Cryptonemia rotunda (Halymeniales) and Schizymenia apoda (Nemastomatales), two new records of red algae from Korea

Here we report the first finding of Cryptonemia rotunda and Schizymenia apoda in Korea based on plastid rbcL sequence analysis and morphological observations. C. rotunda occurred in the subtidal in Hongdo and Namyeodo on the south coast. Thalli have short stipe with foliose, membranous blades with dentate margin, a length of 2-8 cm. S. apoda occurred in many locations along the coast of Korea. Thalli are foliose, simple or broadly lanceolate blades with irregular lobes and proliferations from the margins, sometimes undulate, soft fleshy, and slippery, up to 32 cm tall, and have cystocarps with ostioles on the blade. Plastid rbcL sequences reveal that each of these species is clearly separated from other species of their respective genera. S. apoda was well resolved within the genus. However, C. rotunda was distantly related to other members of the genus, and this result needs an urgent revision of the genus.
Seasonal variation in kelp phlorotannins in relation to grazer abundance and environmental variables in the Alaskan sublittoral zone

Phlorotannins are common metabolites produced in kelps that can have deterrent functions against grazers. The factors dictating seasonal patterns of phlorotannin content in northeastern Pacific kelps are not well understood. This study assessed density and grazing of the gastropod Lacuna vincta on the annual canopy-forming kelp Nereocystis luetkeana and the perennial understory species Agarum clathratum, Saccharina latissima and S. groenlandica in Kachemak Bay, Alaska. In addition, we assessed seasonal patterns of environmental variables as possible drivers of phlorotannin concentrations. Phlorotannins occurred in all species, with overall lowest levels in N. luetkeana, and with different seasonal patterns among the four species. Lacuna vincta was most dense on N. luetkeana thalli in the summer and had highest grazing rates on this low-phlorotannin species. However, correlations between L. vincta density and phlorotannin content of each kelp species were not significant. Except for N. luetkeana, there were no correlations between phlorotannin levels and environmental variables. We suggest that kelp life history traits may be more important for phlorotannin patterns in these kelp species than grazers or environmental drivers.

We report an oxidative burst triggered by prostaglandin
in the brown algal kelp Laminaria digitata, constituting the first such discovery in an alga and the second finding of an oxidative burst triggered by a prostaglandin in a living organism. The response is more powerful than the oxidative burst triggered by most other chemical elicitors in Laminaria. Also, it is dose-dependent and cannot be inhibited by diphenylene iodonium, suggesting that another source than NAD(P)H oxidase is operational in the production of reactive oxygen species. Despite the very strong oxidative response, rather few effects at other levels of signal transduction pathways could be identified.
does not increase lipolysis (free fatty acids) in Laminaria, and only one oxylipin (15-hydroxyeicosatetraenoic acid; 15-HETE) was found to be upregulated in Laminaria. In a subsequent set of experiments in the genome model Ectocarpus siliculosus, none of 5 selected candidate genes, all established participants in various stress responses, showed any significant differences in their expression profiles.
Population ecology of Palmaria palmata (Palmariales, Rhodophyta) from harvested and non-harvested shores on Digby Neck, Nova Scotia, Canada

Population ecology of Palmaria palmata is described from the intertidal zone of Digby Neck and adjacent islands of Nova Scotia. The primary objectives were: to evaluate the difference in habitat specialization and population structure of P. palmata between harvest and non-harvest shores, and to characterize differences in thallus structure and frond sizes between epilithic and epiphytic populations. Harvest shores were gently sloping boulder fields with boulders typically about 0.5-1.0 m with dense cover of P. palmata on many of the rocks. Non-harvest shores (with or without P. palmata) consisted of boulders that were smaller or larger than harvest shores, or bedrock; when P. palmata was present on nonharvest sites it was typically epiphytic on other algae (e.g., Fucus spp., Mastocarpus stellatus, Devaleraea ramentacea). Harvestable epiphytic populations occurred only in high current areas. While there was little difference in average cover of P. palmata harvest and non-harvest shores (
standard deviation [SD]), the cover of P. palmata on harvest shores was highly skewed such that individual boulders often had >90% cover while adjacent rocks had little. Frond length of large fronds was greater on harvested shores, and mean frond density (
) was three times higher than the mean density on the non-harvested shores. Frond lengths of entire epiphytic and epilithic frond complements of 119 thalli from harvest beaches showed no difference in mean size of the largest fronds, and no difference in frond number per holdfast when epiphytic and epilithic thalli were compared.

A mathematical model of the commercial harvest of Palmaria palmata (Dulse) is presented based on a logistic model and field data collected on Digby Neck, Nova Scotia from 14 harvested shores during May to August, 2010. Field observations used to estimate model parameters included cover of Dulse before and after harvest from Dulse dominated boulders for which surface area was estimated, and from which fresh biomass of harvested Dulse was weighed. Over all the surveys the average harvest fraction was about 50%, and the total resource was about
. With growth rates in excess of 4% per day and a 50% harvest of the standing crop each month, the model suggests that the Dulse resource is sustainable at current harvest levels.
Molecular characterization of a lectin, BPL-4, from the marine green alga Bryopsis plumosa (Chlorophyta)

A novel lectin specific to N-acetyl-D-galactosamine as well as N-acetyl-D-glucosamine was isolated from Bryopsis plumosa and named as BPL-4. Sodium dodecyl sulfate polyacrylamide gel electrophorese (SDS-PAGE) and matrix-assisted laser desorption / ionization-time of flight (MALDI-TOF) mass spectrometry data showed that this lectin was a monomeric protein with molecular weight 12.9 kDa. The N-terminal amino acid sequences of the lectin were determined by Edman degradation and the full cDNA sequence encoding this lectin was obtained using the degenerate primers designed from the amino acid sequence. The size of the cDNA was 414 bp containing single open reading frame (ORF) encoding the lectin precursor. The homology analysis showed that this lectin might belong to H lectin group. BPL-4 showed high sequence similarity (60.6%) to BPL-3, which is a previously reported lectin from the same species. The comparative analysis on the lectin's primary structure showed two conserved domains including one possible active domain of H lectin group.
Evaluation of the sub-lethal toxicity of Cu, Pb, bisphenol A and polychlorinated biphenyl to the marine dinoflagellate Cochlodinium polykrikoides

Algae are sensitive to a wide range of pollutants, and are effective bioindicators in ecotoxicity assessments. Here, we evaluated the sub-lethal sensitivity of the marine dinoflagellate Cochlodinium polykrikoides upon exposure to copper (Cu), lead (Pb), bisphenol A (BPA), and Aroclor 1016 (polychlorinated biphenyl, PCB). Toxic effects were assessed by observations of the reduction in cell counts and chlorophyll a levels after exposure to each toxicant. C. polykrikoides displayed dose-dependent, sigmoidal responses when exposed to the tested chemicals.
-72 h values for Cu, Pb, BPA, and PCB were 12.74, 46.70, 68.15, and
, respectively. PCB, which is an endocrine-disrupting chemical, was the most sensitive, proving its toxic effect on the dinoflagellate. This study provides baseline data on the toxic effects of commonly used heavy metals and endocrine-disrupting chemicals to a marine dinoflagellate.
