Effects of Nitrogen and Growth Regulators on Dormancy, Protein and Nucleic Acids Contents in Mulberry Varieties

Winter dormancy of the mulberry(Morus species) in Suwon was investigated with regards to mulberry varieties, such as Kaeryangppong, Daeryunppon, Yongchionppong and Hongolppong application of fertilizers and growth hormone. In general, It initiated at late September and it subsequently became deeper and reached the highest degree through late October to early November. After that early November it gradually turned into the breaking state and was terminated by late November. Intensity and duration of dormancy were lower and shorter in Kaeryangppon. The standard application of N. P. K(30-13-18kg/10a) affects it delayed, but terminated earlier. On the other hand, the double amount of nitrogen affects the dormancy fast, but terminated late. The treatments of GA3 at the early and termination stages increased the bad sprouting. The contents of RNA and protein in the bark gradually increased as the dormancy becomes deeper.
Effects of Number of Days After Urea Foliar Spray on Mulberry Leaf Components and Silkworms

The effect of short term duration(1, 2 and 3 days) after urea foliar spray on leaf quality was investigated by chemical analysis of leaves and silkworm rearing with leaves treated with urea in autumn. Foliar spray increased water content by 0.4~0.7% and total nitrogen by 0.19~0.51% in leaves compared to the conventional treatment. Foliar spray increased NO3-N concentration in leaf 503~1107 ppm more than the conventional treatment. NO3-N concentration in leaf decreased with days after foliar spray. Whole cocoon weight and cocoon shell weight increased by 0.02~0.14g and 0.5~3.1 cg, respectively in foliar spray than in the conventional treatment. Though foliar spray raised NO3-N concentration in leaf, leaves harvested one day after foliar spray were no harmful to silkworm.
A Study on Seasonal Fluctuation of Mineral Contents in Mulberry Trees(Morus)

This study was carried out to understand the seasonal changes in mineral contents of mulberry trees. The shoot barks and the leaves of three mulberry varieties(Kaeryangppong, Shinilppon, Yongcheonppong) were analyzed every 10 days during the year from January to December of 1993, Phosphorus content of the shoot barks and leaves gradually decreased until October in three varieties and increased thereafter. Especially, that of Yongcheonppong decreased 0.18% from 1.33% in January to 1.15% in October. Potassium content of the shoot barks increased 0.7~1.1% from April to August but decreased 0.7~0.9% during winter. That of leaves increased from May to October. Magnesium content of the shoot barks increased until August but decreased after then. And it increased again after October. That of the leaves decreased from May to October. Calcium content of the shoot barks decreased 0.4~0.6% from January to September but increased 0.1~0.4% from October. That of the leaves increased from May to October.
Phenotypic Expression of the Non Molting Gene in The 'Non-molting of Nho' of Bombyx mori

Studies were carried out to investigate physiological and biochemical analysis of recessive lethal mutant "nmn" and to observe the cuticle formation and dermal gland. All nmn homozygous larvae continued to eat a few mulberry leaves, and died without entering into molt. In case of artificial hatching, eggs had a higher nmn individual segregation ratio than in hibernating eggs. The percentage of nmn individuals with the hatching days was alike during 3days period. As a result of histological observation, nmn mutants dermal gland was different from normal dermal gland in form and size. Normal was formed new cuticle in the middle of the molting, but not in nmn mutant. Total protein content in haemolymph of the normal larva was more than that of nmn smutant and there was a difference between normal and nmn mutant protein components.omponents.
Genetic Analysis for Weight of Matured Silkworm and Number of Eggs Laid in Hybrid Population of the Silkwom, Bombyx mori

The genetics analysis for weight of matured silkworm(WMS) and number of eggs produced per moth(NEM) was studied by the seven parents diallel. Mean squares of additive effect, dominant effect, maternal effect and reciprocal effect were significant for two characters observed. The component of genetic variance analysis for WMS and NEM showed that dominant effect was higher than additive effect. Narrow sense heritability(h2ns) estimates were 0.773 and 0.228, in the WMS and NEM. The estimate of broad sense heritability(h2bs) value was higher than that of h2ns because of the low importance of dominance effect. Incomplete dominance was shown by Vr-Wr graphic analysis in the weight of matured silkworm and overdominance in the number of eggs produced per moth. In general combining ability effect, Jam 107 and Jam 124 was showed positively high for WMS and Jam 107 and S1 was expressed positively high for NEM. In specific combining ability effect, hybrids in S1XC51, S1XJam124 and Jam 107XJam 108 were exhibited positively high for WMS and Jam 107XN63, S1XC51, N74XJam 108 and Jam 107XJam 108 were found positively high for NEM.
Sex-Discrimination of Silkworm Pupa, Bombyx mori with Image Analyzer

