BRAZILIN MODULATES THE IMMUNE FUNCTIONS IN NORMAL CBA FEMALE MICE

Brazilin, the main constitutent of Caesalpinia sappan, was examined for its immunomodulating activities in normal CBA mice. Mitogen induced proliferation and production of ConA induced T-cell growth factors (TCGF) of splenocytes were significantly reduced in brazilin treated group, cmpared to control group. It was also found that suppressor activities of splenocytes in brazilin treated group was significantly increased compared to those in normal control group.
Hydroxybrazilin was examined for its effects on glycogen synthesis in primary cultured rat hepatocytes.

Hydroxybrazilin was examined for its effects on glycogen synthesis in primary cultured rat hepatocytes. At 10-6 M hydroxybrazilin, glycogen synthesis was increased in basal state, but not in insulin stimulated state. However, any signiFicant changes were nor observed at 10-5 M hydroxybrazilin in both states. The glycogen synthesis was rather suppressed at 10-5M hydroxybrazilin. It was also observed that hydroxybrazilin increased insulin sensitivity but not insulin responsiveness at 10-5M concentration. These results suggest that hydroxybrazilin might exert hypoglycemic action through its effects on insulin receptor and post receptor events.
A STUDY ON ANTIGENICITY OF GRANULOCYTE-MACROPHAGE COLONY STIMULATING FACTOR(LBD-005) IN MICE AND GUINEA PIGS

Antigenicity of Recombinant Granulocyte macrophage colony stimulating factor(LBD-005), a newly developed drug for hematopoietic growth, was investigated in mice and guinea pigs. 1. Mice showed production of antibodies against LBD-005 (100mg/kg) with alumin]m hydroxide gel(alum) as an adjuvant, judged by the heterologous anaphylaxis(PCA) test using rats. On the other hand, antibodies against ovalbumin(OVA) inoculated with alum were definitely detected. 2. In the studies with guinea pigs, both the inoculation of LBD-005 only and of LBD-005 with complete Freund's adjuvant(CFA) as an adjuvant did not produce positive reactions in homologous passive cutaneous anaphylaxis (PCA).
FERTILITY STUDY (SEGMENT 1) OF RECOMBINANT GRANULOCYTE-MACROPHAGE COLONY STIMULATING FACTORS (LBD-005) IN RATS

The recombinant glycoprotein, LBD-005 (Lucky R & D Center, Biotechnology)stimulates the growth of stem cells and activates the development of the hematopoietic cell in a similar manner to the naturally occurring GM-CSF. This test was conducted to investigate if LBD-005 had any effect on fertility in male and female rats. Sprague-Dawley rats(88 of each sex) bred at KRICT, were dosed subcutaneously, at a volume of 2ml/kg body weight with LBD-005 at 0, 250, 500 or 1, 000mg/kg body weight. The male rats were dosed, from 6 weeks of asge, for 60 days prior to mating.
ACUTE TOXICITY STUDY OF RECOMBINANT GRANULOCYTE-MACROPHAGE COLONY STIMULATING FACTOR (LBD-005) IN RATS

The actue toxicity of a recombinant granulocyte macrophage colony-stimulating factor (code name: LBD-005) was evaluated in both sexes of Sprague-Dawley rats, 4 weeks old, by the oral, subcutaneous and intravenous routes of administration. LBD-005 in the acute toxicity study in the rats was not considerde to induce any toxicological effect on the rats in mortalities, clinical findings, body weights and gross findings. It is suggested that
values in rats would be >48 mg/kg in the oral route and >12 mg/kg in the subcutaneous or intravenous route.
4-WEEK SUBCUTANEOUS TOXICITY STUDY OF RECOMBINANT GRANULOCYTE-MACROPHAGE COLONY STIMULATING FACTOR (LBD-005) IN RATS

Recombinant granulocyte-macrophage colony stimulating factor was subcutaneously administered to both sexes of Sprague-Dawley rats at the doses of 0, 0.5, 1 and 2mg/kg of body weight five days per week for 4 weeks to evaluate the subchronic toxicity. There were decreased 1 and 2 mg/kg. In the serum, changes were decreased alkaline phostatase(ALP) in the female groups dosed at 1 and 2mg/kg, and increased glutamic oxaloacetic transaminiase (GOT)in the male groups dosed at 0.5 and 2.0mg/kg.
EVALUATION FOR THE CONVULSIVE LIABILITY OF VARIOUS QUINOLONE DERIVATIVES IN MICE

The present study was performed to evaluate whether the application of Fenbufen is reasonable for predicting the convulsive liability of the quinolone derivatives and to examine whether pentylenetetrazole (PTZ) can be used as a screening tool for their Central Nervous System (CNS) toxic pontential. The convulsive activity of the quinolones was markedly potentiated by the pretreatment of Fenbufen. In combination with Fenbufen, enoxacin (ENX), norfloxacin (XFLX), and ciprofloxacin (CPFX) provoked convlusions and subsequent death at the intravenous doses of 0.5mg/kg, 10mg/kg, and 40mg/kg, respectively, whereas ofloxacin (OFLX) and pefloxacin (PFLX) did not induce convulsions and death even at a relatively high dose of 100mg/kg iv.
EVIDENCE FOR HE CHANGES IN THE DOPAMINERGIC ACTIVITY AFTER THE SUBACUTE ADMINSTRATION WITH PHYSOSTIGMINE

