Establishment of Hanwoo(Korean Cattle, Bos taurus, Linn.) Traceability System Using Radio Frequency Identification(RF-ID)

This study was conducted to develop a traceability system for systematic animal identifi- cation to increase efficiency of animal production, post production and processing to ensure that quality and sanitary meat products reach the consumers of the entire country. The new animal identification traceability system was designed for easy management, data collection and storage considering bar code, registration number and farm number.Specifically the system aimed 1) To provide standard radio frequency technology identification (RF-ID) for livestock management, standard of ear tag, microchip and ear tag identification equipment, 2) To guarantee meat product label thereby safeguarding the consumers 3) To give incentives to farmers for producing quality meat products and 4) To provide unified system for national livestock management.Results of the research which started in July 2003 which was used by several farmers’ cooperatives and Department Store revealed the feasibility of using the RF-ID system although much will be done to conform to global nomenclatures.
Practical Use of DNA Polymorphisms in the Avian Immunoglobulin Light Chain Constant Domain for Species-specific PCR

Species-specific polymorphisms in chicken, pheasant, turkey, and quail were identified by cloning and sequencing of the immunoglobulin constant domain (IgLC). A set of species-specific primers were then designed on the basis of polymorphisms in the IgLC between species, as well as two additional sets of primers for the cytochrome b and tapasin genes, for the purpose of species identification. Together, the primers successfully distinguished specific species from chicken by species-specific PCR. This simple but unambiguous method may be used to screen avian inter-species germline chimeras, which are valuable models for the conservation of endangered species.
Estimation of Genetic Parameters via Gibbs Sampler using Animal Model for Economic Traits in Pigs

Heritability and genetic correlation for growth traits in Duroc pig breed were estimated using Bayesian method via Gibbs sampling. The data set consisted of 3,526 performance records at National Institute of Animal Science. For estimating those parameters using Gibbs sampling, 5,000 cycles of ‘burn-in’ period were discarded among a total of 55,000 samples. Out of the remaining 50,000 samples, 5,000 estimates by each parameter were retained and used for analyses to avoid any correlation among adjacent samples. The growth traits considered in this study were average daily gain at 30kg(ADG1), average daily gain at 90kg(ADG2), backfat thickness(BF), days to 90kg(D90) and feed conversion ratio(FC). The estimated heritabilities and their standard deviation using Gibbs sampler were 0.43±0.04, 0.49±0.038, 0.31±0.040, 0.48±0.039 and 0.62±0.086, respectively. Genetic correlations were －0.02, －0.13, －0.55 and －0.15 between ADG1 with ADG2, BF, D90 and FC, respectively, 0.16, －0.73, －0.32 between ADG2 with BF, D90 and FC respectively, 0.01, －0.08 between BF with D90, FC, respectively, and 0.23 between D90 with FC.
Estimation of Genetic Trend on Racing Time of Thoroughbred Racehorses

The objective of this study was to estimate genetic trend on racing time of Thoroughbred racehorses in Korea, using a total of 209,725 racing records of 9,934 racehorses collection from January, 1990 to December, 2006. Phenotypic trends for all distances were negative at a rate of －0.148, －0.137, －0.137 and －0.139 second per race year for distances of 1,000m, 1,400m less than, 1,700m more than and overall dataset, respectively. Environmental trends were similar to phenotypic ones in all distances and trends in permanent environmental and jockey effects by race year were not found. Average genetic improvements for racing time were －0.037 and －0.030 second per race year at the 1,000m and overall dataset, respectively, which is low. But Genetic trends were decreased consistently. There is need to establish the genetic improvement program for quality of racehorses.
Estimation of Environmental Effect and Genetic Parameter on Reproduction Traits for On-farm Test Records

The purpose of this study was to estimate the genetic parameters and trend of Landrace and Yorkshire pigs, which were raised on private farms from 1999 to 2005 and tested for their reproductive performance by the Korea Animal Improvement Association. Prior to analysis, records without pedigree or having value with larger than±3×standard deviation for the Total number of born were excluded. The effects of breed and environmental factors were estimated with least square method(Harvey, 1979), and estimation of breeding values and genetic parameters were performed on the data of 1’st litter only with GIBBSF90(Misztal, 2001) which was programmed according to Gibbs Sampling method based on Bayesian Inference by Gianola and Fernando(1986), Jensen(1994) and others. Gibbs sampling was performed 50,000 times for each parameter, and the first 5000 samples were regarded as those in burn-in period and thus, excluded for post hoc analysis. Total number of born and total number of accident were statistically significant(p<0.01) for the breed, farrowing year, farrowing season and parity effects, and the number born alive at birth was statistically significantp<(0.01) for the breed, farrowing year, farrowing season and parity effects. No particular trend was observed in the genetic and phenotypic improvement of the total number of born and number born alive at birth before 2001, when the piglet registration system started, but the tendencies of increasing for the total number of born and number born alive and decreasing for the total number of accident were observed since 2001. Somewhat higher heritability estimates of our study seems to be attributed to the situations that first parity records with poor farrowing performances were used in the analyses and it was impossible to obtain accurate reproductive performance due to the absence of criteria for record keeping at the level of individual farms.
Postnatal Ontogeny of Expression of Monocarboxylate Transporters(MCTs) and Two Regulatory Proteins, Basigin and Embigin, in The Epididymis of Male Rat

