Synergistic antitumor activity of ST1571 and camptothecin in human cancer cells

The in vitro activity of ST1571, an inhibitor of the Abl group of protein-tyrosine kinases, alone or in combination with camptothecin (CPT), a specific topoisomerase I inhibitor, was evaluated against human cancer cells with different metastatic capacity and drug resistance potency. These cell lines showed different sensitivity to ST157 on growth inhibition, and the expression of DNA-dependent protein kinase (DNA-PK), which interacts constitutively with c-Abl, was significantly decreased in drug sensitive CEM and MCF-7 cells and poorly metastatic PC3 and KMl2 cells as compared with that of multidrug resistant CEM/MDR and MCF-7/MDR cells and highly metastatic PC3-MM2 and KM/L4a cells, respectively. These results suggest differential modulation of DNA-PK by ST1571 treatment in drug resistance and metastatic degree dependent manner. We showed that CPT as well as ST1571 significantly inhibits the expression of DNA-PK. The combined treatment with ST15fl and CPT revealed synergistic effect, and the effect was accompanied by inhibition of cell proliferation due to significant reduced expression of DNA-PK components, which resulted in CPT sensitizes human cancer cells resistant to ST1571. Therefore, the results of our study suggested that the suppression of DNA-PK using combination of ST1571 and CPT could be a novel molecular target for against drugresistant and metastatic cancer cells.
The classification and comparison of genetic diversity of genus Malus using RAPD

Cenus Malus is a long-lived woody species primarily distributed throughout Asia. Many species of this genus are regarded as agriculturally and ecologically important. The phynetics and genetic diversity among eight species of genus Malus were reconstructed using the random amplified polymorphic DNA (RAPD) markers. In a simple measure of intraspecies variability by the percentage of polymorphic bands, the M. micromalus exhibited the lowest variation (34.7%). The M. pumila showed the highest (50.0%). Mean number of alleles per locus (A) ranged from 1.347 to 1.500 with a mean of 1.437. The phenotypic frequency of each band was calculated and used in estimating genetic diversify (H) within species. The mean of H was 0.190 across species, varying from 0.155 to 0.220. In particular, two cultivated species, M. pumila and M. asiatica, had high expected diversity, 0.314 and 0.307, respectively. On a per locus basis, the proportion of total genetic variation due to differences among species ranged from 0.388 to 0.472 with a mean of 0.423, indicating that 42.3% of the total variation was found among species. The phylogenetic tree showed three distinct elates. One includes M. sieversii, M. pumila, and M. asiatica. Another includes three M. baccata taxa. The other includes M. sieboldii, M. floribunsa, and M. micromalus. One variety and one form of M. sieboldii were well separated each other. RAPD markers are useful in germ-plasm classification of genus Malus and evolutionary studies.
Heterochromatic Knob Number And Karyotype in Korean Indigenous Waxy Corn by Giemsa C-banding Pattern of Mitotic Chromosome

A Giemsa C-banding method was used for the identification of somatic chromosomes and heterochromatic knob position in Korean indigenous waxy com (Zea mays L.). 5 inbred stocks were examined and their heterochromatic knob numbers ranged from 6 to 12. In comparison of homologous chromosomes of two stocks of YS-1 and MY-1, knob numbers, knob positions, arm ratios and relative length of chromosomes were different between the genotypes. The length of homologous chromosomes in YS-1 were generally larger than those of MY-1. The Giemsa method was proved to be useful for the identification of somatic chromosome and a C-banded diagram showing knob positions, arm ratios and relative length of chromosome could be used as a good tool to compare the characteristics of chromosomes of Korean indigenous waxy corn stocks.
Cooperative stimulation of cisplatin-mediated apoptosis by hepatitis B virus X Protein and hepatitis C virus core Protein

The co-infection with hepatitis B virus (HBV) and hepatitis C Virus (HCV) is associated with a more severe liver disease and increased frequency in the development of hepatocellular carcinoma com-pared to those with single infection. Here, we demonstrated that HBV X protein (HBx) and HCV Core cooperatively up-regulated the level of p53 in human hepatoma HepG2 cells. The elevated p53 subsequently stimulated the expression of proapoptotic Bax whereas it repressed the expression of antiapoptotic Bcl2. These effects, however, were not observed in p53-negative Hep3B cells. Consistently to their cooperative regulation of apoptotic effectors, HBx and HCV Core additively stimulated cisplatin-mediated apoptotic cell death of HepG2 but not of Hep3B cells. These results may help to explain the development of a more severe liver disease in patients co-infection with HBV and HCV as well as some contradictory results on the roles of HBx and Core in apoptosis.
Identification and expression of leuD Gene in Rice (Oryza sativa L.)

