Production of Single-Cell Protein on Petroleum Hydrocarbon
The Korean Journal of Microbiology, volume 9, issue 3, 1971, Pages 95~102
The growth characteristics of Candida tropicalis KIST 351 on gas oil substrate under different culture conditions were investigated and the preliminary animal feeding experiments using this yeast as a partial substitute of fish meal was also conducted. The yeast assimilates effectively n-paraffins in gas oil ranging from
with its maximum cell growth at
and pH 5.5 with aeration of 3 vvn and agitation of 900 rpm. The optimal concentrations of nitrogen sources,
were 4, 2 and 0.5g/1, respectively. Ferrous sulfate, manganese sulfate and zinc sulfate showed positive effect to cell growth with the optimal range of 5-10 ppm. In the feeding experiment with 3 and 5% incorporation of the gas oil grown yeast, neither adverse effects on growth of chicks nor toxic effect were observed. Protein content of the dried cell was 58.8% and its amino acid composition compared well with other single-cell protein products and FAO reference protein.
Study on brewing of sweet potato starch
The Korean Journal of Microbiology, volume 9, issue 3, 1971, Pages 103~120
We have been studied on brewing sweet starch. We obtained the results as follows ; 1) 5 strains, T-T-2, T-T-4, T-K-2, T-T-18, T-T-1, were the most available in view of fermentative power by capacity of
. 2) 5 strains, T-T-4, T-T-2, T-T-1, T-T-3, T-K-2, produced capacity of alcohol more than 5.78%. 3) 6 strains, T-T-2, T-K-2, T-T-4, T-S-2, T-I-3, T-I-1, are available not only taste and flavour, but productive power of alcohol in sweet potato starch. 4) The form of 6 strains are long oval and round and most of them are similar to the other yeast in size. 5) In giant colony the color was cream color and cream buff, and T-K-2 was formed by
on diameter and by 3.5mm on high. 6) Optimum temperature of most of all strains is 25~
but T-K-4 is 28-30.deg.C. 7) Optimum pH is 3.4-4.6. 8) T-S-2 was died off at 65.deg.C, the other strains died
. 9( Making Bun-kok with non-heated wheat bran .alpha.-amylase was more increased by 4.5-13.5 mg of glucose in reaction solution and .betha.-amylase more 1.6-3.4ml of N/10-
Solution than Bun-kok with heated wheat bran. 10) It seems that mycellium grows better than original in substance containing 0.4 ~ 1.2% of HCl. 11) Making Bun-kok to add 0.8% HCl, .alpha.-amylase was increased 9.93-20.7mg of glucose and .betha.-amylase ws increased 2.6~4.3ml of N/10-
solution to reaction solution. 12) 1.2%-HCl, or higher concentration, acts as inhibitor, in the meanwhile the concentration between 0.4~0.8% of HCl acts as activator. 13) We must make Bun-kok for 42 hours, at 28~
) After we made Bun-kok using S-O-II and R-J-I one by one, Bun-kok which mix each other in equal quantity is increased more than original on enzyme acrivity. 15) Oxidation is the best way of refining sweet potato starch in N/10-phosphate buffer solution (pH 7.5). 16) When we prepared sweet potato starch, first pH was 3.0.
Studies on the Bacterial Leaching of Low-grada Copper Mlnerals
The Korean Journal of Microbiology, volume 9, issue 3, 1971, Pages 121~129
The purpose of this study was to investigate the proprieties of practical application and possibility of bacterial leaching method to the low copper minerals in Korea. Therfore, ore samples and mine water of the Talsung, Ilkwang, Taekwang, Yongho and Siheung were analyzed chemically, mineralogically, and bacteriologically. The results obtained were as follows ; 1. The range of pH value was 2.6-3.8. 2. Copper contents in ore samples of Talsung and Ilkwang were 0.38% and 0.63%, and those in mine water of Talsung nad ilkwang were 584 .
g and 15.
g per ml, respectively. 3. Ferric sulfate contents in mine water of Tlsung and Ilkwang were 27.2.
g and 10.5.
g per ml, respectively. 4. Ferrobacillus ferroosxidans was isolated and identified from Talsung and Ilkwang mine water and specially Thiobacillus thiooxidans was isolated from Ilkwang mine water.
General characters and applications of photosynthetic bacteria
The Korean Journal of Microbiology, volume 9, issue 3, 1971, Pages 130~138
In order to investigate the production of aflatoxin in various conditions such as pH, moisture and temperature, 27 smaples were inoculated with Aspergillus flavus, and in addition 3 smaples were inoculated with the mixture of Aspergillus flavus and Bacillus subtilis and cultured under the conditions such as 20.deg.C and 30% moisture contents. The following results were obtained : 1) Aflatoxin production was the highest at pH 5.0 and relatively high at pH 7.0. Its production was decreased significantly when pH reached 9.0. 2) The yield of aflatoxin was shown comparatively high level at 30% moisture contens. The higher moisture contents was, the lower aflatoxin production was. 3) The highest level of aflatoxin production was at 20.deg,C, and comparatively high level was at 30.deg.C. However, its production was fairly low at 40.deg.C. 4) The highest crude aflatoxin production was 5,093ppm (B
, 1.912ppm ; B
, and the lowest one 2.197 ppm (B
, 0.793 ppm : B
, 0.185 ppm : G
,0.102 ppm G
, 0.381ppm) at 63% moisture, pH 9.0 and 40.deg.C. 5) When Aspergillus flavus and Bacillus subilis were cultured together under the conditions such as 20.deg.C and 30% moisture, aflatoxin production was decreased by 27% comparing with the culture of Aspergillus flavus alone.
