Recombinant human BMP-2/-7 heterodimer protein expression for bone tissue engineering using recombinant baculovirus expression system

Bone morphogenetic proteins (BMPs) are essential growth factors for bone formation, skeletal development and bone regeneration. The BMP-2/7 heterodimer is known to have remarkable effects on osteogenic induction that are even stronger than the BMP-2 or BMP-7 homodimers. We designed a recombinant human BMP-2/7 (rhBMP-2/7) heterodimer protein with four glycine residues between BMP-2 and BMP-7 protein to facilitate free bond rotation of domains. The Baculovirus Expression Vector System (BEVS) is routinely used to produce recombinant proteins in the milligram scale. In this study, the BEVS was used to express the rhBMP-2/7 protein whrer the recombinant baculovirus was recovered in the host Sf9 cells. To confirm the biological activity of rhBMP-2/7 protein secreted from the BEVS as an osteogenic differentiation and induction factor, we measured the BMP-induced ALP activity. rhBMP-2/7 could be used as an alternative to BMPs to overcome limitations like short half-life and requirement for high concentrations. Furthermore, rhBMP-2/7 may be an efficient tool for various application studies such as bone regeneration and skeletal development.
Breeding of biparental sex-limited larval marking yellow cocoon variety "Hanbyeolnue"

All of silkworm varieties reared in farmhouses are first generation hybrids, for the production of these hybrid silkworms precise and labor-saving sex discrimination is necessary. The new variety "Hanbyeolnue" is biparental sex-limited larval marking yellow cocoon variety which was bred from biparental sex-limited strain of Japanese originated Jam 319 and Chinese originated Jam 320. Productivity test of Hanbyeolnue in 2015 showed high healthiness and short larval period. The Hanbyeolnue was evaluated as an excellent variety which can be utilized for special purpose silkworm such as male pupae or Cordyceps production.
Effects of olfactory self- and cross-adaptation on perceiving odor in a moth

Pheromone orientation in moths is an exemplar of olfactory sensitivity. To avoid cross mating, the responses of males to pheromone blends must be high specificity and temporal resolution. We tested the effects of olfactory self- and cross-adaptation of pheromone compounds and mixtures in Spodoptera litura moths by electroantennogram (EAG) recordings. The challenge of S. litura antennae to a pulse train of its own pheromone blends of Z9,E11-14:OAc and Z9,E12-14:OAc with 200 ms on/off and 1 s on/off indicated that the repetitive stimulation by 200 ms on/off with high dosages resulted in greater adaptation than that by 1 s on/off with low dosages and the adaptation index of Z9,E11-14:OAc in all treatments is significantly larger than that of Z9,E12-14:OAc, suggesting that high dosages with more frequent stimulation prefer to induce sensory adaptations and a different odor coding exist between the two components in the antennal periphery in this moth. The cross-adaptation EAG test among the two pheromone compounds and Z7-12:OAc and Z9-14:OH from congeneric species of S. litura showed that each of these compounds adapted the antenna more to that specific compound. The significantly higher adaptation to Z7-12:OAc and Z9-14:OH than to the pheromone components of S. litura induced by themselves suggested that both of them are coded by specific odor receptor neurons which are different from those tuned to the pheromone components of S. litura . Thus, we proposed that Z7-12:OAc and Z9-14:OH may play an important role in avoidance of heterospecific mating between S. litura and its sympatric moth species.
Effects of agricultural byproducts, DDG and MSG, on the larval development of mealworms

Distillers dried grain (DDG) and makgeolli spent grain (MSG) are agricultural byproducts to produce alcoholic beverage. However, they are known to contain enough nutrients. Mealworm is a promising insect resource for an animal feed ingredient as well as alternative human food. With low cost, DDG and MSG were investigated as a feed ingredient for rearing high quality mealworms. DDG and MSG were mixed with wheat bran and compared to control feed (only wheat bran) for its effects on larval survivorship, larval weight, duration for developmental period, pupation rate, and pupal weight. When DDG added, larval survivorship was reduced to 50~70% compared to the control group. Larvae fed on DDG were heavier from third to sixth week. Especially, larvae with 50% DDG were 28% heavier than the control group at the third week. For the larval period, the 50% DDG group was 11% less than that for the control. The pupal weight for the 30% DDG group was 7% heavier than that for the control group. Pupation rates for all the DDG groups were higher than 90%. When compared to the control, larval survivorship for the 70% MSG group was low, but the 50% and 70% MSG groups were high during the seventh and eighth weeks because of delayed development. After the eighth week, larvae with 70% MSG showed the highest larval weight increase as 9~18% compared to the control group. Except 70% MSG group, all of MSG groups showed more than 90% pupation rates. We confirmed that adding 30~50% of DDG or MSG to conventional wheat bran have a strong potential to replace the conventional wheat bran insect feed for quality insect production.

The major structural proteins of porcine reproductive and respiratory syndrome virus (PRRSV) are derived from ORFs 4, 5, and 6. They have been considered very important to arouse the humoral and cellular immune responses against PRRSV infection and proposed to be the excellent candidate proteins in the design of PRRS bioengineering vaccine. However, the PRRSV structural proteins are produced in low levels in the infected cells because it forms insoluble protein and possesses several transmembrane regions. To overcome this problem, we fused the ORF4, ORF5, and ORF6 with SUMO (small ubiquitin-related modifier). The resulting fusion protein SUMO-ORF4, -ORF5, and -ORF6 were highly expressed in Bm5 cells. The level of protein expression using the Bombyx mori larvae was higher than that using Bm5 cells. In addition, fusion to SUMOstar, which is not processed by native SUMO proteases, significantly enhanced protein expression levels compared to SUMO fusion. This study demonstrated that SUMO or SUMOstar, when fused with PRRSV structural proteins, was able to promote its soluble expression. This may be a better method to produce PRRSV structural proteins for vaccine development.
