A cDNA Encoding 3-ketoacyl-CoA Thiolase of Soybean and Its Expression in Escherichia coli

The gene encoding a 3-ketoacyl-CoA thiolase from soybeans was cloned, sequenced, and expressed in Escherichia coli. It encoded 446 amino acids with a molecular mass of 47 kDa. The deduced amino acid sequence of the 3-ketoacyl-CoA thiolase showed a high degree of similarity to other plant thiolases: pumpkin, squash, cucumber, Arabidopsis thaliana, Japanese rice, mango, and cotton. The recombinant 3-ketoacyl-CoA thiolase was expressed to a major protein in E. coli and purified by affinity chromatography. This protein could be useful in studying the mechanism of the
-oxidation of fatty acid in soybeans.
Effects of Salt Addition in Sugar Based Osmotic Dehydration on Mass Transfer and Browning Reaction of Green Pumpkin

The effects of immersion temperature and time, sugar concentration, and salt addition on water loss, solid gain, molality and rate parameter of green pumpkins during osmotic dehydration were analyzed by a response surface methodology, and osmotic dehydration effects on browning reduction of air-dried green pumpkins were also evaluated. The water loss, solid gain and molality are fast in the beginning of process and then increase slowly during remainder of the process. After 20 min of immersion time, water loss and solid gain of 100 g wet green pumpkin at
Brix sugar solution with 0, 2, 4% salt addition were 50.5, 54.8 and 58.6 g water loss and 10.4, 10.0 and 9.7 g solid gain, respectively. Based on the coefficients of determination, all second degree polynomial regression models were highly fitted to the experimental data at the level of 0.01%. Rate parameter, K, values were increased as the immersion temperature and sugar concentration increased. Browning degree was decreased as the immersion temperature, time and salt concentration increased. Minimum browning degree of dried green pumpkins (O.D. = 0.076) could be achieved using
binary solutions (
Brix sugar solution with 4% salt addition) with 20 min of immersion time compared to control (O.D. = 0.364) and
blanching with 20 min of immersion time (O.D. = 0.122).
Antibacterial Activity of Oriental Medicinal Plant Extracts toward Helicobacter pylori

The antibacterial activity of the methanol extracts from 64 oriental medicinal plant species toward Helicobacter pylori ATCC 43504 was examined using an impregnated paper disc bioassay. At 1 mg
, good anti-H. pylori activity was observed with the extracts of Alpina officinarum rhizome, Alpina oxyphylla fruit, Angelica tenuissina root, Asiasarum heterotropoides root, Lindera strychnifolia root, and Polygonum cuspidatum rhizome. These plants merit further study as potential H. pylori eradicating agents.
Growth Inhibitors of Soybean Seedling from Bacillus sp. IJ-31

The growth-inhibitors of soybean seedlings were isolated from the culture broth of Bacillus sp. IJ-31 by the consecutive purification using solvent partition and silica gel chromatography. The active principles were identified as indole 3-propionic acid (IPA) and indole 3-acetic acid (IAA) by spectral analyses including MS and NMR. At 10 ppm, the exogenouse IPA inhibited 83% of the root elongation and 73% of shoot growth against water-treated control. Eighty-two percent of the root length and 59% of the shoot growth were reduced by the exogenous addition of the IAA at the same concentration. One liter of the culture produced 47.6 mg of IPA and 34.9 mg of IAA into the broth.
A Rapid Screening for Alcohol Detoxification Constituents of Hovenia dulcis by Microplate Reader

Hovenia dulcis is one of the earliest developed medicinal plants used in traditional Chinese medicine. It has many profound pharmacological actions, including alcohol detoxification activity. The aim of the present study was to discover the medicinal sites and active constituents of the alcohol detoxification activity of H. dulcis. In this paper, alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) were first measured by a microplate reader to assay the alcohol detoxification effect. The screening results suggested that n-butanol extract of fruits and ethyl acetate extract of stem of H. dulcis have relatively higher ADH activity; also, the n-butanol extract, hot water extract of fruits, and water extract of stem of H. dulcis have relatively higher ALDH activity. The experiments indicate that microplate reader screening is a quick, accurate, and effective method to assay ADH and ALDH in vitro. This method is also suitable for carrying out the screening work of a large numbers of in vitro samples.
Acaricidal Activity of Aromatic Medicinal Plants against Dermatophagoides spp. (Acari: Pyroglyphidae)

The acaricidal activity of the methanol extracts of 26 aromatic medicinal plants against D. farinae and D. pteronyssinus was examined using direct contact application. There was more than 70% mortality against D. farinae and D. pteronyssinus at 0.08 mg

Soil Chemical and Biological Characteristic of the Continuous Cropping Area of Garlic or Onions

This study researched the soil chemical properties and microflora in upland fields that had been cultivated with either garlic or onion crops for a long period of time. The soil samples were collected from 60 spots in 5 experimental sites at the final (20 May to 10 June) garlic and onion harvests in 2000. Then the soil chemical and biological properties were determined. The soil pH and organic matter content significantly decreased when the garlic and/or onion crops were continuously grown in the same area, while EC and phosphate increased. The number of bacteria and actinomycetes in the soil that continuously grew with garlic was lower when compared to the soil from onion crops. For the fungal population, however, no significant difference was found between the soils from the different garlic and onion crops. These results indicate that the continuous cropping of garlic and onions produced a nutrient imbalance in the soil.