In the present study, real-time PCR was performed to evaluated expression of several isoforms of monocarboxylate transporters(MCTs) and two known MCT regulatory proteins, basigin (Bsg) and embigin, in the epididymis of the male reproductive tract during postnatal development. In addition, ERα�-mediated regulation of MCT1 expression in the epididymis was determined with estrogen receptor(ER) α� knockout(α�ERKO) mice by immunohistochemistry. Results from the current study demonstrated differential expression of MCT isoform(MCT 1, 2, 3, 4, and 8), Bsg, and embigin mRNAs in rat epididymis according to postnatal age and epididymal region. In addition, immunohistochemical study of MCT1 revealed the limited localization of MCT1 at apical area of corpus and caudal epididymis. The present study also showed that expression of MCT1 was not directly regulated by ERα�. The findings from the current study suggest that MCTs would involve in establishing adequate microenvironment for sperm maturation and storage in the epididymis, eventually leading to maintenance of male fertility.
Effects of Dietary Pine Cone Meal on Growth Performance, Serum Cholesterol, Carcass Quality and Fatty Acid Composition and Cholesterol Content of Meat in Broiler Chickens

This study was conducted to evaluate effects of dietary pine cone meal on growth performance, serum cholesterol, carcass quality and fatty acid composition and cholesterol content of meat in broiler chickens. Three treatments were assigned to 480(2 days) Arbor Acre broiler chicks with eight replications, allocating 20 broiler chicks in each replication. Dietary treatments included 1) CS(1% cottonwood sawdust), 2) PCM0.5(0.5% cottonwood sawdust + 0.5% pine cone meal) and 3) PCM1.0(1% pine cone meal). During the overall period the difference of growth performance was not significant among treatments (P>0.05). Also, cholesterol profile(total, HDL, LDL cholesterol and triglyceride) in serum and carcass characteristic were not affected by treatments(P>0.05). TBARS of leg and breast meat was significantly decreased (P<0.05) in PCM1.0 treatment compared to CS treatment. In fatty acid contents of leg meat, C20:0 was significantly higher in PCM0.5 treatment than PCM1.0 treatment(P<0.05). Also, C18:3n3 was significantly lower in PCM1.0 treatment than other treatments(P<0.05). Cholesterol contents of leg, breast meat and abdomen fat were not significant among treatment(P>0.05). In conclusion, results of the experiment were affected by dietary supplementation of pine cone meal on TBARS of leg and breast meat.
Effects of Feeding Dehulled Soybean Meal on Growth Performance, Carcass Characteristics and Meat Quality Parameters in Berkshire Pigs

A total of 360 Berkshire(hog) weighing 25kg were fed experimental diets containing either dehulled soybean meal(DSBM) or non-dehulled soybean meal(NDSBM) as a major protein source until they reach 80kg. The pigs were then fed the finisher diets either high(3,265 kcal/kg) or low(2,940 kcal/kg) in ME to evaluate the effects of dietary energy level for finisher phase on the growth performances, carcass and meat quality parameter. In growth performance, daily weight gain(kg/day) was higher(P<0.05) in T1 than in other groups. Feed conversion(feed/gain) was lower(P<0.05) in other groups. In carcass parameter, carcass percentage was higher(P<0.05) in T2 than in T1 and T4. Backfat thickness was lower(P<0.05) in T2 than in T1 and T3. The ratio of up B grade in carcass grade tender to be higher in T2 than in other groups. In meat quality, chemical composition was similar between groups. WHC was slighter higher in T1 than in other groups. Shear force was tender to be higher in T1 than in other groups. Hunter L and a in meat color were tender to be higher in T2 than in other groups. Hunter L in backfat color was thender to be higher in T1 than in other groups. The results indicate that growth performance and carcass parameter of Berkshire were improved by the dehulled soybean meal as comparison with non-dehulled soybean meal during growing and finishing periods.
Effects of Dietary Escherichia coli Phytase Supplementation on Growth Performance, Carcass Quality and Excretion of Copper and Zinc Concentrations in Finishing Pigs

