Fractionation of Gelatin Hydrolysates with Antioxidative Activity from Alaska Pollock Surimi Refiner Discharge

This study was conducted to obtain the gelatin fraction with a high anti oxidative activity from Alaska pollock surimi by-products using a two-step enzymatic hydrolysis and ultrafiltration. Among gelatin hydrolysates from refiner discharge of Alaska Pollock surimi, the highest antioxidative activity (81.5%) resulted from gelatin hydrolysate sequentially treated with Pronase E and Flavourzyme each for 2 hr. However, no difference was seen in the anti oxidative activity of the second hydrolysate (Pronase E-/Flavourzyme-treated hydrolysate) when compared to the permeate fractionated through a 10-kDa membrane. The results suggest that the Pronase E-/Flavourzyme-treated hydrolysate from refiner discharge gelatin of Alaska pollock surimi can be used as a supplementary raw material for improving health functionality.
Genetic Phylogeny among Three Species Red Seabream, Black Seabream and Rock Bream Based on Mitochondrial DNA Sequences

The Perciformes include approximately 40% of all bony fishes and are the largest order of vertebrates. This order includes some of the most economically relevant marine fishes, particularly the red seabream, black seabream and rock bream. A 409 bp fragment of the cytochrome b (cyt b) gene and 403 bp and 518 bp fragments of ribosomal RNA (12S and 16S rRNA, respectively) were sequenced from five populations of natural and cultured red seabreams, natural black seabream, and natural and cultured rock breams. The mitochondrial DNA sequences were utilized for the genetic identification and population structural analyses of these three species. Phylogenetic relationships of intra- and inter-species were elucidated using three types of molecular genetic markers from three species of the order Perciformes in Korea. We noted no significant differences in the intra-specific variation of the cyt b and rRNA genes in each population however, inter-specific divergences were greater than intra-specific variation. Inter-specific variation was induced more by transition than transversion type in the cyt b and rRNA genes. The cyt b gene and rRNA genes make it possible to determine the inter-species divergence. The rRNA genes have more conserved sequences than the cyt b gene. Therefore, these genes are expected to prove useful among species belonging to the different genera or families.
Hepatic Expression of Cu/Zn-Superoxide Dismutase Transcripts in Response to Acute Metal Exposure and Heat Stress in Hemibarbus mylodon (Teleostei: Cypriniformes)

Hemibarbus mylodon (Cypriniformes) is an endemic freshwater fish species in the Korean peninsula, for which urgent conservation efforts are needed. To understand their stress responses in relation to metal toxicity and thermal elevation, we performed a real-time RT-PCR-based expression assay of hepatic copper/zinc-superoxide dismutase (Cu/Zn-SOD), a key antioxidant enzyme, in response to experimental heavy metal exposure or heat treatment. The transcription of hepatic Cu/Zn-SOD was differentially modulated by acute exposure to Cu, cadmium (Cd), or Zn. Exposure to each metal at
for 24 h revealed that Cu stimulated the mRNA expression of Cu/Zn-SOD to a greater extent than the other two heavy metals. The elevation in Cu/Zn-SOD transcripts in response to Cu exposure was dose-dependent (0.5 to

A Myostain-like Gene Expressed Highly in the Muscle Tissue of Chinese mitten crab, Eriocheir sinensis