A rice OsLeuD gene for small subunit of 3-isopropylmalate isomerase (IPMI) (EC 184.108.40.206) has been isolated. OsLeuD gene is located on 109.3 cM of chromosome 2. OsLeuD gene was expressed abundantly in metabolically active organs including leaves and developing seeds, indicating that OsLeuD gene expression is developmentally regulated. The cDNA of OsLeuD gene was coded for 257 amino acids which showed 58% and 48% homology to small subunits of IPMI in OsLeuD genes of cyanobacteria and green sulfur bacteria, respectively. The molecular character of OsLeuD is closely related to those of photosynthetic bacteria rather than those of eukaryotes including fungi and yeast. This suggests that OsLeuD gene in chromosomal genome of plants may possibly be originated from chloroplast genome.
NADPH oxidase inhibitor diphenyleneiodonium induces p53 expression and cell cycle arrest in several cancer cell lines

The Diphenyleneiodonium (DPI) is widely used as an inhibitor of flavoenzymes, particularly NADPH oxidase. In this study, we investigated the effect of DPI on the cell growth progression of human colon cancer cells HCT-116 (wild-type p53), HT-29 (p53 mutant) and human breast cancer cells MCF-7 (wild-type p53). DPI treatment in cancer cells evoked a dose- and time-dependent growth inhibition, and also induced the cell cycle arrest in C2/M phase. The peak of cell population arrested in C2/M phase was observed at12 hr after treatment of DPI. In addition, DPI significantly induced the expression of p53, which induces proapoptotic genes in response to DNA damage or irreparable cell cycle arrest, at 6 hr in DPI-stimulated cells. However, a catechol apocynin, which inhibits the assembly of NADPH oxidase, did not induce p53 expression. This suggest that p53 expression induced by DPI is not associated with the inhibition of NADPH oxidase. In conclusion, we suggest that DPI induces the expression of wild-type p53 by ROS-in-dependent mechanism in several cancer cells, and upregulated p53 may be involved in regulatory mechanisms for growth inhibition and cell cycle arrest at C2/M phase in DPI-stimulated cells.
Chromophore formation and phosphorylation analysis of constitutively active rhodopsin mutants

G protein coupled receptors (GPCRs) transmit various extracellular signals into the cells. Upon binding of the ligands, conformational changes in the extracellular and/or transmembrane (TM) domains of CPCRs were propagated into the cytoplasmic (CP) domain of the molecule leading to the activation of their cognate heterotrimeric C proteins and kinases. Constitutively active GPCR mutants causing the activation of C Protein signaling even in the absence of ligand binding are of interest for the study of activation mechanism of GPCRs. Two classes of constitutively active mutations, categorized by their effects on the salt bridge between Ell3 and K296, were found in the TM domain of rhodopsin. Opsin mutants containing combinations of the mutations were constructed to study the conformational changes required for the activation of rhodopsin. Rhodopsin chromophore regenerated with 11-cis-retinal showed a thermal stability inversely correlated with its constitutive activity. In contrast, rhodopsin mutants exhibited a binding affinity to an agonist, all-trans-retinal, in a constitutive activity-dependent manner. In order to test whether the conformational changes responsible for the activation of trans-ducin (Gt) are the same as the conformation required for the recognition of rhodopsin kinase, analysis of the mutants were carried out with phosphorylation by rhodopsin kinase. Rhodopsin mutants containing combinations of different classes of the mutations showed a strong synergistic effect on the phosphorylation of the mutants in the dark as similar to that of Gt activation. The results suggest that at least two or three kinds of segmental and independent conformational changes are required for the activation of rhodopsin and the conformational changes responsible for activating rhodopsin kinase and Gt are similar to each other.
Effect of Solvent Fractions from Doenjang on Antimutagenicity, Growth of Tumor Cells and Production of Interleukin-2

