The Widespread Distribution of the Venomous and Poisonous Blue-lined Octopus Hapalochlaena spp., in the East/Japan Sea: Possible Effects of Sea Warming

The geographical distribution of the toxic blue-lined octopus (commonly known as the blue-ringed octopus), Hapalochlaena spp., around the East/Japan Sea was investigated. Observation records of the octopus were gathered using commercial search engines on the Internet. A questionnaire to complement and enhance the base data was conducted that targeted fishermen from areas where the octopus was most likely to occur, i.e., the southeast coast and islands of Korea in the East/Japan Sea. Overall, 32 observational records of the blue-lined octopus were found from Korea and Japan. In Korea, only one record, from 2003, was found on a website; none of the 240 fishermen who participated in the questionnaire reported seeing blue-lined octopus. However, a total of 31 observations of the blue-lined octopus from 2004 to July 2010 were found from 17 different regions in the East/Japan Sea and neighboring waters in Japan. Twenty-two cases were from coastal Honshu Island, and nine were from the west coast of Kyushu Island, Japan. The northern distributional boundary of the blue-lined octopus on the Japanese coast was off Fukui Prefecture around latitude
10' N. Our results indicate that the blue-lined octopus is distributed extensively along the Japanese coast, at a low frequency, in the East/Japan Sea.
Characterization of Edible Film Fabricated with Channel Catfish Ictalurus punctatus Gelatin by Cross-Linking with Transglutaminase

The objectives of this research were to improve the film-forming properties of Channel Catfish Ictalurus punctatus skin gelatin (CSG) by cross-linking with transglutaminase (TG), determine and optimize the TG reaction time, and characterize the mechanical and barrier properties of CSG edible film. Cross-linking of CSG was performed by TG for 0, 10, 20, 30, and 40 min at
, and the reaction was confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The color and mechanical and barrier properties of edible films fabricated with CSG cross-linked with TG were characterized. Gelatin yields from the extraction ranged from 18.2% to 23.3%. SDS-PAGE exhibited dark bands at 120 and 250 kDa, indicating successful TG-mediated cross-linking. The color of CSG film was not affected by TG cross-linking. The tensile strength of CSG films cross-linked with TG decreased from 42.59 to 21.73 MPa and the percent elongation increased from 42.92% to 76.96% as reaction times increased from 0 to 40 min. There was no significant difference in water vapor permeability of CSG films.
Gamakamide-E, a Strongly Bitter Tasting Cyclic Peptide with a Hydantoin Structure from Cultured Oysters Crassostrea gigas

A new cyclic peptide (six-membered amino acid), gamakamide-E (L-Leu-L-Met (SO)-L-Me-Phe-L-Leu-D-Lys-L-Phe), was isolated as a strongly bitter tasting compound from cultured oysters, Crassostrea gigas. The molecular formula of
was deduced from high resolution fast atom bombardment mass spectrometry (HR FAB-MS) (
= -2.4 mmu). Its unique structure including a hydantoin structure was firstly elucidated by nuclear magnetic resonance (NMR) analysis. Stereochemistries of constituent amino acids were determined by chiral high performanced liquid chromatography analysis of natural and synthesized peptides.
Metabolic Activity of Desalted Ground Seawater of Jeju in Rat Muscle and Human Liver Cells

