A Study of Okchun-San Formulation for Freeze Drying Capsulation

Objective : Okchun-San(OCS) is known as an effective herbal medicine on Type 2 diabetes. We performed to change OCS formulation for freeze drying capsulation. Methods The mixtures of OCS were extracted with water. finally, the filtered solution were evaporated and lyophilized to dry granules. The various ratio of excipients were studied to determine the formation for capsulizing. The samples were inspected for any difference in color, taste and appearance. Results: The prepared form of OCS were dried and weighted 260kg. The lyophilized dry powder yielding 40kg. The suitable ratio of OCS-dry powder and excipient was 10:1. The average weights of On and placebo capsules were
, respectively. There was no notable change in color and appearance for both capsuled samples throughout the study period. Conclusions: Therefore in can be concluded that freeze drying capsulation is appropriate form of OCS.
Protective effect of injinhotang and its components on acetaminophen-induced hepatoxicity in rats

Acetaminophen, which causes acute liver min in humans and animals, has made useful inducer of hepatoxicity for studying hepatopreventive drugs. Injinhotang is known as one of the hepatopreventive drugs. However, its mechanism of recovery of hepatoxicity treated with acetaminophen is poorly understood. this study was performed to observe the antioxidative effect of injinhotang extract and its several combination groups. The results were obtained as follows:1. In the study on free radical scavenging effect in vitro(the suppressing effect on peroxidation of linoleic acid on concentration, the scavenging effect of DPPH radical, inhibitory effect of superoxide in xanthine-xanthine oxidase system and the inhibitory effect on lipid peroxidation reaction by hydroxy radical in H2O2-Fe2+system, injinhotang have more effect than its components groups relatively. 2. In the study on antioxidants system in vivo(the level of serum LPO, the level of hepatic LPO, catalase, GSH, GST), only injnhotang has a significant effect. 3. In the study on hepatotoxicity(GOT, GPT,
-GTP, ALP, LDH, b ilirubin), only injinhotang has a significant effect. These results suggest that injinhotang has the protective effect on acetaminophen-induced hepatoxicity. The mechanisms of these are supposed to be involved in antioxidant and three drugs' cooperative synergy effect.
Effect of the neuroprotetion and anti-Alzheimer's disease in CT99-induced PC12 cells by Gakamgobonhwan water extract

Alzheimer's disease(AD) is a geriatric dementia that is widespread in old age. In the near future AD will be the biggest problem in public health service. It has been widely believed that
peptide devided from APP causes apoptotic neurotoxicity in brain. However, recent evidence suggests that n99 may be an important factor causing neurotoxicity in AD. Mouse PC12 cells expressed with n99 exhibited remarkable apoptotic cell damage. We invesgated the protective effects of Gagamgobonhwan water extract(GKG). Findings from our experiments have shown that GKG inhibits the activities of CT99, which has neurotoxicities and apoptotic activities in cell line. In addition, treatment of GKG(
24 hours) partially prevented CT99-induced cytotoxicity in PC12 cells. As the result of this study, in GKG group the apoptosis in the nervous system was inhibited, the repair against the degerneration of PC12 cells by CT99 expression is promoted. Taken together, GKG exhibited inhibition of CT99-induced apoptotic cell death. GKG may be beneficial for the treatment of AD.
The Experimental Study of Sunkihwalhyul-Tang against Inhibitive Effects on the Brain Ischemia

