Future Prospects and Health Benefits of Functional Ingredients from Marine Bio-resources: A review

The marine ecosystem represents a vast and dynamic array of bio-resources attributed with its huge diversity and considered as potential untapped reservoirs for the development of functional foods for future health markets. Basically, marine microorganisms, sponges, algae, invertebrates such as crustaceans and mollusks along with marine fish species can be considered as marine bio-resources, which can be utilized to obtain different health benefits for humans, directly or after processing. Most of the bio-molecular components, such as lipids and proteins from these marine bio-resources, which can be extracted in large scale using the modern and advanced biotechnological approaches, are suitable drug candidates for the pharmaceutical industry as well as functional food ingredients for the food industry. Moreover, the furtherance of high throughput molecular biological techniques has already been incorporated with identification, mining and extraction of molecular components from marine bio-resources. In this review, potential marine bio-resources with respect to their extractable bio-molecules were described in details, while explaining the present and prospective methods of identification and extraction, which are integrated with advanced techniques in modern biotechnology. In addition, this provides an overview of future trends in marine biotechnology.
The Effect of Cryoprotectants on the Properties of Pacific Sand Lance Ammodytes personatus Girard Surimi During Frozen Storage

We investigate the effects of cryoprotectant mixtures on the quality of sand lance surimi (SLS) during storage at
. We monitored freeze-induced denaturation of myofibrillar protein in SLS and examined the texture profile of SLS gel. Freeze-induced denaturation was assessed by evaluating SLS
-ATPase activity. SLS gels prepared with sorbitol or sucrose and a mixture of both as cryoprotectant. Higher concentrations of cryoprotectants resulted in significantly higher residual SLS
-ATPase activity at the same storage time (P < 0.05). Residual
-ATPase activity of SLS prepared with sorbitol was higher than that of sucrose when cryoprotectant concentration and storage period were same. A blend of sorbitol and sucrose resulted in a stronger cryoprptective effect of SLS myofibrillar protein than did sorbitol or sucrose alone. The presence of a phosphate compound in SOP (3% sorbitol + 0.2% phosphate compound) resulted in higher SLS
-ATPase activity than that of did 5% sorbitol. The hardness, brittleness, and elasticity values and a folding test of the SLS gels were significantly affected by cryoprotectant concentrations and the storage time. Preference scores and acceptance for texture in a sensory evaluation of the SLS gels increased with increasing sorbitol or sucrose concentration.
Physicochemical Properties of Gelatin from Jellyfish Rhopilema hispidum

The objective of this study was to elucidate the physicochemical characteristics of gelatin extracted from jellyfish Rhopilema hispidum. We investigated the proximate composition, amino acids, gel strength, gelling/melting points, dynamic viscoelastic properties, and viscosity of jellyfish gelatin. Jellyfish gelatin contained 12.2% moisture, 1.5% lipid, 2.1% ash, and 84.8% protein. Glycine, hydroxyproline, proline, and alanine were the predominant amino acids. The gelatin showed a gel strength of 31.2 kPa, a gelling point of
, and melting point of
. The gelatin was composed of
-chain. During cooling and heating process, jellyfish gelatin showed lower elastic modulus (G') and loss modulus (G") values than mammalian gelatin. Jellyfish gelatin did not show superior rheological properties to mammalian gelatin, like other fish gelatin; however, it can be used in various food and cosmetic products not requiring high gel strength.

Dieckol is a polyphenol compound isolated from brown algae that has anti-oxidant, anti-inflammatory, and anti-tumor activity. We examined the anti-angiogenic effects of dieckol in endothelial cells under hypoxic conditions. Treatment with
, a hypoxic mimetic agent, increased proliferation, adhesion, migration, and tube formation in HUVECs, as well as vessel sprouting in rat aortic rings, which correlated well with increased expression of hypoxia-inducible factor 1-alpha (

-induced adhesion, migration, and tube formation in HUVECs and vessel sprouting in rat aortic rings. Dieckol treatment decreased

-integrin/Fak, Akt/eNOS, and p38 MAPK. These results suggest that dieckol is a novel angiogenesis inhibitor and a potential treatment for angiogenesis-dependent diseases in humans, such as malignant tumors.
Bioaccumulation of Heavy Metals in the Mussel Mytilus galloprovincialis in the Changseon area, Korea, and Assessment of Potential Risk to Human Health

