Anti-oxidative capacity of mulberry genetic resources

Much attention has been focused on the activity of the natural antioxidants present in fruits and vegetables, because potentially these components may reduce the level of oxidative stress. Especially, mulberry leaves containing many natural components are considerable resource for natural antioxidants. The antioxidant capacity of mulberry leaves was investigated with minilum L-100 device and ARAW-KIT (anti-radical ability of water-soluble substance), in comparison to the ascorbic acid. The antioxidant capacity of 16 varieties was 3303.4 nmol at opening stage of five leaves in spring. The highest stage of antioxidant capacity (3708.0 nmol) and yield rate was just before the coloration stage with anthocyanin in fruits, whereas the lowest stage was middle of June (2231.6 nmol) and about two months growing stage after summer pruning (2064.6 nmol). But after summer pruning, the antioxidant capacity of mulberry leaves increased gradually until just before fallen leaves stage. Even if samples were same variety, antioxidant effect of those showed different results according to collected regions. Also, antioxidant effect of mulberry leaves were higher than that of branches. The antioxidant capacity of yield-type mulberry leaves and fruits (Morus alba L., M. bombycis Koidz, and M. Lhou (Ser.) Koidz) collected from In-je, Won-ju and Yang-yang regions, Kang-won province, Korea, was investigated. The results indicated that total antioxidant capacity of yield-type mulberry leaves was 2711.2 nmol. In the antioxidant capacity analysis of Jeollabuk-Do genetic resources, autumn's mulberry leaves showed higher antioxidant capacity than that of spring's it. To investigate the effect of tea on antioxidative capacity, five kinds of tea(coffee mix, green tea added brown rice, mulberry leaf tea, Polygonatum odoratum tea and black tea added lemon) were selected and analyzed. Their's anti-oxidative capacity were 2,531.01 nmol, 1,867.42 nmol, 1,053.72 nmol, 292.71 nmol and 188.91 nmol, respectively. The antioxidative capacity of drinking water soaked with mulberry leaf showed 891.96 nmol.
Fasting conditions and dietary phenamena of edible cricket (Gryllus bimaculatus)

This study aimed to determine a feed withdrawal duration time of Gryllus bimaculatus (Gb), for removal of intestinal feces. Finally, this species of cricket has been temporarily approved for use as a human food source in Korea. To commercialize Gb as an edible insect, we attempted to prevent the proliferation of micro-organisms and improve taste of Gb by removing intestinal feces of live crickets. Our study showed that weak crickets were damaged in 2 days after feed withdrawal in farm. The microscope findings also revealed that feces of strong crickets' intestine almost disappeared at 1 day after feed withdrawal and instead, some of arms and legs of weak crickets were observed in the former's intestine. The results suggest that 1 day is deemed to be an adequate time for feed withdrawal, when it usually takes 1 day to deliver live crickets from the farm to a destination.
Mulberry leaf yield and optimal amount of silkworms rearing in different mulberry cultivars for mulberry fruit production