A complete cDNA, which encodes for a myostatin-like protein (Es-MSTN), was isolated from the Chinese mitten crab, Eriocheir sinensis. Es-MSTN was composed of 2,397 nucleotides and the open reading frame (ORF) specified a protein containing 468 amino acids. Es-MSTN exhibited 32% amino acid sequence identity and 52% similarity to human myostatin. Multiple sequence alignment analysis indicated that Es-MSTN possessed the conserved proteolytic cleavage site (RXXR) for maturation of the protein and nine cysteine residues for disulfide bridges. Besides the conserved structural features, Es-MSTN also exhibits its unique characters; a longer N-terminal domain which is involved in protein folding and latent form of myostatin and absence of the cleavage site for BMP-1/tolloid family of metalloproteinase to activate mature myostatin. Phylogenetic analysis suggests that Es-MSTN showed the closely related to both vertebrate myostatin and GDF11. Es-MSTN is expressed highly in the claw muscle, leg muscle, thoracic muscle and heart, and moderately in the hindgut suggesting that Es-MSTN may play important roles in the muscle tissues. As homolog of mammalian myostatin and GDF11, Es-MSTN may be involved in development of muscular tissue and further study will help to produce high-quality seafood.
Contrasting Correlation in the Inhibition Response of ADP-induced Platelet Aggregation and the Anti-coagulant Activities of Algal fucoidans Derived from Eisenia bicyclis and Undaria pinnatifida sporophylls (Mekabu)

Sulfated fucans are known to have both anti-thrombotic and anti-coagulant activities. In this study, the variation in platelet aggregation and anti-coagulant activities was investigated in vitro with regard to administered dose, molecular weight distribution, sulfate content, and sugar composition in two algal fucoidans from Eisenia bicyclis and Undaria pinnatifida sporophylls (Mekabu). The anti-coagulant activity largely correlated with sulfate content and with molecular weight distribution in a dose-dependent manner. However, both fucoidans demonstrated inhibitory responses to ADP-induced platelet aggregation in dose- and structure-dependent manners that contrasted with the anti-coagulant activity. Neither molecular weight distribution nor sulfate content greatly affected platelet-aggregation inhibition (PA-inhibition) by the fucoidan fractions, whereas anti-coagulant activity was sensitive to these structural factors. Interestingly, an E. bicyclis fucoidan fraction exhibited almost complete PA-inhibition at a treatment dose of 500 mg/mL while retaining weak anti-coagulant activity. In conclusion, these observations suggest that fucoidan may be a useful anti-thrombotic or anti-platelet agent in various arterial thrombotic disorders, including post-vascular intervention with controlled bleeding complications, due to its anti-coagulant modulating activity.
Effects of Sources and Levels of Dietary Carbohydrate on Growth and Body Composition of Juvenile Sea Cucumbers, Apostichopus japonicus

A factorial feeding trial of different levels of three carbohydrate sources (wheat flour,
-potato starch, and sodium alginate) was conducted to determine proper sources and levels of dietary carbohydrate for juvenile sea cucumbers. Three replicate groups of juvenile sea cucumbers (770 mg average weight) were fed the experimental diets once a day for 10 weeks. After feeding trial, survival of sea cucumbers was not significantly different between diets. The body weight of sea cucumbers was significantly (P<0.01) affected by both source and level of dietary carbohydrate, tending to decrease as dietary carbohydrate level increased. Body weight of sea cucumbers fed a 25% wheat flour diet was the highest but was not significantly different from that of sea cucumbers fed diet containing 10% wheat flour with 15% sodium alginate (P>0.05). No significant differences were observed in contents of moisture, crude protein, crude lipid and ash of sea cucumbers among dietary treatments. These results suggest that sea cucumbers utilize wheat flour and sodium alginate more efficiently than they do
-potato starch, and that a formulated diet containing 43-60% carbohydrate may be suitable for juvenile sea cucumber culture.
Survival of the Ark Shell, Scapharca subcrenata and Physiological and Histological Changes at Decreasing Salinity

We examined physiological and histological responses related to the survival, oxygen consumption, excretion, and O/N ratio of the ark shell, Scapharca subcrenata, as a result of salinity changes. The 20-day
(median lethal salinity) at
was 13.87 practical salinity units (psu; confidence limits 10.30-18.74 psu), whereas the 14-day
was 12.59 psu (confidence limits 8.03-18.16 psu). In conditions of decreasing salinity, the osmolarity of individuals acclimated within 5 h above 26.4 psu but required more than 60 h below 13.2 psu. Oxygen consumption and ammonia excretion rates varied irregularly as salinity decreased. The O/N ratio was 19 and 27 at water temperatures of
, respectively, but decreased to 1-10 as salinity declined. The effects of decreasing salinity were observed in the histological changes to each organ of S. subcrenata. As salinity decreased, cilia fell off, the epithelial layer underwent necrosis and vacuolation, the connective tissue layers of the mantle and visceral mass were destroyed, and hemocytes increased in the gills. The results of this study could prove important in investigating causes of mass mortality and managing shellfish aquaculture farms.
Seasonal Variation in the Biomass of Eelgrass (Zostera marina) and Epiphytic Algae in Two Eelgrass Beds around Namhae Island in Korea