Ground seawater in the east area of the volcanic Jeju Island contains abundant minerals. We investigated the metabolic activity of electrodialyzed, desalted ground seawater (EDSW) from Jeju in both cultured cells and animals. The addition of EDSW to the culture medium (up to 20%, v/v) reduced the leakage of lactate dehydrogenase and increased MTT activity in CHO-IR cells. EDSW (10%) promoted insulin-induced glucose consumption in L6 muscle cells as well as the activities of the liver ethanol-metabolizing enzymes, alcohol dehydrogenase and aldehyde dehydrogenase. Moreover, EDSW suppressed palmitate-induced intracellular fat accumulation in human hepatoma
cells. Activities of AMP-stimulated protein kinase and acetyl CoA carboxylase, enzymes that modulate fat metabolism, were altered by EDSW in
cells toward the suppression of intracellular lipid accumulation. EDSW also suppressed hepatic fat accumulation induced by a high-fat diet in mice. Taken together, EDSW showed beneficial metabolic effects, including the enhancement of ethanol metabolism and insulin-induced glucose consumption, and the suppression of intrahepatic fat accumulation.
An Anti-inflammatory Peptide Isolated from Seahorse Hippocampus kuda bleeler Inhibits the Invasive Potential of MG-63 Osteosarcoma Cells

Osteosarcoma is the most common primary malignancy of bone, and patients often develop pulmonary metastasis. The mechanisms underlying osteosarcoma metastasis remain to be elucidated. Recently, anti-inflammatory agents were shown to be useful in the treatment of tumor progression. We previously isolated a natural anti-inflammatory peptide from the seahorse Hippocampus kuda bleeler. Here, we examined the antitumor metastatic activity of this peptide and investigated its mechanism. The peptide significantly inhibited 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced invasive migration of human osteosarcoma MG-63 cells. Its inhibitory effect on invasive migration was associated with reduced expression of matrix metalloproteinases (MMP1 and MMP2). In addition, TPA stimulation increased intracellular reactive oxygen species (ROS) generation and small GTPase Rac1 expression, whereas the peptide decreased ROS generation and Rac1 activation. Taken together, these results suggest that the peptide inhibits invasive migration of MG-63 osteosarcoma cells by inhibiting MMP1 and MMP2 expression through downregulation of Rac1-ROS signaling.
Growth Response to a GH-Autotransgenesis in Common Carp Cyprinus carpio

Autotransgenic manipulation with a growth hormone (GH)-construct is a potential approach to improving the growth rates of farmed fish. Here, we present the generation of GH-autotransgenic common carp Cyprinus carpio carrying a transgene comprised of the carp homologous GH gene and a
-actin regulator. Autotransgenic carp showed similar viability to their non-transgenic siblings. Early growth characteristics of founder autotransgenic carp up to 50 days postfertilization were highly variable among individuals; i.e., some fish exhibited significant growth depression, while others showed dramatic acceleration of growth, achieving greater than sixfold increases in body weight relative to their non-transgenic counterparts. Stimulated growth performance became more notable with age and many transgenic individuals of the largest class reached 5 kg within 8 or 9 months, which is at least 10 times heavier than the average body weight of communally grown non-transgenics. Four of six founder transgenic males were successful in passing the transgene to their
offspring with frequencies ranging from 19 to 36%. Growth stimulations were also persistent in all
progeny groups examined.
Growth Performance of Transgenic Mud Loach Misgurnus mizolepis Carrying a GH Transgene Driven by Mud Loach C-Type Lectin Regulator

Growth hormone (GH) transgenesis in fish has the potential to improve aquaculture efficiency and capacity. However, many fast-growing transgenic fish have experienced side effects caused by excess GH expression. To overcome this unwanted issue associated with several GH transgenic mud loach Misgurnus mizolepis lines carrying GH construct driven by a strong
-actin regulator (
-actGH), we performed an alternative version of GH autotransgenesis using a weaker but more stable regulator, the mud loach lectin promoter. GH transgenesis with a pmlectGH construct consisting of the mud loach GH gene driven by the 2.3-kb lectin promoter exhibited significant growth stimulation. However, the extent of the growth acceleration in pmlectGH transgenics (six times maximum when assessed 2 months post hatching) was much less than that in transgenic individuals carrying the
-actGH construct. Additionally, the extraordinary gigantism that was common in
-actGH-transgenic mud loaches was diminished in transgenic loaches harboring the pmlectGH construct. Transgenic founders (pmlectGH) successfully transmitted their transgene into the next generation with up to 41% frequency. Growth stimulation also persisted in the transgenic F1 strains, with a seven-fold increase in maximum body weight at 6 months of age.
Genetic Variation in the Asian Shore Crab Hemigrapsus sanguineus in Korean Coastal Waters as Inferred from Mitochondrial DNA Sequences

