Seasonal Variation of Lipids and Fatty Acids of Sharp Toothed Eel(Muraenesox cinereus)

Seasonal variation of uraenesox cinereus) muscle was investigated. Crude lipid content varied from 3.85 to 12.59 g/100g, comprising the highest content in November. The major fatty acids of total lipid, neutral lipid, and phospholipid were C16:0, C23:0, C16:1, C18:1, C20:5, and C22:6, but in phospholipid, Cl8:3n-6 was also the major fatty acid. The C22:6 content of the neutral lipid was much lower compared to that of the total lipid and phospholipid. The content of polyunsaturated fatty acids (ranged from 73.93 to 66.23%) in phospholipid was higher than that of any other lipid fraction. In glycolipid, C20:1 and C14:1 were higher compared to those of any other lipid fraction, but C20:5 and C22:6 were lower. The annual average ratio of n-3 to n-6 of total lipid, neutral lipid, phospholipid, and glycolipid was 10.82, 12.27, 6.63, and 6.50, respectively. The particular trend of seasonal variation of fatty acid composition was not showed in total and neutral lipid. However, the samples caught in September and November had a high crude lipid content with a significantly lower content of polyunsaturated fatty acids in phospholipid. Also, the content of monounsaturated fatty acids in glycolipid was lower in samples of September, November, and January.

Application of Species-specific DNA Probe to Field Samples of Alexandrium tamarense (Lebour) Balech

Fluorescent species-specific DNA probe (AT1) of toxic dinoflagellate Arexandrium tamarense was tested on several other species, on comparison of binding activity at different preservatives for fixation of the cells, at different culture age and estimation of cell density by light microscope or epifluorescent microscope using whole cell hybridization. Th AT1 probe specifically bound to Alexandrium tamarense, whereas it did not bind to other phytoplankton, in particular Alexandrium catenella, morphologically similar to Alexandrium tamarense, could not react to AT1 probe. When cells were fixed with all three preservatives, labeling cells of Alexandrium tamarense emitted strong fluorescent signal intensity. In addition, regardless culture days, binding activity with AT1 probe was strong. The tell densities estimated by epifluorescent microscope were than those estimated by light microscope. The enumeration and identifying of Arexandriurn tamarense using DNA probe method will be contributed to a new biotoxin monitoring and prediction system in field.
Hybrid Vigor and Reciprocal Effect of Several Growth and Yield Characters in Fls crossed between Hulled and Naked Barley

This study was carried out to estimate the degree of heterosis and heterobeltiosis, and the heritability and genetic correlation in F
hybrid produced by reciprocal crosses between two six-rowed hulled barley (Gangbori and Olbori) and three six-rowed naked barley (Saechalssalbori, Saessalbori, and Moodeongssalbori). Heterosis was observed positively in traits such as cults length, spike length, awn length, leaf length, leaf width, number of spikes per plant, number of grains per spike, and 1000 grain weight. Heterosis and heterobeltiosis from the crosses with hulled barley as a material parent showed higher performance in culm length, leaf length, leaf width, number of spike per plant, number of grains per spike. The hulled grain characteristics is dominant over the naked one. The highest heritability was confirmed in leaf width with a value of 94.5% and overall high heritability was observed in most traits with the range from 76.7% to 94.5%. In an analysis of correlation coefficient, higher genetic correlations were observed between leaf length and number of spikes per plant(0.970), between cults length and number of grains per spike(0.963), and between awn length and leaf length(0.862) in the forward crosses in which hulled barley was maternal. In the reverse crosses, higher genetic correlation was observed between culm length and spike length(0.926), between awn length and leaf length(0.922), and between number of spikes per plant and number of grains per spike(0.713).
Cloning and Expression of Antifungal Protein (PR5) Genes from Hot Pepper (Capsicum annuum L.)

We have isolated and artificially expressed three cDNA clones of Capsicum annuum PR5 genes for elucidating the antifungal activity against Phytophthora capsici which contracted a hot pepper root rot in field condition. Three divergent PR5 proteins from hot pepper were designated as CAPR5-1 and CAPR5-2 from susceptible cultivar (Subicho) as well as CAPR5-3 from resistant cultivar (CM331) in response to P. capsici. The cDNA similarity was found over 80% of identity among the three CAPR5s, and deduced amino acid sequence was characterized that all of CAPR5s contained 16 cysteine residues which possibly had a significant role in the structural formation. The result of genomic DNA blot showed that CAPR5-1 and CAPR5-2 existed as single copy in the Subicho genome. Three recombinant CPARs in E. coli were identified by SDS-PACE, and each expressed protein was treated on the PDA medium which contained cultured pathogens. Although three CAPR5 proteins did not affected the hyphal growth of Glomerella glycines and Colletotrichum fagenarium, CAPR5-1, CAPR5-2, and CAPR5-3 showed a specific antifungal activities against P. capsici.
Phenylalanine Ammonia Lyase and Cinnamic Acid 4-Hydroxylase Activities of Rice and Pepper in response to UV and Wounding

