DETECTION OF DNA SINGLE-STRAND BREAKS AND UNSCHEDULED DNA SYNTHESIS INDUCED BY PROCARCINOGENS IN PRIMARY CULTURES OF RAT HEPATOCYTES

Procarcinogen induced DNA single-strand breaks and unschduled DNA synthesis were measured in primary rat hepatocytes culture. For DNA single-strand breaks assay, rat liver DNA was prelabeled by injection 3H-thymidine during the peak of DNA synthesis following partial hepatectomy. Hepatocytes were isolated from the rat 2 weeks after surgery by a collagenase perfusion techinique and maintained as monolayers in serum free medium on collagen-coated culture dishes. DNA sigle-strand breaks were measured by the alkaline elution techinique.
REPRODUCTION STUDIES WITH PRANOPROFEN, A NONSTEROIDAL ANTI-INFLAMMATORY AGENT IN RATS-TERATOLOGY TEST

Teratological study on pranoprofen, as antiinflammatory agent, was conducted by oral intubation in Sprague-Dawley rats. Pranoprofen was administered doses of 1.0, 2.5 and 5.0mg/kg/day and doze of 0.5mg/kg/day of Indomethacin was used as positive control. The rats were dosed from day 7 to 17 of gestation. At necropsy on day 20 of gestation, pathologically changes of gastroin-testinal system, liver and adrenal gland were examined at the high dose administered group. There were no differences between control and treated group on the number of implantations, the number of alive and dead fetuses, tail length, and external visceral and skeletal malformations.
ACUTE TOXICITY STUDY OF HEPACCINE-B(HEPATITIS B VACCINE)

Acute toxicity study was conducted on a Hepatitis B vaccine (Hepaccine-B-inj.) with mice, guinea pigs, and rabbits, in accordance with the norms suggested by the F.D.A. in U.S.A. Dose ranges were 2 doses/mouse, 5 doses/guinea pig, 10 doses/rabbit. They received the vaccine subcutaneously and intraperitoneally. Thereafter, all animals injected were observed of general signsdaily, and of body weight for two weeks. At the end of the observation period (or at the time of death), all animals received the highest dose group were autopsied and gross observation was made on various organs and tissues. No significant toxicity was noted.
EFFECT OF GLYCYRRHETINIC ACID ON PYRIDINE TOXICITY IN MOUSE

The present work was undertaken to investigate the effect of glycyrrhetinic acid on pyridine toxicity. When glycyrrhetinic acid was pretreated, pyridine-induced CNS depression and mortality were decreased. A striking enhancement of serum transaminase activities after pyridine administration was markedly decreased by glycyrrhetinic acid pretreatment. It was also observed that the hepatic microsomal aniline hydroxylase activity, which is concerned with pyridine metabolism, was significantly increased in glycyrrhetinic acid pretreated mice.
THE TERATOLOGICAL EFFECTS OF NITROFEN ON FETUSES IN PREGNANT RATS

This paper describes the effects of nitrofen (Hi-TOK), a herbicide on the fetuses of rats. The results were observed as follows: The internal soft tissue anomalies were classified as diaphragmatic hernia, cardiac malformation (T.G.V., V.S.D., S.V.), dilatation of ventricle in brain, dilatation of renal pelvis, underdevelopment of fetal lung, shortening of cortex length and increasing of immatured glomeruli counts in the fetal kidney. The heart and diaphragm appear to be the target organs.
