Genetic Diversity of Agaricus bisporus Strains by PCR Polymorphism

Twelve Universal fungal PCR fingerprint (UFPF) primers that were modified from Universal rice primer (URP) were used to assess genetic diversity of 64 Agaricus strains including 45 A. bisporus strains and other 19 strains of other Agaricus spp. Eight primers, UFPF1, UFPF2, UFPF3, UFPF7, UFPF9, UFPF10, UFPF11, and UFPF12 produced PCR polymorphic bands within and between the Agaricus species. Primer UFPF7 produced specific PCR polymorphic bands that are distinct Korean strain from different strains. Ninety five PCR polymorphic bands were inputted for UPGMA cluster analysis. Forty five strains of A. bisporus are genetically clustered into 8 groups, showing coefficient similarity from 0.75 to 0.9 among them. The varieties, Saea, Saedo, Saejeong and Saeyeon that have recently been developed in Korea were involved in the same group with close genetic relationship of coefficient similarity over 0.96, whereas, other Korean strains were genetically related to A. bisporus strains that were introduced from USA, Eroupe and Chinese.
Distribution of Higher Fungi in Wolchulsan National Park

The result of the survey on higher fungi in the Wolchulsan National Park from April 2009 to October 2011 is as follows. During the survey, total of 1 kingdoms 2 divisions 5 classes 18 orders 56 families 133 genera and 298 species were surveyed, Basidiomycota has 12 orders 47 families 120 genera 278 species, Ascomycota 6 orders 9 families 13 genera 20 species. As for Basidiomycota, Agaricomycetes has 46 families 118 genera 276 species. The most occurred fungi were Russulaceae with 37 species, followed by Boletaceae, Amanitaceae, Agaricaceae and Polyporaceae. Various species of most higher fungi occurred during the period with average temperature of 26.0~27.9, max. 32.0~34.9, min. 24.0~25.9, and over 100 mm of precipitation.
Isolation and Diversity of Yeasts from Fruits and Flowers of Orchard in Sinam-myeon of Yesan-gun, Chungcheongnam-do, Korea

Several kinds of yeasts from fruits and flowers of orchard in Yesan-gun of Chungcheongnam-do, Korea were isolated and identified by comparison of nucleotide sequences for PCR-amplified D1/D2 region of 26S rDNA using BLAST. Fourty eight yeast strains of twenty five species and one hundred eight yeast strains of fourty eight species were isolated from fruits and flowers of orchard in Sinam-myeon of Yesan-gun, Chungcheongnam-do, respectively. Among total one hundred fifty-six yeast strains, only sixteen species were overlapped from fruits and flowers.

Various yeasts from wild flowers of Ulleungdo in Gyeongsangbuk-do and Yokjido in Gyeongsangnam-do, Korea were isolated and identified by comparison of nucleotide sequences for PCR-amplified D1/D2 region of 26S rDNA using BLAST. Forty eight yeast strains of twenty two species and sixty yeast strains of twenty five species were isolated from wild flowers of Ulleungdo and Yokjido, respectively. Only seven species were overlapped from the two different islands areas: Cryptococcus albidus, Cryptococcus laurentii, Metschnikowia reukafii, Pichia scolyti, Rhodotorula glutinis, Rhodotorula graminis and Rhodotorula mucilaginosa. Among forty species from two different islands, other thirty three species were restricted to specific collection site suggesting that each area has distinctive yeast flora.
Diversity of Arbuscular Mycorrhizal Fungi in Rhizospheres of Camellia japonica and Neighboring Plants Inhabiting Wando of Korea

