The Selection and Sensitivity to Environmental Mutagen of Silkworm Reared Artificial Diet in a Screening System Using Specific Locus Mutation of the Silkworm, Bombyx mori

We investigated the selection and sensitivity to environmental mutagen of silkworm reared artificial diet to develop all-year-round operation system using a specific locus mutation of Bombyx mori. In the system, mutagenicity could be detected by the egg colour manifested by the pe and/or re genes, which is a kind of recessive visible mutation of the insect. Among, hi, Backokjam, C5, and N12, varieties of silkworm, AT was higher than other varieties in eclosion rate of female, and C5 and N12 were higher in fertility of male. Bakokjam was higher in eclosion rate of female, rate of moth to lay eggs normally in female and male, no. of eggs layed in female and fertility of female. As above results, Bakokjam was finally selected as the most fitness one among varieties of silkworm reared artificial diet. But the sensitivity to mutagen was lower than other varieties. In the sensitivity to mutagen, AT was the most sensitivity to mutagen in tested varieties of silkworm. To use AT variety in this system, AT was improved major characteristics, eclosion rate, fertility, rate of moth to lay eggs normally, and so on, by crossing of other varieties including Bakokjam.
Structural Analysis of Natural Indigo Colorants Extracted from Polygonum tinctorium

Natural indigo colorants were prepared by extraction of Polygonum tinctorium which was harvested just in the blooming season(in the late of July). The components were analyzed by TLC and HPLC, and its structures were analyzed by FT-IR, EI-mass. The results obtained are summarized as follows; The natural indigo powder was dissolved in DMSO and developed in eluent,
(8.5:1.5 v/v) by means of TLC for its quality analysis. It was segregated into indirubin as a red colour and indigo as a blue colour. In case of HPLC analysis,. FT-IR spectrum of indirubin showed a peak for NH residue between 3200 and
spectrum for indigo displayed AA'BB' spin system caused by indole structure between 6.5 and 7.7ppm of H4, 5, 6 and 7, and -NH proton for indirubin showed an singlet between 10.88 and 11.0ppm. EI-mass spectrum of indigo and indirubin both disclosed their molecular size as 262 and it implies that these two substances are isomer.
Breeding of New Silkworm Variety Golden silk, a Yellow Cocoon Color for Spring Rearing Season

A new silkworm variety Golden silk for spring rearing season is
hybrid between Jam311, a Japanese race bred from introduction breeding and Jam312, a Chinese race from introduction breeding. In the local adaptability test performed at 8 local areas in spring of 2006, "Golden silk" was showed 7 percent higher in hatchability than authorized silk-worm races laval. "Golden silk" was receded higher 1.7 percent, 2.2 percent in pupation rate, best cocoon rate respectively than Chunsujam. The larval period shortened for 1.12 day even though low record to the several items including productivity than Chunsujam. "Golden silk" was showed 1,031 m, 2.97 denier in length of cocoon filament, weight of cocoon filament respectively. The concentration of Deoxynojirimycin(DNJ) was measured 4.34 mg in "Golden silk", high then to "Chunsujam"(3.81 mg). In the Paecilomyces tenuipes production ability test, the pupal weight of "Golden silk" was 1.20g.
Anti-oxidative Capacity of Mulberry Leaf and its Tea

The importance of genetic resources conservation and utilization for economy and the health and well-being of societies in general is recognized all over the world. Mulberry genetic resources contain many natural components and are considerable resource for functional sericulture. We carried out antioxidative capacity analysis of mulberry leaves that were collected from Korea and some of foreign countries and preserved in the greenhouse. The mean antioxidant capacity of 34 accessions was 892.30nmol(ascorbic acid equivalent). Also we evaluated fruity characteristics and selected 4 accessions as breeding resources for the production of mulberry fruit. To investigate the effect of tea on antioxidative capacity, five kinds of tea(coffee mix, green tea added brown rice, mulberry leaf tea, Polygonatum odoratum tea and black tea added lemon) were selected and analyzed. Their's antioxidative capacity were 2,531.01nmol, 1,867.42nmol, 1,053.72nmol, 292.71nmol and 188.91nmol, respectively. The antioxidative capacity of drinking water soaked with mulberry leaf showed 891.96nmol.
Fabrication Process of Natural Silk Including Ag Nano-particle

Silkworm fed on the mulberry leaf mixed with silver nanoparticle to produce silver-nanoparticle embedded cocoon. Comparative analysis of silver content of cocoon shell, percentage of pupation and percentage of cocoon-shell weight showed that the optimum concentration and the feeding period of mulberry leaf mixed with silver nanoparticle were 500 ppm and the period from 3 day 5 instar to mounting of silkworm. The silver content of cocoon was observed variously by silkworm breedings. C212 variety makes pale yellow cocoon with the highest silver content(69%). Using the scanning electron microscope, we showed that the size of silver nanoparticles in silk was observed from 26.98 to 99.81nm. Silver-nanoparticle embedded silk is expected to use as high valuable application owing to the different functional properties including antibiotic characteristics and mechanical and electronic properties. The applicable fields expected is antistatic and/or electronic products with biological degradable natural materials.
Effect of Silkworm Hemolymph Collected Large-scale Bleeding Method in Insect Cell Culture

This study was carried out to investigate the utilization of hemolymph of silkworm, Bombyx mori, as a substitute for fetal bovine serum(FBS) in the insect cell culture. Hemolymph is collected on a small scale by clipping the abdominal leg; however, this method is not appropriate for large scale collection. The hemolymph was collected from 5 th instar larva by centrifugation after cutting of the abdominal legs was more appropriate procedure for large scale collection. The cell growth in the medium supplemented with hemolymph(Baekokjam) collected in large scale was almost same as that in the medium hemolymph supplemented with hemolymph collected in small scale. However, the mutant(
) hemolymph collected in large scale was still less effective in the cell growth, as compared to the Baekokjam hemolymph collected in large scale. The optimum centrifugation condition for large-scale bleeding was 500 rpm and 15 min.
