.

Objectives : The plans such as Selaginellales, Equisetales and Psilotale have been used for treatment of diseases. Therefore, we examined their effects and distributions. Methods : We examined herbalogical books and research papers which published at home and abroad. Results : 1. There were totaled to 5 genera and 33 species in Selaginellales. Equisetales, Psilotales in Korea and among them medicinal plants are 4 genera, 21 species, about 64% in total. 2. Lycopodium genus is main kind enough that it has 13 species among 33species in the Selaginellales, Equisetales, Psilotales, of which medicinal plants are 10 species. 3. The herb is the main medicinal parts of medicinal plants in the Selaginellales, Equisetales, Psilotales which is used in 21 species. 4. On the bases of nature and flavor of medicinal plants in the Selaginellales, Equisetales. Psilotales, they were classified into neutral 15 species; bitter taste 15 in the order. 5. On the bases of meridian propism of medicinal plants in the Selaginellales, Equisetales. Psilotales, they were classified into liver meridian 11 species. 6. On the bases of the properties and principal curative action, they were classified into drugs for styptic 15 species, drugs for expel wind 14, drugs for drain damp 13 in the order. 7. It has been shown that one species in the Selaginellales, Equisetales, Psilotales is toxic. Conclusion : There were totaled to 5 genera and 33species in Farinales in Korea and among them medicinal plants are 4 genera, 21 species, About 63% in total.
A Herbological study on the plants of Farinales in Korea.

Objectives : The plans can be used for medicinal purposes, among Farinales in korea and examined their effects and distributions. Methods : The examined herbalogical books and research papers which published at home and abroad. Results: 1. There were totaled to 8 genera and 24 species in Farinales Korea and among them medicinal plants arc 8 genera, 11 species, some 46% in total. 2. Eriocaulon is main kind enough that it has 11 species among total 24 species in the Farinales, of which medicinal plants are Commelina and Monochoria that they have each 2 species 3. The herb is the main medicinal parts of medicinal plants in the Farinales which is used in 9 species. 4. According to nature and flavor of medicinal plants in the Farinales, they were classified into cool 9 species; sweet taste 9 in the order. 5. According to meridian propism of medicinal plants in the Farinales, they were classified into liver meridian 6 species. 6. According to the properties and principal curative action. they were classified into drugs for detoxicant 12, drugs for antifebrile 11 species, drugs for drain damp 8 in the order. 7. It was noticed that 1 medicinal plant in the Farinale is toxic. Conclusion: There were totaled to 8 genera and 24 species in Farinales in Korea and among them medicinal plants are 8 genera, 11 species, some 46% in total.

Conclusion: These results suggest that the Ginseng Radix plus Crataegi Fructus extract may be effective for the prevention and treatment of Alzheimer's disease. Investigation into the clinical use of the Gin-CHF extract for Alzheimer's disease is suggested for future research.

A study of 'Asari Herba should not exceed Jeon(錢, Qian, 3.0 gram)'

Objectives: Asari Herba comes from the dried perennial herbaceous plant, Asarum sieboldii
., A. heterotropides
. SCHMIDT var. mandshuricum(
. and A sieboldii
., family Aristolochiaceae. This medicine is pungent in flavor(味辛), warm in nature(性溫), mildly toxic and affects the lung, kidney, and heart meridians. It is able to repel wind dispel cold(祛風散寒), remove the obstruction of the nose(宣通鼻竅), relieve pain(止痛) and warm the lungs to remove the retention of fluids(溫肺化飮). In a clinic it is prescribed for exterior syndrome of wine-cold type(風寒表證) manifested as headache and general pain. It is indicated for headaches, toothaches, bisyndrome of wind-cold-dampness type(風寒濕痺), etc. For many generations medical books have warned doctors that the dosage of Asari Herba should not be excessive because the pungent flavor has the effect of dispersing exopathogens from superficies of the body. The principle, 'Asari Herba should not exceed Jeon(錢, Qian, 3.0 gram)' is still considered to be relevant. Methods : Extensive research of several ancient medical texts has lead to the following conclusion. Results : The statement 'Asari Herba should not exceed Jeon' is a dosage which is used to prepare single recipe and powder. Conclusion : The definition of Jeon is not 1 Jeon(3.0 gram), but 1 Jeonbi(錢匕, Qianbi, 1.0 gram), moreover, the usage, collection. and refinement process are not in accordance with the contents of the ancient medical texts. A deeper study of Asari Herba's dosage in the future is expected to have interesting results.
Scavenging Activity of Reactive Oxygen Species and Inhibitory Effect of Cytochrome P450 from Circium japonicum Extract

