Effect of Plant Based Essential Oil on Immune Response of Silkworm, Antheraea assama Westwood (Lepidoptera: Saturniidae)

Botanicals have been regarded as sustainable solution of pest control because of their nominal effect on nontarget pest, biodegradability and other environment friendly properties. However, study on impact of botanicals against beneficial insects is less. In this investigation effort has been made to study the effect of essential oils of Ocimum sanctum, Ocimum gratissimum and Ageratum conyzoides against non-mulberry silkworm Antheraea assama in terms of percent mortality, total haemocyte count (THC) and phenol oxidase (PO) enzyme activity. After application of essential oil of the plants by contact residual film technique, age dependent decrease of percent mortality was recorded. Topical treatment of oil at higher dosages (
) caused 100% mortality of late instar larvae at 24 hour but the lowest dose (
) did not cause any mortality till 48 hour of treatment. Initial increase of total haemocyte count (THC) was recorded at early hours of treatment (0.25 hour) at all dosages (1, 0.5,
) after topical treatment. A dose dependent increase was observed in phenol oxidase enzyme activity and this might indicate ability of the essential oils to cause induction of immune response in Antheraea assama larvae.
Estimation of Hybrid Vigor of Some Egyptian Single Local Hybrids of Mulberry Silkworm, Bombyx mori L.

Fifteen races resulted from silkworm breeding program at Sericulture Research Department (SRD) were used for hybridization. Fourteen hybrids were obtained and coded as; Giza C, D, R, S, T, U, A, V, W, P, H, L and Qanater 1, 2. The traits of cocoon weight, cocoon shell weight, pupal weight, cocoon shell ratio, silk productivity, fifth instar duration, total larval duration, number of cocoon per liter and pupation ratio were evaluated. Data were analyzed by using three formulae of heterosis over better, mid and check parent values. Hybrids of Giza V, C, N, Qanater 1 and 2 are promising and could be used in commercial cocoon production. Generally, there are some new hybrids can be exploited in commercial scale. Also, the local races can be evolved using the hybridization, inbreeding and selection program.
Inhibitory Effect of the Culture Broth Extract of Aspergillus tamarii on Nitric Oxide Production and Its Antioxidative Activity

Many studies have explored suppression of aflatoxin produced by Aspergillus Genus. On the other hand, this study examined the inhibitory effect of the culture broth extract (CE) of A. tamarii obtained from dead silkworm on nitric oxide (NO) production and its antioxidative activity. The culture broth was extracted with EtOAc, dried, and then used in this experiment. As a result, CE did not show cytotoxicity on RAW 264.7 cells at any concentration. Moreover, CE suppressed lipopolysaccharide (LPS)-induced NO production of RAW 264.7 cells in a dose-dependent manner. The total phenol content according to the Folin-Dennis method, the antioxidative activity by DPPH, and the nitrate radical scavenging capacity of CE were increased in a dose-dependent manner. Thus, many of the phenolic compounds were considered to represent the antioxidative activity.

Establishment of Pest Forecasting Management System for the Improvement of Pass Ratio of Korean Exporting Pears

A decrease in pass ratio of Korean exporting pears causes several negative effects including an increase in pesticide dependency. In this study, we attempted to establish the pest forecasting management system, composed of weekly field forecasting by pear farmers, meteorological data obtained by automatic weather station (AWS), newly designed internet web page (
) as information collecting and providing ground, and information providing service. The weekly field forecasting information on major pear diseases and pests was collected from the forecasting team composed of five team leaders from each pear exporting complex. Further, an abridged weather information for the prediction of an infestation of major disease (pear scab) and pest (pear psylla and scale species) was obtained from an AWS installed at Bonghwang in Naju City. Such information was then promptly uploaded on the web page and also publicized to the pear famers specializing in export. We hope this pest forecasting management system increases the pass ratio of Korean exporting pears throughout establishment of famer-oriented forecasting, inspiring famers' effort for the prevention and forecasting of diseases and pests occurring at pear orchards.
Studies on the Utilization of Bivoltine Foundation Cross Males in the Preparation of Cross Breed Eggs of the Silkworm, Bombyx mori L.

In order to study the feasibility of utilizing bivoltine foundation cross (FC2) males of the silkworm as male components with Pure Mysore (PM), six foundation crosses viz,CSR2 x CSR27, CSR27 x CSR2, CSR27 x CSR50, CSR50 x CSR27, CSR2 x CSR50 and CSR50 x CSR2 along with CSR2 pure breed were assessed for performance of parental crop,grainage, rearing and reeling performance. The foundation crosses exhibited better performance than CSR2 as a parental crop and male component which resulted in higher egg recovery. Though there was not much difference between PM crossed with FC2 and CSR2 males pertaining to important bioassay parameters, PM x CSR2 exhibited superiority in reeling traits particularly with reelability and raw silk %. This study reveals that FC2 seed cocoons can be utilized during exigency when there is a dearth for CSR2 seed cocoons but not regularly.
Studies on the Artificial Rearing of Mole Cricket, Gryllotalpa orientalis (Orthoptera : Gryllotalpidae)

