Herbal Formula Science, volume 12, issue 2, 2004, Pages 19~30
This thesis aims to provide an analytical basis for existing or new prescriptions based on the theory for properties and tastes of Korean oriental herbal medicines. To this end, prescriptions presented in the Various Changes in the Dominations of the Six Energies and their Relations with the Diseases of Plain Questions were analyzed based on the theory for properties and tastes of Korean oriental herbal medicines in Yellow Emperor's Canon of Internal Medicine. This thesis focused on symptoms and prescriptions based on the theory for properties and tastes of Korean oriental herbal medicines, which were described in the Various Changes in the Dominations of the Six Energies and their Relations with the Diseases of Plain Questions with regard to the year when Taeyang is affecting the earth energy and the cold energy is partially over-abundant. Those symptoms and prescriptions originate from the theory of the five elements and six climates. The symptoms were analyzed from the perspective of physiology. The prescriptions were first analyzed based on the theory for properties and tastes of Korean oriental herbal medicines in Yellow Emperor's Canon of Internal Medicine. Then, a mix formula of oriental medicines pursuant to those prescriptions was studied. Lastly, established prescriptions, which were in conformity with the above prescriptions, were selected. From the physiological point of view, symptoms with regard to the year when Taeyang is affecting the earth energy and the cold energy is partially over-abundant can be, based on the theory of the five elements and six climates, diagnosed as the symptoms from impaired heart due to excessive cold energy. Established prescriptions pursuant to prescriptions based on the theory for properties and tastes of Korean oriental herbal medicines can be broken down into the following: Cassia Twig, White Peony and Anemarrhena Decoction (Gyejijakyakjimotang); Rhemannia Bolus with Eight Herbs (Palmijihoanghoan); Back to the Left Pill (Joaguihoan); Bone-Strengthening Pill (Hojamhoan); Major Eum-Replenishing Pill (Daeboeumhoan). Therefore, symptoms of six kinds of weather presented in the Various Changes in the Dominations of the Six Energies and their Relations with the Diseases of Plain Questions can be analyzed from the physiological point of view. As a result, in addition to the methodology that analyzes existing prescriptions within the boundary of the theory for properties and tastes of Korean oriental herbal medicines in Yellow Emperor's Canon of Internal Medicine, it is expected that a theoretical basis for new prescriptions can be provided by analyzing established prescriptions based on prescriptions from the theory for properties and tastes of Korean oriental herbal medicines.

Wang Cheong-Im(1768-1831) in the Ching dynasty conducted the therapy of blood statis syndrome using the methods for promoting blood circulation and removing blood statis in accordance with the description in the book Uirimgaechak(醫林改錯). Thus, this study examined his methods for promoting blood circulation and removing blood statis, therapy of blood statis syndrome and mechanism of function. Since the blood statis symptoms varied on the parts where the blood statis were occurred, the therapy for promoting blood circulation and removing blood statis applied different treatments and drugs depending on the symptoms. The therapy also valued much of qi and blood. Qi is important to the human body so that the therapy replenished qi and simultaneously promoted the blood circulation as mainly focusing on the regulation of the qi flow. Moreover, the therapy adjusted the level of removing blood circulation and rate of replenishing qi. The degree of prescription and drug was differentiated in treating a variety of blood statis because of the difference in the seriousness and characteristics of blood circulation, amount and application of drug and combination of drugs. The therapy was careful about the regulation of qi for the replenishment of qi and the nurishment of blood for the promotion of blood circulation. If the blood circulation was blocked, the blood statis was formed. Accordingly, the phlegm and the blood statis were closely related. Then, the phlegm and blood statis were simultaneously treated. The therapy divided the properties of blood statis into the stagnant excess syndrome, the stagnant deficiency syndrome, the stagnant cold syndrome and the stagnant heat syndrome and treated the patients suitable for each symptom. The function of mechanism in the treatment using the methods for promoting blood circulation and removing blood statis was divided into the stagnation of blood and interruption in blood circulation. The therapy laid stress on promoting blood circulation and removing blood circulation. With such emphasis, the therapy facilitated the circulation in the bloood vessel, prevented the coagulation of blood, removed the blood statis and promoted the metabolism. Then, the physiological function and pathological change internal organs were improved. Furthermore, the therapy strengtened the heart and promoted the blood circulation by improving the systemic blood circulation. Moreover, the therapy facilitated the micro-circulation by adjusting the balance of body.

Depression is a sort of mental disorder which is very common. To treat depression, many drugs such as TCA, MAOI are developed and used. But they have a lot of side effects, so it needs to develop drugs without side effects or with less side effects. Herbal medicines have been used to treat diseases not only physical but also mental and have less side effects. therefore, it has been thoght the need to develop herbal medicine with antidepressant effect. The purpose of this study was to reseach antidepressant effect and influence on monoamines of chunwangboshimdan thought to have antidepressant according to ancient medical book- donguibogam- and recent reports. We used 'forced swimming test(FST)' to know antidepressant effect of chunwangboshimdan and HPLC to check the influence on monoamines and their metabolites(norepinephrine, dopamine, DOPAC, HVA, serotonin, 5-HIAA) of chunwangboshimdan after divided into cerebral cortex, striatum, hypothalamus and hippocampus. The results were obtained as follows: In the study of antidepressant effect by 'forced swimming test(FST)'method, chunwang boshimdan had a significant antidepressant effect. In the study of influence on monoamines by HPLC, chunwangboshimdan mainly increased dopamine among monoamines and their metabolites(norepinephrine, dopamine, DOPAC, HVA, serotonin, 5-HIAA) significantly in 4 parts of rat's brain above-mentioned. Calculated by turnover ratio formulae of monoamine, chunwangboshimdan has more results than Imipramine. These results suggest that chunwangboshimdan has antidepressant effect that is related with the increase of monoamines by suppressing their metabolism as its mechanism.