Objectives: Our previous studies have clearly demonstrated that the scavenging activity of reactive oxygen species (ROS), protective effect of lipid peroxidation (LPO), and inhibition of cytochrome P450 isozymes (CYPs) from the Circium japonicum aqua-acupuncture solution (CJAS). But, Circium japonicum water extracted solution (CJWS) was weakly reported in cardiovascular diseases such as oxidative stress-mediated atherosclerosis or its value evaluated. Methods: CJWS was assessed to determine the mechanism of its scavenging activity of ROS and inhibitory effect of CYP 2E1. Results: CJWS exhibited a concentration-dependent scavenger of DPPH and superoxide anions radicals using different assay systems. In addition, CJWS showed dose-dependent free radical scavenging activity, including hydroxyl radicals, peroxynitrite, and nitric oxide. The CJWS was also found to be effective in protecting rat liver homogenate against LPO. Futhermore, the CJWS showed significant inhibition of CYP 2E1 induced by pyrazol in a rat liver microsome. Conclusion : ROS and CYPs may play a role in several diseases, such as cardiovascular disease and heart failure. Our study demonstrated that the CJWS has excellent scavenging activity of ROS. Hence, it is worthwhile to investigate the potential effectiveness of CJWS in preventing oxidative stress-mediated cardiovascular diseases.
Experimental Effects of Acanthopanax sessiliflorus
Extracts Following Gamma-ray Irradiation on Immuno-stimulating and anti-tumor activity in mice

Objectives : This experimental study was designed to investigate the effects of Acanthopanax, sessiliflorus SEEM extracts following gamma-ray irradiation on immuno-stimulating and anti-tumor activity in terms of proliferation of tumor cells, thymocytes, splenocytes, and NO production from peritoneal macrophages in mice. Methods: 10AS and 100 AS were the bark powders of Acanthopanax sessiliflorus
stems which were exposed in 10 kGy or 100 kGy of electron beam respectively. Results : Treatment with either 10AS or 100AS increased proliferation rates of thymocytes and splenocytes significantly, and treatment with l0AS also decreased proliferation rates of tumor cells significantly. Treatment with either l0AS or l00AS promoted NO production from peritoneal macrophages significantly. Conclusion : These results suggested that AS has direct inhibition effect of tumor growth and immuno-stimulating activity. In conclusion, we demonstrate that AS could be used to treat cancer patient as complementary or alternative medicine to typical anti-cancer medication.
A study on external and internal morphology in 4 kinds of Uie-Suel Radix

Objectives : This study was designed to establish a characteristic discrimination of internal and external morphological standard of original plants and herbal states in Polygoni Multiflori and Cynanchi Wilfordii Radix. Methods : In this studies, the external-internal morphological standards were determined by using stereoscope and butanol series. Results: 1. The external characteristics of original plants: Polygonum Multiflorum has alternate leaves, and its flower with white color blooms at the shoot apex or leaf axil. In the other hand, Cynanchum wilfordii has opposite leaves, and its flower with yellowish green color blooms at the leaf axil. 2. The physical characteristics of herbal slates: Polygoni Multiflori Radix is red-brown in outer surface pink-brown in section. In the other hand, Cynanchi Wilfordii Radix is earthly-brown in outer surface greyish white in section. 3. The physical characteristics according to the place of production: Demonstrable difference according to the place of production is not seen. Polygoni Multiflori Radix is brown or dark brown in outer surface, soft
brown in section. In the other hand, Cynanchi Wilfordii Radix is white in outer surface,
red in section, and has power type. 4. The internal characteristics: Polygonum multiflorum has hetero-vascular bundle and lignification of cork layer is progressive. In the other hand, Cynanchum wilfordii has not hetero-vascular bundle, and lignification of cork layer is weak. Conclusion: In the future, many fundamental study such as how to discriminate between Polygoni Multiflori Radix and Cynanchi Wilfordii Radix in origin and efficacy will be necessary. Also the standard of discrimination must be specific and distinct in that several kinds of fo-ti has been traded. It is considered the results of this study will be furnish κ I the basis Lo succeeding studies and it is needed to extensive comparative study for the same genus-degree of relatedness.
Effect of Angelicae Dahuricae Radix on Expression of Cytokines and Chemokines Levels in Human Mast Cells (HMC)

Objectives: Angelicae Dahuricae Radix (Baek-Ji in Korean, BJ) is well known to be used as a medicine for cold, headache, supraorbital pain, nasal congestion, and toothache. Little is understood about the roles of BJ in the cytokine and chemokine secretion by immune cells. This study was designed to find out the effects of BJ on the cytokine and chemokine secretion in human mast cells (HMC). Methods : We treated BJ according to consistency on HMC and measured cytokines and chemokines levels using flow cytometry CBA system. Results: In BJ treated group. the expression of interferon-inducible protein 10 (IP-l0), monocyte chemoattractant protein-1 (MCP-1), chemokine (C-X-C motif) ligand 9 (MIG), and interleukin 10 (IL-l0) levels were decreased significantly and chemokine (C-C motif) ligand 5 (RANTES), IL-8,
), and tumor necrosis factor alpha (TNF-a) were decreased significantly. Conclusion : The results of this experiment supposed that the treatment of BJ will ameliorate the secreting levels of some chemokines or cytokines such as IP-10, MCP-1, MIG, IL-10, RANTES, IL-8,
, and TNF-a.
Ethanol extract of Bojungbangam-tang (EBJT) prevents Cisplatin-induced Apoptosis in mice.