The metabolites related to phenylpropanoid pathway play an important role in the self-defense of plants and induced by environmental stress like wounding, pathogen attack, UV-irradiation and so on. The mRNA level of rite phenylalanine ammonia lyase (PAL) was increased at 12 h to 48 h, however it was gradually decreased 48 h to 60 h after UV irradiation. The PAL enzyme activities in rice were peaked at the time of 24 h after UV irradiation, on the other hand, it was not affected by wounding. The PAL enzyme activities in pepper were raised high at 24 h and 10 h by UV irradiation and wounding respectively. The cinnamic acid 4-hydroxylase (C4H) activities were increased by wounding treatment and were detected from 12 h to end time point of experiment, while UV-irradiation didn't affect the C4H activity in rice and pepper. These results were assumed that the action of isoflavonid has an alternative effect on the defenses which include wounding and UV irradiation and on the diverse roles in rice and hot pepper.
Biotransformation of Arylnitroso Compound by Mammalian 1,4-Benzoquinone Reductase

Quinone reductase was purified to homogeneity from bovine liver and the purified enzyme catalyzed the reduction of phenanthrenequinone as well as benzo- and naphthoquinones. The enzyme catalyzed the biotransformation of arylnitroso nitroso compound and the reaction product was identified by TLC, GC, CC-MS and NMR. The reaction was almost entirely inhibitable by Cibacron blue 3GA or dicumarol, potent inhibitors of mammalian quinone reductase.
Characterization of Remazol Black B-Biodegradation by Stenotrophomonas maltophilia EJ-211

A reactive dye-degrading bacterium was isolated from textile waste water and it was identified as Stenotrophomonas maltophilia based on its biochemical characteristics. The effects of additional carbon and nitrogen sources were investigated for the development of an optimal medium for biodegradation of Remazol Black B by 5. maltophilia. The optimal pH and temperature were determined to be 6.5 and 3
, respectively. Remazol Black B with the initial concentration of 50 ppm could be degraded up to 86% within 28 h.
Morphological Study on Differentiation of Hepatocytic Stem Cell by Intrasplenic Transplantation after Partial Hepatectomy

This study was performed to evaluate regenerative effects of intrasplenic stem cell transplantation after partial hepatectomy. To evaluate the regenerative effects, Sprague Dawley rats were used. In vivo the embryonic stem cells of blastocysts were collected from superovulated rats on day 3.5 after the vaginal plug checked. The embryonic stem cells were cocultured with hepatocytes for 8 days, they were transplanted into the spleen. After the intrasplenic transplantation of cultured stem cells, they were initially distributed near the periarterial lymphatic sheath after transplantation in the hematoxylin-eosin staining. Their number were formely increased and their size enlarged at forming small lobules. The embryonic stem cells in the culture proliferated and initially proliferated around the periarterial lymphatic sheath and later they around the trabecula with blood vessels. After the transplantation of stem cells, their cell organelles were well developed rough endoplasmic reticulum at the 20th with prominent epidermal growth factor reaction, developed smooth endoplasmic reticulum at the 30th day, well differentiated bile canaliculi with increased transforming growth factor-
and apoptosis reactions.
Effect of Transplantation of Intravascular Cultured Neural Stem Cell upon Peripheral Nerve Regeneration

The ultrastructural change of sciatic nerve and immunohistochemical changes of NGF, PCNA were studied at the transplanted segment of intravascular cultured neural stem cell in the rat sciatic nerve by 5 months after the sciatic nerve transection. The transplanted intravascular neural stem cells were differentiated into Schwann reals at the 20th day and these cells began to regenerate by the proliferation and hypertrophy. There were many remyelinating Schwann cells in the transplanted nerve in term of stimulation. According to NGF finding, we suggest preexisting Schwann cells may induce the differentiation of neural stem cells into regenerating Schwann cells. Electron microscopic changes were the remyelinating appearance, the increase of intraaxonal microtubules and enlarged mitochondria and contacting tell processes each other.
Differential expression of glycoconjugates, estrogen receptor-α, c-fos and c-jun in the vagina of normal and ovariectomized rat

