Effect of Water Stress on Yield and Quality of Ligusticum chuanxiong Hort.

The experiment was conducted to investigate the effect of water stress on yield and quality of Ligusticum chuanxiong Hort. The water stress treatment was imposed artificially on seedling, flowering and rhizome enlargement stage of the plant. The root yield rate decreased to 19.1%, 18.2% and by the water stress treatment at rhizome enlargement, seedling and flowering stage, respectively. Portion of the products having commercial quality grade (above 20g of rhizome weight) was 93.4% at control plot, while it was 85%, 81.7% and 78.3% when stressed for water at seedling, flowering and root enlargement stage, respectively. Content of extract was the higher in the order of control, water stressed at rhizome enlargement, flowering and the seedling stage. Postive correlationship was found between yield of rhizome and rootlet yield or economic production ratio, and between dry weight of stem and rootlet yield.
Effect of Plant Growth Regulators on the Adventitious Root Formation from Bupleurum falcatum Callus

Calli induced from the leaf segment of Bupleurum falcatum were cultured on Mu-rashige and Skoog's(MS) medium supplemented with 2, 4-D, IBA, IAA and NAA of 0.1 mg/l , The induction of adventitious roots from callus was the best in MS medium supplemented with 0.1 mg/l 2, 4-D and the lateral root was the same. The pretreatment of 0.1 mg/l 2, 4-D for 120 hours was most effective for the formation and grwoth of adventitious roots. The number of adventitious roots per micro callus pre-treated with 0.1 mg/l 2, 4- D was 5. 3 which was the highest level. The callus subcultured for 4 weeks were best for the adventitious root formation. The callus subcultured for more than 4 weeks decreased the adventitious root formation and turned to brown in color.
Factors Affecting Anther Culture in Schizandra chinensis

To increase the efficiency of the callus induction in anther culture of Schizandra chinensis, the effects of culture stage, low temperature pretreatment, growth regulators, sucrose and gelling agents were tested on Murashige and Skoog's medium. And the effects of ABA and
on organogenesis were investigated. The most suitable stage for anther culture was at the middle to late uninucleate stage, of which the flower size was
, and the frequency of callus induction was 13.3%. The effect of low-temperature pretreatment on callus induction was highest as 12.5% in the trearment for 8 days at
. The combination of 1.0 mg/L 2, 4-D and 0.25 mg/L kinetin for callus induction was most effective as 8.3% among various media. The frequency of callus induction was excellent as 10.8% in 4% sucrose. Effect of gelling agents on callus induction was highest as 9.0% on 0.6% Gelrite medium. The prevention of callus browning was effective on the media supplemented with ABA and
and rooting was promoted on medium supplemented with 5 mg/L ABA. But shoot was not developed.
Effect of Thidiazuron on the Formation of Micro-tubers and Plantlet Regeneration of Pinellia ternata T.

This experiments were conducted to determine the effect of thidiazuron on forming tuberlets and plant regeneration of Pinellia ternata T. by tissue culture. The addition of
TDZ to the medium had better regeneration than that of any other treatments of NAA and TDZ. At the combination treatments of NAA and TDZ, as the level of thidiazuron increased, the rate of shoot regeneration was incresed while the increment of NAA concentration inhibited the rate of shoot regeneration. The supplement of
thidiazuron produced the best number of micro-tubers per explant and the number of micro-tuber formed was 25 in MS medium and 29 in MG medium on 30 day culture, respectively. Microtuber formation was the best on MG medium with 1.0 mg/l NAA and
thidiazuron. MG medium was superior to MS and B5 medium for the growth of tuberlets. Half strength of MS medium with NAA 2 mg/l was the most effective for root formation. Rooting ability on nursery soil of plantlets produced in in vjtro was good as a 80% after 3 weeks.
Environment and Ecological Characteristics in Euphorbia ebracteolata Hayata Flora

