HEPATO-AND RENAL TOXICITY OF AQUEOUS EXTRACT OF A MUSHROOM, AMANITA VOLVATA IN MICE

Toxic effects of a mushroom, Amanita volvata, were studied in respect to biochemical and histological changes induced in the liver and kidneys of mice. The changes in biochemical parameters characteristically appeared 12 hrs after oral administration of an aqueous extract of the mushroom. The hepatic glycogen decreased markedly to 17% of the control level and a concemitant decrease in blood glucose was also observed. The activity of serum glutamic oxaloacetic transaminase (SGOT) was elevated by 2.5-fold and the level of blood urea nitrogen (BUN) increased by 3-fold, respectively, 12 hrs after administration of the mushroom, their levels being maintained up to 24 hrs.
EFFECT OF BUTYLATED HYDROXYTOLUENE (BHT) AND ITS METABOLITE ON THE UPTAKE OF TAUROCHOLATE IN PRIMARY CULTURE OF ADULT RAT HEPATOCYTES

The effect of butylated hydroxytoluene (BHT) and its major metabolite, 3, 5-di-tert-butyl-4-hydroxybenzoic acid (BHT-acid) on the uptake of taurocholate into hepatocytes was studied using the primary culture of rat hepatocytes. Hepatocyte were isolated by an in situ collagenase perfusion technique and maintained as a monolayer in serum-free meadia for 24 hours before use. The uptake of taurocholate was saturable with an apparent Km of 12.8+2.8 MuM and Vmax of 0.18+0.01 nmol/mg/min. Both BHT and BHT-acid inhibited the hepatocellular uptake of taurocholate when they were added to the culture.
DIFFERENTIAL INDUCTION OF RAT LIVER MICROSOMAL CYTOCHROME-DEPENDENT MONOOXYGENASE AND UDP-GLUCURONOSYLTRANSFERASE ACTIVITIES BY VARIOUS NARCOTIC DRUGS

Chronic adminstraction of morphine to adult male rats has long been known to lower hepatic cytochrome p-450 content and its dependent mixed-function oxidase activity. Following the treatment of adult male rats with morphine, pethidine pentazocine and codeine and also by concomitant adminstration of naloxone activities of microsomal electron transfer in the adult male rats were examined. In present study, the acute treatment of mature male rats with a dose of narcotic drugs higher than that used chronically also reduces their hepatic cytochrome p-450.
MICROCIRCULATORY ABERRATIONS IN THE ISOLATED PERFUSED RAT LIVER INDUCED BY SODIUM CYANIDE, ANOXIA OR ACETAMINOPHEN

When acetaminophen (25mM) was introduced into the perfused rat liver, the hepatic O2 uptake was rapidly inhibited first and then later slow-down. The rapid inhibition was found to be due to mitochondrial blockade, whereas the so-called slow inhibition" was associated with microcirulatory aberrations as evidenced by inhomogneous staining of the liver tissue by trypan blue infusion (0.1%). NaCN (0.5mM) also caused rapid and slow respiratory inhibitions, giving heterogeneous trypan blue staining.ning.
EFFECTS OF DICHLOROMETHANE ON CARBON TETRACHLORIDE HEPATOTOXICITY IN RATS

A non-hepatotoxic dose of dichloromethane (DCM) was examined for potential effects on the hepatotoxicity of carbon tetrachloride (CT) in adult male rats. A concomitant treatment of DCM (0.45ml/kg, po) significantly potentiated the hepatotoxicity of CT at varing doses (0.06 to 0.63 ml/kg, po) as determined by increase in SGOT and SGPT activities 24 hn following the treatments. The carboxyhemoglobin (COHb) saturation induced by DCM was significantly decreased by CT treatments. The potentiation of CT hepatotoxicity by DCM does not appear to be associated with increased metabolism of CT.
STUDY ON THE MERCURY CONTENTS IN KOREAN SCALP HAIRS

The average value of total mercury contents in scalp hair of Korean male was found to be 1.66+1.10 ppm (S.D.) and that of methylmercury was 1.02+0.72 ppm (61.0% of the total mercury). In case of total mercury contents of female was 1.06+0.46 ppm and that of methylmercury was 0.51+0.27 ppm (48.4% of total mercury level). When compared with data reported by Japanese, the levels were about half. The levels of mercury in scalp hair of male subjects were significantly different with their occupations, but in case of female, the variation was not so much.
ACUTE MAMMALIAN TOXICITY OF O-CHLOROBENZYLIDENE MALONONITRILE(CS)

Acute inhalation intoxication of CS (O-chlorobenzylidene malononitrile) occurred among the 192 animals in confined animal cages of farm as the result of prolonged exposure. A total of 8 animals (3 silver foxes, 3 fitches and 2 minks) died in 15 hours after the exposure. Distinct evidences of pulmonary atelectasis were observed as with hepatorenal damages. The lethal toxicity of CS was considered to be due to early severelung damages leading to asphyxia, accompanying acute toxic hepatitis and nephritis.
EFFECT OF PATULIN ON THE GROWTH OF BACTERIOPHAGE M13

A mycotoxin Patulin, isolated from apple juice medium cultured with Penicillium patulum NRRL5259, was purified through acid aluminum column using ethyl ether as eluent. The yield of purified patulin crystal was 3mg/ml culture medium after 8 days of shaking culture at 28C. The growth rate of Escherichia coli K12JM103 infected with bacteriophage M13 was decreased by patulin at the concentration range of 1Mug/ml to 10Mu/nl. ED50 of patulin for the bacterial growth was 4.5Mug/ml and 10Mug/ml of patulin caused maximum inhibitory effect (60% inhibition) on the growth.
TERATOGENICITY STUDY OF FOOD ADDITIVES CHEMICALS IN THE DEVELOPING CHICK EMBRYO

To evaluate possible teratogenicity of food additives such as sodium propionate and monosodium glutamate, 300 haching eggs were subjected to potential mutagenicity assay by administration of low and high doses of the materials via york and air sac. The results are summarized as follows: 1. Mortality rate of chick embryo was increased significantly by dosage related in the group of sodium propiorate and monosodium glutamate when compared to placebo and non-treated group. 2. Frequency of embryo with malformation in treated group was not increased significantly when compared to placebo and non-treated group.
