Anti-skin-aging effects of Paeonia Suffruticosa Andrews on maintaining skin collagen in STZ-induced diabetic rats and inhibiting MMP-1 systhesis in human skin fibroblasts

Objective : Skin aging is commonly observed in patients with diabetes mellitus, which can be accessed by the amount of skin collagen and matrix metalloproteinase-1 (MMP-1). In the present study, anti-skin-aging effects of Root Cortex of Paeonia Suffruticosa Andrews (PSA), which has been widely used to treat diabetes mellitus, are investigated. Methods : Streptozotocin (STZ) was intraperitoneally injected to rats to induce diabetes. Body weights, feed intake, organ weights, blood glucose, and other biochemical index are determined in both normal and diabetic rats. In order to study the effect of PSA on skin aging, the amount of skin collagen was measured in diabetic rats after PSA treatments. Also, MMP-1 synthesis in UVB-irradiated human skin fibroblasts was investigated. Results : 1. When PSA was administered to STZ-induced diabetic rats, feed intake was significantly increased and blood glucose and total cholesterol were decreased in a dose-dependent manner. However, there are no differences in individual organ weights, GOT, and GPT. 2. A decrease of skin collagen in diabetic rats was significantly suppressed when PSA was treated. 3. PSA also inhibited MMP-1 synthesis in UVB-irradiated normal human skin fibroblasts, similar to retinoid, a well-known effective anti-skin-aging substance. Conclusion: PSA suppressed a collagen decrease in diabetic rats and inhibited MMP-1 synthesis in UVB-irradiated human skin fibroblasts. Therefore, the treatment of PSA is very effective to slow down the skin aging process.
The Effects of Acorus gramineus on Changes of Nasal Tissue in Allergic Rhinitis Model

Background & Objectives : Rhinitis is an inflammation of nasal mucosa. The major symtoms are watery rhinorrhea, sneezing, itchy nose, and nasal obstruction. Allergic rhinitis is an immune reaction by allergen. So, we aimed to determine therapeutic effects of Acorus gramineus by observing changes in IL-4,
and the nasal mucosal tissue. Materials and Methods : Fifteen BALC/c mice were divided into three groups : m group(which ate low concentrated herbal medicine ), M group(which ate high concentrated herbal medicine) and control group. Control and experimental group were induced allergic rhinitis by Ovalbumin as the method of Levin and Vaz. Experimental group was orally administered the Acorus gramineus extract for 28days. We observed changes in IL-4,
and trans aminase(AST, ALT) in blood and nasal mucosa and submucosa. Results : There were no significant changes statistically in IL-4 and
in blood(p<0.05). And there were no hepatotoxicity with Acorus gramineus extract. Histologically, almost no inflammatory response in treatment group(m,M) against that there were inflammatory response(increased goblet cells, dilated vessels, edema of bowman's glands and injured olfactory hairs) in control group. Conclusion : According to above results, it is supposed that Acorus gramineus has no immunological effects on allergic rhinitis.
Effects of NaisoOkseol-Tang (NOT) on solid tumor and immunity

Objective : The present study was carried out to investigate the effects of NaisoOkseol-Tang (NOT) on solid tumor in mice in terms of immune-potentiating and direct cytotoxic action of NOT in vitro and vivo study. Methods: The present author investigated thymocyte proliferation and NO production to confirm immune-potentiating activity of NOT and also investigated body/tumor weight and survival rates in tumor bearing mice. In this study, administration of NOT decreased tumor/weight ratio significantly, and prolonged survival duration compared to non-treated control. Treatment with NOT also elevated proliferation rates of thymocytes isolated from tumor bearing mice. Results: In vitro study, treatment with NOT suppressed proliferation rate of Sarcoma 180 (S-180) cells. Contrary, treatment with NOT elevated proliferation rate of thymocytes. These results were co-related with in vivo study. In addition, NO production from abdominal macrophages isolated from normal mice was not affected by treatment with NOT. Conclusion : NOT is useful to treat patient with solid tumor, because NOT has direct toxic action for tumor cell and immune-potentiating action for T cells.
Effects of Pinelliae Rizoma (PR) on asthma induced intra-nasal instillation of ovalbumin in mice

Objective : This study was designed to investigated effects of Pinelliae Rizoma (PR) on asthma Methods : Detecting antigen specific antibody isotypes, cytokines in serum and bronchoalveolar lavage fluid (BALF). In addition, the present author also investigated changes in weight of spleen and proliferation rates of splenocytes. Finally, histopathological observation of the lung tissue was also investigated. Results : Oral administration of PR lowered OVA specific IgE levels in serum, IgG1 levels in serum and BALF. PR also decreased production levels of IL-4 in BALF. In addition, total cells in BALF were decreased by oral administration of PR effectively. In histapathological observation, PR group showed downward tendency of inflammatory cell infiltration around small vessels. Conclusion : PR is useful to treat patients with asthma and the mechanisms are related in suppression of Th2 skewing reactions.
Effects of Gleditsia spina (GS) water extract on Gene Expression of Human Melanoma cells, by using Microarry technique