This study was carried out to develop the silkworm rearing technique in unused mulberry leaves after harvesting mulberry fruit. The growth of Gwasang No. 2 and Suhyang was very good compared to control cultivar Chungil in leaves size and new branch growth, but new branch and leaves of Daeshim was similar or small to control cultivar Chungil. The number of leaves of Gwasang No. 2 and Suhyang was lower than Chungil, but weight of leaves per tree was heavyer than Chungil. Mulberry leaf yield was Gwasang No. 2 521 kg/10a, Suhyang 189 kg/10a, Daeshim 73 kg/10a, Chungil 1,095 kg/10a. Content of mineral element of all three mulberry cultivars leaf for mulberry fruit production was higher than Chungil in N, P, K, Ca etc. Feeding quantity of silkworm of Gwasang No. 2 and Suhyang was much more than Chungil. Feeding quantity was highest at Suhyang in 96 kg/box. Growth duration of silkworm larvae was not different in all four mulberry culivars but weight of silkworms (5th instar 3rd day) was heavy at Gwasang No. 2 (2.07 g/head) and Suhyang (2.11 g/head) compared to control cultivar Chungil (1.92 g/head). Mortality of silkworms was 14.6% (Gwasang No. 2), 13.3% (Suhyang), 13.9% (Daesim), 12.6% (Chungil) and than higher at elder instar stage. The production amount of silkworm (5th instar 3rd day) was 35.4 kg/box (Gwasang No. 2), 36.6 kg/box (Suhyang), 35.0 kg/box (Daeshim), 33.6 kg/ box (Chungil). Amount of possible rearing silkworms was estimated 3.4box/10 a (Gwasang No. 2), 1.3box/10a (Suhyang), 0.5box/10a (Daeshim), 8.7box/10a (Chungil).
Effect of silkworm varieties on Paecilomyces tenuipes culture

There is no specific data on Entomogenous fungus (DongChungHaCho) in accordance with the silkworm varieties, despite of very high value industrial use as functional materials at domestic and abroad. In this study, we investigated culture condition and characteristics of Paecilomyces tenuipes in 3 silkworm cultivars and 2 specific silkworm breeds. Infection rate of P. tenuipes for silkworm varieties was the highest Kumokjam, followed by Baegokjam, Daeseongjam, Golden silk, Yeonnokjam in that order. in planting density per pupa. Also, we have developed annual production technologies of P. tenuipes. First, in order that the moisture content of the infection pupa became 3% or less in the natural condition. Second, the dried pupa must be vacuum-packed and was kept under
. Finally, by restoring the moisture content of the dried pupa and culture in optimal condition, the technique for annual production of the P. tenuipes was achieved. Therefore, we expect that the use of Kumokjam and annual production technique will contribute significantly to farm income.
Cell proliferation of silk proteins obtained from Bombyx mori silkworm varieties

It is known that silk protein supports effectively proliferation of cell such as insect cell and hybridoma cell. Although there are many varieties of Bombyx mori silkworm, the effect of silkworm varieties on cell proliferation has not been considered in detail. We studied that characteristics of silk cocoon obtained from Baegokjam, Kumokjam, Daeseongjam silkworm varieties and whether silk protein affected cell proliferation or not. Silk sericin was prepared under high temperature and high pressure condition. Silk fibroin was prepared using
with different dissolution time. As a result, there are differences in silk cocoon from different silkworm varieties about cell proliferation. The proliferation was accelerated in the presence of Baegokjam silk sericin and Kumokjam silk fibroin with 5hr dissolution time. We expect that silk proteins could be a preferable culture medium supplement for stimulating the proliferation of cell. Then, this results suggest silk as a new material for medium supplement replacing with fetal bovine serum.
Structure and mechanical properties of Korean commercial silkworm cocoon

Korean commercial Bombyx mori silkworm cocoon made by Kumokjam and Daesungjam was examined to characterize for application. Amino acid analysis showed that the main amino acid of Kumokjam and Daesungjam cocoon was glycine, alanine, serine, tyrosine, and aspartic acid. The content of three major amino acids was about 67% and the content of long chain amino acids was about 37%. Stress-strain curve showed that breaking stress of Kumokjam cocoon, 50 MPa, was higher than that of Daesungjam cocoon, 48 MPa. Breaking strain of Kumokjam cocoon, 37%, was also longer than that of Daesungjam cocoon, 30%. These results might be used as basic information for development of non-textile materials using Bombyx mori silkworm cocoon.