In this study, the community structures of arbuscular mycorrhizal fungi (AMF) in rhizospheres of Camellia japonica and neighboring woody plants in Wando, Korea were investigated. Rhizospheres of C. japonica and other woody plants were dominated by the same species, Acaulospora mellea, but Shannon's index, species richness and total spore numbers of the AMF communities were higher in non-C. japonica than in neighboring plants. Regardless of host plant species, the frequency of A. mellea was significantly high comparing with other AMF species. The community similarity of AMF within C. japonica was significantly higher than between C. japonica and neighboring plants or neighboring plants (p<0.005). Results showed that AM fungal communities in rhizospheres of C. japonica have unique community structure and are different from that of neighboring host plants, suggesting that community structure of AMF could be influenced by host plant species.
Mycelial Growth and Extracellular Enzyme Activities of Wood-decaying Mushroom Strains on Solid Media

The mycelial growths of wood-decaying mushroom strains collected from Korean forests were investigated on solid media under different culture media and temperatures. Most of strains showed the higher mycelial growth on potato dextrose agar (PDA) than malt extract agar (MEA) or sabouraud dextrose agar (SDA) plates. Except for a few strains, they grew well on PDA at
and showed a poor growth at low temperature (
) than high temperature (
). All strains showed the carboxymethylcellulase (CM-cellulase) and laccase activities on solid media containing the specific substrates for two different enzymes.
Screening and Characterization of LTR Retrotransposons in the genomic DNA of Pleurotus eryngii

Transposable elements (TEs) are mobile DNA elements that often cause mutations in genes and alterations in the chromosome structure. In order to identify and characterize transposable elements (TEs) in Pleurotus eryngii, a TE-enriched library was constructed using two sets of TE-specific degenerated primers, which target conserved sequences of RT and RVE domains in fungal LTR retrotransposons. A total of 256 clones were randomly chosen from the library and their insert sequences were determined. Comparative investigation of the insert sequences with those in repeat element database, Repbase, revealed that 71 of them were found to be TE-related fragments with significant similarity to LTR retrotransposons from other species. Among the TE sequences, the 70 TEs were Gypsy-type LTR retrotransposons, including 20 of MarY1 from Tricholoma matsutake, 26 of Gypsy-8_SLL from Serpula lacrymans, and 16 of RMER17D_MM from mouse, whereas a single sequence, Copia-48-PTR, was found as only Copia-type LTR retrotransposon. Southern blot analysis of the HindIII-digested P. eryngii genomic DNA showed that the retrotransposon sequences similar to MarY1 and Gypsy-8_SLL were contained as high as 14 and 18 copies per genome, respectively, whereas other retrotransposons were remained low. Moreover, both of the two Gypsy retrotransposons were expressed in full length mRNA as shown by Northern blot analysis, suggesting that they were functionally active retrotransposons.
Selection of Superior Strains from Collected Ear Mushrooms for Artificial Cultivation and Their Optimal Condition of Mycelial Growth

This study was carried out to obtain morphological and physiological characteristics of ear mushrooms for an artificial cultivation. Eighteen strains were cultivated with bag culture and classified into mainly five groups such as brown, black, white, purple and others group. The highest yield was shown in 43007 strain as 98.3 g/bag and 43009, 43016, 43025 and 44035 strains were more than 60 g/bag. Among collected strains, 43007, 43009 and 43035 were selected in this study as superior strains. Three selected strains were investigated for optimal mycelial growth conditions. MCM and GPYM media were selected for the favorable culture medium. The carbon sources of 43007, 43009 and 43035 on mycelial growth were maltose, fructose and glucose, respectively and peptone was selected as a nitrogen source. Highest mycelial growth was observed when the C/N ratio was 10 for 43007 and 20 for 43009 and 43035.
Infection of Mycovirus in Imported Lentinula edodes

Up to date several mycoviruses including Lentinula edodes Spherical Virus (LeSV) have been reported. As fungal virus was spreaded by infested hypae and spores it could be important to use virus-free spawns to eradicate the mushroom virus disease in the culture farm. We tested the imported spawns of Lentinula edodes by PCR whether LeSV was infested them or not. The primer set targeting the RdRp gene of LeSV was prepared based on partial sequence of the LeSV genome. The RT-PCR analysis showed that 87 among 88 imported spawns of L. edodes were infested by LeSV.
Identification of Fungi Occurred on Silicon of Washbasin

