Antibacterial, cytotoxic and neuropharmacological activities of Cerbera odollam seeds

The MeOH extract of seeds of Cerbera odollam Gaertn. (Apocynaceae) was screened for its antibacterial, cytotoxic and neuropharmacological activities. The extract showed moderate anti-bacterial activity against Salmonella typhi, Streptococcus saprophyticus, and Streptococcus pyogenes. It exhibited high level cytotoxicity against brine shrimp (
). The extract potentiated pentobarbital induced sleeping time in mice which was further supported by the exploratory behavior test at dose of 25 mg/kg. The overall results tend to suggest the antibacterial, cytotoxic and CNS depressant activities of the extract.

The aim of the study is to investigate the antioxidant activity through, reducing power, 2, 2-diphenyl-1-picryl hydrazyl radical (DPPH), nitric oxide radical (NO), superoxide radical, hydrogen peroxide radical (
) scavenging activity and the amount of total phenolic compounds of chloroform, ethyl acetate, methanol and aqueous extracts of the leaves of Talinum portulacifolium. Chloroform extract of leaves of T. portulacifolium showed highest antioxidant activity, with a direct relationship between activity and concentration of extracts (
). Among all the extracts, the highest amount of the total polyphenolic compounds was found in the chloroform extract. Chloroform extract of T. portulacifolium showed an important free radical scavenging activity towards the DPPH, NO, Superoxide and

, respectively. In the lipid peroxidation assay, extracts of chloroform and ethyl acetate showed a remarkable inhibitory activity. The extracts showed significant activity in all the experiments but lower than the standard antioxidant, ascorbic acid.
Anti-Salmonella activity of a flavonone from Butea frondosa bark in mice

Butea frondosa has been used traditionally as a topical formulation in the treatment of many diseases and disorders. Two compounds [BF-1 (crystalline flavonol quercetin) and BF-2 (tannin) from ethyl acetate fraction of ethanolic extract] were isolated from the bark of Butea frondosa. The stereostructures of the compounds were determined on the basis of chemical and physicochemical evidence. BF-1 and BF-2 were screened in vitro for possible antibacterial property against 112 bacteria comprising 3 genera of Gram-positive and 12 genera of Gram-negative types. It was found that both BF-1 and BF-2 exhibited inhibitory activity against several bacteria. Most of these strains were inhibited by BF-1 at

In vivo studies with this extract showed that it could offer statistically significant protection (p < 0.01) to mice challenged with a virulent bacterium. The inhibitory activity of Butea frondosa against Gram-positive and Gram-negative bacteria indicates its usefulness in the treatment of common bacterial infections. The potentiality of BF-1 as an antibacterial agent may be confirmed further by pharmacological studies.
Anti-diabetic activity of Thespesia lampas Dalz & Gibs on alloxan induced rats

Anti-diabetic effect was observed with Thespesia lampas Dalz & Gibs (Family: Malvaceae) when given as a root extract in normal as well as alloxan induced diabetic rats. The effects, however, were more pronounced in diabetic animals in which administration of plant extract for 15 days after alloxan induced diabetes, significantly reduced blood glucose levels. After alloxan induced diabetes it was observed that both standard drug (glibenclamide) and aqueous extract of Thespesia lampas were significantly superior to control in reducing blood sugar on long term treatment (15 days). The aqueous extract of T. lampas (300 and 600 mg/kg) reduced the blood glucose levels from
, respectively. The data suggested that T. lampas could be of beneficial in diabetes mellitus in controlling blood sugar. The present investigation established pharmacological evidence to support the folklore claim as an anti-diabetic.
Effect of an herbal formulation on DOCA-salt and fructose induced models of hypertension in rats

The present study was carried out to investigate the antihypertensive effect of a folklore herbal formulation (HF) (300mg/kg/day; p.o.) in deoxycorticosterone acetate (DOCA)-salt induced and fructose induced hypertensive rats. In DOCA model, DOCA (15 mg/kg, s.c., twice a week) was administered to unilateral nephrectomized rats for 4 weeks. In fructose model, drinking water was replaced with 10% fructose solution for 6 weeks to induce hypertension. Systolic blood pressure (BP) was measured once every week during the treatment schedule. After completion of treatment schedule, BP and vascular reactivity to various agonists like Noradrenaline, Adrenaline, Phenylephrine and Serotonin (5-hydroxytrptamine; 5-HT) were recorded in rats of both models. A cumulative concentration response curve of 5-HT was carried out in isolated rat fundus strip of the DOCA-salt induced and fructose induced hypertensive rats. The results tend to suggest that HF possesses antihypertensive activity.
Nootropic Potential of Murraya koenigii leaves in Rats

Murraya koenigii leaves commonly known as 'curry patta' are routinely added to Indian gravy and vegetarian dishes by south Indian as a favourate condiment. The present study was undertaken to investigate the effects of Murraya koenigii leaves (MKL) on memory in rats. Elevated plus-maze and Hebb-Williams maze served as the exteroceptive behavioral models for testing memory. Diazepam-, scopolamine- and ageing-induced amnesia served as the interoceptive behavioral models. MKL fed orally to various groups of young and aged rats with diet containing 2, 4 and 8% w/w of MKL for 30 days consecutively were investigated. The MKL diets produced a significant dose-dependent improvement in memory scores of young and aged rats and significantly reduced the amnesia induced by scopolamine (0.4 mg/kg, i.p.) and diazepam (1 mg/kg, i.p.). Therefore, it would be worthwhile to specifically investigate the therapeutic potential of MKL in the management of dementia patients.