Seasonal variation in the biomass of eelgrass (Zostera marina) and epiphytic algae in two eelgrass beds (Dongdae and Aenggang Bay) around Namhae Island was investigated throughout 2005. Shoot density and eelgrass biomass differed across months and locations. Peak shoot density occurred from April to August 2005, whereas eelgrass biomass was higher in July and August 2005. Shoot density as well as eelgrass biomass were higher in Dongdae Bay compared to Aenggang Bay. A total of 21 epiphytic algal species (4 Chlorophyta, 2 Phaeophyta, and 15 Rhodophyta) were collected, and dominant species included Polysiphonia japonica, Lomentaria hakodantensis, Symphyocladia latiuscula, Champia sp., and Heterosiphonia japonica. Seasonal variation in both the species composition and biomass of epiphytic algae was substantial: peak epiphytic algal biomass occurred in January and December 2005. We also observed high epiphytic algal biomass in the eelgrass bed of Dongdae Bay. Seasonal changes in the biomass of eelgrass and epiphytic algae were primarily influenced by water temperature, whereas those of the epiphytic algal community were also correlated with eelgrass (substrate) morphology and growth, the life cycle of epiphytic algae, and physical characteristics within eelgrass beds. The spatial variation of eelgrass density and biomass were also limited by sediment characteristics.
Abundance of the Toxic Dinoflagellate Alexandrium catenella in Jinhae Bay, Korea as Measured by Specific Real-time PCR Probe

The marine toxic dinoflagellate Alexandrium catenella has been implicated in numerous paralytic shellfish poisoning (PSP) events in many countries. Due to difficulties in rapidly identifying A. catenella, field-based study of this species has been problematic. The present study developed a TaqMan format A. catenella-specific probe for real-time PCR assay (specific to Korean genotype) based on LSU rDNA sequence information for studying geographic and temporal distribution of the species in surface sediments and water columns of Jinhae Bay, Korea. The field survey from 2007 to 2008 revealed that A. catenella occurred in most seasons at low densities, mostly below 1 cell
, and was more abundant in spring (maximum cell density of 2 cells
) when shellfish exceed the quarantine toxin level for PSP toxins in Jinhae Bay.
New record of the snake eel Ophichthus asakusae (Ophichthidae: Anguilliformes) from Korea

We describe Ophichthus asakusae from Korea based on a single specimen (426.0 mm TL) collected from the southern sea off Jeju Island. The species is characterized by a pointed, hard, sponge-like tail tip without a caudal fin and 126-132 total vertebrae. We propose a new Korean name, "Je-ju-ba-da-baem" for the species.
Distribution and Abundance of Planktonic Shrimps in the Southern Sea of Korea during 1987-1991

Distribution and abundance of planktonic shrimps were studied in the southern sea of Korea from 1987 to 1991. Nine species of five genera belonging to three families were identified. Of these, Leptochela sydniensis and Lucifer intermedius were predominant species and Lucifer penicilifer and Lucifer typus were common species. Remaining five species, Acetes chinensis, Leptochela gracilis, Lucifer chacei, Process wheeleri and Sergestes similis occurred sporadically and rarely. The distribution pattern of planktonic shrimps varied with species and sampling periods. L. intermedius occurred abundantly along inshore waters. L. penicilifer and L. typus appeared in both inshore and offshore waters, but more abundantly in offshore waters under influence of the Tsushima Current. In conclusion, the distribution patterns of planktonic shrimps may be related with hydrographic characteristics and seasonality in the southern sea of Korea.
