Synergistic Antibacterial Activity of Ecklonia cava (Phaeophyceae: Laminariales) against Listeria monocytogenes (Bacillales: Listeriaceae)

In an effort to discover alternative antimicrobials against Listeria monocytogenes, several marine algae were screened. The methanolic extract of Ecklonia cava exhibited the highest antibacterial activity against L. monocytogenes, with the ethyl acetate (EtOAc) soluble fraction of E. cava methanolic extract having a MIC value of
and a MBC value of
. The MIC values of streptomycin in combination with the EtOAc fraction were markedly reduced up to 64-fold, suggesting that the antibacterial activity of the antibiotic was restored when combined with the EtOAc fraction. The interaction between streptomycin and the EtOAc fraction was assessed by fractional inhibitory concentration (FIC) indices. The combination of streptomycin and the EtOAc fraction against L. monocytogenes resulted in

Evaluation of the Quality of Canned Seafood with Added Spice-oil Extract

The influence of spice (cinnamon, allspice, black pepper)-oil extract on canned seafood quality was studied. During the processing of canned seafood, the substitution of spice-oil extract for vegetable oil (refined sunflower, corn, soybean and olive oil) resulted in a decrease in the heat resistance of spore microorganisms, making it possible to reduce the duration of sterilization for canned food to 5-10 min at
. This reduction in the sterilization duration of canned seafood with spice-oil extract inhibited residual microflora in the product, thus reducing the deleterious effect of heating on the main food compounds while preserving protein digestibility.
Nutritional Value and Bioactive Properties of Enzymatic Hydrolysates prepared from the Livers of Oncorhynchus keta and Oncorhynchus gorbuscha (Pacific Salmon)

Calculated chemical scores (computed in relation to the FAO/WHO reference protein) for salmon liver protein hydrolysates indicated that all amino acids (other than methionine and threonine) were present in adequate or excess quantities; thus, the raw liver material is a good source of essential amino acids. The hydrophobic amino acids contents in hydrolysates prepared from Oncorhynchus keta and O. gorbuscha were 38.4 and 39.1%, respectively. The proportion of released peptides exceeding 500 kDa was reduced when hydrolysates were treated with the commercial enzyme Alcalase, although proportions in the following MW ranges were elevated: 100-500 kDa and <50 kDa. The optimal conditions for enzymatic hydrolysis were as follows: pH 7.0,
, and a reaction time of 1 h. Of the different proteases tested, Alcalase was the most efficient for production of salmon liver hydrolysate with the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity. The hydrolysates prepared from salmon liver had a balanced amino acid composition. The liver protein hydrolysates contained low molecular weight peptides, some of which may be bio-active; this bio-active potential should be investigated. Inhibition of the DPPH radical increased with increased degree of hydrolysis (DH), regardless of protease type. DPPH radical scavenging abilities, antithrombotic effects and
-glucosidase enzyme inhibition effects of O. keta liver hydrolysate increased in a dose-dependent manner. Thus, salmon liver hydrolysate may be useful in functional food applications and as a source of novel products.
Thermal Oxidative Stability of Various Vegetable Oils used for the Preparation of the Seasoned Laver Pyropia spp.

Seasoned laver Pyropia spp. is a traditional Korean seafood that has gained popularity worldwide because of its unique taste, texture, and health benefits. It is prepared by roasting a sheet of dried laver, to which vegetable oils have been applied, at an ultra-high temperature (UHT) of
. Therefore, the oxidative stability of the oils is the most important factor in determining the shelf life of seasoned laver products. In this study, we investigated changes in the thermal oxidative stability of six major vegetable oils (sesame, perilla, sunflower, rice bran, canola, and olive) during the seasoned laver processing. The oxidation induction time of each oil from the seasoned laver products was decreased compared with the fresh oil. These results indicate that the UHT treatment (
, 10 s) induced thermal oxidation of the oils. Among the six seasoned laver oils, the induction times of olive (OL, 8.02 h) and sesame (SE, 5.31 h) oils were significantly higher than the other oils. The acid values (AVs) of OL and SE oils from the seasoned laver were 0.49 and 0.79, respectively. On the other hand, perilla oil had the overall worst thermal oxidative properties (induction time: 0.35 h, AV: 2.82). Our results provide useful information about seasoned laver products for researchers or manufactures.
Monitoring and Optimization of the Effects of the Blending Ratio of Corn, Sesame, and Perilla Oils on the Oxidation and Sensory Quality of Seasoned Laver Pyropia spp.