This study was conducted to evaluate effects of dietary Escherichia coli phytase supplementation on growth performance, carcass quality and excretion of copper and zinc concentrations in finishing pigs. The total of seventy two [(Landrace×Yorkshire)×Duroc] pigs(65.43±0.72 kg in average initial body weight) were used in 56 days assay. Dietary treatments included 1) CON (basal diet) 2) P5(basal diet+phytase 0.05%) and 3) P10 (basal diet + phytase 0.1%). There were three dietary treatments with six replicate pens per treatment and four pigs per pen. During the overall periods, ADFI(Average daily feed intake) was increased (P<0.05) in phytase supplementation treatments compared to CON treatment. At the 5th week, dry matter, nitrogen and ash digestibilities were higher in P5 treatment than in CON and P10 treatments(P<0.05) and CON treatment showed the lowest effect on nutrition digestibility(P<0.05). However, phosphorus digestibility was higher in both phytase supplementation treatments than in CON treatment(P<0.05). The pH of M. logissimus dorsi was significantly higher in CON treatment than phytase supplementation treatments(P<0.05). L* value of M. logissimus dorsi muscle color was significantly increased(P<0.05) in P10 treatment compared to CON and P5 treatments. Also, a* value was increased (P<0.05) in CON treatment compared with phytase supplementation treatments. However, excretion of copper and zinc concentrations were no significant difference among the treatments. In conclusion, the effect of Escherichia coli phytase showed in ADFI, digestibilities, pH and color of meat in finishing pigs.
Effects of the Combination Feeding of β-glucan and Multi-species Probiotics on Growth Performance and Microflora in Broiler Chickens

This experiment was carried out to investigate the combination feeding of β-glucan and multi-species probiotics on growth performance, various lipid concentrations of serum, antibody production and cecal microbial profiles in broiler chicks. A total of six hundred 1-d-old male broiler chicks were divided into five groups, placed into four pens per group(30 birds per pen) and fed one of five non-medicated experimental diets(T1, 0.15% multi-species probiotics; T2, 0.1% β-glucan+0.15% multi-species probiotics; T3, 0.3% multi-species probiotics; T4, 0.1% β-glucan+0.3% probiotics or devoid them as control) for 5 wk. There was no significant difference in feed intake among the groups. The average weight gains and FCR in groups fed diet containing 0.3% probiotics were significantly improved(p<0.05) than control in finisher period(22-35d). The concentration of serum cholesterol ester in groups fed 0.3% probiotics were significantly lowered(p<0.05) as compared to that of the control. Relative weights of liver, spleen, bursa of Fabricius, breast and leg were not influenced by the dietary treatments. The average ND or IB antibody titers in groups fed diets containing β-glucan and probiotics were tended to be increased, but not significantly. The number of cecal lactic acid bacteria was significantly increased(p<0.05) by the dietary β- glucan and probiotics. These results indicated that dietary β-glucan and probiotics exerted a growth- promoting and immune-enhancing effects on broiler chicks. In addition, yeast derived β-glucan, and multi-species probiotics modulated the profiles of cecal microflora, reflecting potential alternative substances to replace antibiotics for feeding broiler.
Effect of Cryoprotectants on Quality Properties of Chicken Breast Surimi Manufactured by pH Adjustment

This study was conducted to determine the effect of pH adjustment and the addition of cryoprotectants on the storage properties of chicken breast surimi. Surimi as a control was prepared with Alaska pollack by two times of washing treatment and addition of cryoprotectants(4% sugar, 5% sorbitol and 0.3% polyphosphate). Three types of surimi were manufactured by different cryoprotectant conditions(T1: 5% sorbitol+0.3% polyphosphate, T2: 4% sugar+5% sorbitol+0.3% polyphosphate, T3: 2% salt+4% sugar+5% sorbitol+0.3% polyphosphate) and frozen after adjusting the surimi to pH 11.0. The amino acid and saturated fatty acid composition were significantly higher in the control. EAA(essential amino acid), FAA(amino acid in relation to flavor), SAAA(amino acid in relation to saccarinity) and FRAA (fragrant amino acid) were significantly higher in the control. The TBARS(thiobarbituric acid reactive substances) and VBN(volatile basic nitrogen) were increased with storage times. TBARS and VBN values were significantly higher in the control and T3 than the other treatments. The VBN was significantly higher in control and T3 than other surimi samples. Chicken breast surimi adjusted to pH 11.0 had higher in microorganisms than the control, and T1 sample had the highest total plate count. Sensory evaluations were significantly higher in the control and T3 than the other samples. Especially, the overall acceptability of T3 was similar to the control.
Effects of Lipolysis and Fatty Acid Composition on Off-flavor in Goat Milk