This Study was designed to investigate the effect of Sunkihwalhyul -Tang extract(SHT) on the change of cerebral hemodynamics [regional cerebral blood flow(rCBF), pial arterial diameter(PAD) and mean arterial blood pressure(MABP)] in normal and cerebral ischemic rats, and further to determine the mechanisms of action of SHT on hemodynamics. In addition, this study was designed to investigate whether SHT inhibits lactate dehydrog enase(LDH) activity in neuronal cells and cytokines production in serum of cerebral ischemic rats. The results were as follows 1. SHT significantly increased rCBF and PAD in a dose-dependent manner, but MABP was not changed by injecting SHT. These results suggest that SHT significantly increases rCBF by dilating PAD. 2. The SHT-induced increase in rCBF was significantly inhibited by pretreatment with indomethacin(IDN, 1 mg/kg, i.p.), an inhibitor of cyclooxygenase and methylene blue(MTB,
, i.p.), an inhibitor of guanylate cyclase. 3. The SHT-induced dilation in PAD was significantly inhibited by pretreatment with IDN and MTB. 4. The SHT-induced some increase in MABP was significantly increased by pretreatment with IDN. These results suggest that the mechanism of action of SBT is mediated by guanylate cyclase. 5. Both rCBF and PAD were significantly and stably increased by SHT(10 mg/kg, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. 6. SBH significantly inhibited LDH activity in neuronal cells. These results suggest that SHT prevents the neuronal death. 7. In cytokine production in the senlm drawn from femoral artery 1 hr after middlecerebral arterial occlusion, sample group showed significantly decreased production of IL-1
production, decreased production TNF-
and increased Production of IL-10 compared with control group. 8. In cytokine production in the serum drawn femoral artery 1 hr after reperfusion, sample group showed significantly decreased production of IL-1
as wellas significantly increased production of IL10 compared with control group. These results suggest that SHT mediated by guanylate cyclase has inhibitive effect on the brain damage by inhibiting LDH activity, IL-1
production, and by accelerating IL-10 production. The present author thinks that SHT has an anti-ischemic effects through the improvement of cerebral hemodynamics and inhibitive enects on the brain damage.
The Effects of Chelidonii Herba Extract on Apoptosis in Human Stomach Adenocarcinoma Cell Line

Chelidonii Herba (Baekgulchae in Korean: CHE), a commonly used herb in Korea, Japan and China, is widely used in the treatment of stomach cancer, jaundice, gastric ulcer, edema and pain of stomach. In the present study, we demonstrated that CHE induces apoptosis in AGS cells, human stomach adenocarcinoma cell line. One of the most important recent advances in cancer research is the recognition that apoptosis plays a major role in both tumor formation and treatment response, In this study, CHE caused a decrease of viability in AGC cells. When AGS cells were treated with CHE, cells showed dose-dependent manner apoptotic cell death. Increased apoptotic cell death, exposured to CHE, resulted from induction of Bad translocation to mitochondria, cytochrome-c release from mitochondria to cytosol, activation of caspase-3, 8, 9, and PARP cleavage. These results suggest that CHE may be potential therapeutic approach in the clinical management of stomach adenocarcinoma.
Study on Antihyperlipidemic Effect of Kamiondamtang

Kamiondamtang(KODT) has been used in oriental medicine as stress by many medical practitioner for the study of KODT, we had fed mice divided to three groups(basal diet, hyperlipidemic diet, hyperlipidemic diet + KODT), and observed the change of weight, total cholesterol, triglyceride, SGOT, SGPT, HDL-cholesterol, LDL-cholesterol on the serum per every 7days for 6weeks. To help comparison with the results above, we had tested endothelial cells and liver. The results of this Study were obtained as fallows ; 1. Total cholesterol, triglyceride and LDL-cholesterol were decreased significantly by KODT. 2. HDL-cholesterol was increasd significantly by KODT. 3. SGOT was intended to decrease by KODT, 4. %W was decreased significantly by KODT. 5. In case of supplying KODT, WBC attatched on endothelial cells and vacuoles in muscular layer were not observed. 6. In case of supplying KODT, fatty degeneration was not observed in liver portal area.

Base on these findings, IGA may be beneficial for the treatment of AD.
Studies on the Liver Toxicities with different Dosage of Wild Aconiti Tuber Decoction