In the present study, various extracts of Clerodendrum infortunatum leaves have been studied for its anthelmintic, antimicrobial and antipyretic activities. In the anthelmintic and antipyretic study it was observed that the benzene extract was more potent than the other two extracts (ethanol and aqueous) even though all the three extracts were endowed with both the properties. The study reveals antimicrobial activity of the extracts against the tested strains of microorganisms between concentration ranges of
and shows effectiveness more against gram-positive bacteria than the gram-negative bacteria.
Immunomodulatory effect of Tinospora cordifolia and Centella asiatica and its modulation on cyclophosphamide challenge

Ethanolic extracts of T. cordifolia and C. asiatica were evaluated for immunostimulatory effect in mice against sheep RBCs as antigen by three models viz. delayed type hypersensitivity reaction, ercent change in neutrophil count and haemagglutination titre. Immunostimulatory effect in the presence of immunosuppressant agent, cyclophosphamide (100 mg/kg, i.p.) was also investigated. T. cordifolia and C. asiatica significantly (p < 0.001, p < 0.05 respectively) enhanced foot pad thickness when measured after 24 hours of sheep RBC antigen challenge. Both the plant materials increased foot pad thickness even after being subjected to immunosuppressant exposure. T. cordifolia revealed enhanced neutrophil counts, while C. asiatica had no significant effect on neutrophil counts. T. cordifolia exhibited significantly (P < 0.01) elevated neutrophil levels even in the presence of cyclophosphamide administration. Both the plants exhibited humoral antibody response, as haemagglutination titre values were significantly high as compared to control. T. cordifolia and C. asiatica could combat immunosuppressant effect of cyclophosphamide (P < 0.01). This suggests that T. cordifolia and C. asiatica can be regarded as biological response modifiers and can be utilized for the development of immunostimulating agent among plant sources.
Pharmacological and Biochemical studies on Telescopium telescopium - a marine mollusk from the Mangrove regions

The tissue extract (TTE) of a marine snail Telescopium telescopium, collected from the coastal regions of West Bengal, India, was extensively screened for pharmacological and biochemical properties. Telescopium telescopium (TTE) produced significant lysis of washed rat erythrocytes (both direct and indirect), produced haemorrhagic lesions in the skin and also released haemoglobin (in vitro tissue damage) from different tissue samples. TTE was found to produce pro-inflammatory effects when injected into the rat hind paw and also increased peritoneal vascular permeability. Furthermore, intravenous administration of TTE produced a decrease in blood pressure (hypotensive effect) in anaesthetized rats. The extract produced potent esterase activity, as was evident from the breakdown of FDA with subsequent release of fluorescein (in vitro). TTE also demonstrated prominent cholinesterase, phospholipase, phosphatase and protease activities.

Methanol and aqueous extract of leaves of Trema orientalis Linn. were subjected to the potential antioxidant and antibacterial activities. The pharmacological interest of this plant coupled with traditional use (antidiarrhoeal, antiseptic, analgesic etc) prompted to test for antioxidant and antibacterial activities. The antioxidant potential of the methanolic extract was determined on the basis of their scavenging activity of the stable 1,1-diphenyl-2-picryl hydrazyl free radical.

which indicated the strong antioxidant activity of the plant. However the aqueous extract showed mild antioxidant activity. In case of antibacterial activities test, the extract was subjected for its effectiveness against both Gram-positive and Gram-negative bacteria in agar diffusion method. The zones of inhibition produced by the crude methanol and aqueous extract against few sensitive strains were measured and compared with those of standard antibiotic Gentamycin. It is evident that both extracts are active against the bacteria at low concentrations. The obtained results provide a support for the use of this plant in traditional medicine and suggest its further advance investigation.

The aim of the present study is to investigate the anti-inflammatory and antimicrobial activity of petroleum ether and ethanol extracts of Scutia myrtina (Family: Rhamnaceae). In anti-inflammatory activity carrageenan and histamine induced paw oedema and cotton pellet induced granuloma for acute and chronic inflammatory models were studied in Wister albino rats. Based on the results of the present study it can be concluded that petroleum ether and ethanol extract of Scutia myrtina at 400 mg/kg has potential anti-inflammatory effect and they act in a dose dependent manner. Both extracts of Scutia myrtina showed antimicrobial activity against all bacterial and fungal strains tested at the concentration of
. From the result, it can be concluded that the Scutia myrtina contain antibacterial and antifungal principle. Further more, besides the confirmation of the popular use; the obtained results demonstrate this herbal drug to represent a new source of antimicrobial and anti-inflammatory agent.
Search towards an insight for comparative anti-tumour effects of Wrightia tomentosa leaf & bark in ehrlich ascites carcinoma bearing mice

