The Changes in Structural Carbohydrate on Crown Daisy and Butterbur

Change on the structural carbohydrate(several fiberous components) was determined by vegetables(crown daisy and butterbur)-cultivated in Ulsan, Kyungnam, Korea-as its stage of maturity developed. Samples were separated into leaf and stem, which were dried at 7
for 24hr, and ground to pass a 1mm screen. They were subjected to moisture, crude protein, crude fat and several dietary fiber-DF(dietary fiber, include unavaible components), NDF(neutral detergent fiber), ADF(acid detergent fiber), lignin, hemicellulose, cellulose and protein corrected NDF(c-NDF), IDF(indigestible fiber, include lignin, hemicellulose and cellulose). In general, structural carbohytrate(several dietary fiber) of vegetable was affected by the growth stage. In case of crown daisy and butterbur, dietary fiber in leaf was higher than DF in stem.
Organogenesis by Combined-Dose of Activin A and IGF-1 In Xenopus Presumptive Ectoderm

The trend of organogenesis in Xenopus presumptive ectoderm was studied by combined dose of activin A and IGF-1(insulin-like growth factor-1). In reference study of Asashima and his colleagues, the inductive patterns of various organs were reported with activin, the potent mesoderm inducing factor. In present study, the inducing pattern was cleared with combined-dose of concentration 1-100 ng/ml activin A and IGF-1. In addition, the result from single treatment of activin A was compared with former study. As a result, eye was differentiated in 5-20% of explants at 10 and 50 ng/ml concentrative combination of activin A. Otic vesicle was appeared in the entire concentrative combination of IGF-1. Pronephric duct was induced 19-38% of explants at the concentration of activin A 100 ng/ml by adding IGF-1. The comparison of single treatment of activin A was showed some difference in dose-dependent inducing pattern.
Isolation of Aerobic Bacteria and Its Efficacy for the Treatment of Korean Food-Wastes

For the treatment of Korean food-wastes, three mesophilic and one thermophilic bacteria were isolated from soil and fermented fertilizers. The thermophilic Streptomyces sp. strain WF021 produced two enzymes which were a protease and a lipase at 55

. The bacteria were grown in media containing 6% NaCl at least and did not have antagonism each other. The four isolates treated much more food-wastes than referance strains did. In a flask without aeration, three reference strains treated 15.4% of food-wastes, while four isolates treated 23.7% of food-wastes. In a flask with aeration, food-wastes were treated 67.3% by four isolates, and 64.3% by three reference strains, but 53.9% without bacteria. However, food-wastes were treated about 78% in a 200
-reactor made by Siwon Co., while 65.8% in a 20
-reactor made by Sanyo Co.
Study on the Reproductive Biology of the Giant Freshwater Prawn, Macrobrachium rosenbergii(De Man) IV. Oviposition, Fertilization and Brooding

Timing of oviposition, nuclear maturation of oocytes, egg-sperm contact and brooding were observed in the giant fresh-water prawn Macrobrachium rosenbergii reared in the laboratory. When a pre-spawning molted female was transferred to the male, the female oviposited commonly at 5 to 10 hr after mating. The first polar body were extruded within 2-3 min after spawning. The egg chromosome progressed to release second polar body by 30 min and mitotic nuclear division occurred in 8 hr after spawning. Thumbtack-shaped spermatozoa were seen peneterating egg membrane with their spike directed forward. Transformation of spermatozoa and fertilization membrane could not be observed by light microscopy. The elapsed hours from oviposition to the end of brooding wee taken in 5-7 hr. Brood size for female 9 cm and 18 cm in body length was estimated to be 10,000~15,000 and 100,000 eggs, respectively and is proportional to the total length of the berried females. The ovigerous females reared at chlorinity of 2.21~4.25
Cl. showed a normal egg development up to hatching.
Isolation of Bacteria from Freshwater Plant Roots

The isolation and identification of bacteria from freshwater plant root system were performed. Twenty four different strains were identified by microbiological identification system. Gram negative bacteria were isolated three times more than Gram positive ones. The ratio of rod and coccus was 11:1. The similarity above 0.5 was 37.5% from the tested samples in identification. Among these isolated bacteria, three dominant strains Pseudomonas cepacia JH10, Xanthomonas maltophilia JH12, Aeromonas salmonicida JH13 were selected to investigate biochemical properties and optimal growth conditions. These strains reached maximum growth after 24-36 hr of incubation in peptone broth, and their optimal temperature was 28
and that of pH was between 7 and 8.
Construction and Analysis of cDNA Library from Porphyra yezoensis

