Pollen morphology of subfamily Euphorbioideae (Euphorbiaceae)

Pollen morphology of 20 species of Euphorbioideae and one from Crotonoideae was examined with light, scanning and transmission electron microscopy. The pollen morphology presented here did not support the monophyly of Euphorbioideae, and the unique characters of tribe Stomatocalyceae such as absence of aperture margins, and thick and continuous endexine suggest the exclusion of Stomatocalyceae from Euphorbioideae which is in accordance with previous phylogenetic studies and wood anatomical data. Aforementioned pollen features in this tribe support that it is closely related to Acalyphoideae. Two subtribes of Stomatocalyceae were well recognized in terms of pollen morphology: Hamilcoinae includes species with reticulate or microreticulate exine patterns, and Stomatocalycinae includes those with perforate patterns with small supratectal elements. Three subtribes of Euphorbieae, classified in terms of pollen morphology, were further divided into three different types including perforate in Euphorbiinae, microreticulate in Neoguillauminiinae and reticulate in Anthosteminae, strongly supporting the Webster`s subtribal system. Hureae and Hippomaneae, having perforate sculpturing pattern with smooth margo, were similar in pollen morphology. Especially, the presence of small supratectal elements in Hura crepitans pollen suggests the close relationships with Excoecaria species of tribe Hippomaneae. According to the previous studies, unique intine thickness along the aperture margin in Euphorbieae was not reported elsewhere in Euphorbiaceae. However, in this study we found the same intine thickness along the aperture margin in Maprounea brasiliensis and Hura crepitans. We hypothesize that the similar intine thicknesses found in Euphorbieae and above two species are the evidence of close relationships among them.
Taxonomic position and genetic differentiation of Korean Astragalus mongholicus Bunge

To clarify the taxonomic position for Astragalus nakaianus and provide correct scientific name for A. mongholicus cultivar in South Korea, we examined external morphological characters and sequence variations from ITS and five cp non-coding DNA regions. Genetic structure was also analyzed for 61 individuals from three populations using nine microsatellite loci. We found no significant difference between the South Korean cultivar and A. mongholicus var. dahuricus when morphology and ITS sequences were considered. Morphologically, A. nakaianus specimens varied somewhat from A. mongholicus var. mongholicus and var. dahuricus in habit, plant height, and lengths of leaf axis and leaflet. Although sequence data from ITS and cp noncoding DNA regions could not distinguished A. nakaianus from A. mongholicus, microsatellite analysis revealed strong structuring between the cultivar and A. nakaianus. Therefore, we conclude that the South Korean A. mongholicus cultivar should be treated as A. mongholicus var. dahuricus and that A. nakaianus should be merged into A. mongholicus as a variety, i.e., A. mongholicus var. nakaianus.

Androcorys pusillus (Ohwi & Fukuy.) Masam. (Orchidaceae) was first found from Mt. Baekdu of the Korean peninsular and was known to distribute from Taiwan and Japan. This species is distinguished from the related taxa by its denticulated sepal margin, petal width which is as broad as dorsal sepal, and larger bract size.
Isolation and characterization of microsatellite markers for Hydrangea luteovenosa (Hydrangeaceae), an endangered species in Korea

Hydrangea luteovenosa is a critically endangered plant species of Jeju Island in Korea, though it is widely distributed in western Japan. We isolated and characterized five microsatellite loci in this species. The number of alleles ranged from 3 to 27, observed heterozygosity from 0.27 to 0.86, and expected heterozygosity from 0.34 to 0.91. The markers described here will be useful for investigating the genetic diversity, genetic structure, and gene flow of H. luteovenosa, and the genetic findings would contribute to the establishment of effective conservation measures for this species in Korea.
A systematic study of the Polygonum amphibium L. complex (Polygonaceae) based on chloroplast DNA sequences