From 2008 to 2013, the mussel Mytilus galloprovincialis were collected from a major area of mussel production (Changseon area), which is a designated shellfish-cultivating area for export, located on the southern coast of Korea. The samples were analyzed for mercury (Hg) using a direct Hg analyzer and for other metals, such as cadmium (Cd), lead (Pb), arsenic (As), chromium, copper, nickel, and zinc, using inductively coupled plasma mass spectrometry. The concentrations and bioaccumulation of the heavy metals were determined, and a potential risk assessment was conducted to evaluate their hazards towards human consumption. The concentration and bioaccumulation ratio of Cd were the highest of the three hazardous metals (Cd, Pb, and Hg). The concentrations of hazardous metals in all samples were within the limits set by Korea and other countries. The estimated dietary intake (EDI) was compared to the provisional tolerable daily intake (PTDI) adopted by the Joint FAO/WHO Expert Committee on Food Additives and the U.S. Environmental Protection Agency. The EDIs of all heavy metals tested for mussel samples ranged from 0.01 to 4.99% of the PTDI; the highest value was measured for As. The hazard index (HI) can be used to assess the risk of heavy metal consumption associated with contaminated food. The HI for all samples was far less than 1.0, which indicates that the mussels produced in the Changseon area do not represent an appreciable hazard to humans and are fit for consumption.
Effects of the Dietary Microbial Phytase Supplementation on Bioavailability of Phosphorus in Juvenile Olive Flounder Paralichthys olivaceus Fed Soybean Meal based Diets

An 8 weeks feeding trial was carried out to evaluate the effects of dietary microbial phytase (P) supplementation on nutrient digestibility, and body composition in juvenile olive flounder Paralichthys olivaceus fed soybean meal-based diets. Seven experimental diets were formulated to be isonitrogenous and isocaloric to contain 50.0% crude protein (CP) and 16.7 kJ of available energy/g with or without dietary phytase supplementation. White fish meal (FM) provided 92.4% of the total protein in the basal diet (
), in the other 6 diets, 30% or 40% FM protein was replaced by soybean meal: 70% FM + 30% soybean meal (
); 70% FM + 30% SM + 1000 U phytase/kg diet (
); 70% FM + 30% SM + 2000 U phytase/kg diet (
); 60% FM + 40% SM (
); 60% FM + 40% SM + 1000 U phytase/kg diet (
); and 60% FM + 40% SM + 2000 U phytase/kg diet (
). After two weeks of the conditioning period, triplicate groups of 25 fish initially averaging
) were randomly distributed into the aquarium and were fed one of the experimental diets for 8 weeks. After feeding trial, supplementation of phytase significantly improved the apparent digestibility coefficients of phosphorus in flounder diets (P<0.05) containing 30% and 40% soybean meal regardless the levels. However, phytase had no significant influence on growth performance and whole body composition of fish. Based on the experimental results, we conclude that dietary supplementation of phytase could improve the apparent digestibility coefficient of phosphorus in olive flounder.
The Functional Relevance of Prepro-melanin Concentrating Hormone (pMCH) to Skin Color Change, Blind-side Malpigmentation and Feeding of Oliver Flounder Paralichthys olivaceus

To assess the functional structure of prepro-melanin-concentrating hormone (pMCH), we isolated and cloned pMCH (of-pMCH) mRNA from the brain of the olive flounder, Paralichthys olivaceus, and compared its amino acid sequence with those from other animals. In addition, to examine whether activation of the brain of-pMCH gene is influenced by background color, density, and feeding, we compared pMCH mRNA activities against different background colors (bright and dark) and at different densities (100% PCA and 200% PCA). To examine whether the pMCH gene is related with malpigmentation of blind-side skin and appetite, we compared pMCH gene expression between ordinary and hypermelanic flounders, and between feeding and fasting flounders. The of-pMCH cDNA was 405 bp in the open reading frame [ORF] and encoded a protein of 135 amino acids; MCH was 51 bp in length and encoded a protein of 17 amino acids. An obvious single band of the expected size was obtained from the brain and pituitary by RT-PCR. In addition, of-pMCH gene activity was significantly higher in the bright background only at low density (< 100% PCA) making the ocular skin of fish whitening, and in ordinary fish. However, the gene activity was significantly decreased in dark background, at high density (>200% PCA), and in hypermelano fish. These results suggest that skin whitening camouflage of the flounder is induced by high MCH gene activity, and the density disturbs the function of background color in the physiological color change. Moreover, our data suggest that a low level of MCH gene activity may be related to malpigmentation of the blind-side skin. In feeding, although pMCH gene activity was significantly increased by feeding in the white background, the pMCH gene activity in the dark background was not influenced by feeding, indicating that the MCH gene activity increased by feeding can be offset by dark background color, or is unaffected by appetite. In conclusion, this study showed that MCH gene expression is related to ocular-skin whitening camouflage and blind-skin hypermelanosis, and is influenced by background color and density.
Development of Clamping Probe for Rare DNA Detection using Universal Primers