The present study is to develop the rearing method of Gryllotalpa orientalis. In total 429 of G. orientalis were collected from the field rearing cage (
) in 2012. Its sex ratio was 1: 1.15(Female : Male). Survival rate of the mole crickets was 94.4~86.1% with the artificial diets formulated for the present study. Successful oviposition rate was 20, 20 and 80% for one, two and three pairs of adult crickets, respectively, from the indoor rearing. The mean number of hatchlings was
, and the mean number of dead hatchlings 1.2, 1.7 and 1.2. The mortality of nymphs on horticultural soil and clay sand mixed with ocher was 18.3 and 10.0%, respectively. The mortality of nymphs in circular and rectangular cages was, respectively, 60 and 40%.
Recombinant Expression and Enzyme Activity of Chymotrypsin-like Protease from Black Soldier Fly, Hermetia illucens (Diptera: Stratiomyidae)

Chymotrypsin serine protease is one of the main digestive proteases in the midgut of and is involved in various essential processes. In a previous study, a gene encoding a chymotrypsin-like protease, Hi-SP1, was cloned from the larvae of Hermetia illucens and characterized. In this study, we produced the recombinant chymotrypsin-like protease Hi-SP1 in Escherichia coli cells. The molecular weight of the recombinant Hi-SP1 was estimated to be approximately 26 kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Western-blotting. Chymotrypsin activity was detected when AAPF was used as the substrate. Examination of the effects of temperature and pH revealed that the proteolytic activity of recombinant Hi-SP1 decreased markedly at temperatures above
, and the optimum pH was found to be 10.0.
Molecular Cloning, Gene Structure, Expression, and Enzyme Activity of a Serine Protease from Water Scorpion, Laccotrephes japonensis (Hemiptera: Nepidae)

Serine proteases are major insect enzymes involved in the digestion of dietary proteins and in the process of blood meal digestion. In this study, cDNA was constructed using the whole body of Laccotrephes japonensis. The flanking sequences of the 5- and 3- end of this gene were characterized by RACE-PCR. Sequence analysis showed that this gene contained a 963-bp ORF encoding 320 amino acids. The deduced amino acid sequence showed 62% identity with the Creontiades dilutus serine protease, 58% with the Lygus lineolaris trypsin precursor, and 54% with the Triatoma infestans salivary trypsin. To assess the expression of the L. japonensis serine protease (JGsp), the JGsp gene was cloned into a baculovirus transfer vector, pBac-1, and expressed in Sf9 cells (Spodoptera frugiperda). SDS-PAGE and western blot analysis have shown that the JGsp recombinant protein was a monomer with a molecular weight of about 32 kDa. Recombinant JGsp has shown activity in the protease enzyme assay using gelatin as a substrate.
Purification and Identification of Paenibacillus sp., Isolated from Diseased Larvae of Allomyrina dichotoma (Linnaeus, 1771) (Coleoptera: Scarabaeidae) in Insect Farms

In reared populations of Allomyrina dichotoma, commercial insects, the skin of last instar larvae was changed softer with opaque white, and infested grubs eventually died. To clarify the cause of the symptom, we collected the larvae of A. dichotoma from five farms and examined their intestinal bacterial florae using pyrosequencing technique. From those results, a member of Paenibacillus was found only in the larvae showing the symptom of disease. Through PCR analysis using a Paenibacillus specific primer set, we obtained the partial 16S rRNA gene sequence and confirmed the microbe as Paenibacillus sp. For clear identification, a whole guts was extracted from each larva showing the sign of the disease and incubated at

The 16S rRNA gene sequence analysis for the isolated bacteria showed that they were closely related to P. rigui(97.9% similarity), to P. chinjuensis(96.1% similarity), and to P. soli(95.3% similarity). Additional tests including API test and cellular fatty acid composition analysis were performed, but the strain couldn't be identified at species level, suggesting it may represent novel species of the genus Paenibacillus.
Cocoon Characteristics of Antheraea pernyi Silkworm Reared in Korean Oak Field

Antheraea pernyi silkworm is a well known wild silkworm to produce a valuable silk fiber. A. pernyi silkworm was reared in Korean oak field and examined the cocoon characteristics, such as cocoon weight, cocoon shell weight, and percentage of cocoon shell weight. Degumming loss was also measured after alkali degumming process. A. pernyi silkworm spins tawny color cocoon in oval shape. Cocoon shell weight of A. pernyi silkworm, 0.78 g, was heavier than that of B. mori silkworm, 0.51 to 0.63 g. Cocoon shell percentage of A. pernyi silkworm, 32.8%, was higher than that of B. mori silkworm, 23.4 to 25.2%. Degumming loss percentage of A. pernyi silkworm, 17.1%, was lower than that of B. mori silkworm, 25.0%. SEM showed that the surface of the cocoon filament was coarse and oriented with longitudinal direction.
General Characteristics of Korean Propolis

The propolis was collected from the whole part of Korea Peninsula, such as central, southern, and Jeju island, to analyze the general composition including total flavonoid and phenolic contents, heavy metals, colors, amino acids, and crude lipid. The total flavonoid and phenolic contents of middle region in Korea Peninsula (4.26% and 13.06 g/100 g gallic acid) were higher than southern region (2.53%, 10.9 g/100 g gallic acid) and Jeju island (0.03%, 7.22g/100 g gallic acid), respectively. The heavy metals contents showed that Zn contents were found in
and a harmful heavy metal such as Cr, Cd, Pb, Ni, Cu, As, and Hg were not detected at all in our experiment. The colors of extracted propolis showed a various spectrum from yellow to red purple. Various amino acids were also detected as proline, valine, methionine and the average crude lipid contents of propolis were 42.4%. All data collected in South Korea were very first executed in our institution for the purposes of utilizing and developing the industrial insects, honeybees as well as silkworm.