Objective : This study was carried out to evaluate the liver toxicities of Wild Aconiti Tuber decoction. Methods : The amounts of aconitine in the methanol extract of Wild Aconiti Tuber was measured by HPLC. Safeties was studied by LD50 in mice. Liver toxicities were evaluated histologically and by CBC, blood chemistry after 2 weeks of 0.4g/kg/day clinical dosage oral administrations in rat. Results : 1. The amounts of aconitine in the methanol extract of Wild Aconiti Tuber is
. But aconitine was not detected in the water decoction of Wild Aconiti Tuber. 2. To evaluate LD50 and safeties of Wild Aconiti Tuber decoction, ICR mice were given high dose of 2, 5, 10g/kg for single time and were observed for 2 weeks. There were no dead animal and abnormal clinical sign and no abnormalities at the autopsy. So, LD50 was admitted to higher than 10g/kg. 3. After 2 weeks of 0.4g/kg/day clinical dosage oral administrations in rat, there was no significant change in the CBC and blood chemistry. 4. In the liver tissues of clinical dosage, mitotic figures, apoptosis and individual cell death were observed, but clear liver toxicities like fatty liver or necrosis were not observed. the liver tissues of high dose in mice, hydropic changes were getting severe as dose grows. Conclusions : According to the results, though aconitine was not detected in the Wild Aconiti Tuber decoction, 0.4g/kg/day 2 weeks p. o (clinical dosage) group showed weak changes in the liver tissues and high dose group showed liver toxicities like hydropic changes.
The Effects of Juglandis Semen Extract on the Experimental Asthma induced by Ovalbumin

Objective : Juglandis Semen has a function that to invigorate the lung and kidney. And It is commonly used as a supporting agent in the treatment of coughing and bronchitis. This study was performed to investigate the effect of oral administration of Juglandis Semen Extract (JSE) against the experimental asthma induced by ovalbumin. Methods : Asthma was induced to Balb/c mouse by i.p. injection and aerosol immunization with ovalbumin. It was observed the change of the cell number in the BAL fluid. Concentrations of IL-4, IL-5 in splenoc yte were assessed by ELISA, IgG and IgE from serum were calculated by same method. Results : 1. Number of macrophage in BAL fluid was significantly decreased in JSE group compared with control group, but not eosinophil and lymphocyte. 2. Levels of IgG and IgE in serum were significantly decreased in JSE group compared with control group, respectively. 3. Concentration of IL-4 in culture supernatant of splenocyte was significantly decreased in JSE group compared with control group, but there was no significant in IL-5. Conclusion : We found that the effect of JSE extract in asthma was implicated in reduction of IL4released from Th2 cell, and decreases of IgG and IgE from plasma cell. These findings suggest that JSE can produce anti-asthmatic effect, which may play a role in allergen-induced asthma therapy.
A study on Applications of prescriptions including Fructus Schisandra as a main component in Donguibogam

This report describes 47 studies related to the use of Fructus Schisandra main blended prescriptions from Donguibogam. The following conclusions were reached through investigations on the prescriptions that use Fructus Schisandra as a key ingredient. 1.34% of a cough, 10.6% of a consumptive disease, recorded the largest number of clinical frequency of the prescriptions in therapeutic use when Fructus Schisandra was taken as a monarch drug in prescriptions 2. Prescriptions that utilize Fructus Schisandra as the main ingredient are used in the treatmeant of a cough, a consumptive disease, an exogenous febrile disease, a carbuncle, and cellulitis, and they are also used for treating 11 different types of diseases. 3. The prescriptions are compounded with Fructus Schisandra as a monarch drug can apply to a deficiency syndrome of the lung a deficiency syndrome of both the lung and the stomach, a deficiency syndrome both the spleen and the lung a deficiency syn-drome of the kidney, a hypofunction of the bladder with cold syndrome, a cold of insufficiency type, a deficiency syndrome of the heart, a heat syndrome of the stomach, an affective by cold, an invasion by wind, a consumptive disease. 4. The dosage of Fructus Schisandra is 5pun(about 1.88g) to 5jeon(about 18.75g), however 1jeon(about 3.75g) has been taken the most for clinical application. 5. When Fructus Schisandra is combined with base prescriptions such as Ijintang Chungliongsan, Saengmaksan, it applies symtoms of cough. In addition, when Fructus Schisandra is combined with base prescriptions such as Liukmizihwangtang, Ssangbohwan, Sipjeondaebotang, it utilizes a consumtive disease.
Studies on Application of Perilla frutescens Main Blended Prescription in Donguibogam