To produce F1 hybrid of silkworm sex discrimination has to be followed at the pupal stage by sexual organ. However it requires a lot of labour and may bring about a wrong classification. In these regards, this study has been implemented to find out an effective measure for the pupal sex discrimination by use of variation of cocoon weight and image analysis of cocoon. As a result, it was found that in case of the pupal weight the percentage of a wrong classification fell on 0.3% and in case of single cocoon weight 0.4%. The discrimination rate was 99% in the weight variables of cocoon but analysis by single cocoon weight and cocoon shape variables, it was 98.7%. Efficiency of discrimination was increased by 2.7% as compared to variable of single cocoon weight. The minimum cocoon sampling size may be 15 cocoons sexual-wise.
The Study on the Weight loss Finishing for the Mixture of Silk/Polyester I. The Weight loss Finishing for the Raw-silk/Polyester

The weight loss for the mixture(raw-silk/PET) was tested to investigate the effects of weight loss accelerator on the degumming and weight loss. The degumming loss of raw-silk was decreased while the weight loss polyester(PET) was increased depending on the increase of accelerator concentration. The difference of the weight loss was not significant in raw-silk but the weight loss of PET was increased with the increase of the alkali concentration. The proper degumming of raw-silk was obtained and simultaneously the weight loss of PET was high for the raw-silk/PET "A" and "B" type. The proper degumming of raw-silk was obtained but the weight loss of PET was low for the raw-silk/PET "D" and "E" type. It showed that the typical degumming curve in raw-silk part and the weight loss of PET was increased depending on the treatment time in the weight loss for the fabric composed of raw-silk and PET.
Determination of Plasmids Encoding Crystal Toxic Protein Gene in Bacillus thuringiensis var kurstaki HD-1

The objective of this study is to identify plasmids of Bacillus thuringiensis var. kurstaki HD-1(B. t k HD-1) toxic to lepidopteran larvae. The results from agarose gel electrophoresis indicated that the bacterium contained 9 plasmids with approximate sizes of 1.4, 4.9, 5.4, 9.3, 10, 29, 44, 52, and 150 megadaltons(Md). By treating the wild type of B. t k HD-1 with either SDS or EtBr as curing agent, 26 cured mutants of the bacterium were obtained, 9 of them were crystallifereous(cry+) and the others acrystallifereous(cry-). Plasmids from B. t k HD-1 were transferred to B. cereus 569 strR cry- recipients(Bc569 M1). Among 13 isolates of Bc569 M1 transcipient, 11 of them were capable of producing the crystal toxic proteins. The plasmid patterns of Bc569 M1 transcipients and partially curved mutants of B. t k HD-1 on agarose gel electrophoresis suggested that the 29 and 44Md plasmids should be involved in the production of crystalline toxic proteins.
Cloning and Expression of the Bacillus thruingiensis var. kurstaki HD-1 Crystal Protein gene in Eschelichia coli

The 44Md plasmid of Bacillus thruingiensis var. kurstaki HD-1(B. t k HD-1) was partially digested with Sau3AI and the fragments were cloned into E. coli HB101 on vector pBR322. Of 2, 950 clones with a recombinant pBR322, only one clone KC1 was determined to have the gene for crystal toxic proteins from the 44Md plasmid of B. t k HD-1 at the BamHI site of pBR322. The recombinant pBR322 was named pKC1 and its molecular size was 12kb. The KC1 produced a protein which was toxic to the silkworm and antigenically similar to the crystal toxic protein of B. t k HD-1. Also, electrophoretic mobility of the KC1 protein was apparently the same as that of the crystal toxic protein of B. t k HD-1.
