EFFECTS OF AQUEOUS EXTRACT OF A POISONOUS MUSHROOM, AMANITA PANTHERINA ON MICE AND ASSAY OF TOXIC ISOXAZOLE DERIVATIVES BY HIGH PERFORMANCE LIQUID CHROMATOGRAPHY

In order to elucidate the mechanism of toxic action of a pisonous mushroom, Amanita pantherina, biochemical effects of the mushroom extracts on mice were studied. A hotwater extract of Amanita pantherina injected intraperitoneally into male ICR mice evoked signs similar to those observed clinically upon acute poisoning by the mushroom and also changed the levels of component enzyme activities in blood, liver and urine. The serum cholinesterase activity was decreased significantly during 1-3 h after injection.
A POSSIBLE MECHANISM OF POLYACETYLENE: MEMBRANE CYTOTOXICITY

The effects of polyacetylenes on living membrances, rat erythrocyte and murine leukemic L1210 cell as well as artificial lipid bilayer were determined to investigate the cytotoxic mechanism of polyacetylenes against cancer cell lines. As results, panaxydol and panaxynol caused erythrocyte hemolysis dose-dependently while panaxytriol had no lysis. For liposomes composed of phosphatidyl choline (PC) and phosphatidic acid(PA), all three polyacetylenes supressed the osmotic behavior at the same degree.

The effects of altering the hepatic mixed-function oxidase(MFO) activities on the acute toxicity of parathion were examined in female rats. Phenobarbital sodium pretreatment (50mg/kg/day, i.p.) for 4 consecutive days has resulted in significant decreases in the toxicity of parathion (2 or 4 mg/kg, i.p.) as determined by lethality and cholinesterase activities wheras the toxicity arising from a single dose of CCl4(2 mmol/kg, i.p.) 24 hr prior to parathion challenge was potentiated.
QUANTITATION OF OXANDROLONE(A SYNTHETIC ANABOLIC STEROID) IN HUMAN URINE BY GC/MS

A sensitive method for the quantitation of oxandrolone in urine was developed using GC/MS. After oral administration of 10mg oxandrolone, oxandrolone excreted in urine as unchanged form was extracted in ether and derivatized to its O-TMS. Oxandrolone excreted in urine as glucuronide conjugated form was extracted after enzymatic hydrolysis and derivatized to its O-TMS. The amounts of oxandrolone-O-TMS was measured in GC/MS with selected ion monitoring. Calusterone, a structurally similar anabolic steroid, was employed as internal standard.
USE OF A MIXED METABOLIC ACTIVATION SYSTEM IN THE SALMONELLA REVERSE MUTATION TEST OF CHEMICAL CARCINOGENS

The post-mitochondrial liver fractions (S-9) were prepared from rats and hamsters which have been treated with Aroclor 1254 (PCB) and the capacities of these S-9 fractions to generate mutagenic metabolites from several well known procarcinogens have been compared. Benzo(a)pyrene (B(a)P), 3-methylcholanthrene (3-MC), Aflatoxin B1(AFB1), 2-acetylamino-fluorene(AAF), and 2-aminofluorene (AF) were employed as promutagens in the Salmonella reverse mutation tests. Results showed that the rat and hamster S-9 fractions had differential abilities to produce mutagenic metabolites from a given promutagen.
AN EVIDENCE FOR THE INVOLVEMENT OF CYTOLYSIN IN VIBRIO VULNIFICUS DISEASE

Cytolysin produced by Vibrio vulnificus ATCC 27562 was partially purified by sequential ammonium sulfate precipitation, gel filtration with Sephadex G-200, and ion exchange chromatography with DEAE-Sephadex. The partially purified cytolysin was inactivated by cholesterol. More than one molecule of the cytolysin was required to lyse a single erythrocyts. The antiserum against cytolysin enhanced the survival ratio of mice infected with low dose of V. vulnificus
IMMUNOMODULATING ACTIVITIES OF BRAZILIN AND HEMATOXYLIN IN NORMAL YOUNG MICE

Experiments were undertaken to evaluate the effects of brazilin and hematoxylin on immune functions in normal young mice. Brazilin and hematoxylin decreased the circulating leucocyte counts. Brazilin and hematoxylin decreased the cirulating leucocyte counts. Both compounds did not produce a significant change of immunoorgan weights in comparison with those in the control group. Brazilin decreased significantly IgM plaque forming cells. Brazilin and hematoxylin decreased significantly Arthus reaction at 3 hours-post challenge with respect to aggregated bovine serum albumin.
INHIBITORY EFFECTS OF RED GINSENG EXTRACT ON PRENEOPLASTIC NODULE FORMATION IN THE RAT LIVER OBSERVED BY RAPID BIOASSAY OF CARCINOGENESIS

This study was carried out to investigate the inhibitory effects of red ginseng extract on preneoplastic nodule formation in the rat liver using a rapid bioassay of hepatocarcinogenesis. Wistar male rats in test groups were fed diets containing 3.0, 0.3 or 003% of red genseng extract for 6 weeks after 2 week of dosing with diethylnitrosamine (DEAN, 200 mg/kg B.W., I.P.) or saline solution. The rats in the control groups were treated with DENA or saline alone. Three weeks after the beginning of the experiment, the two-thirds partial hepatectomy was performed on all animals. The inhibitory effects were investigated by measuring the number, total area and size of hyperplastic nodules enhanced by the 2/3 partial hepatectomy following induction with DENA. The results obtained were summarized as follows: 1. In general, the body weight gains of the rats fed diets containing red ginseng extract were higher than those of other groups.
INFLUENCE OF LOW OR HIGH ENDOTOXIN-SUSCEPTIBLE RABBIT ON PYROGEN TEST

This study was carried out to investigate whether the use of the high endotoxin-sensitive or the low endotoxin-sensitive rabbits (New Zealand White strain) are more suited for the pyrogen test regulated by the Korean Pharmacopeia and United State Pharmacopeia. The results obtained were as follows. 1. The ferbrile responese were increased according to increased endotoxin doses used (i.e., 0.1, 1, 5, 10ng/kg) and the febrile responses reached to its maximum between 90 mins and 120 mins after the intravenous injection of endotoxin.