Seasoned laver Pyropia spp. is one of the most well-known Korean traditional seafoods, and is becoming more popular worldwide. Various mixed oils are used in the preparation of seasoned laver; however, there is no information available regarding the effects of the blending ratio of oils on the quality of seasoned laver. In this study, the effects of the blending ratio of corn, sesame, and perilla oils on the oxidation and sensory quality of seasoned laver were monitored and optimized using a response surface methodology. An increase in the proportion of corn and sesame oils resulted in an excellent oxidation induction time, whereas a high ratio of perilla oil reduced the thermal oxidative stability of the mixed oil. In the sensory test, the seasoned laver with the highest proportion of sesame oil was preferred. The optimal blending ratio (v/v) of corn, sesame, and perilla oils for both oxidation induction time (
) and sensory score (
) was 92.3, 6.0, and 1.7%. Under optimal conditions, the experimental values of
points, and were similar to the predicted values (4.34 h and 5.13 points). Our results for the monitoring and optimization of the blending ratio provide useful information for seasoned laver processing companies.
Evaluation on Anticancer Effect Against HL-60 Cells and Toxicity in vitro and in vivo of the Phenethyl Acetate Isolated from a Marine Bacterium Streptomyces griseus

We previously identified Streptomyces griseus as an anti-cancer agent (Kim et al., 2014). In this study, we isolated compounds from S. griseus and evaluated their anticancer effect and toxicity in vitro and in vivo. Preparative centrifugal partition chromatography (CPC) was used to obtain three compounds, cyclo(
-Pro) and phenethyl acetate (PA). We chose PA, which had the highest anticancer activity, as a target compound for further experiments. PA induced the formation of apoptotic bodies, DNA fragmentation, DNA accumulation in
phase, and reactive oxygen species (ROS) formation. Furthermore, PA treatment increased Bax/Bcl-xL expression, activated caspase-3, and cleaved poly-ADP-ribose polymerase (PARP) in HL-60 cells. Simultaneous evaluation in vitro and in vivo, revealed that PA exhibited no toxicity in Vero cells and zebrafish embryos. We revealed, for the first time, that PA generates ROS, and that this ROS accumulation induced the Bcl signaling pathway.
Effects of Salinity Changes on Hematological Responses in Juvenile Ship Sturgeon Acipenser nudiventris

A 10-day trial was conducted to evaluate the salinity tolerance and hematological responses of juvenile ship sturgeon Acipenser nudiventris under different salinity levels. A total of 240 fish averaging
SD) were randomly distributed into 12 tanks in groups of 20, and each tank was then randomly assigned to one of three replicates of four different salinities at 0, 4, 8 and 12‰. At the end of 10 days of experimental period, plasma protein value of fish exposed to 0‰ and 4‰ salinities was significantly higher than those of fish exposed to 8‰ and 12‰ salinities (P < 0.05). Significantly higher values for plasma cortisol and glucose were obtained in fish exposed to 12‰ salinity compared to the fish exposed to 0‰ and 4‰ salinities (P < 0.05). Plasma potassium and sodium values increased significantly by increment of salinity levels (P < 0.05). Therefore, these results indicated that in natural environments, juvenile ship sturgeon, A. nudiventris in migratory populations could be able to migrate and adjust successfully into brackish water with a salinity of up to 8‰ without any short-term hematological stress responses.
Effects of Photoperiod Manipulation on Growth Performance and Hematological Responses of Juvenile Caspian Roach Rutilus rutilus caspicus

A 8-week trial was conducted to evaluate the effects of photoperiod manipulation on the growth performance and hematological parameters of juvenile Caspian roach, Rutilus rutilus caspicus (average weight
SD) reared under five photoperiods (24 h Light, 24L; 18 h Light & 6 h Dark, 18L:6D; 12 h Light & 12 h Dark, 12L:12D; 6 h Light & 18 h Dark, 6L:18D; 24 h Dark, 24D) with constant light intensity 1,500 lx on the water surface. Triplicate of 20 fish were allocated into each of 15 fiberglass tanks of 50 L capacity and they were fed three times per day with the commercial feed (SFK, Co., Sari - Iran) contains 50.0% protein and 10.5% lipid. At the end of experimental period, final body weight, weight gain and specific growth rates of fish exposed to 24L were significantly higher than those of fish exposed to 12L:12D, 6L:18D and 24D (P < 0.05). Red blood cell and hemoglobin of fish exposed to 24L were significantly higher than those of fish exposed to 24D. No significant difference observed in hematocrit, white blood cell and plasma glucose among the different treatments groups. Therefore, these results demonstrated that the growth performance of juvenile Caspian roach can be significantly stimulated by using 24L and 18L:6D photoperiods without any measurable significant stress response such as plasma glucose concentration.
Effects of Rearing Density on Growth of the Polychaete Rockworm Marphysa sanguinea