This study was investigated to make sure the usage of prescriptions in which Perilla frutescens is used as a main herbal medicine in Donguibogam. Through the investigation on main treatment, pathology, etiology and nature of disease, dosage in the relevant prescriptions, The consequences are as follows; 1. The Perilla frutescens is used in 15 treatment fields which contain the cough, cold chapter ect. 2. The Perilla frutescens is used as a main heral medicine in 30 natures of disease such as cough-relative disease, an asthma, an external wind and cold, etc. 3. The Perilla frutescens is used for various pathology in the exogenous etiological factor, endogenous etiological factor, exo-endogeneous etiological factor and non-exo-endogeneous etiological factor. 4. The Perilla frutescens is used in a range of
in relevant prescription. The main dosage is 4g. 5. The Perilla frutescens is used with various crude herbs in accordance with the pathogeny. The Perilla frutescens has been used to reduce heat from the lungs, to relieve asthma, to promote qi's movement and to soothe fetal movement etc. According to the results, 1 suggest to use the Perilla frutescens in a various pathogenic fields. The Perilla frutescens is able to remove not only pathogenic heat from the cough-relative disease, but also pathogenic asthma, an external winds and colds, etc.
Study on Application of Lignum akebiae Main Blended Prescription from Dongeuybogam

This report describes the remedial fields, symptoms, pathology, dosage, prescriptional constitution of 46 studies related to the use of Lignum akebiae main blended prescriptions from Dongeubogam. The following conclusions were reached through investigations on the prescriptions that use Lignum akebiae as a key ingredient. Lignum akebiae blended prescriptions are utilized 17.4% of prescriptions appear in the chapter of urine. Prescriptions that utilize Lignum akebiae as the main ingredient are used commonly in the treatment of the symptoms of a urine and a cystic disease related to the urinary organs, or the diseases of eye, ear, nose, throat and the symptoms of a tumor, abscess. Lignum akebiae is utilized for an etiological and pathologenic fators related to heat, dampness, phlegm and summer heat such as excessive fever of heart and bladder, fever of small intestine, lower energizer dampness and heat, fever due to blood deficiency, phlegm fire, affection by summer heat and dampness etc. The clinical dosage of Lignum akebiae is 1 don(about 3.75gram), according to symptoms, it has ranged at a wide variety of amounts from 3 pun(about 1.13gram) to 1.5 don(about 11.25gram). Lignum akebiae is widely applied with base prescriptions such as Dozuksan, Moktongchajuntang, Manjunmoktongtang. etc.
A Clinical Report of Patient of Autoimmunity disease treated with Gwackryungtang

About the case of the patient who has diagnosed Systemic Lupus Erythematosus and admissed to our hospital from 27. January. 2005 to 28. February 2005, we could controll by herb medications userd differently patients-to-patients. SLE does not exactly correspond to any specific category of oriental medicine. But, according to previous reports, it can be controlled by oriental differential diagnosis of symptoms and signs. Because we diagnosed and treated her as at the point of Oriental Medicine and got the improvement, so we report. We think that the patient must get the screen test exactly and treat the disease proper ly, and if we apply this result to clinical cases at the point of Oriental Medical base from gathering and researching more cases, it will arouse sympathy-the excellence of Oriental Medicine and make the necessity of the further research and report from now on.
A Clinical Study on the Postpartum Depression Treated by Gami-Boheo-tang

As woman receive many stress during pregnancy and delivery, they happen to contract to psychopathy like a postpartum depression. Postpartum depression is a serious problem if they are not treated early enough the unhappy seriousness can be resulted. A 32-yeared-old female patient with postpartum depression was taken to Dongshin University oriental medical hospital on April, 28, 2003. We diagnosed this patient as postpartum depression, which originated from and efficiency of vital energy and blood. We treated this postpartum depression patient with herb medicine(Cami-Boheo-tang),acupuncture, moxibustion, and psychical therapy. After the oriental medical treatments, the patient was improved in all complains of the postpartum disease. After this, further study and approach will be needed about the disease of the postpartum depression.
A case report of postpartum patient with panic disorder

Purpose The purpose of this study is to report the effect of oriental treatments for the postpartum patient who is diagnosed as panic disorder. Methods : The patient had been in admitted to Dongshin Oriental Hospital from 2004. 10. 28 to 2004. 11. 27 This patient was treated with oriental methods such as herb medication, acupuncture, moxibustion therapy etc. Results : As a result of oriental treatments based on planing treatment according to diagnosis, the symptom of postpartum disease with panic disorder was improved. Conclusion : It is thought that the clinical study about the treatment of the postpartum disease with panic disorder based on planing treatment according to diagnosis, should be required.
