Alpha-glucosidase Inhibition Activity of Methanol Extracts Obtained from Nine Pteridophyte Species Native to Korea

This study was conducted not only to analyze
-glucosidase inhibition activity with fronds and rhizomes of nine Pteridophyte species, but also to select the plant materials suitable for natural
-glucosidase inhibitor. Harvested rhizomes and fronds were washed, freeze-dried and grinded. After conducting ultrasonification extraction for 30 minutes in ultrasonic water tank with 100% methanol solvent, and vacuum filtration,
-glucosidase inhibition activity was measured. Acarbose was used as the positive control. After mixing

-glucosidase enzyme solution into
of extract and reacting them at

). The extracts of fronds and rhizomes showed higher value than acarbose by 1.55~100.98 and 6.87~109.33 times each. Especially,
-glucosidase inhibition activities of Pyrrosia lingua in fronds and Osmunda cinnamomea var. fokiensis in rhizomes were the highest. The necessary biomass of fronds and rhizomes for inhibiting 50% of
-glucosidase activity showed the lowest value, 0.35, 0.27 mg each, in O. cinnamomea var. fokiensis.
value of P. lingua was the highest among fronds of nine Pteridophyte species, but content of soluble solids was 2.4 times less than O. cinnamomea var. fokiensis. So frond of O. cinnamomea var. fokiensis is more economic in comparison with P. lingua. As the result of this study, O. cinnamomea var. fokiensis showed high
-glucosidase inhibition activity even with small biomass. Therefore it was considered to be high-valued economic material as natural
Improving Effects of Platycodon Extracts Jelly on β-amyloid-induced Cytotoxicity and Scopolamine-Induced Cognitive Impairment Animal Models

The aim of this study was to examine improving effect of Platycodon extracts (PE) and/or Platycodon extracts jelly (PEJ) on cognitive impairment in vitro and in vivo. PC12 (Pheochromocytoma) cells were pretreated with PE for 1hr and than incubated with
for additional 48hr. Cell viability was assessed by WST-1. Animals for Morris water test and passive avoidance test were divided into normal, control and two Platycodon extracts treated groups that were named Normal (n=7), Control (0 mg/kg, n=7), PE (300 mg/kg, n=7), PEJ (10 g/kg, n=7). Cognitive impairment was induced by scopolamine (1 mg/kg/body weight, i.p.) in the three experimental groups but not the normal group. Pretretment of PE (0.01-1 mg/mL) were not induced cytotoxicity but observed in high dose-treated group (5 and 10 mg/mL) in PC12 cells. Protective effects of PE against
-induced cytotoxicity were increased in dose dependent manner in PC12 cells. Administration of PE and PEJ were significantly reduced escape latency time on Morris water maze test and passive avoidance test in copolamine-induced cognitive impairment animal model. These results suggest that Platycodon extracts and its related product available to ameliorative purpose for cognitive ability impairments.
Evaluations on Antioxidant Effect of Methanol Extract from Immature Cotton Boll

The results of the content of total polyphenol and flavonoid, DPPH (1-1-diphenyl-2-picryl-hydrazyl) radical scavenging activity, ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)] radical scavenging activity, nitrite scavenging activity and SOD-like activity of methanol extracts from immature cotton boll are follows. The contents of total polyphenol and flavonoid compound were higher in small size cotton boll, and DPPH and ABTS radical scavenging activity also showed a relatively high activity in the small size. These results indicate that there is a correlation between phenol content and DPPH radical scavenging, ABTS radical scavenging. The test concentrations of immature cotton boll extract for measuring DPPH and ABTS radical scavenging activities were set as 1.25, 2.5, 5, 10 and 20 mg/ml. Immature cotton boll has high radical scavenging activity at the concentration of 1.25~20 mg/ml and the result showed a tendency to increase in a concentration-dependent. The nitrite scavenging activity showed high activity in the pH 1.2, and the result in the pH 4.2 showed progressively less active, and in the pH 6.0 near neutral was confirmed that does not affect the nitrite scavenging. In addition, SOD-like activity showed somewhat lower activity compared with ascorbic acid, but tended to be higher when compared with the results of the other natural substances. Through this experiment, we confirmed that immature cotton boll was excellent antioxidant activity. Therefore, it is demonstrated that the cotton suggest the possibility of development of new material for cosmetic product or functional food in the future, and is expected to make a greater usability.
Comparative Study of Native Flowers for Anti-oxidative Effects in Korea