Objective : This study was designed to investigated effects of Gleditsia spina (GS) on human derived melanoma cells Methods : The genetic profile for the effect of medicine on human derived melanoma cells of SK-MEL-2, was measured by using microarray technique, and the functional analysis on these genes was conducted. The network of total protein interactions was measured by using cytoscape program. Results : Total 253 genes were up-regulated and 439 genes down-regulated in cells treated with GS. Genes induced or suppressed by GS were all mainly concerned with metabolic process, regulation of biological process and protein binding. Conclusion : Suggest the possibility of GS as anti-cancer drug and cosmetic agent, and also suggest that related mechanisms are involved in regulation of intra-cellular metabolism in melanoma cells.
The Effects of Baickbujasan Extract on the Skin Damage and Pigmendation Induced by Ultraviolet Irradiation

Objective : The purpose of this study is to examine the effects of Baickbujasan(BB) on the skin damage and depigmentation. Method : The inhibition of tyrosinase activity, melanogenesis and cell viability in cultured B16 melanoma cells were measured. In order to test effects of reduction of melanogenesis, B16 F-10 mouse melanoma stem line was employed to extract melanin from cultured cell, where BB was added or not, and was dissolved in alkali for colorimetric analysis. Also, in order to test skin alteration in C57BL/6 after UV irradiation, the animals were grouped into a UV urradiation group and UV irradiation after BB application group. Dopa oxidase tissue staining was excuted to invesitage the change in the distribution of active melanin cell. The distribution of active melanin cell in inner skin of iNOS after damage from UVB irradiation and the manifestation condition of P53 which takes part in natural death of keratinocyte were examined. Result : The results indicate that BB has significant effects on tyrosinase activity, and melanogenesis in vivo test. BB seems to reduce C57BL/6, external dermatological damage, for instance, erythematous papule, eczema, loss of keratinocyte, reduction in pus, and relieves dermatological damages. Conclusion : BB can be applied externally for UV protection and depigmentation.
The Treatment Effect of Younggamgangmisinhayin-tang through Control of Th 2 cell Differentiation in Allergic Rhinitis

Objectives : Younggamgangmisinhayin-tang is clinically used for the treatment of the aller -gic rhinitis. The aim of this study is to investigate the effects of Younggamgangmisinhayin -tang on allergic rhinitis. Methods : The thirty rats were divided into three groups : control group, AR(allergic rhinitis eliciated) group, YT(Younggamgangmisinhayin-tang treated after allergic rhinitis elicitation) group. To induce allergic rhinitis in AR group, YT group rats were sensitized intraperitoneally with 0.1% ovalbumin(OVA) solution 3 times at intervals of 1 week. Then intranasal sensitization was performed by diffusing 0.1% ovalbumin(OVA) solution 3 times at intervals of 2 days. After that time rats in YT group were oral administration treated by Younggamgangmisinhayin-tang for 7days. The change of the amounts of esinophil, PAS, ZO-1, MIP-2, COX-2, IL-4 were observed in each group. Th 2 skewed condition in EL4 cell was induced by using PMA and OPl. Younggamgangmisinhayin-tang was added into EL4 cell by density. And then IL-4 mRNA was observed. Results : In esinophil study YT group was proved significant inhibitary effect. In PAS and ZO-1 YT group were preserved well. In MIP-2, COX-2 study YT group was proved significant inhibitary effect. In IL-4 and IL-4 mRNA study YT group was proved significant inhibitary effect. Conclusion :According to the above results, it is considered that Younggamgangmisinha- yin-tang has inhibitory effects on the allergic rhinitis rat models.
The effcect of Gorynju and a Soju extract Psoraleae fructus on Melanin synthesis of B16 melanoma cells

Objective : This research was carried out to compare the effect of Gorynju and a Soju extract Psoraleae fructus on melanin synthesis of B16 melanoma cells. Methods : To investigate melanin synthesis of B16 melanoma cells, this research was measured cell survival, tyrosinase activity, melanin synthesis, western blot. Results : Both Gorynju and Soju extract Psoraleae fructus, cell toxicity depended on the density. Tyrosinase activity depended on the density of Gorynju extract Psoraleae fructus and statistic was showed significant(0.5, 1, 2, 3
), in a Soju extract Psoraleae fructus, 1
were showed significant. Melanin synthesis was showed significant in a Soju extract Psoraleae fructus(3, 4
). Western blot was showed to depend on the density of Gorynju and a Soju extract Psoraleae fructus. Conclusions : In a tyrosinase activity and a melanin synthesis, the intermediate alcohol of Gorynju and a Soju may be suitable to use.
Effect of Ethanol Extract of Belamcandae Rhizoma on Dermal Bioactive Properties