Black molds were occurred on silicon of washbasin in a bathroom in apartment. Blocks of silicon where the fungi grew, were placed on MEA, DG18, DRBC, PDA+PS media, and 18 strains of fungi were isolated. They were identified as Phoma herbarum, Peyronellaea calorpreferens, Cladosporim halotolerans based on morphological and molecular methods. Ph. herbarum and Pe. calorpreferens were isolated with high frequency, but C. halotolerans was isolated with low frequency. The fungal strains were deposited in Korean Agricultural Culture Collection (KACC), and they shall be used for further related researches.
Identification of Fungal Strains Isolated from Salami Casing

The fungi play an important role in smell and taste during the fermentation of salami. In this study, the fungi from the salami casing during the fermentation process in Korea were isolated. They were examined by morphological and molecular methods and were identified as Aspergillus cibarius, Penicillium echinulatum, Cladosporium sphaerospermum. Of these, A. cibarius formed white or light green conidial heads on salami casing and it was widely distributed through almost whole casing. P. echinulatum occurred sparsely on the casing of the salami and formed white masses. C. sphaerospermum occurred rarely and formed black spots. It is considered that additional studies on their roles on salami are required.
Diversity Analysis of Endophytic Fungi Isolated from the Roots of Coastal Plants in Taean Peninsula

Forty-two endophytic fungi were isolated from the roots of coastal plants, such as Aster sphathulifolius Maxim., Aster tripolium L., Phragmites australis, and Puccinellia nipponica Ohwi, naturally growing in Taean peninsula. The identity of the endophytic fungal strains was analyzed using the sequence of internal transcribed spacer regions. All fungi belonged to the phylum Ascomycota, and they were classified into eight orders (Botryosphaeriales, Capnodiales, Diaporthales, Dothideales, Eurotiales, Helotiales, Hypocreales, and Pleosporales) and thirteen genera (Alternaria, Aureobasidium, Cadophora, Cladosporium, Davidiella, Diaporthe, Fusarium, Gibberella, Macrophomina, Metarhizium, Neosartorya, Penicillium, and Phoma). Among the analyzed fungi, the fungi belonging to the genus Penicillium in Eurotiales were the most widely distributed. The host plant Aster tripolium L. was found to contain the most diverse endophytic fungal species among the coastal plants.
Biochemical Characterization of Agaricus bisporus Dikaryon Strains

Button mushroom (Agaricus bisporus) strains from diverse origins were compared in this study to obtain basic information on their growth and biochemical properties that are helpful for breeding. Among 31 dikaryotic strains tested, most strains showed better mycelial growth on oatmeal agar than on MEA and PDA. Mycelia of the mushroom strains revealed
-glucosidase activity the most clearly among the seven extracellular enzymes tested. All the strains showed protease activity, but
-glucosidase activity was found in 27 strains and xylanase activity was found in 30 strains. The activity of avicelase, CM-cellulase, amylase, and pectinase was detected in less than 20 strains. These results implied that enzymatic characteristics could be used as a criterion of strain selection for breeding study.
Occurrence of Fusarium Surface Rot on Sweet Potato Caused by Fusarium commune

In June 2013, surface rot symptoms were observed on sweet potatoes (Ipomoea batatas) in Yeoju city, Korea. The lesions were circular, light to dark brown, firm, dry, and superficial. The diseased area became sunken in older tissues, and the symptoms usually did not extend deeply into the flesh. Seven isolates of Fusarium species were isolated from diseased sweet potatoes. All isolates were identified as Fusarium oxysporum based on morphological characteristics on CLA medium. To confirm the identification, molecular analysis of elongation factor 1 alpha gene was conducted. Among the isolates, however, four isolates were F. commune, and three isolates were F. oxysporum based on the DNA sequence data. Pathogenicity was tested using agar block inoculation on wounded or unwounded sweet potato pieces. Lesions were observed on wounded sweet potato pieces after 7 days inoculation in only F. commune isolates. This is the first report that F. commune causes Fusarium surface rot of sweet potato in Korea.