We studied the inhibitory effect of solvent fractions from doenjang on mutagenicity using Salmonella typhimurium TA100 in Ames test. We also investigated the effect of solvent fractions from doenjang on the growth of tumor cells and the production of interleukin-2 (IL-2). The treatment of dichlorormethane and ethylacetate fractions (2.5 mg/assay) from doenjang to Ames test system inhibited aflatoxin B
) induced mutagenicity by 96% and 97%, respectively, and showed a higher antimutagenic effect than other solvent fractions. In case of N-methyl-N`-nitro-N-nitrosoguamidine (MNNG) induced mutagenicity, the ethylacetate fraction showed the highest inhibitory effect (by 75%) among the other sol-vent fractions, although the inhibitory effect was not stronger compared to AFB
induced mutagenicity. The treatment of dichloromethane and ethylacetate fractions markedly inhibited the growth of Yac-1 (by 80% and 94%, respectively) and sacroma-180 cancer cells (by 60% and 96%, respectively) after 4 days of incubation at 37
C. To elucidate the immunological mechanism of antitumor activity of doenjang, spleen cells of Balb/c mouse were exposed to the dichloromethane and ethyl-acetate fractions for 24 hours at 37
C . The culture supernatants following the treatment of djchloromethane and ethylacetate factions to spleen cells increased the production of IL-2. These results indicated that the anticarcinogenic effect of doenjang was mediated by the production of IL-2.
Optimal Culture Conditions on the Tyrosinase Inhibitor Production by Actinomycetes F-97

A Actinomycetes F-97 producing tyrosinase inhibitor was isolated from soil samples. The optimum culture condition for 쇼rosinase inhibitor production was investigated and the results were as follows. The best carbon source for tyrosinase inhibitor production was shown as soluble starch, the optimum concentration was 3.0%. The best nitrogen source for tyrosinase inhibitor production was shown as peptone, the optimum concentration was 0.36%. As effect of metal ions on the production of tyrosinase inhibitor, K
was shown the best and the optimum concentration was 0.1 mM. The optimum pH and temperature was shown 7.0 and 30
C, respectively. And the highest tyrosinase inhibitor production was observed at 70hr cultivation under optimum conditions in jar fermentor scale.
ISSR을 이용한 고추나물 집단의 유전적 다양성과 계통학적 연구

Inter simple sequence repeat (ISSR) markers were performed in order to analyse the phylogenetic relationships of eight Hypericum electum populations in Korea. The six primers were produced 37 reproducible ISSR bands. Analysis of ISSR from individual plants of Korean H. erectum resulted in 22 polymorphic bands with 59.5%. Across populations, the mean number of alleles per locus was 1.348 and Shannon`s information index was 0.203.Population Mt. Gyeryong had the highest expected genetic diversity (0.175) among all populations. When species were grouped by eight populations, within group diversity was 0.140 (Hs), while among group diversity was 0.472 (G
) on a per locus basis. The estimated gene flow (Nm) for H. erectum was very low (0.561). It is suggested that reproductive isolation by the isolation of geographical distance among H. electum populations and genetic drift may have played roles in shaping the population structure of this species. In phonetic tree, all populations were well separated from each other. Thus, ISSR markers are very effective in classifying natural population levels of genus Hypericum in Korea.
Isolation and Antimicrobial Susceptibility of Campylobacter jejuni from Diarrhea Patients

ln this study we isolated 27 isolates of Campylobacter jejuni from stool samples of 882 diarrheal patients. The seasonal distribution of patients was highest at July (11.7%). All the isolates of C. jejuni hydrolyzing sodium hippurate were serotyped on basis of heat-stable antigens, and identified with the use of passive hemagglutination assay. A total of 59.3% among 27 C. jejuni isolates were identified into 6 different serotypes, which serotype HS2, HSl/44, and HS2l were dominant. Antibiotics resistant rates of C. jejuni isolates were shown to be 100%, 63.0%, 51.9%, 37.0%, 33.3%, 25.9% and 7.4% to cephalothin, trimethoprim- sulfamethoxazole, tetracycline, ciprofloxacin, ampicillin, gentamycin and clindamycin, respectively. All isolates were sensitive to the erythromycin and imipenem.
nhibition of Melanin Synthesis by Mycelial Culture Broth of Paecilomyces japonica in the Mulberry