Nine kinds of flowers were selected by its antioxidative activities evaluated. DPPH(1,1-diphenyl-2-picryl hydrazil), reducing power, total phenol contents, total flavonoid contents, and antimicrobial activity inhibitory effects of nine natural flower varieties were examined using ethanol extract (80%, v/v). DPPH radical scavenging of Agastache rugosa (fisch.&Mey.) kuntze (
) and solidago virga-aurea var. asiatica (
) showed higher antioxidant activity compared with those of the other varieties. Reducing power of Agastache rugosa (fisch.&Mey.) kuntze (
) had higher antioxidant activity. Agastache rugosa (fisch.&Mey.) kuntze showed the highest content of total phenol (134.6 mg GAE/g). However, total flavonoid (554.6 mg QE/g) exhibited the lowest. These results suggest that nine kinds of flower with 80% ethanol extracts have significant antioxidant activity.
Screening of Useful Plants with Anti-inflammatory and Antioxidant Activity

This study was conducted to select some useful plants as functional material candidates. A total of 38 plants were preliminarily screened for the anti-inflammatory and antioxidant activities. The preliminarily selected 8 plants were further investigated to verify the in vitro inhibitory effect on inflammation and oxidative stress. Boehmeria platanifolia (root), Carpinus coreana (branch), and Eupatorium japonicum (leaf) inhibited the expression of inducible nitric oxide synthase (iNOS) in lipopolysaccharide (LPS)-treated RAW 264.7 cells. Eupatorium japonicum (leaf) suppressed the expression of cyclooxygenase-2 (COX-2), whereas Boehmeria platanifolia (root) and Prunus yedoensis (branch) inhibited the transcription of nuclear factor-kappa B (NF-
). Treatment with the extracts (
) of Abutilon theophrasti (leaf, flower/seed) and Hemistepta lyrata (stem) did not show toxicity on RAW 264.7 cell proliferation, but treatment with
of Boehmeria platanifolia (root) exhibited cell toxicity. Carpinus coreana (branch) and Prunus yedoensis (branch) showed potent scavenging activities on peroxynitrite. Akebia quinata (flower), Carpinus coreana (branch), and Prunus yedoensis (branch) effectively inhibited reactive oxygen species (ROS). Abutilon theophrasti (leaf), Boehmeria platanifolia (root), Carpinus coreana (branch), and Eupatorium japonicum (leaf) exhibited strong inhibitory capacity with regard to nitric oxide (NO) production. The results suggested that Abutilon theophrasti (leaf) has in vitro anti-inflammatory and antioxidant activities, and that is a useful functional material candidate.
Studies on the Production and Optimal Drying Condition of Curcuma longa L.

We investigated the optimal cultivation system and dry processing method of rhizomes from Curcuma longa L. in Suncheon, the southern area of Korea. The experimental results are summarized as followings. Curcma longa rhizomes were transplanted on Apr. 30th, May 10th, and May 20th. The plant height, stem diameter, number of leaves and yield were highest in transplanted on May 10th. The fresh rhizome weight of Curcma longa was the heaviest in November 20 and the optimal dry temperature was
. In sensory evaluations, there were significant differences among the Curcuma longa pills in terms of flavor, color, color and texture and overall acceptability, and honey added groups received significantly higher scores than the other groups.
Taxonomic Identity of Echinochloa crus-galli (L.) Beauv. var. crus-galli in Dokdo

Molecular study were conducted to evaluate taxonomic identities of Echinochloa crus-galli (L.) Beauv. and Echinochloa crus-galli var. echinata (Willd.) Honda in Dokdo. Echinochloa crus-galli complex of two species 26 individuals analyse based on nuclear ribosomal DNA (ITS region) and cpDNA (trnH-psbA, trnL-F). At a result, two species were same sequence. Characters the length of the lemma and the length of the awn traits were identity of the species was unclear. According to, Taxonomy treatments that is based on existent morphological characters should thinks again. On the other hand, in the case of ITS, Echinochloa crus-galli (L.) Beauv. and Echinochloa crus-galli var. echinata (Willd.) Honda at the Dokdo forms from other clades with individuals that is collected at land area and Ulleungdo. These result is showing that is flowing independent evolution trends.
Vascular Flora of Namhan River

This study was carried out to investigate the vascular plants of Namhan River. The period of survey was from March to November on 2012. Vascular plants based on voucher specimen were summarized as 430 taxa including 86 families, 250 genera, 383 species, 2 subspecies, 39 varieties and 6 forms. The rare plant species designated by Korea Forest Service were 13 taxa including Penthorum chinense Pursh, Polygonatum stenophyllum Maxim, and Sparganium erectum L., etc. And the endemic plant species were 8 taxa including Salix koriyanagi Kimura, Clematis trichotoma Nakai, and Carex gifuensis Franch., etc. Furthermore, 56 taxa were listed as specific plant species based on phytogeographical and based on the list of plants which have been approved for overseas delivery, 27 taxa were recorded in the investigated area. The naturalized plants were recorded as 58 taxa, and their Naturalization Ratio and Urbanization Index were recorded as 13.5%, and 18.1%, respectively. 430 taxa listed consists of 174 taxa(40.5%) of pasturing plant, 141 taxa(32.8%) of edible plants, 112 taxa(26.0%) of medicinal plants, 48 taxa(11.2%) of ornamental plants, 11 taxa(2.6%) of stain plants, 7 taxa(1.6%) of fiber plants, 4 taxa(0.9%) of timber plants, 2 taxa(0.5%) of industrial plants and 109 taxa(25.4%) of unknown plants.
Development of EST-SSR Markers and Analysis of Genetic Diversity Using Persimmon (Diospyros kaki Thunb) Cultivars Collecting from Domestic