We investigated the compositional changes of glycoconjugates (GCs) and expression of estrogen receptor (ER)-
, c-fos and c-jun in the vagina of normal and ovariectomized rats by histochemical and immunohistochemical methods. The mucinous transformation of the superficial layer that occurred from late diestrus to proestrus was accompanied with extensive enrichment of GCs. According to the cyclic changes of the vagina, distinct reactivity patterns such as SBA affinity in the diestrus and Con A affinity in the diestrus and estrus phase was observed. However, weak staining for GCs was detected in the atrophied vaginal epithelium of ovariectomized rats. ER-
immunoreaction was mainly demonstrated in the basal layer of epithelium and estrus cycle-related variation in the number of ER-
immunoreaction were not pronounced. But the stromal cells showing ER-
immunoreaction were abundantly observed from diestrus to estrus phase. The most numerous c-fos immunoreactive cells were observed in the basal and intermediate layer of epithelium and stromal fells from the proestrus to estrus phase and c-jun in the basal layer of epithelium during estrus phase. The c-jun immunoreaction of stromal cells expressed only in the estrus phase. In the ovariectomized rats, a few of ER-
, c-fos and c-jun immunoreactive cells were observed in the vaginal epithelium and no immunoreaction were found in the stromal cells. ER-
and c-fos immunoreaction fully expressed in the proestrus coincident with the cell proliferation, mutinous transformation and cornification of vaginal epithelium. These data indicate that vagina epithelium and stromal reals express multiple protein such as ER-
, c-fos and c-iun by estrogen that may function in process of cells proliferation and differentiation of vagina epithelium.
Antioxidative activity of flavonoid rich extract of Oenothers odorate Jacquin on oxidation of low density lipoprotein

There is growing interest ill understanding the role and mechanisms of flavonoid as antioxidant on LDL. The antioxidative activity of flavonoid typically present in Oenothers odorate Jacquin was investigated in vitro using a human LDL oxidation assay. In present work, LDL was incubated with increasing concentrations of extracts of extracts of Oenothers odorate Jacquin and LDL oxidation was started by adding CuSO
to the media. Substances in leaves extracts of Oenothers odorate Jacquin are capable of inhibiting the initiation and the propagation of LDL oxidation. They inhibit LDL oxidation, monitored by thiobarbituric acid-reactive substances(TBARS), as well as modification as shown through direct measurement of electrophoretic mobility, diene conjugates. Inhibition is a dose dependent effect that becomes already apparent at concentration of extracts as low as 40
/mL. Inhibition is almost complete at 80
/mL. Extracts of Oenothers odorate Jacquin were more potent antioxidative activity than either ascorbic acid and dl-a -tocopherol.
Protective Mechanism of Flavonoids Isolated from Rhus verniciflua on the Biliary Liver Fibrosis in Rat

The pathogenesis of cholestatic liver injury as well as the modulation of hepatic fibrogenesis is causally associated with involvement of reactive oxygen species and free radical reactions. In this study, we investigated whether flavonoids (fustin, sulfuretin) which were isolated from Rhus verniciflua Stokes (RCS) have antioxidant and antihepatotoxicity effect under the biliary liver fibrosis condition. After surgery (control) and posttreated RCS methanol extract (250mg/kg), ethyl acetate extract (250mg/kg) and flavonoids were administered p.o. 10mg/kg/day in two weeks for control groups. The concentration of clinical parameters and product of hepatic lipid peroxidation and the hydroxyproline content were significantly increased in liver fibrosis developed rats. Among the clinical parameters of serum, value of ALT, AST, SDH, total bilirubin and
-GT in posttreated RCS components-group showed significantly lower than in control-group. The content of hydroxyproline in posttreated RCS components-group showed lower than in control group and then the value of MDA in posttreated RCS components-group was also significantly reduced to 40~60% of that in control group. The hepatic xanthine oxidase and aldehyde oxidate activities were posttreated RCS components-group showed significantly lower than in control-group. The hepatic SOD and glutathione peroxidase activities were posttreated RCS components-group showed significantly higher than in control-group. Hence we concluded that active components of fustin and sulfuretin which were isolated from R. verniciflua Stokes were hepatoprotective effect in experimental liver fibrosis.
Effects of some edible plants on changing chromosomal morphology, and apoptosis of MCF-7 cell