This study was carried out to investigate the whitening effect of mycelial culture broth of P. japonica in the mulberry leaf extract. The culture broth inhibited melanin synthesis in vitro in the concentration dependent manner of 10 to 50%. The mulberry leaf extract itself has a cytotoxicity to the mouse melanoma cell, B16BL6 with the value of IC
of 7% concentration. But the culture broth of P. japonica in the mulberry leaf extract did not show any cytotoxicity up to 50%. Therefore, we concluded the mycelial culture broth of P. japonica in the extract of mulberry leaf can be used as a whitening cosmetic resource.
Efficient plant regeneration from cotyledon and primary leaf explants of lettuce (Lactuca sativa L.)

The efficient system for plant regeneration from cotyledon and primary feat explants of lettuce was established. Plant regeneration efficiency was shown 91.3% from cotyledon and 85.9% from primary leaf explants of variety `Jungtongpogi` in KN medium. Plant regeneration efficiency was also estimated with various plant regeneration media in variety` Chungchima`, which was lowest plant regeneration efficient showing 35.4% from cotyledon and 30.3% from prima leaf explants in KN medium. Kl medium increased 77.9% and 80.7% of plant regeneration efficiencies from cotyledon and primary leaf explants of variety `Jungtongpogi` were cultured on KN medium. In case of varie쇼 ‘Chungchima`, efficient plant regeneration was shown when primary leaf explants were cultured on SH and KI media.
Control effect of the newly developed insecticidal protectant on Sycamore lace bug, Corythucha ciliata (Hemiptera : Tingidae)

To determine the effects of newly developed insecticide (Ultra) on Sycamore lace bug (Corythucha ciliata), ten leaf discs were sprayed with the Ultra solution including the other selected commercialized insecticide solutions such as Carbaryl and Pirimicarb, and also with a water-sprayed control. According to the results, the control effect of Ultra on the adults and immatures was respectively 98.9% and 97.5% after 48hr, and the corresponding values were higher than that of the other tested insecticides (i.e Carbaryl showed 84.6% for adults and 85.6% for immatures; Pirimicarb showed 83.0% for adults and 90.7% for immatures). For the Susceptibility test, LC
value of Ultra showed the lowest as 1.5ppm, the value of Carbaryl and Pirmicarb were 3.7 ppm and 30.4 ppm, respectively. Therefore, the insecticidal effects were better for the newly developed insecticide than the commercialized insecticides.
Effect of Plant Crowth Regulators on Production of Good Quality Plant for Korean Gerbera Lines

This study was carried out to obtain information about the valuable combination and concentration to produce good plants of cut gerbera flowers of 19 domestically - bred gerbera cultivars. BA and kinetin combination is more effective than NAA and BA combination to produce good plants and mass propagation of domestically - bredgerbera cultivars. Normally almost of 19 cultivars, mass propagation was more effective on the medium containing 1.0 mg/L BA + 0.5, 1.0 mg/L kinetin. But some cultivars, `Sunnyeo`, `Oksaem`,`Piny`, and `Pink Light`, vitrified plants were induced on MS medium high level of BA concentration(BA 1.0 mg/L), in comparison with those on the medium with low level of BA(0.1 mg/L).Fresh and dry weight, more effective on the medium containing BA 0.5 mg/L . Kinetin 0.1, 0.5 mg/L. Anatomical investigation of vitrified leaf, stomata of vitrified leaves were circular and inflated, where-as those of normal leaves.
Study on the Mineral Contents of Commercial powdered infant formula