Genetic variation in the Asian shore crab Hemigrapsus sanguineus was determined from partial mitochondrial DNA (mtDNA) sequences of the cytochrome b (Cytb) gene. Samples included 143 crabs from six localities along three coastlines in South Korea. A nucleotide sequence analysis revealed 38 variable sites in a 470-bp sequence, which defined 37 haplotypes. The haplotypes were not associated geographically and had a shallow genealogy. Pairwise
tests and a two-dimensional scaling analysis revealed no significant genetic differentiation among most of the populations. The low pairwise comparison values, but significant genetic differentiation of a northeastern population from all other populations, might have been influenced by a restriction in gene flow caused by hydrographic conditions such as ocean boundaries. The high haplotype diversity, low nucleotide diversity, and time since H. sanguineus expansion in Korean coastal waters indicate rapid population growth and a recent, sudden expansion in the Late Pleistocene.
Survival Strategies of the Rotifer Brachionus rotundiformis for Coexisting with the Copepod Apocyclops borneoensis in Laboratory Culture

Interspecific relationship between a euryhaline rotifer Brachionus rotundiformis and a cyclopoid copepod Apocyclops borneoensis was investigated in the laboratory culture. In a mixed culture of B. rotundiformis and A. borneoensis, population growth of B. rotundiformis was suppressed from day 10, while growth in a monoculture population continuously increased throughout the experimental period. However, the population growth of A. borneoensis in the mixed culture did not markedly differ from that in a monoculture population. Suppression of B. rotundiformis growth coincided with a decrease in the numbers of both non-egg-bearing and egg-bearing females, and increasing resting egg formation. Growth of A. borneoensis was not affected by the presence of the rotifer. However, relative growth index of ovisac bearing females in the mixed culture was 1.62 times higher than that in the monoculture. Presence of the copepod did not greatly reduce the food available to the rotifer population. The rotifer B. rotundiformis responded in a unique way, to stresses such as physical damage (filtering by A. borneoensis) with the production of many resting eggs to increase its chances of survival.
Comparative Studies of Bile Acid Release in the Mature Male Lampreys

A comparative study of bile acid components from four lamprey species revealed that mature male chestnut lamprey Ichthyomyzon castaneus can produce 3 keto petromyzonol sulfate (3kPZS) while mature males of Pacific lamprey Lampetra tridentata, river lamprey Lampetra fluviatilis and American brook lamprey Lethenteron appendix produce petromyzonol sulfate (PZS). Identification of 3kPZS from a group of ancient lamprey species and of PZS from recently derived species led to a speculation that differentiation of bile acid biosynthetic systems has taken place during the course of evolution. Further studies on the biological functions of different bile acids in the adult lampreys are required to understand the evolution of chemical communication in lampreys.
Effects of Dietary Nutrient on the Biological Index and Serum Chemistry of Juvenile Olive Flounder Paralichthys olivaceus Achieving Compensatory Growth

Effects of dietary nutrient content on the biological index and serum chemistry of olive flounder Paralichthys olivaceus achieving compensatory growth were investigated. Six treatments were prepared in triplicate. Fish were hand-fed with the control (C) diet twice daily for 8 weeks (8W-C) or fish were starved for 2 weeks and then hand-fed with the C, high protein (HP), high carbohydrate (HC), high lipid (HL), or intermediate protein, carbohydrate and lipid (IPCL) diets for 6 weeks, referred to as 6W-C, 6W-HP, 6W-HC, 6W-HL, and 6W-IPCL, respectively. Weight gain of fish in the 8W-C, 6W-HP, and 6W-IPCL treatments was higher than that of fish in the 6W-C treatment. Condition factor (CF) of the fish in the 6W-HP, 6W-HC and 6W-IPCL treatments was higher than that of fish in the 8W-C and 6W-C treatments. The hepatosomatic index (HSI) of fish in the 6W-HC, 6W-HL and 6W-IPCL treatments was higher than that of the fish in the 8W-C, 6W-C and 6W-HP treatments. Serum chemistry except triiodothyronine (
) was not significantly different among the treatments. In conclusion, CF and HSI of the fish could be indices reflecting compensatory growth, whereas
seemed to play a partial role in achieving compensatory growth.
In Vivo Antipyretic, Analgesic, and Anti-inflammatory Activities of the Brown Alga Ecklonia cava Extracts in Mice