PCR amplification with universal primer is a useful tool for speciation of symbionts in marine eukaryote coupled with robust separation method such as denaturing high performance chromatography (DHPLC). To overcome the biased amplification, clamping PCR is recommended to suppress the amplification of host gene. In this study, we evaluated the efficiency of rare gene detection for two kinds of clamping probes which were successfully utilized for eukaryotic symbiont analysis: C3 linked nucleotide (C3) and peptide nucleic acid (PNA). PNA was 3-4 orders of magnitude higher than that of C3 tested in clamping efficiency and rare gene detection. This represented that PNA could be a more competent clamping probe for the enhancement of PCR amplification for rare symbiont genes.
Combined Effects of Copper and Temperature on Antioxidant Enzymes in the Black Rockfish Sebastes schlegeli

Copper has been widely used to control algae and pathogens in fish culture ponds. However, its toxic effects on fish depend not only on its concentration in the water but also on the water quality. A laboratory experiment was conducted to assess copper toxicity in the black rockfish Sebastes schlegeli using a panel of antioxidant enzymes, including glutathione (GSH), glutathione S-transferase (GST), glutathione peroxidase (GPx), glutathione reductase (GR) and superoxide dismutase (SOD), at different levels of copper at three water temperatures (WT, 18, 23,
) for 4 days. After exposure to two copper concentrations (100 and
), GSH levels and GST activities increased significantly, depending on WT (P < 0.05) in the liver, gill, and kidney of the black rockfish. GPx and SOD activities decreased significantly with both increasing WT and copper treatment in the organs of black rockfish (P < 0.05). These changes can be seen as initial responses to temperature stress and as a sustained response to copper exposure. This also indicates that GSH and related enzymes activities were sensitive indexes to stress by toxicants such as copper. The present findings suggest that simultaneous stress due to temperature change and copper exposure can accelerate changes in enzymes activities in the black rockfish. This provides another example of synergism between environmental temperature and pollutants, which may have important implications for the survival of fish in polluted environments during seasonal warming and/or global climate change.
First Record of Two Siriella Species of Mysids (Crustacea: Mysida) from the Korean Waters

Two pelagic species Siriella izuensis Murano and Fukuoka, 2008 and Siriella japonica Ii, 1964, are reported for the first time from the Korean waters, together with descriptions and illustrations. S. izuensis can be distinguished from its closest relative, Siriella japonica, by the normal inner flagellum of the antennule of males, as opposed to the swollen proximal part in S. japonica, and by the slightly more distinct tendency of alternate arrangement of spines on the distal third lateral margin of the telson than in S. japonica. S. izuensis and S. japonica differ distinctly from other related species by a combination of the triangular rostral plate, the moderate size of the eyes, the form and arrangement of the lateral spines of the telson, and six or seven graded spines on the distal outer margin of the uropodal exopod. A taxonomical key is given for the genus Siriella from the Korea waters.
Target Strength Measurements of Live Golden Cuttlefish Sepia esculenta at 70 and 120 kHz