This study was conducted to investigate mineral contents of commercial powdered infant formula for obtaining basic data on infant nutrition. 11 Commercial infant formula based on cow`s milk were collected and the contents of macro minerals (calcium, phosphorous, sodium, potassium and magnesium) and trace minerals (iron, zinc and copper) were compared with Dietary Reference Intakes for Koreans (KDRIs). The overall mineral contents in 100 g and in 100 kcal of infant formula satisfied the recommended formula regulation and Codex. In infant formula during 0-5 monthly age, calcium, phosphorous, sodium, potassium, magnesium, zinc and copper could supply 233.1%, 273.5%, 156.7%, 142.0%, 150.8%, 209.3%, 171.1% of recommended daily mineral intakes, respectively. The content of iron in 0-5 monthly age formula supplied2842.6% of recommended daily iron intakes. In infant formula during 6-11 monthly age, calcium, phosphorous, potassium, magnesium, iron, zinc and copper satisfied their recommended daily intakes. However, sodium only supplied 76.6% of its recommended daily intake. Intake ratio between Ca/P, Ca/Mg, Ca/Fe, Na/K and Zn/cu in infant formula during 0-5 monthly age were 1.7±0.2, 11.0±2.4,64.9±10.0, 0.3±0.1, and 9.6±1.0, respectively. Intake ratio between Ca/P, Ca/Mg, Ca/Fe, Na/K and Zn/cu in infant formula during 6-11 monthly age were 1.7
13.8, 0.3, and 9.4
1.1,respectively. From this study, evaluation of mineral contents of commercial infant formula was established, which could strengthen the basic information on infant nutrition.
SEREX; discovery of tumor antigens

The identification of tumor antigens is essential for the development of anticancer therapeutic vaccines and clinical diagnosis of cancer. SEREX (serological analysis of recombinant cDNA expression library)has been used to identify such tumor antigens by screening sera of cancer patients with cDNA ex-pression libraries. SEREX-defined antigens provide markers for the diagnosis of cancers. SEREX is also a powerful method for the development of anticancer therapeutics. The development of anticancer vaccines requires that tumor antigens can elicit antigen-specific antibodies or T lymphocytes. This re-view provides information on the application of SEREX for discovery of tumor antigens.

The endoplasmic reticulum (ER) is an important intracellular organelle for folding and maturation of newly synthesized transmembrane and secretory proteins. The ER provides stringent quality control systems to ensure that only correctly folded proteins exit the ER and unfolded or misfolded proteins are retained and ultimately degraded. The ER has evolved stress response both signaling pathways the unfolded protein response (UPR) to cope with the accmulation of unfolded or misfolded proteins and ER overload response (EOR). Accumulating evidence suggests that, in addition to responsibility for protein processing, ER is also an important signaling compartment and a sensor of cellular stress. In this respect, production of bio-functional recombinant-proteins requires efficient functioning of the ER secretory pathway in host cells. This review briefly summarizes our understanding of the ER signaling developed in the recent years to help of the secretion capacities of recombinant cells.

The aging is related to numerous changes in our body system which declines functional ability. The functional ability in advanced age can be evaluated by the 4 components of fitness (cardiorespiratory endurance, muscular strength and endurance, flexibility and body composition). The understanding and knowledge of changing 4 components of fitness and the role of exercise on the process of aging is of paramount importance. Despite exercise and physical activity have been shown to delay the process of aging, the exercise guidelines and recommendations for old adults are not well published. This review is to presents the age-related changes in each component of fitness and to present current guidelines and recommendations for exercise in older adults.
The inhibitory effect on the melanin synthesis in B16/F10 mouse melanoma cells by Sasa quelpaertensis leaf extract

Effects of hot-water extract from Sasa quelpaertensis leaf (HWES) on melanogenesis were investigated in B16/F10 mouse melanoma cells. HWES inhibited cellular tyrosinase activity and melanin biosynthesis in a dose-dependent manner. Western blotting analysis showed that HWES dose-dependently inhibited tyrosinase and tyrosinase related protein-1 expression. Also, HWES suppressed sustained ERK activation in a concentration-dependent manner, suggesting that HWES inhibits the melanin biosynthesis through the suppressive effect against pathway involving sustained ERK activation.
