Purification and Partial Characteriza6tion of the Storage Protein-like Protein from the 5th Instar Latval Haemolymph of the Chinese Oak Silkworm, Antheraea pernyi.

The storage protein-like protein has been purified from the 5th instar larval haemolymph of the Chinese osk silkwom, Antheraea pernyi, and the preparation was shown to be homogeneous by 7.5% native-PAGE. The molecule was consisted of a single subunit with a molecular weight of 80K, but the number of the subunits was not determined. The protein was defied as glycoprotein by Schiff's regent stining. Rabbit antibody prepared against the purified protein crotein crossreacted with the 5th instar larval haemolymph proteins of Antheraea pernyi and antheraea yamamai, but not with those of Bombyx mori and Bombyx mandarina.
Major Hemolymph Proteins and Vitellogenin in Mulberry Longicorn Beetle, Apriona germari Hope

Hemolymph proteins and vitellogenin from mulberry longicorn beetle, Apriona germari HOPE, were indentified, and their changed were analyzed during the larval-pupal-adult development and in the newly laid eggs. Thres major hemolymph proteins were observed in the hemolymph during the larval-pupal-adult development and the intensity of their proteins was clearly observed during the pupal stage. From SDS-[olyacrylamide gel electrophoresis analysis, molecular weights of three major hemolymph proteins were approximately 74 kDa, 78kDa and 85kDa. Vitellogenin in A. Germati appeared in the hemolymph of only abult female and is considered to be a product synthersized within 10 days after adult emergence. The molecular weight of vitellogenin was consited of a heavy subunit (165 kDa) and a light subunit (40 kDa).
Light and Scanning Electron Microscope Observatt-ons on Sexual Dimorphism in Pupa of Mullberry silkworm, bombyx Mori Linn (Lopidoptera : Bombycidae)

Under Light and Scanning electron microscope, the pupal morphology of mulberry silkworm Bombyx mori Linn. revealed the prothorax and metathorax, well developed mesothorax, less defined last pair of sporacle, well exposed prothoracic femora and wing pads approaching the anterior margin of Ab III. The important sex separating characters viz, wegiht, antennal elevations, intersegmental lines and genings have discussed. Further, two separate openings bursa copulatrix and ovipositional opening were observed, performing different functions in abult moth.

Twelve pure silkworm lines identified based in higher G.C.A values from a line
Tecter crossing programme, were crossed with twelve breeds showing cocoon shell ratio>22% and hybrids prepared, evaluated in a Multiple index method. Seven hydrids showing average index value>50 in all the 10 economic characters were seclcted and evaluated in the laboratory. However, two hybrids viz., A3
935E and A3
916B were finally selected based on the superiority of the breeds in one day shorter larval duration and with and with higher reeling characters compared to control KA
NB4D2. The breeding procedure involed in evolution hybrids are discussed.
Analysis of Promoter Strength of Autographa californica Nuclear Polyhedrosis Virus IE1 Gene by Using Rreconmbinant Baculovirus

To analysis a promoter strength of Atographa californica nucler polyhedrosis virus (AcNPV) IE1 gene, an immediate viral gene,
-glactosidase gene as a reporter gene was introduced under the control of the IE1 promoter. The restriction fragment containing IE1 promoter and
-galctosidase gene from pAcIE1-gal were inserter into pBacPAK9 to yield transfer vector pAcNPV-IE1-gal. The pAcNPV-IE1-gal was cotransfected with AcNPV genomic DNA BacPAK6 into Sf9 cells to produce recombinant baculovirus AcNPV-IE1-gal. In addition, recombinant bacvulovirus AcNPV-gal, which express
-galac-tosidase under the control of the polyhedrin promoter, was constrer, was constructed to compared with AcNPV-IE1-gal. The recombinant viruses were respectively infected into Sf9 cells and characterized by the virus titer and expression of
-galactoxidase in Sf9 cells. The promoter strength of IE1 and polyhedrin promoters was determined by the amount of
-galactosidase secreted into medium by viral infection. The titer of AcNPV-IE1-Gal determined by plaque assays in Sf9 cells was similar to that of AcNPV-gal. However, expression level of
-galactosidase by AcNPV-IE1-gal was significantly lower than that by AcNPV-gal. In conclusion, promoter strength of IE1 was approximately 25-fold lower than that of polyhedrin.
Imporoved Method for the Preparation of Silk Fibroin Hyoysates

An improvement of methods in fibroin hydrolysates preparat significantly enlarges their applications to practical use. Acidic hydrolysis by hydrohloric acid is one of the methods for silk fibroin depolymeriza6tion. A low yield of final product and long time of the process are the demerits of this method. Possibility of preparation of water-soluble silk hydrolysates with more yield and less expenses is investigated in this study. Such possibility is occurred with the increasing tratment temperature and simultaneously decreasing treatment time, concentration of hydrochoric acid respectively, the concentration of sodium hydroxide used for neutraliza6tion of hydolysates after hydrolysis. Colour is decreased in this case and a small amount of activated, too. Protection of hydrolysates against precipitation after neutraliza6tion, and separation and during concentrating process is the other merit of this method. Creamy-coloured insoluble silk powder is the remainder of hydrolyzed fibroin. This is the only the immobiliza6tion of enzymes and other physiological active substances. Fine particles of this powder can be used as additives for artificial diets and cultural media, as well as raw materials for polymer membranes, etc.
Oxime Generation of Silk Fibers by Hydroxlammonium choride treatment

This study was aimed to explain the essence of Hydroxylammonium hydrochloride(H.A.) effect on degummed silk fiber increasing the colour sites due to oxime generating reaction. H.A. in aqueous solution caues to increase the amount of [H+] and reduce pH values as the concentration of H.A. increases. The rate of [H+] absorption of silk fiker in acidic solution differs on the basic of solution pH and shows a specific uptake in each pH, the lower the pH of solution, the higher the amount [H+] absorption. The pH of solution after treating of silk fiber in H.A. and HCl, showed more remaining [H+] in H.A. solution due to [H+] releasing under the procedure of oxime production. Also it was revealed that in higher concentration of H.A. the reaction for oxime fixation in silk fiber carried out stonger and as a result the bigger gap with acid uptake curve appeared. FT-IR analysis of silk fiber treated with H.A. revealed the creating of intermolecular H-bond at the 2,981-2.930 cm-1, which was not appeared for nontraeted silk fibers and shows H-bond between N-OH group in oue chain and C=) group in another chain of silk protein. Colourimetry of dyed silk fiber after H.A. tratment showed that the silk fiber treated with the high concentration of H.A. compare to low concentration, absorbed more dyeing molecules and so Showed less percontage of Whiteness.