In the present study, the ethanolic leaf and bark extract of Wrightia tomentosa were tested for comparative in vivo antitumour properties against Ehrlich ascites carcinoma (EAC) tumour bearing mice at 100 and 200 mg/kg body weight doses given orally once daily for 16 days. The EAC mice receiving 100 and 200 mg/kg ethanolic leaf and bark extract showed a dose dependent elevation in tumour, free survival and a highest number of survivors were observed at 200 mg/ kg for leaf extract of ethanol, which was considered as an optimum dose for its anti neoplastic action. The Median survival time for this dose was approximately 44 days when compared with 23 days of non-drug treated controls. The results indicate that the administration of leaf extract not only increased the survival of animals with ascites tumour and reduced packed cell volume and viable tissue cell count, but also altered many hematological parameters changed during tumour progression, indicating the potent antitumour nature of leaf extract than the bark extract. Statistical analysis also reveals that the leaf extract showed highly significant anti tumour potency (p < 0.001) when compared with control.
Evaluation of anti-inflammatory activity of Solanum trilobatum roots

This study evaluated the anti-inflammatory potential of the crude alkaloidal fraction (CAF) of methanol extract of Solanum trilobatum Linn. (Solanacea) root in animal models of inflammation. Crude alkaloidal fraction at doses of 25, 50 and 100 mg/kg significantly (p < 0.01) reduced carrageenan induced rat paw volume at 3 h after carrageenan challenge as compared to control group of animals. CAF (25, 50 and 100 mg/kg) significantly (p < 0.01) and dose dependently suppressed cotton pellet induced granuloma formation. Topical application of CAF (1, 5 and 10 mg/ear) markedly inhibited multiple application of TPA in mice. CAF elicited pronounced inhibitory effects on formaldehyde and adjuvant induced arthritis in rats. These results indicate that CAF of methanol extract of the Solanum trilobatum has anti-inflammatory activity in acute and chronic inflammation.
Anti-inflammatory and analgesic activities of methanol extract of Triphala - a poly herbal formulation

Ayurveda, an ancient System of Indian Medicine, has recommended a number of drugs from indigenous plant sources for the treatment of inflammation. To evaluate the anti-inflammatory and analgesic activities of methanol extract of Triphala were investigated in Wister albino rats and mice. The methanol extract of Triphala were found to encompass substantial anti-inflammatory effect in acute and sub-acute models and analgesic effect. Animal models of carrageenan induced edema and cotton pellet induced granuloma in albino rats were used. Extract of 200 mg/kg shows significantly reduced paw edema. Analgesics activity of Triphala with 200 mg/kg shown by significant reduction of writhing. These report shows to support the use methanol extract of Triphala in relieving inflammation and pain.
Effect of Chungpaesagan-tang on ischemic damage induced by MCAO in spontaneously hypertensive rats

Chungpaesagan-tang (CPSGT) is most frequently used to treat ischemic brain injury in tradition Korean medicine. Clinically, cerebral ischemia is likely to be accompanied by preexisting or complicating disease. However, animal models used to examine the effects of herbal medicines on cerebral ischemia have not given this issue sufficient consideration. The present study was undertaken to determine the effects of CPSGT on focal cerebral ischemia in normal and SHR rats subjected to transient middle cerebral artery occlusion (MCAO). Animals were divided into four groups: Normal (Sprague-Dawley) rats subjected to MACO (the NC+MCAO group), normal rats subjected to MCAO and then administered CPSGT (NC + MCAO + CP), SHR rats subjected to MCAO (SHR + MCAO), and SHR rats subjected to MCAO and then administered CPSGT (SHR + MCAO + CP). MCAO was performed using the intraluminal method. CPSGT was administrated orally twice (1 and 4 h) after MCAO. All animals were sacrificed at 24 h postoperatively. Brain tissues were stained with hematoxylin & eosin, to examine the effect of CPSGT on ischemic brain tissues. In addition, changes in TNF-
expression in ischemic areas were examined by immunostaining. CPSGT was found to significantly reduce infarction areas in normal and SHR rats and infarction volumes in SHR rats. Similarly, CPGST markedly increased neuron numbers and sizes in all treated groups, except cell sizes in SHRs. Furthermore, CPSGT reduced TNF-
expression in MCAO administered SHR rats. The findings of the present study suggest that CPSGT effectively ameliorates neuron damage caused by MACO-induced cerebral ischemia, and that it has a significant neuroprotective effect after cerebral ischemia in SHR.

Preliminary study on the central nervous system depressant effect of Picrorhiza kurrooa Royle. (Scrophulariaceae) in mice models

Picrorhiza kurrooa Royle. is a well known medicinal plant among the indigenous medical practitioners of India. Present study is the first time to report the activity on the central nervous system. Preliminary study of the hot water extract showed significant depressant activity on the hole board test as evidenced from the ambulation and head dipping scores. The extract showed better activity compared to diazepam on the duration of pentobarbital induced sleeping time.