As an attempt to preserve resources in marine biological organisms, we first constructed a cDNA library from the red alga Porphyra yezoensis. The library construction method from P. yezoensis consists of three steps; those include protoplast presparation, RNA isolation, and phage library construction. Protoplast was prepared in order to remove much of the carbohydrate compounds which are characteristics of algal cell walls. Carbohydrate contamination in the purified RNA may inhibit further enzyme reactions, those carbohydrates should be removed. RNA samples prepared from protoplast still seemed to contain residual amount of carbohydrate because mRNA isolation with conventional method failed. We therefore developed a method with Poly ATtract mRNA isolation system. The constructed phage library was tested by analyzing cDNA insert in phage vector from randomly picked ten independent white plagues. All of the phages contained cDNA inserts with sizes ranging 0.5kb and 2.0kb.
Effects of Sea Tangle (Laminaria japonica) and Fucoidan Beverages on Sociopsychological Stress

This study was designed to investigate the effects of sea tangle (Laminaria japonica) extract (Dasi-Ex group: dry base 4.0%) and fucoidan-added (Fuco-I, II, III group: fucoidan of 1.0%, 2.0%, 3.0% added to Dasi-Ex) beverages on the anti-stress action. ICR male mice (20
2g) were fed basic experimental diets and given free through water bottle filled with these beverages instead of water for 18 days including sociopsychological stress. Body weight gains were consistently lower in Dasi-Ex and Fuco-I,II,III groups compared with control group, expecting in a inhibitory effect of obesity. Dasi-Ex group resulted in a significant decrease of 25% in serum corticosterone (CS) secretion, while Fuco-I,IIand IIIgroups resulted in a marked decreases of 45~55% in serum CS secretion compared with control group. Noradrenaline (NA) secretions were significantly increased about 15% in Dasi-Ex group, and 20~22% in Fuco-I,II,III groups compared with control group. Significant differences in brain MHPG-SO4 levels of Dasi-Ex group could not be obtained, but Fuco-I,II,III groups resulted in a marked decreases of 20~25% in brain MHPG-SO4 ratio of brain, but Fuco-I,II and III groups resulted in a marked increases of 45~60% in NA/MHPG-SO4 ratio of brain compared with control group. These results suggest that fucoidan beverage may play a effective role in a ridding of the sociopsychological stress by pivotal anti-stress effect of fucoidan.
Effects of Ganoderma lucidum Extract on Memory and Oxidative Stress of Senescence-Accelerated Mouse

Long-term effects of Ganoderma lucidum (GL) on memory and oxidative stress of senescence-accelerated mice (SAM) were investigated. Senescence-resistant (R1) and prone (P8) strains of SAM were fed GL diets, premixed with low (20 mg/kg/day, T1) or high (200 mg/kg/day, T2) levels of GL powder for 9 months starting from young (3 months of age) or for 5 months starting from old (7 months of age). After the final feeding at 12 months of age, all animals were subjected to passive avoidance test for the evaluation of memory function. In addition, the changes in hepatic thiobarbituric acid-reactive substance (TBARS) and glutathione were analyzed. SAMP8 fed GL diets from old age (7 months) exhibited the improvement of memory, although GL rather inhibited the memory function of both SAMR1 and SAMP8 mice fed diets from young (3 months of age). Hepatic TBARS contents were decreased in SAMP8 fed high GL diet for 9 months and in SAMR1 fed low GL diet for 5 months. Hepatic glutathione content was also remarkably increased in SAMR1 following both feeding periods, and less extent in SAMP8 fed diet for 5 months of age. Taken together, it is proposed that GL extracts may play an anti-aging role through antioxidant action, and thereby may improve the senescence-related memory dysfunction.
Expression of Human Immunodeficiency Virus Type 1 Gag Protein in Escherichia coli

Presence of antibody to the capsid protein p24 is the main diagnostic criterion, since this reflects reliable antibody response to HIV infection. However, it takes about 6-8 weeks for antibody production after infection and people who are infected but antibodies are not produced yet are classified as seronegative. Therefore, there is a strong need for an improved diagnostic method for better health security. As a first step for developing such an improved diagnostic system, gag protein of human immunodificiency virus type 1 was expressed in E. coli DH5
. The gag fragment of HIV-1 (including a portion of p17 and whole p24) was amplified by polymerase chain reaction (PCR) and BamH I/EcoR I sites were created during PCR. The amplified DNA fragment was cleaved with BamH I/EcoR I and was subcloned into the GEX-2T vector which had been digested with BamHI/EcoRI, resulting gene fusion with gst gene of pGEX-2T. for 3h. Recombinant gag protein or GST-gag fusion protein was purified with glutathione-sepharose 4B bead and migrated as a single band when analyzed by 10% polyacrylamide gel. These proteins were confirmed by immunoblotting with anti-GST goat sera or Korean AIDS patients sera. The results of this study establish the expression and single step pulification of HIV-1 gag protein which can specifically bind with Korean AIDS patients sera.
Establishment of Regeneration System and Antibiotic Sensitivity Test for Transformation of Various Vegetable Crops