Biosystematic study on Euphorbia ebracteolata Hayata was carried out by comparing ecology and morphology between wild and cultivated ones. As the result of this study, cultivation method about this medicinal plant was established, and ecologically, the shape of leaf or root of this plant appeared to be dependent on ecological environment while the hair of ovary and the involucel were considered to be fixed characters. From the investigation of natural conditions of habitats, this plant was considered to be a facultive sciofytes and a day neutral plant.
Effects of Colchicine and EMS Dropping to Apical Meristem on Induced Mutation and Plant Protein Pattern in Wasabia japonica Matsum

This study was investigated on the effects of colchicine and EMS. induced mutation, plant on the germination and polypeptide pattern and peroxidase activity monitored by two dimensional gel analysis in Wasabia japonica Matsum (wasabi). Germination rate of Muju was higher than that of Ulrung-Do and optimum concentration for germination was appeared 100 ppm GA3 containing with 10 ppm BAP in these cultivars. Survived plants rate of Muju was higher than that of Ulrung-Do after colchicine and EMS treatment. Peroxidase activity and plant height were decreased by mutagen treatments, while incresed in root and stem thickness. The number of protein spots and pattern showed difference between Muju and Ulrung-Do . The plants treated mutagen increased polypeptide spots, especially EMS treatment showed more different polypeptide pattern compared to control.

Callus Formation and Plant Regeneration from Immature Embryos of Eleutherococcus senticosus

This study was conducted to establish mass propagation system from the tissue culture using immature embryos in Eleutherococcus senticosus. Immature embryos from seeds were removed under the microscope and placed on MS,
medium containing several plant growth regulators. While the calli were well formed on media containing 2 mg/l of 2, 4-D, 2 mg/l of 2, 4-D and 0.7 mg/l of TDZ, shoot regeneration was better on MS medium with combinations of high concentrations of TDZ and low concentrations of 2, 4-D. Treatment of 2, 4-D alone was better than treatment of TDZ alone in callus induction, but plant regeneration was reversed. The results of callus formation and shoot regeneration on
media were similar to those of MS media. The rate of callus formation was nearly 100% when 2, 4-D was added to
, medium on concentration of 2 mg/l or 0.7 mg/l. TDZ showed very significant effect on the formation of multiple shoots.
Morphological and Growth Characteristics of Collected Coix lacryma-jopbi mayuen STAF in Korea

This research was conducted to investigate morpological and growth characteristics of 358 Coix lacryma-jopbi mayuen STAF collected in Korea. The test collections contained 76% medium wide-type leaf, 59% medium size-type seed, 34% large size-type seed, 70% elliptical-type shell, 50% brown shell color, 92% low stem color and hardness of seed coat averaged

Effect of Growth Regulator C-MH Application on Growth and Root Yield in Bupleurum falcatum L.

This experiment was conducted to evaluated the effect of choline salt of maleichydrazide (C-MH) application on growth and root yield components of Bupleurum falcatum L. Flowering was delayed by C-MH application
days. Twice application was more effective to delay flowering. Application of C-MH 2,000ppm at 30cm to 40cm of plant height reduced plant height 15cm, but increaced highly branches and the growth of root. The dried root yield per 10a was 27% higher in twice application of C-MH 2,000ppm than control plot.
Effects of Planting Density on Agronomic Traits and Yield in Bupleurum falcatum L.

Bupleurum falcatum is one of the oldest medicinal crop spontaneously generated as well as cultivated by man and successfully grown during the summer season in Korea. This study was conducted to examine the effect of planting density in the row spacing and hill spacing system using machine seeder on major agronomic traits, yield and labor saving. In 1993 and 1994, a spilt plot design was used with row spacing as the main plot and hill spacing as subplot. Labor saving efficency of the machine seeder reduced 96% than the hand seeding in sowing time required. Number of seeding stand increased, but stem height and number of nodes did not affect with different density in each row and hill spacing. There were reduced with narrow hill spacing on stem diameter, number of branches, shoot weight and root traits. There was no significant interaction between row and hill spacing for any major traits studied except shoot weight. Yield was more increase at 20cm than at 30cm row spacing. For these studies the optiomum density appears to be between drilling or 5cm hill spacing with 20cm row spacing. A highly significant positive correlation was appeared between number of seedling stand and root yield.