Equisetum arvense L. and Lactuca dentata Makino. var. Flaviflora Makino. of samples relatively showed anticancer effects on MCF-7 mammary gland adenocarcinoma cell. The most active plant among the samples was Capsicum annuum L. var angulosum Mill. We studied that MCF-7 cells were changing chromosomal morphology and apoptosis on these samples. Capsicum annuum L. var. angulosum Mill. of samples relatively showed good anticancer effects. The cells became vague after 2 days and then destroyed. The supernatant of the cells including medium was measured by UV absorbance. The results showed that Capsicum annuum L. var. angulosum Mill also exerted high level. We also used electrophoresis in order to observe apoptic characterization of DNA fragmentation. The cells treated with Capsicum annuum L. var. angulosum Mill showed the apoptotic characterization. The chromosome of the cells were observed on those samples. The cells treated with Capsicum annuum L. var. angulosum Mill among them were shown the fastest changes. The cells were aggregated and destroyed by treatment with some edible plants. Especially, the case of Capsicum annuum L. var. angulosum Mill, it led MCF-7 cell to apoptosis faster than others. And we can observe chromosomal changes and dispersion by PI staining. These results showed that each sample exerted anticancer effects on MCF-7 cells. Especially Capsicum annuum L. var. angulosum Miff exerted significant anticancer effects.
The Effects of Panax Ginseng Extract on Antioxidative Enzyme Activity in Ovariectomized Rats

In order to observe the bioactivity of ovariectomized rats, nonovariertoized (sham) group, ovariectomized (Ovx) group, ovariectomized ginseng total saponin (GTS)-treated (Ovx+ GTS) group and ovariectomized ginseng water extract (GW)-treated (Ovx+CW) group were made. We measured AST (L-aspartate aminotransferase) and ALT (L-alanin aminotransferase) in sera, and MDA (malondialdehyde:lipid peroxidation), SOD (superoxide dismutase), catalase, total-glutathione (GSH + GSSG) and GPx (glutathione peroxidase) in liver tissue total homogenates of rat. AST activity of serum in Ovx group was 2.11 times increased, but ALT activity was not changed compared to Sham group. In AST activity, they tend to decrease significantly in each substance such as GTS and GW administered group. Lipidperoxides of each fraction in Ovx group were highly increased compared to Sham group. Extracts of ginseng-treated group markedly inhibited lipid peroxidation by 62% ∼72%. And as the result of the measurements of SOD, catalase, total-glutathione and GPx which are antioxidant enzyme, antioxidant enzymes in Ovx group much lower than in Sham group. But they were significantly increased in each substance such as GTS and GW, administered group. Based on the results, it is supposed that more produced free radicals decreased antioxidant enzyme. And it is also thought that extracts of ginseng can inhibit aging by reducing antioxidant enzyme.
Study of Functional Chungkukjang contain Fibrinolytic Enzyme

A bacterial strain showing the fibrinolytic activity was screened from korean traditional soybean products. For the identification, the strain was investigated morphology and biochemical characteristics and it was classified to Bacillus subtilis. The strain had high fibrinolytic activity in Chungkukjang. The optimum fermentation condition of temperature and time were 37
and 24hour. The pH in Chungkukjang was gradually alkalized during fermentation. The fibrinolytic enzyme in Chungkukjang stable at heft treatment; After heating at 6
for 30 min, the fibrinolytic activity remained 75% and 40%, respectively.
Quality Assessment of Traditional wine and viable cell count by Lumitester

To evaluate the quality of korean traditional wine, 31 different type of wine were selected and examined for the following properties; ethanol contents, pH, acidity, total sugar contents, reducing sugar contents. In addition ATP contents of traditional wines were measured using lumitester and the result were compared with these obtained by viable cell counts. The physical and chemical properties of traditional wines are summarized as follow.; pH were in the range of 3.59~4.76, Total acidities were in the range of 4.23~16.21, Total sugar and reducing sugar contents were in the range of 0.04~1.13 g/100 mL and 0.006~0.999g/100 mL, respectively, ATP contents were in the range of 10
RLU(Relative light unit).
Production of Microbial Pesticides by Soybean Curd Waste-water in Bacillus thuringiensis subsp. kurstaki HD-1

The waste-water from the industry for production of a soybean curd (the soybean curd waste-water) was investigated to use for the substrate to produce the endotoxin of Bacillus thuringiensis subsp. kurstaki HD-1 used as one of well known microbial pesticides. The pH of the soybean curd waste-water was 9.8 and its chemical oxygen demand (COD), total nitrogen (TN) and phosphate (TP) were 276.0, 71.1 and 5.5mg/
, respectively. The higher was the concentration of the soybean curd waste-water in the medium, the more endotoxin was produced. Maximal sporulation occurred at which concentration of
in the medium supplied with the soybean curd waste-water was 1% (w/v). Production of the endotoxin with the optimized medium supplied with the soybean curd waste-water was 1.5 times higher than that without the soybean curd waste-water. The soybean curd waste-water was found to be suitable substrate for production of the endotoxin of Bacillus thuringiensis subsp. kurstaki HD-1.