Rats were treated with physostigamine acutely and subacutely for 7 days. The duration of tremor, striatal acetylcholinesterase(AChE) activity and the occurrences of dopaminal antagonist-induced catalepsy were measured. During the daily treatment with 0.75mg/kg of physostigmine, the durations of tremor were markedly reduced. However, the tremors were ot occurred in the continuous infusion up to 0.20mg/kg/hr. Striatal AChE activities were significantly inhibited(8.25%) early hour, but twenty-four hour after daily administration with physostigmine AChE activities during the continuous infusion with physostigmine were also significantly inhibites (30-60%).
EFFECTS OF DIETARY AFLATOXIN B1 ON PERFORMANCE, ON HEMATOLOGIC, PATHOLOGIC AND IMMUNOLOGIC CHANGES IN BROILER CHICKENS

The influences of dietary aflatoxin B1 on performance, on hematologic, pathologic and immunologic changes in broiler chickens were studied. One hundred and fifty hatched broiler chickens were fed with diet containing aflatoxin B1 (1.0 ppm and 2.5 ppm) fot three weeks. Blood samples, serum, and immune organs were obtained to investigate hematological, clinico-chemical, and histopathologial changes. Body weight gain and feed intake were significantly decreased. The liver and kidney were increased, whereas the bursa of Fabricius, spleen and thymus were decreased.

Basolateral and brush border membrance (BLM and BBM) vesicles of renal proximal tubules were prepared from adult male New Zealand White rabbits to develop an experimental for assessment of nephrotoxicity. PAH uptakes using BLMV, and glucose and leucine uptakes using BBMV were measured in the rabbits treated uranyl acetate. In addition, urinalysis and histopathological studies were performed to investigate the correlationship with the membrance vesicle uptakes.
Streptozotocin에 의한 랫드 적혈구 손상에 관한 연구

Streptozotocin (STZ) is a naturally occurring nitrosoamide used extensively to produce diabetes in experimental animals. Since streptozotocin reportedly decrease deformability of red blood cells (RBC), we sought to investigate its potential toxicity at RBC. In addition to elevation blood glucose, 100mg/kg iv streptozotocin caused significant RBC hemolysis in female Sprague-Dawley at 48 hrs post treatment. Streptozotocin induced hemolysis was found to be dose and time-dependent` complete hemolysis required a relatively high streptozotocin concentration (500 mM, 4 hrs incubation), which is much more than in vivo dose.
몇가지 중금속이 배양 섬유모세포에 미치는 세포독성에 관한 연구

Present study was carried out to investigate the cytotoxicity of cadmium, chromium and mercury on cultured rat fibroblasts. The colorimetric assays of neutral red (NR) and tetrazolium MTT, the measurement of total content of protein and electron microscopic studies were performed on the fibroblasts cultured in the media containing various concentrations of cadmium, chromium and mercury. The results are as follows' 1. In cadmium-treated group, the NR and MTT values were dose-dependent increase. The NR90, NR50, MTT90 and MTT50 values of cadmium were 0.2mM, 19.5mM 1.0mM and 60.0mM, respectively.
THE STUDY ON CARCINOGENICITY OF COPTIS RHIZOME AND CROTON TIGLIUM L.

Sprague-Dawley rats aged six weeks divided into four groups and group 1, 2, 3 and 4 of rats were given an intrapertioneal injection of diethylnitrosamine at 200 mg/kg body weight. Group 4 was Control. Two weeks after beginning of the experiment, group 1 of rats were begun to feed on water containing 0.05% phenobarbital sodium as a promoter for six weeks, and CP-2 were intrapertioneally given to rats of group 2(20mg/kg) and group 3(1mg/kg). Three weeks after beginning of the experiment, partial hepatectomy was performed in all rats. Preneoplastic foci were identified histopathologically by glutathione S-transferase placental form (GST-P) activity. In the Immunohistochemical quantitative analysis of carcinogen-induced foci, it was concluded that CP-2 was not carcinogen.
ANTIGENICITY TEST OF COPTIS RHIZOME AND CROTON TIGLIUM L. EXTRACTS(CP-2) IN GUINEA PIG AND RABBITS

Antigenicity tests-ASA(Active Systemic Anaphylaxis), PSA(Passive Systemic Anaphylaxis), PCA(Passive Cutaneous Anaphylaxis)-of CP-2(Green cross Co). were performed according to the established regulations of National Institute of Safety Research. The results were as follows. 1. No specic clinical signs related anaphylaxis were observed, therefore, it was considered that CP-2 had not antigenicity in guinea pigs and rabbits. 2. No blue spots were observed on the back of guinea pig in the PCA test` CP-2 related IgE was not produced.
THE OVERVIEW OF FEED ADDITIVES AND VETERINARY DRUGS USED IN JAPAN AND THEIR RESIDUAL ANALYSIS IN LIVESTOCK PRODUCTS

The residue of drug in foods of animal origin has increasingly become of interest to the entire livestock industry as growing consumer health concerns. The current overvie of feed additives and veterinary drugs used in Japan and their residual analysis has been reviewed. High performance liquid chromatographic technique(HPLC) with various detectors can be expected to be successfully applied for the routine analysis of residual feed additives and veterinary drugs including anabolic agents in livestock products.