This study was conducted to determine the concentrations of plant growth regulators required for regeneration and the concentrations of antibiotics for the selection of transformed regenerants from lettuce, musk melon and tomato. The optimal concentrations of plant growth regulators for shoot formation were NAA 0.1 mg/

for lettuce, NAA 0.01 mg/
+BA 2.0 mg/
for melon and NAA 0.1 mg/
+BA 0.5 mg/
for musk melon. Shoot induction from tomato, lettuce and melon was completely inhibited by 30 mg/
or higher concentrations of kanamycin. Shoot formation from mu나 melon was not affected by kanamycin up to 40 mg/

and completely inhibited by 100 mg/
. Shoot formation of all four crops was completely inhibited by higromycin at 10 mg/
. Both carbenicillin and cefatoxinme did not show any negative effects on shoot formation.
Isolation and Identification of Influenza Virus from Pusan, 1998-1999

Investigate the epidemics for influenza outbreaks. The outbreak pattern of the internal patients housed in the 10 designated hospitals was monitered to investigate and the characteristics of the virus isolates are as follows. 232 strains of influenza virus was isolated from the oral specimen of 1,320 respiratory disease patients in Pusan from Oct. 1998 to Jun. 1999. Among these isolates, 222 strains were A-type and the rest were B-type. The outbreak pattern for sex-and age-groups is as follows. The male outbreak was similar to the female outbreak: male outbreak, 47.4% and female outbreak, 52.5%. Most of the patients were less than 10 years old. The monthly influenza outbreak was consistent from Dec. 1998 to Apr. 1999. and The 113 strains from the A-type isolates were A/ Sydney/05/97(H3N2)-like, the 109 strains were A/Beijing/262/95(H1N1)-like, and all of the 10 B-type isolates were B/Harbin/07/94-like.
A Study on the Development of Analytical Method for Micro-Odorous Compounds in Water

Even if odorous compounds remained very low concentration in water, it caused strong odor. Because Geosmin and most of odorous compound had very vaporization, those were difficult to analyze with GC/MSD and Purge & Trap. So, we needed pre-treatment method for decreasing amounts of extracting solvents, improving recovery efficiencies and increasing analytical efficiencies. This study developed efficient technology for analyzing odorous compounds, using various adsorbents and extracting solvents. The optimum adsorbent was XAD resins. Especially, XAD-2, XAD-7 and XAD-2010 were superior, but XAD-2 of these and MTBE was the optimum extraction solvent. Other extraction solvent's efficiency was in order of MTBE>Dichloromethane>n-Hexane>Diethylether. The optimum NaCl dosage for increasing efficiency was 5g in liquid-liquid extraction method. The shaking time(0∼24hr) had no concern with adsorption efficiency. The optimum adsorbent was XAD-2 resin and extraction solvent was MTBE. Dosing NaCl, adsorption efficiency was increased in liquid-liquid extraction method, but NaCl has no effect on liquid-solid extraction method. In this experimental results, this method will apply to not only Geosmin but other well-known odorous compounds (2-MIB, IBMP, IPMP, TCA) and algae toxins (Mycrocystin, Anatoxin etc)
A Study on Geosmin Removal of Algae Byproducts by Ozonation and Photocatalysis

This study was carried out to compare ozonation with photocatalysis degradation for removal Geosmin of algae byproduct. The change of pH was decresed from 7.02 to 2.8 after contact time 480 minute for ozonation. In case of UV-germicidal lamp, pH was very quickly increased from 7.02 to 7.5, but Halogen lamp did very slowly change pH. Geosmin degradation ratio was as following, UV-germicidal lamp/TiO2(100mg/L) O3>UV-germicidal lamp/TiO2(50mg/L)>UV-germicidal lamp(10W)>halogen lamp(50W). Instead of TiO2 suspension solution, Geosmin degradation ratio was very low using hollow bead and pellet as coated TiO2. As a result of identifing byproducts, ozonation generated three species of aldehyde such as 3-Heptanone and three species of alcohol such as Heptanal, but photocatalysis formed 1, 14-Tetradecanediol infinitesimally.
A Study on Ozone Oxidation of Algae