Effects of rearing density on growth and survival of the polychaete rockworm Marphysa sanguinea have been investigated in order to develop rearing techniques for this species. This study was examined over a nine-month period in the Fisheries Science and Technology Center of Pukyong National University. Three rockworm densities, 500, 1,000 and
with weight ranges of < 0.5 g, 0.6-1.5 g, and 1.6-2.5 g, and the no feed control treatment, were stocked in triplicate
boxes with sand bottoms. Growth rates were checked with 15 randomly sampled rockworms from each box at months 3, 6 and 9. Results showed that SGRs in all treatments were higher during the first period (0-3 months) than the second (3-6 months) and third periods (6-9 months) for all treatment densities, while SGRs decreased with increasing density. However, survival and growth of worms at high density was not better than low density, but daily biomass production in medium and high density groups was

. Results showed that M. sanguinea can be one of the most suitable species to commercially exploit in a farming system. In particular, specified densities permit elevated pure production.
Expression Patterns of the chgH:rfp Transgene in Response to 17α-Ethinylestradiol (EE2) Exposure in Marine Medaka Oryzias dancena

The functional utility of a transgenic marine medaka Oryzias dancena strain carrying the red fluorescent protein (RFP) gene driven by an endogenous choriogenin H (chgH) promoter was evaluated for its ability to detect waterborne
-ethinylestradiol (EE2), a synthetic estrogen derivative. The chgH:rfp transgenic marine medaka larvae showed an age-dependent tendency in the efficiency of EE2-mediated transgene expression, in which transgenic larvae older than 6 days post-hatching displayed a more effective response in their transgene expression to EE2 than did younger hatchlings. During experimental exposures to high concentrations of EE2 (200 to 1,000 ng/L), the transgenic responses in the hatchlings were broadly dose- and duration-dependent. With exposures using lower doses of EE2 (25, 50 and 100 ng/L), EE2-induced transgenic RFP was also observed in the transgenic larvae, although the lower doses required exposure of longer duration. Under the EE2 exposure and microscope assay conditions used in our study, transgenic marine medaka larvae exhibited a similar degree of EE2-mediated RFP phenotype expression at various salinity levels (0, 15 and 30 ppt).
Concatemer-Associated Transgene Expression Patterns in Transgenic Marine Medaka Oryzias dancena Strains

To examine the interrelationship between transgenic insertion patterns and transgene expression profiles in established transgenic fish lines, four stable transgenic marine medaka Oryzias dancena germlines harboring
-actin regulator-driven RFP reporter constructs were selected. The established transgenic strains were characterized with regard to their transgenic genotypes (insertion pattern, concatemer formation, and transgene copy number based on genomic Southern blot hybridization and qPCR assay) and expression characteristics at the mRNA (qRT-PCR), protein (western blot), and phenotypic (fluorescent appearance) levels. From comparative examinations, it was found that transgenic expression at both the transcription and translation levels could be significantly downregulated in transgenic strains, potentially through methylation-mediated transgene silencing that was particularly associated with the formation of a long tail-to-head tandem concatemer in the chromosomal integration site(s). When this occurred, an inverse relationship between the transgene copy number and fluorescence intensity was observed in the resultant transgenic fish. However, with the other transgenic genotype, transgenic individuals with an identical Southern blot hybridization pattern, containing a tandem concatemer(s), had very different expression levels (highly robust vs. low expression strengths), which was possibly related to the differential epigenetic modifications and/or degrees of methylation. The concatemer-dependent downregulation of transgene activity could be induced in transgenic fish, but the overall pattern was strain-specific. Our data suggest that neither a low (or single) transgene copy number nor tandem transgene concatemerization is indicative of strong or silenced transgene expression in transgenic fish carrying a ubiquitous transgene. Hence, a sufficient number of transgenic lineages, with different genotypes, should be considered to ensure the establishment of the best-performance transgenic line(s) for practical applications.
Factors Affecting Primary Cultures of Abalone Haliotis discus hannai Ovary-dissociated Cells and General Culture Aspects