The average milk fat content in goat milk was 3.88% on yearly basis. The milk fat content of 3.8% during summer season was lower than 4.2% during winter season. Total solid content increased in proportion to milk fat. When goat milk was stored at 4℃ for 24 hr, short-chain FFA(C4:0~C10:0) and medium- and long-chain FFA(C12:0~C18:1) increased about 106% and 203%, respectively. Induced lipolysis of goat milk by homogenization increased short-chain FFA and medium- and long-chain FFA by 22% and 199%. When goat milk was treated with calf lipase, there was increase of short-chain FFA by 9 times greater than increase of medium- and long-chain FFA by 5.6 times. Treatment with lipases from Candida rugosa and Pseudomonas fluorescens resulted in increase of medium- and long-chain FFA by 34 and 162 times, respectively, which was greater than increase of short-chain FFA by 6 and 14 times, respectively. Lipolysis in goat milk stored at 4℃ for 24 hr was correlated with LPL activity in goat milk(r=0.5635). Off-flavor of goat milk was correlated with LPL activity(r=0.5777). Milk fat content was negatively correlated with LPL activity(r=－0.4627). Palmitic acid content in goat milk was correlated with off-flavor(r=0.7226).
The Effects of Pig Breeds on Proximate, Physicochemical, Cholesterol, Amino Acid, Fatty Acid and Sensory Properties of Loins

A total of 100 pigs were used to investigate the effects of pig breed on proximate, physicochemical, cholesterol, amino acid, fatty acid and sensory properties of loins. Crossbred pigs were alloted into one of five experimental groups [
×Berkshire (YBB), British Berkshire(BB), Kagoshima Berkshire(KB), Korean native black pig×Wild boars(KW; Sus coraanus)]. Pigs were slaughtered at 110 kg live weight. Moisture content was significantly(p<0.05) higher in KW than those of other pig breeds. Crude protein content was significantly(p<0.05) higher in YBB and KW than in LYD, BB, and KB. Cholesterol content was significantly(p<0.05) higher in YBB than those of other pig breeds. WHC and pH value had lower and shear force had higher(p<0.05) in YBB than those of other pig breeds. In color, LYD was significantly(p<0.05) higher in L* than those of other pig breeds. There was no significant different in a* and brittleness value among the breeds. In fatty acid and amino acid composition, KW had a higher(p<0.05) UFA/SFA, EFA/UFA ratio and EAA content but lower (p<0.05) SAAA. Total amino acid content was significantly(p<0.05) higher in LYD(20.44%) and BB (20.81%) than those of other pig breeds. In fresh meat, breeds affected meat color, drip loss, marbling score, and overall acceptability(p<0.05). Nevertheless, no significant differences were found among the breeds in the sensory parameters evaluated in the cooked meat, except for flavor
Effects of Sweet Persimmon Powder Type on Quality Properties of Low Salted Pork Patties during Cold Storage

Four different pork patties were made with two levels, HP/FP-3% and HP/FP-6%, containing 3.0 and 6.0% HP(hot air dried sweet persimmon powder)/FP(freeze-dried sweet persimmon powder), respectively. After manufacture, the meat patties were packaged with polyvinyl wrap and stored at 4℃ for 8 days. CTL(control) and HP-3% meat patties were significantly(p<0.05) higher in moisture content than the other samples. FP-6% was found higher in protein content than the other treatments. FP-3% had a higher fat content than other meat patty samples. However, ash contents were not found significantly(p>0.05) different among the meat patty samples. The value of pH, L* and a* values were decreased as the cold storage time increased in all treatments(p<0.05). WHC(water holding capacity) of CTL and HP-6% and cooking loss of HP-3% were significantly(p<0.05) decreased with increased storage period. The diameter and thickness of all meat patties decreased with increasing the storage period. VBN(volatile basic nitrogen) values of all meat patties were increased(p<0.05) with increased storage period. TBARS (thiobarbituric acid reactive substance) of treatments were higher than that of CTL during whole storage time. The number of microorganisms(Total plate counts, Escherichia coli.) were maintained below 4.61 log10 CFU/cm2 during the whole storage period. In sensory evaluation, treatment groups had higher(p<0.05) scores in aroma, flavor, color and overall acceptability.