Dichloromethane, ethanol, and boiling water extracts of the brown alga Ecklonia cava were examined in vivo for their antipyretic, analgesic, and anti-inflammatory activities in mice. These activities were evaluated by yeast-induced pyrexia, tail-flick test, and phorbol myristate acetate-induced inflammation (edema, erythema, and blood flow). Ethanol extract of E. cava (0.4 mg/ear) inhibited the inflammatory symptoms of mouse ear edema, erythema, and blood flow by 82.6%, 69.0%, and 65.4%, respectively. This extract also demonstrated potent analgesic activity. No acute toxicity was observed after p.o. administration of each extract (5 g/kg bw). These in vivo data are in agreement with the claims of the health care industry and indigenous medicine that E. cava is an effective remedy for inflammation-related symptoms.
Toxicity of Cryoprotectants to Gametophytic Thalli of Red Algae Porphyra yezoensis

We assessed the toxicity of cryoprotectant agents (CPAs) to gametophytic thalli of red alga Porphyra yezoensis at room temperature. The CPAs used were: dimethyl sulfoxide (DMSO), ethylene glycol (EG), glycerol (GC), 1,2-butanediol (1,2-BD), 1,3-butanediol (1,3-BD), 2,3-butanediol (2,3-BD), 1,3-propanediol (1,3-PD) and propylene glycol (PG). CPA concentrations of 10, 15, 20, 25, 30, 35, 40, 45, and 50% were employed with 30 or 60 s immersion. The toxicity of the eight CPAs to gametophytic thalli of P. yezoensis was in the order: 1,3-BD < DMSO
EG < GC < 1,3-PD
1,2-BD. All thalli were more sensitive to high CPA concentrations, and most (>75%) thalli survived exposure to 10-25% CPA for 60 s. These data will facilitate selection of the optimal cryoprotectant concentration for cryopreservation of P. yezoensis thalli.
Isolation and Characterization of Flavobacterium johnsoniae from Farmed Rainbow Trout Oncorhynchus mykiss

Flavobacterium johnsoniae was isolated from farmed rainbow trout Oncorhynchus mykiss in Korea, and its biochemical and molecular characterization was determined. Yellow-pigmented bacterial colonies were isolated from 18 of 64 fish samples (28.1%) on trypticase soy agar plates, and their biochemical profiles were characterized by API 20E and API 20NE test kits. F. johnsoniae was identified by biochemical phenotyping of factors including rapid gliding motility, Gram-negative condition, oxidase- and catalase-positive status, Congo red absorption, nitrate reduction,
-galactosidase production, acid production from glucose, and gelatin and casein hydrolysis. PCR and subsequent sequencing of 16S rRNA confirmed that the yellow-pigmented colonies were most similar to F. johnsoniae. The alignment analysis of 16S rRNA sequences also showed that all 18 rainbow trout isolates had highly similar homologies (97-99% identity). One isolate was selected and named FjRt09. This isolate showed 98% homology with previously reported F. johnsoniae isolates, and in phylogenetic analysis was more closely grouped with F. johnsoniae than with F. psychrophilum, F. columnare, or F. branchiophilum. This is the first report on the occurrence and biochemical characterization of F. johnsoniae isolated from rainbow trout in Korea.