We investigated factors affecting primary cultures of Pacific abalone Haliotis discus hannai ovary-dissociated cells to identify general aspects of their early-phase culture. Ninety-seven cell populations derived from 30 individuals were cultured in different media with varying compositions of medium supplements, and initial attachment, subculture, and survival for
weeks were assessed according to medium composition and individual. We also examined the time required for subculture and the rate of cell death according to both culturing period and passage number within 10 weeks. A lack of fetal bovine serum (FBS) and hemolymph significantly inhibited the growth of cultured cells, while we detected no significant effect of medium composition on initial cell attachment. Through data reallocation, with the omission of data from cell populations cultured in FBS-free and hemolymph-free media, we showed that growth inhibition was also affected by individual differences among the abalones used. During the culture, we observed four different types of cell morphology. Moreover, considerable time was required for subculture-18.4 and 19.5 days for first and second subcultures, respectively-and cell death did not occur within 30 days or for passage 0. Our results will provide valuable information for developing universal cell culturing guidelines in abalone species and suggest the feasibility of culturing abalone ovary-dissociated cells.
Magnetic Nanoparticles as Carrier of Cisplatin for Drug Delivery

A synthesis method for a chitosan-coated magnetic drug-delivery system of cisplatin is proposed. Here, cisplatin was conjugated to the surface of Magnetite (
) nanoparticles via a (3-Aminopropyl)-trimethoxysilane (APTS) coupling agent. To reduce the cytotoxic effect of cisplatin, the magnetic drug was then encapsulated in chitosan (CS-cisplatin-
) through the water/oil (W/O) emulsion method. The CS-cisplatin-
nanoparticles were synthesized in a spherical shape with a diameter of 190 nm. The cytotoxicity assay was performed using HeLa cells. The cisplatin uptake of the cells was determined using High Performance Liquid Chromatography (HPLC) to calculate the drug content. The controlled release of cisplatin was demonstrated by regulating the dissolution and diffusion of the drug through the chitosan matrix.
A Natural Hybrid of Intergeneric Mating between a Female Pungtungia herzi and a Male Pseudorasbora parva (Cypriniformes: Cyprinidae)

A natural hybrid of a probable intergeneric mating between the striped shiner Pungtungia herzi and the stone morocco Pseudorasbora parva (Cypriniformes: Cyprinidae) was captured in the Geumho River, a tributary of the Nakdong River basin in Korea. Morphological characters and DNA sequences were analyzed to verify its hybrid state and identify the parentage of its parent species. The hybrid exhibited a phenotypic intermediacy between the two parent species in the number of vertebrae and the mouth shape. Out of 1,488 base pair (bp) positions of the nuclear recombination activating gene 1 gene (rag1), which has a biparental mode of inheritance, 41-bp substitutions were detected between the two parent species, whereas an electropherogram of the hybrid displayed polymorphic double peaks at all of the base positions, along with one additional one, strongly indicating its hybrid state. Meanwhile, sequence comparison of the mitochondrial cytochrome b gene (mt-cyb) (1,140 bp), which has a maternal mode of inheritance, showed only 5-22-bp differences (97.6-99.5% identities) between the hybrid and Pu. herzi, but as many as 158-168-bp differences (85.2-86.1% identities) between the hybrid and Ps. parva, clearly indicating Pu. herzi as the maternal species. Thus, combined analyses of independent data sets (i.e., morphology and nuclear and mitochondrial DNA sequences) offered convincing evidence for the hybrid state of a naturally occurring hybrid resulting from intergeneric mating between a female Pu. herzi and a male Ps. parva.
New Record of Scolecenchelys fuscogularis (Anguilliformes: Ophichthidae) Leptocephali from Korea, as Revealed by Morphological and Molecular Analyses

Three leptocephali (22.2, 22.7, 56.0 mm in total length) collected from the East/Japan Sea were identified by morphological and genetic analyses as belonging to the genus Scolecenchelys (Anguilliformes, Ophichthidae). Morphologically, the specimens were characterized by 148-158 myomeres, 10 gut swellings, dorsal fin origin above middle of the body, and 6 postanal melanophores between the anus and the caudal margin. An analysis of an 849-base pair 12S rRNA sequence of mitochondrial DNA showed that sequences are concordant with those of adult Scolecenchelys fuscogularis (genetic distance = 0.001). Furthermore total number of myomeres is consistent with the total number of vertebrae in adult S. fuscogularis. This study provides the first description of the morphological characteristics of S. fuscogularis leptocephali and their variations with size. The Korean name of S. fuscogularis is "Ga-neun-mul-baem", established by Ji et al. (2012).