Persimmon (Diospyros kaki Thunb) fruit is one of the most important fruit and have been cultivated from ancient times in Korea. In this study, we found 16 EST-SSR markers that contained one or more EST-SSR sites from 246 cDNA sequences. The developing of EST-SSR marker analysis from 42 persimmon cultivars was compared by genetic relationships and morphological relationships using 6 qualitative traits (fruit related 6 traits) and 19 quantitative traits (flower related 19 traits). In this study, 25 primer sets were tested to identify PCR polymorphism and 14 potential EST-SSR primer pairs were selected. The result of morphological relationship EST-SSR marker analysis showed that the coefficient 0.02 was difficult to categorize in several groups. And then, coefficient 0.77 of genetic relationship showed that the group was classified as four groups. The result of correlation distance between genetic relationship and morphological relationship were investigated was low significance (-0.03). Our results also provided an optimized method for improvement of breeding efficiency and introduce of superior character at persimmon cultivars using EST-SSR markers which was useful for further investigation.
Effect of Initial Seedling Size and Root Pruning Intensity on Above-ground and Root Development in Quercus serrata Seedlings after Transplanting

This study was conducted to determine the optimum intensity of root pruning and the initial seedling size of 1-year-old Quercus serrata for producing 2-year-old healthy seedlings. Large( > 26 cm hight) and small size ( < 24 cm hight) seedlings were pruned their root by the length of 5, 10, and 15cm and then transplanted on nursery. Stem height, root-collar diameter(RCD), height to RCD ratio(H/D ratio), shoot dry weight(DW) and root development were measured during first growing season after transplanting. Seedling size and pruning intensity influenced on shoot growth such as height, RCD, H/D ratio and DW after transplanting. In addition, the development of lateral roots such as number, length, DW and diameter were affected by the seedling size. Taproot pruned by 15cm was shown excellent above-ground growth in large seedling group, but pruned by 10 and 15cm showed respectively better shoot growth in small seedling group. Large seedlings increased their above-ground growth, taproot regrowth, and number of lateral roots more than those of the small seedlings. But the length and DW of lateral roots were increased in the small seedlings. In conclusion, in order to produce high quality 2-year-old seedlings, it is important to produce large size of Q. serrata seedlings. Also root pruning length of 1-year-old Q. serrata was reasonable on 15 cm in taller than 26 cm or 10cm in less than the height.
Histological Characteristics of Somatic Embryos in Melon (Cucumis melo L.)

Hypocotyls explants of melon seedling were cultured on Murashige and Skoog's (MS) medium supplemented with 1 mg/L 2,4-dichlorophenoxy acetic acid (2,4-D) and 0.5 mg/L benzyl aminopurine (BA) for 6 weeks to produce somatic embryos. In somatic embryos produced through intervening bright yellow friable (BYF) from the explants, somatic embryos with two-cotyledon (26%) and horn-type cotyledon (74%) were observed. The procambial strand of cotyledons was originated from circular procambial tissues of lower hypocotyls. The circular procambial independently divided into two procambial strand at the edge of cotyledonary-node, and then connected to each cotyledon to form somatic embryos with two-cotyledon. When cotyledon was horn-type, the circular procambial strand in lower hypocotyls would continuously remain connected to the cotyledon. However, somatic embryos with two or horn type cotyledon formed an abnormal shoot apex without the tunica-corpus structure or dome shape in the inter-cotyledonary area. These results demonstrated that the variation of cotyledon in somatic embryos was closely related to procambial tissue differentiation and shoot apical formation.
New Cultivar 'White Edge' of Leaf Variegated Hosta minor

A new leaf variegated cultvar 'White Edge' was developed by ethyle methane sulfonate(EMS) treatment on seed of Hosta minor. Among induced leaf variegated plant, some with white color on the edge of the leaves was selected. They were cultivated via vegetative propagation. 'White Edge' was finally selected through the test of characteristics from 2003 to 2005. Assessment of botanical characteristics was conducted for three years. The major characteristic of these cultivar is that they maintain uniformly all year round. 'White Edge'(Grant No.1880) was registered to the Korea Seed and Variety Service(KSVS) for commercialization in 2007. These cultivar are useful as materials for pot or as ground cover plants.