To extend shelf life of mulberry fruits, usually expensive freeze drying of freezing process is required. In this study an economic to multipurpose agricultural products dryers with mulberry dryer and technique was developed to reduce postharvest processing cost. Fuel and electricity consumption for drying were lowest when drying temperature was
. And various drying conditions (fuel 3 hrs, heat 1 hr, room temperature air velocity 51 hrs) of the developed dryer were analyzed to improve drying performance. Dried mulberry fruits had dark brown color and marketable high quality explain the semi-dried characteristics.
Characteristics of mulberry cultivar 'Sangmaru' (Morus alba L.) for mulberry fruit production

We bred a mulberry cultivar named Sangmaru, through local adaptability test, which is under registration as a new cultivar for fruit production. Local adaptability test had been carried out at three places (Suwon, Cheongju, and Jangseong) for seven years from 2008. This is tetraploidy variety belonging to Morus alba L. made by colchicine treatment on growing point of 'Chungil (Morus alba L.)' winter buds. Sangmaru was high yielding cultivar in fruit productivity by 6% compared to control cultivar 'Chungil (Morus alba L.)' for five years. Although Sangmaru was similar in sugar content of mulberry fruits, it was bigger than that of 'Chungil'. It is adaptable to every where except the places where cold damage and sclerotic disease happen frequently.
Effect of ethanol concentration on the infrared spectroscopic characteristics of silk beads

The structural transition of silk protein has been induced by various method including alcohol treatment. To know the effect of alcohol concentration on silk beads conformation, silk beads were prepared in different alcohol concentration (100%, 70%, and 50%) and then examined the infrared spectra of silk beads. Silk beads prepared in 100% alcohol showed at
conformation and did not showed a characteristic absorption peak from model drug. However, silk beads in 70% and 50% alcohol showed some peaks originated from model drug including

Development of polymorphism genetic marker for identification of the silkworm races

We have previously characterized the complete fibroin light chain gene from one of the silkworm race Baegokjam (Bombyx mori) and found two variable regions (FibL1, intron 2 ~ 3; FibL2, intron 6) with the primer sets designed to cover these variable regions. In this study, we tested the utility of these variable regions as genetic markers for classifying silkworm races. For the purpose, Europian races (Q, PK), Chinese races (C26, C31), Japanese races (N15, N9), and tropical races (SA2, SA5) were used in this experiment. The polymorphism of the FibL1 and FibL2 were divided into two and three types, respectively. The combination of the FibL1 and FibL2 polymorphisms were constant within the same races. The result suggest that the primer sets designed from two variable regions of fibroin light chain gene may be useful as the genetic markers for silkworm races.
Antioxidant activity and antimicrobial activity of the grasshopper, Oxya chinensis sinuosa

In this study, to confirm the physiological activity of the grasshopper (Oxya chinensis sinuosa) antioxidant activity, antimicrobial activity, thermal stability of the antimicrobial substance, pH stability, total polyphenol content of the adult grasshopper was measured. Antibacterial activity in accordance with the extraction solvent showed a strong antibacterial activity in a mixed solvent of ethanol for the E. coli strain. Antimicrobial activity in
hot air drying and a freezedried condition was the highest and there was no difference in the gender. Antibacterial substance was stable to heat and pH. Antioxidant activity of the grasshopper exhibited a high activity in the 50% and 70% ethanol extract. Total polyphenol content was the 12 ~17 mg / 100 g and there was not great difference according to the drying conditions and gender.

The American cockroaches, Periplaneta americana L. was the most important worldwide pest species. It has been an public health problems. We were determinated life cycle and extraction of crude extracts by chemical reagents from cockraches (P. americana L.). The extracted crude solution has been antibacterial activity to gram negative bacteria (Pseudomonas aeruginosa,
), gram positive bacteria (Bacillus subtilis,
), and fungus (Candida albicans,
) using radial diffusion assay. We were analysed of up-regulation of Glutathione-S-transferases (GSTs) stimulation, indicating that antioxidantial protein from various classes are simultaneously expressed in a single insect upon infection or injury. The gene from Periplaneta americana L. were cloned, analysed sequence, and measured protein expression by Real Time PCR (Polymerase Chain Reaction).
