Pattern of Spore Discharge Over Hymenial Surfaces

Considerable variations in spore productivity were detected within the area of single hymenial surfaces of certain polypores. The hymenium has in irregular patterns, areas of low as well as high spore production, common both in the periphery(young) as well as in the inner(old)part. These have been recorded as isospore diagrams showing that the basidia all over the hymenia are at different stages of development.

Each of 103 isolates of Phytophthora capsici was obtained from diseased red pepper plants randomly belonged to either the mating type

Isoates with changed pathogenicity were selected among iprodione-resistant Alternaria mali to investigate any relationship between their pectolytie enzyme activity and pathogenicity. In artificial medium, strongly pathogenic isolates

Isolation of Protoplasts from Flammulina velutipes

To obtain basic information for the genetic analysis and breeding of Flammulina velutipes, some factors affecting the release of protoplasts from the fungus were studied. Potato Dextrose peptone Agar medium was suitable for the growth of the mycelium and the protoplast formation of F. velutipes. The culture age for the high yields of protoplast was 5 days on PDPA. Few protoplasts were formed from the mycelium cultured on Mushroom minimum Media. The highest yield of protoplasts was obtained in enzyme solution containing Novozyme 234 plus cellulase CP at 10 mg
concentration, while a half amount of protoplasts was obtained in enzyme solution containing Novozyme 234 only. The optimal reaction time of the mycelium in the Iytic enzyme mixtures was 3 hours. The best osmotic stabilizer for the protoplast formation of the mycelium was 0.6M sucrose without buffer at pH 6.2.
Interspecific Protoplast Fusion of Ganoderma applanatum and Ganoderma lucidum and Fruit Body Formation of the Fusants

Interspecific fusion products were obtained from fusion of Ganoderma applanatum and Ganoderma luridum. Frequency of fusion was 0.77-1.38%. Fusion products were selected by the comparision of morphology and color of colony. Fusion Products had chracteristics of two parental strains and generally grew faster than the parents. Some fusants were segregated on GCM. Fusion products were confirmed by mycelial morphology and electrophoretics pattern of esterase isozyme from mycelium. Most of fusion products were lack in clamp connection but those fusion products, that were segregated produced mycelium with and without clamp connection. Some of the fusion products produced fruit body on sawdust medium.
Comparison of Electrophoretic Isozyme Band Pattern of Pleurotus spp. in Korea -I. Homogeneous Gel-

Electrophoretic isozyme patterns from mycelia, primordia, cap and stem of Pleurotus spp. collected in Korea were compared. Primordia, cap and stem of fruitbody showed very similar isozyme patterns but mycelial isozyme patterns were different from those of fruitbody. Isozyme patterns of malate dehydrogenase, acid phosphatase in Pleurotus ostreatus collected from different regions in Korea were similar but those of esterase, peroxidase, leucine amino peptidase and superoxide dismutase were different. Interspecific comparison of esterase isozyme patterns among Pleurotus ostreatus P. cornucopiae and, P. florida was very different and may be valuable subsidiary tool to conventional taxonomic techniques for identifying species of Pleurotus. A dendrogram by similarities of isozyme band pattern was presented.

Isozyme band patterns were investigated by isoeletric focusing Esterase and Leucine amino peptidase of Pleurotus spp. in Korea. Esterase patterns of mycelia and fruitbody were distinquished. However, those of primordia, cap and stem were similar. Interspecies differences of Pleurotus ostreatus, P. cornucopiae and P. florida of Esterase zymogram were found. Species identification by electrophoretic zymogram may be a role as an additional taxonomic tool.