Objective : This experimental study was preformed to investigate the effect of ethanol extract of Belamcandae Rhizoma (EBR) on dermal bioactive properties related to cosmeceuticals such as radical scavenging activity and depigmentation. Methods : The free radical scavenging activity of EBR was assayed in cell free systems using 1,1-diphenyl-2-picrylhydrazyl (DPPH). Antibacterial potency was measured by the size of inhibition zone with change of volume. Cell viability was measured by MTT assay and tyrosinase activity was assessed using L-DOPA. Results : EBR showed slightly radical scavenging activity and protected against UVB-induced cell cytotoxicity. Futhermore, EBR showed antibacterial activities on S. epidermidis and S. aureus. Although the proliferation of B16/F10 cells was decreased by EBR, necrosis was not revealed. EBR augmented tyrosinase activity and dendrite outgrowth in B16/F10 cells. Conclusions : These results suggest that EBR has antioxidative ＆ antibacterial activities, and stimulate melanogenesis.
Correlation between Prognosis and Factors on Acne Patients

Objective : The purpose of this study was to explore the correlation between efficacy and various factors of the acne patients. Method : We reviewed 30 acne patients who visited Allergy/Derma-cosmetic Clinic at Kyung Hee Neo Medical Center from June, 2006 to December, 2007 and were treated with herbal medicine and acupuncture for 4 weeks. We investigated premenstrual aggravation of acne, facial burning sensation, constipation, Heart Rate Variability(HRV) results and regional distribution of acne. And we evaluated inflammatory acnes according to the Korean Acne Grading System(KAGS). Results & Conclusions : 1. The average age of total patients was 23.0 years old. The most frequent age group was 21-25 years old. Patients evaluated as KAGS Grade 2 were most common. 2. In total patients, inflammatory acne were reduced significantly after 4 weeks of treatment. 3. The result of analysis on premenstrual aggravation of acne, facial burning sensation and constipation, there weren't any difference of treatment results on statistical meaning. 4. The results show that there were significant correlations between the Low Frequency at HRV and the rate of acne reduction. (Pearson correlation = -0.565) 5. If the distribution of acne did not involve the T-zone of the face, chest, or back of the body, inflammatory acne were significantly decreased after 4 weeks of treatment.
A Clinical Analysis of Inpatient of Dermatology, Ophthalmology & Otorhinolaryngology

Objective : This study was performed to examine the patterns of inpatients that had visited Dept. of Dermatology Hospital of Oriental Medicine, Dongguk University. Methods : We analysed statistic study in 106 patients, who had admitted to the Dept. of Dermatology, Ophthalmology & Otorhinolaryngology Hospital of Oriental Medicine, Dongguk University from July, 2005 to January, 2008. Results : The results were as follows; 1. Males were 50.94% and females were 49.06%. 2. Distribution of aging was 18.87% in twenties and 17.92% in fifties. 3. Common disease group were Facial palsy(33.96%); Atopic dermatitis(21.7%); Tinnitus(7.55%); Sudden Sensorineural Hearing loss(4.72%); Sore throat(5.66%). 4. Average age of Facial palsy patients was 50.9 years old, average hospitalized period were 10.47days and subjective satisfaction rate was 2.56 points on the basis of 4 points. 5. Average age of Atopic dermatitis patient's average age was 22.22 years old, average hospitalized period were 8.35days and subjective satisfaction rate was 3.44 points on the basis of 4 points. 6. 47.2% of all patient were admitted into the hospital on 1st or 2nd outpatient service and 44% of Facial palsy patient were admitted into the hospital via emergency room. 7. All patient's average hospitalized period were 7.5 days. 8. All patient's subjective satisfaction rate were 2.97 points on the basis of 4 points. Conclusion : This study suggests that oriental medical treatments is especially popular in facial palsy and atopic dermatitis. We have to take steps for effective management and treatment for special diseases and ages.
One Case of traffic accident induced paralytic strabismus (oculomotor nerve palsy) which was treated with electroacupuncture at oculomotor muscles

Objectives : We already reported the effectiveness of this treatment method(electroacupuncture at oculomotor muscles) at the adult and young idiopathic paralytic strabismus patients and Miller-Fisher syndrome. Then I explored the possibility for improvement of the traumatic injury paralytic strabismus case. Methods : I treated the case by using the electroacupuncture at both paralytic lateral rectus muscle lesions. The case was treated almost daily and every treatment was enforced 10 minutes. I use the PG-306 electra-acupuncture products(Suzuki Iryoki Co. Japan) and apply the low consequence wave of 1-8Hz. Results : We gained good results from the case and not observed any sides effect or compliant. So I hope to apply this treatment for traumatic and post-operative nerve injury cases and am sure to make the treatment protocol for them in the future.