Objectives : The purpose of this study is to investigate cisplatin-induced apoptosis by ethanol extract of bojungbangam-tang (EBJT). Methods : To evaluate of anti-apoptic effects of EBJT, we examined several kinds of cell populations such as
T cells and macrophages in spleen. Result : 1. EBJT inhibited cisplatin-induced cell death in spleen. 2. EBJT inhibited the death of
T cells. 3. EBJT slightly recovered the number of macrophages by cisplatin. Furthermore, cisplatin-induced upregulation of class II were inhibited by EBJT. Conclusion : EBJT prevented cisplatin-induced cell death, which could provide a clinical basic for side effects of anti-tumor therapeutics.
The Effects of Danggwisaeyeoktang(當歸四逆湯) on the Hypothyroidism of Rat

Objectives : The aim was to study experimentally the effects of Danggwisaeyeoktang(當歸四逆湯) on the hypothyroidism of rats induced by thiouracil. Methods : Spargue-Dawley rat were separated into three groups. Except normal group, the other two groups were treated with thiouracil 50mg/kg/days for 30 days by oral administration. Among the two group, one group was treated DGSYT extract 0.54g/kg/days for 20 days separately. T3-uptake;, T3, T4, TSH, hematological values, enzyme activity, total cholesterol and Na+ content charge in the Serum of rats were measured after medication of solid extract of DGSYT. Result : As a result of this study, in the serum T3-Uptake and T4 level, hematological values and sodium were increased in DGSYT group compared to controls. The serum TSH, total cholesterol and body weight were decreased in DGSYT group compared to controls. Conclusion : According to the above results, it is suggested that DGSYT is effective to cure hypothyroidism.
Preventive effects of water extracts from Artemisiae Iwayomogi Herba on hyperlipiderma and liver damage induced by alcohol

Objectives : This study was performed to prove the preventive effect of water extracts from Artemisiae Iwayomogi Herba on hyperlipiderma and liver damage induced alcohol. Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And Artemisiae Iwayomogi Herba extract was administrated for the same period. We measured the serum component in rat's blood. body weight and weight of liver. Result : At first, we observed preventive effects of Artemisiae Iwayomogi Herba on hyperlipiderma induced by alcohol. Artemisiae Iwayomogi Herba group showed significant decrease of total cholesterol and triglyceride in comparison with those of the control group. Artemisiae Iwayomogi Herba group showed significant increase of body weight in comparison with those of the control group in 4weeks and 8weeks. At second, we observed preventive effects of Artemisiae Iwayomogi Herba on liver damage induced by alcohol. Artemisiae Iwayomogi Herba group showed significant decrease of GOT, ALP and LDH in comparison with those of the control group. Artemisiae Iwayomogi Herba group showed significant increase of liver weight in comparison with those of the control group. Conclusion : Reviewing these experimental results, it appears that water extracts from Artemisiae Iwayomogi Herba have pharmaceutical preventive efficacy on hyperlipiderma and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of Artemisiae Iwayomogi Herba.
Anti-inflammatory effect of extract of Pulsatilla koreana
in LPS-stimulated Murine peritoneal macrophage

Objectives : The purpose of this study was to investigate the anti-inflammatory effects of extract from Pulsatilla koreana
(PK) on the peritoneal macrophage. Methods : To evaluate of anti-inflammatory of PK, we examined cytokines and NO production in lipopolysacchride (LPS)-induced macrophages. Furthermore, we examined molecular mechanism using western blot. Results : 1.Extract from PK reduced LPS-induced NO, tumor necrosis factor-a (
), interleukin (IL)-6 and IL-12 production in peritoneal macrophages. 2.Extract from PK itself does not have any cytotoxic effect. PK inhibited the activation of extracelluar signal-regulated kinase(ERK 1/2) but not another mitogen-activated protein kinases (MAPKs) such as p38, c-Jun NH2-terminal kinase (JNK) and the degradation of inhibitory kappa B a (
) does not any effect in the LPS-stimulated peritoneal macrophages. Conclusion : PK down-regulated LPS-induced NO and cytokines production, which may be provide a clinical basis for anti-inflammatory properties of PK.
