Anti-Oxidation of Cultured Cordyceps militaris Growing on Silkworm Pupa

Cordyceps, one of the well-known traditional Chinese medicines, consists of the dried fungus C. sinensis growing on the larva of caterpillar. It is commonly used as a tonic of restoring vital body functions. In order to evaluate the pharmacological effects of C. militaris, the anti-oxidation and antiaging effect of cultured C. militaris growing on pupa of silkworm Bombyx mori, were investigated. The results showed! that the contents of lipofusin in myocardium were reduced and the SOD activity in the liver were elevated in a dose-dependent manner while no effect was observed for blood GSH-Px activity and liver MDA content in mice perfused with the cultured C. militaris compared to those untreated mice. Furthermore, the average life span, maximum life span and 50% death time of the fruit flies treated with cultured C. militaris were markedly prolonged. The results suggest that the cultured C. militaris growing on pupa of silkworm possesses anti-oxidation and delay senescence activity.
Serichlor, A New Disinfectant in Indian Sericulture

Silkworm diseases are better prevented than cured. Disinfection and hygiene are the two important aspects in silkworm rearing to prevent the diseases. Suitable disinfectant is the primary need to disinfect the rearing house, its surroundings and appliances to eliminate the persistent pathogens from the rearing environment. In this direction, Serichlor, a new disinfectant in Indian Sericulture marketed as Serichlor-60 (contains 60,000 ppm of chlorine dioxide) and Serichlor-20 (contains 20,000 ppm of chlorine dioxide) has been evaluated for its germicidal effect against the pathogens of silkworm, viz., spores of Nosema bombycis, Bacillus thuringiensis, polyhedra of BmNPV and conidia of Beauveria bassiana both in vitro and in vivo. Results indicated that high concentration (2,500 ppm of chlorine dioxide) is required to kill all the pathogens at 100% level. The efficacy of the Serichlor was greatly enhanced by the addition of 0.5% slaked lime solution. 500 ppm of chlorine dioxide in 0.5% slaked lime solution was found effective against all the pathogens tested. This concentration of disinfectant was also found effective for disinfection of rearing house, rearing appliances and silkworm egg surface. The disinfectant is stable, non hazardous, least corrosive and most suitable for Indian Sericulture.
RNA Interference to Prevent Bombyx mori Nuclear Polyhedrosis Virus Infection in Vivo

RNA interference has been used as a powerful tool in preventing virus proliferation in many species. In this study, we injected the dsRNA in vitro transcripts into Bombyx mori to investigate the resistance to B. mori nuclear polyhedrosis virus (BmNPV). Through vivisectional observation and real-time quantities PCR analysis, we found that these dsRNA can prevent the BmNPV to a certain extent, and delay the viruses' proliferation.
Peroxidase Activity during Leaf Infection of Mulberry (Morus alba L.) with Brown Leaf Spot Fungus Myrothecium roridum

Peroxidase activity was measured in brown leaf spot pathogen (Myrothecium roridum) inoculated potted mulberry (Morus alba) during pre-symptomatic to various symptom development stages and compared with corresponding healthy leaf tissues. The enzyme showed a pH optimum of 7.0 and the activity was linearly increased up to 15 min of incubation. The peroxidase had a broad substrate specificity and the rates of oxidation were in the rank of pyrogallol> guaiacol> ascorbate at pH 7.0. Catechol at 10 mM inhibited 89% of guaiacol-peroxidase and 76% pyrogallol-peroxidase activities, indicated higher non-specific peroxidation in pyrogallol dependent assay system in mulberry than guaiacol. The optimum requirement for the guaiacol dependent assay was 0.2 ml (
equivalent of protein) of crude enzyme source. Excepting the 8th leaf from the apex, the peroxidase activity did not vary appreciably in different leaf positions. In pre-symptomatic phases, an initial (1 to 5 min) rise of peroxidase activity was noticed in inoculated leaves, and then maintained a plateau up to 300 min. In contrary, non-infected tissue showed a slightly increased trend of enzyme level up to 420 min. In infected tissue, a sharp transient increase (3.1 fold) of peroxidase activity appeared between 300 - 420 min post infections. Afterwards, significantly different but steady maintenance of enzyme levels were observed in two treatments. On the other hand, during symptom development, a sharp increase in peroxidase activity was noticed up to 4th grade of lesion appearance (25.1 % to 50% of leaf area infection), and then declined slightly. However, in non-infected but same age healthy leaves, such huge fluctuations of enzyme level did not apparent. A high positive correlation
between peroxidase activity and leaf spot development grades was also marked. The result implies that pre-symptomatic burst (between 1 - 5 and 300 - 420 min) and subsequent increased trend of guaiacol peroxidase activity may require for the symptomatic manifestation of Myrothecium leaf spot in mulberry.
Development of a Robust Polyvoltine Breed
of the Mulberry Silkworm, Bombyx mori L.

A breeding programme was initiated during 2001 utilizing two polyvoltine silkworm breeds viz.
, an evolved breed tolerant to high temperature and MAR, comparatively resistant to Bombyx mori nuclear polyhedrosis virus (BmNPV) with the objective to develop robust polyvoltine breeds and hybrids. The breed

, the breed was tested for hybrid forming ability utilizing six bivoltine silkworm breeds viz.

exhibited its superiority by recording 97.2% survival, 1.892 g cocoon weight, 0.406 g cocoon shell weight, 21.5% cocoon shell ratio, 16.6% raw silk percentage and 890 m filament length whereas the control
has recorded 90.2% survival, 1.599 g cocoon weight, 0.304 g cocoon shell weight, 18.9% cocoon shell ratio, 13.1 % raw silk percentage and 768 m filament length. Commercial exploitation of the new
hybrid in sericulture industry has been discussed.
Screening of Some Indigenous and Exotic Mulberry Varieties against Major Foliar Fungal and Bacterial Diseases

Fifty-six indigenous and twenty nine exotic mulberry varieties were screened against powdery mildew, Myrothecium leaf spot, Pseudocercospora leaf spot, sooty mold and bacterial leaf spot for a period of three years under field condition. The percent disease index (PDI) was recorded during peak season of the foliar diseases. Out of eighty-five varieties studied, ten varieties were highly resistant and eight were resistant to powdery mildew; six varieties were immune and seventy-eight varieties were highly resistant to Myrothecium leaf spot; sixty varieties were highly resistant and 21 were resistant to Pseudocercospora leaf spot; forty four varieties were highly resistant to sooty mold and two varieties were immune and fifty-eight were highly resistant to bacterial leaf spot. Lowest cumulatative disease index was observed in M. multicaulis (7.28) followed by Thailand lobed (7.85) and Italian mulberry (8.06).

Influence of Host Age on Development and Morphological Parameters of the Parasitoid Nesolynx thymus Girault (Hymenoptera: Eulophidae), An Ecto-Pupal Parasitoid of the Uzi Fly, Exorista bombycis (Louis) (Diptera: Tachinidae)

The present investigation was conducted to know the effects of host age on development and morphological characteristics of Nesolynx thymus, a gregarious ectopupal parasitoid of the uzi fly, Exorista bombycis, a serious en do-larval parasitoid of the silkworm, Bombyx mori L. For experimentation, 3 - 11 day-old pupae of E. bombycis were exposed to 2 day-old adults of N. thymus at

A highly significant negative correlation between host age and the following development and morphological characteristics of the parasitoid was observed: number of pupae parasitised, total adult recovery, sex ratio and recovery, longevity, body length, wing span, abdomen length, and abdomen width of females. The findings of the present study clearly demonstrate that various development and morphological parameters of N. thymus are influenced by the age of E. bombycis pupae.