This study was carried out to investigate the liver protective effects of extracts from Persicae Semen (WT-003, WT-005, WT-006). The acute hepatic injury was induced by intraperitoneal injection of alpha-naphtylisothiocyanate (75 mg/kg, p.o.) and treated with WT-006 (100 mg/kg/day, 200 mg/kg/day, 400 mg/kg/day). The experimental hepatic fibrosis was induced by bile duct ligation／scission(BDL/S), duration of 4 weeks and treated with WT-003, WT-005 or WT-006 (200 mg/kg/days for 4 week). In acute liver injury, WT-006 (200 or 400 mg/kg) lowered serum alanine transferase(ALT) and aspartate transferase(AST) significantly. In fibrotic rats, WT-006 treatment inhibited the hydroxyproline deposition in liver and lowered serum AST, ALT and ALP, significantly. These results suggest WT-006 extract, which does not contain amygdalin, from Persicae Semen have liver protective and antifibrotic effects in rats.

In order to evaluate the anti-tumor and synergic effect of Soonkiwhajungtang with doxorubicin, the inhibitory concentration(IC),
of single use of doxorubicin and Soonkiwhajungtang with their concomitant treatment against Colon-26(Murine Rectum Carcinoma) was observed using MTT(Microculture Tetrazolium test) assay. In addition, their anti-tumor effects were also observed in the xenograft nude mice models agianst to Colon-26 cell lines. Soonkiwhajungtang has only mimic direct anti-tumor effect against to Colon-26 cell lines but they were decreased general depressed signs induced by implantation of tumor cell lines and increased the total WBC and lymphocyte numbers. So, it is considered or expected that Soonkiwhajungtang extracts were reduced the critical toxicity of doxorubicin and shows favorable synergic effect with doxorubidn and Soonkiwhajungtang extracts.

Chelidonii Herba (CHE, Baek-gul-chae in Korean), which has its original description in Gu-Hwang-Bon-Cho, a classic book of oriental Herbal book, is widely used in the treatment of stomach cancer, jaundice, gasrtic ulcer, edema and stomach pain, in Korea, Japan and China. The present study was conducted to evaluate the effect of CHE on the nitric oxide (NO) production, iNOS and COX-2 expression in lipopolysaccharide - activated Raw 264.7 cells. After the treatment of CHE, NO production was monitored by measuring the nitrite content in culture medium, cell viability was measured by MIT assay. COX-2 and iNOS were determined by lmmunoblot analysis. The production of nitric oxide was significantly inhibited by pretreatment (1h) with CHE (0.1-0.3 mg／ml) on LPS-activated Raw264.7 cells. The expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase 2 (COX-2) protein were up-regulated by LPS, but the increased levels of iNOS and COX-2 were inhibited by pretreatment of CHE (0.1-0.3 mg／ml), respectively. Thus, the present data suggest that CHE may play an important role in adjunctive therapy in Gram-negative bacterial infections.

Nitric oxide(NO) play an important role in normal and pathophysiological cells including as a messenger molecule, neurotransmitter, microbiocidal agent, or dilator of blood vessels and artheriosclerosis, hypertension, myocardial infarction, respectively. To investigate that Ondamtang in the potential contribution of the levels of nitric oxide generated by endothelial nitric oxide synthase (eNOS) and the mechanisms of protection against L-NAME, human ECV304 cells, which normally do not express eNOS, were expressed by L-NAME. L-NAME stimulated rat or cells were found to be resistant to injury and delayed death following the Ondam-tang. Inhibition of nitric oxide synthesis abolished the protective effect against L-NAME, thrombin and collagen exposure. Interestingly, such effects have bee observed during stimulation with agents such as KCl on L-NAME mediate rats, were damaged by the NOS inhibitor NG-nitro-L-arginine methyl ester (L-NAME). Cardiovascular diseases is one of the blood vessels and renin-angiotensin system dynfunction. So we studied on herbal medicine that have a relation of vessels endothelium necrosis. In Oriental Medicine, Ondam-tang has been used for disease in relation to cardiovascular system. We studied on the protection and inhibitory effects of cardiovascular diseases in L-NAME induced rat or ECV304 cell lines through the Cell morphological pattern, Tunel assay, LDH activity, heart rate, blood pressure and immunohistochemistric analysis by Ondam-tang. As the result of this study, In group, the anti-apoptosis and necrosis in the cardiovascular system have a potential capacity for prevented, protected and treating the diseases of cardiovascular system, against the necrosis of rat and ECV304 cells with eNOS and calpain expression by L-NAME is promoted.