Cuttlefish Sepia esculenta are commercially important in Korea. Assessments of their biomass currently depend on fishery-landings data, which may be biased. Towards fishery-independent acoustic surveys of cuttlefish, target strength (TS) measurements at 70 and 120 kHz were made of 23 live cuttlefish, in early May 2010. The fish were caught by traps in the inshore waters around Geojedo, Korea. The TS were measured using split-beam echosounders (Simrad ES60 and EY500, respectively). The cuttlefish mantle lengths (L) ranged from 15.6 to 23.5 cm (mean L=17.8 cm) and their masses (W) ranged from 335 to 1020 g (mean W=556.1 g). Their mean TS values at 70 and 120 kHz were -33.01 dB (std=1.39 dB) and -31.76 dB (std=2.15 dB), respectively. The mean TS at 70 kHz was 0.17 dB higher than the TS-length relationship resulting from a least-squares fit to the data (
= 0.52, N=23). The mean TS at 120 kHz was 0.45 dB higher than the fitted TS-length relationship (
= 0.58, N=23). The differences between the mean TS values and an equation regressed from all of the TS measurements at both frequencies (
= 0.86, N=46) was 0.22 dB at 70 kHz and 0.31 dB at 120 kHz, respectively.
Overview of the Applications of Hydroacoustic Methods in South Korea and Fish Abundance Estimation Methods

I provide an overview of the application of hydroacoustic methods in South Korea to understand the current research status in relation to fisheries acoustics and to determine which areas require further study. One main purpose for using a scientific echosounder, a representative tool using the hydroacoustic method, is to evaluate the abundance of fisheries resources. Thus, two representative methods for abundance estimation are described. The history of fisheries acoustics worldwide is also summarized.
Simultaneous and Systemic Knock-down of Big Defensin 1 and 2 gene Expression in the Pacific Oyster Crassostrea gigas using Long Double-stranded RNA-mediated RNA Interference

RNA interference (RNAi)-mediated transcriptional knock-down of Crassostrea gigas big defensin 1 and 2 genes (Cg-BigDef1 and Cg-BigDef2) was investigated. The cDNA sequences of Cg-BigDef1 and Cg-BigDef2 were identical, excluding an additional fragment of 20 nucleotides in Cg-BigDef1; thus, a long double-stranded RNA (dsRNA) targeting the mRNA of Cg-BigDef2 effectively downregulated both Cg-BigDef2 and Cg-BigDef1. In addition, long dsRNA targeting green fluorescent protein (GFP) did not affect transcription of the two big defensin genes. These results suggest that the transcriptional downregulation of Cg-BigDef1 and Cg-BigDef2 was mediated by sequence-specific RNA interference (RNAi). Despite injection of long dsRNA targeting Cg-BigDef2 into only the adductor muscle, knock-down of Cg-BigDef1 and Cg-BigDef2 was observed in the adductor muscle, hemocytes, mantle, and gills, suggestive of systemic spread of RNAi in C. gigas. Furthermore, the inhibitory effect of dsRNA persisted until 72 h post-injection, indicative of a long-lasting RNAi-mediated knock-down of target genes.
A New Record of a Spot-fin Porcupinefish, Diodon hystrix (Diodontidae, Pisces) off Jeju Island, Korea

A single specimen belonging to the family Diodontidae was collected from Seo-gwi-po, Jeju Island, Korea in July 2013 and was identified as Diodon hystrix Linnaeus, 1758, a species new to the Korean fish fauna. The species is characterized by 14 dorsal fin rays, 14 anal fin rays, spines on the dorsal and ventral surfaces of the caudal peduncle, and many small black spots on all fins except the anal fin. The most similar species, Diodon holocanthus, is well distinguished from D. hystrix by the absence of black spots on the dorsal and caudal fins, and the lack of a spine on the dorsal surface of the caudal peduncle. Accordingly, we describe herein the morphological traits of D. hystrix and suggest its new Korean name, "Jan-jeom-bak-i-ga-si-bok".
Initial Culture Conditions for Primary Cell Populations Derived from Radula Tissue in Abalone Haliotis discus hannai

Abalone immortal cell lines can be used to study the physiological properties and disease mechanisms of abalone at the cellular and molecular level. As a first step for the final goal to establish abalone immortal cell lines, we examined various initial culture conditions for primary cell populations derived from Haliotis discus hannai radula tissue. The survival rate after cell isolation procedures using the enzymatic method was as low as
. Based on three different experimental conditions for H. discus hannai radula-derived cell culture, we found that the salinity of the media and the presence of growth-promoting factors were important to support radula-derived primary cell populations during the initial culture. The growth factor-containing media adjusted to 35 psu salinity could induce 100% (8 out of 8 trials) initial cell attachment, and the rate of cell attachment reached 50-70%. The data obtained from this study will provide useful information for developing immortal cell lines from abalone species.