The Polygonum amphibium complex (Poygonaceae) is a highly polymorphic taxon that can grow in aquatic environments as well as in moist terrestrial habitats. Aquatic and terrestrial plants of the P. amphibium complex vary significantly in morphology and exhibit very complicated patterns of morphological variation, resulting in the description of numerous infra-specific taxa. Principal components analysis of 107 individuals of the P. amphibium complex from Asia and North America using 11 morphological characters showed that the aquatic plants can be discerned from the terrestrial plants by leaf size, shape, and petiole length. In contrast, both aquatic and terrestrial plants collected from the same population or locality shared identical sequences in the matK, psbA-trnH IGS, rbcL-accD IGS and trnL-trnF regions of the chloroplast DNA (cpDNA), suggesting that aquatic and terrestrial forms of the P. amphibium complex are not genetically diverged; morphological differences between the two forms are probably due to the differences in environmental conditions of the habitats. In addition, results from the morphological analysis and the maximum parsimony analysis of the cpDNA data set revealed that the plants from Asia including Korea, Japan, China, Mongolia and Russia Far East are diverged from those in North America and Europe, suggesting that the Asian populations should be recognized as a distinct variety, P. amphibium var. amurense Korsh.
Taxonomic position of Taxus cuspidata var. latifolia endemic to Ulleung Island

The purpose of this study is to review the taxonomic position of Taxus cuspidata var. latifolia endemic to Ulleung Island with related taxa T. cuspidata var. cuspidata, T. caespitosa, and T. cuspidata var. nana based on external morphological characters and DNA barcoding study. T. cuspidata var. latifolia was similar to T. cuspidata var. cuspidata in the arbor, straight trunk, and symmetric arrangement of leaf. But the unique differences between T. cuspidata var. latifolia and T. cuspidata var. cuspidata were leaf size and the exposure of seed from aril. Additionally, sequences of four chloroplast DNA regions including matK, rbcL, trnL intron and trnL-trnF spacer regions were analyzed. Korean Taxus species and their related taxon T. cuspidata var. nana were strongly supported as a monophyletic group in neighbor-joining analysis. Taxus cuspidata var. latifolia showed 100% sequence identity to related taxa. Korean endemic T. caespitosa is also distinguishable from related taxa by prostrate stems and spiral arrangement of leaf. The examinations of external morphology and DNA barcoding study suggest that the taxonomic position of T. cuspidata var. latifolia should be maintained as a variety of T. cuspidata.
Identification of Echinochloa oryzicola (Vasinger) Vasinger and E. oryzoides (Ard.) Fritsch in Korea

Echinochloa oryzoides (Ard.) Fritsch (Na-do-non-pee), was found at paddy fields of Jeonranamdo on a large scale after a first discovery at Masan, Kyeongsangnamdo. This species was not easily distinguished from rice before flowering at the paddy field because of a similar morphology to rice. Leaf variants of Pinus and their ITS DNA sequences

ITS DNA sequences of two variants of Pinus spp. having single fasciculate leaf or two to three fasciculate leaves within an individual were analysed in order to determine their origin. Also, the same DNA locus of P. densiflora, P. rigida and P. koraiensis, distributed at the same region together with the OTUs having leaf variations, were analysed to compare with each other. Aligned sequences including ITS1, 5.8S and ITS2 were 580~584 bp in length. The 5.8S region was well conserved among all the OTUs we tested except for P. koraiensis, which has two nucleotide substitutions. The partial ITS1 region upstream of the 5.8S region showed relatively high sequence diversity compare to the ITS2 and has 181~185 bp in length. In this region, the sequences of the two variants were identical to that of P. densiflora. The ITS2 has identical for all OTUs in length and the two variants also have same sequences compare to that of P. densiflora. These two variants and samples of P. densiflora were grouped together in the UPGMA tree with 100% similarity level. The result strongly suggest that these two variants were originated from P. densiflora.
Floristic study of Mt. Namdeogyu

We investigated the flora of Mt. Namdeogyu, southern regions of Deogyusan National Park, from April of 2011 to September of 2012. The number of vascular plants in this area were summarized as 507 taxa, including 102 families, 296 genera, 444 species, 8 subspecies, 49 varieties, 6 forms. Among the recorded taxa, Korean endemic plants were 9 species and the floristic regional indicator plants were 80 taxa including 3 taxa in grade V, 3 taxa in grade IV, 19 taxa in grade III category. The rare and endangered plants which was designated by Korea Forest Service were 15 taxa comprising 1 endangered species (EN) of Rhododendron tschonoskii, 5 taxa of vulnerable (VU) such as Cynanchum inamoenum, Lilium cernuum, Gastrodia elata, Bupleurum euphorbioides, Paeonia japonica, and 9 least concerned (LC). In addition, the subalpine plants of the region were summarized as 24 taxa, and Mt. Deogyu area inclunding the Mt. Namdeogyu was suggested as the north most limits line of the distribution for 2 taxa, Abies koreana and Rhododendron tschonoskii, and as the south most limit of the distribution for Bupleurum euphorbioides.