The objectives of this research were to estimate variation characteristics of TOC, UV-254 and NH4+-N, etc, if odorous generated algaes flowed into water treatment plant and they contacted with ozone known as typical advanced treatment. It was estimated that pH decreased from initial 7.4 to 2.1 after ozone contact 100min. pH declined to early 10min. suddenly and then pH drop did almost occur. TOC and UV-254 continued to decrease with passed time of ozone contact. NO3--N concentration was much higher than NH4+-N and NO2--N for whole test period. Because ozone oxidized organic matters as various kinds of ion material and ion intensity of sample was extended, conductivity showed high as passed time of ozone contact. Owing to ozone oxidation of algae, color did almost disappear after ozone contact 20min.
Effects of Mind Tonic Drinks on Sociopsychological Stress

The effects of anti-stress mind tonic (ASMT) drink (ASMT-0.1% and ASMT-0.5% concentration) were tested for the anti-stress action. ICR male mice (20
2 g) were fed with basic experimental diets free access to water containing these ingredients for 18 days. Psychological stress/sociopsychological stress were exposed by foot-shock for 1 hour for 3 days. Both ASMT-0.1 and ASMT-0.5 groups in the sociopsychological stress resulted in a significant decrease of 19.1% and 41.9% in plasma corticosterone (CS) secretion compared with psychological stress (control group). Noradrenaline (NA) secretions in the brain were significantly increased 23.4% and 35.9%, respectively, in ASMT-0.1 and ASMT-0.5 groups compared with control group. MHPG-SO4 (3-Methoxy-4-hydroxyphenylethyleneglycol sulfate) levels in the brain resulted in a marked decreases of 27.1% and 19.6%, respectively in ASMT-0.1 and ASMT-0.5 groups. NA/MHPG-SO4 ratios in the brain of ASMT-0.1 and ASMT-0.5 groups resulted in a significantly increase of about 70% compared with control group. These suggest that mind tonic anti-stress drink can effectively ridded the sociopsychological stress.
Effects of Chrysanthemum Extract Drinks on Sociopsychological Stress

The effects of anti-stress chrysanthemum flower (ASCF) extract drink (ASCF-0.1% and ASCF-0.5% concentrations) were tested for the anti-stress action. ICR male mice (20
2 g) were fed with basic experimental diets and given free access to water containing these ingredients for 18 days. Psychological stress/sociopsychologcal stress exposed by foot-shock for 1 hour for 3 days. Both ASCF-0.1 and ASCF-0.5 groups in the sociopsychological stress resulted in a significant decrease of 28.1% and 27.3% in plasma corticosterone (CS) secretion compared with psychological stress (control group). Noradrenaline (NA) secretions in the brain were significantly increased 49.7% and 53.9%, respectively, in ASCF-0.1 and ASCF-0.5 groups compared with control group. MHPG-SO4 (3-Methoxy-4-hydroxyphenylethy-leneglycol sulfate) levels in the brain resulted in a marked decreases of 12.9% and 16.6%, respectively in ASCF-0.1 and ASCF-0.5 groups. NA/MHPG-SO4 ratios in the brain of ASCF-0.1 and ASCF-0.5 groups resulted in a significantly increase of 71.3% and 81.0%, respectively, compared with control group. These results suggest that anti-stress chrysanthemum flower (ASCF) drink can effectively ridded the sociopsychological stress.
Effects of Rose Fruit Extract Drinks on Sociopsychological Stress

The effects of anti-stress rose fruit (ASRF) extract drink (ASRF-0.1% and ASRF-0.5% concentrations) were tested for the anti-stress action. ICR male mice (20
2 g) were fed with basic experimental diets and given free access to water containing these ingredients for 18 days. Psychological stress/sociopsychological stress exposed by foot-shock for 1 hour for 3 days. Both ASRF-0.1 and ASRF-0.5 groups in the sociopsychological stress resulted in a significant decrease of 24.3% and 38.0% in plasma corticosterone (CS) secretion compared with psychological stress (control group). Noradrenaline (NA) secretion in the brain were significantly increased 35.9% in ASRF-0.5 group, but NA secretion in ASRF-0.1 group could not be obtained significant result compared with control group. MHPG-SO4 (3-Methoxy-4-hydroxy-phenylethy leneglycol sulfate) levels in the brain resulted in a marked decreases of 16.5% and 18.9% respectively in ASRF-0.1 and ASRF-0.5 groups. NA/MHPG-SO4 ratios in the brain of ASRF-0.1 and ASRF-0.5 groups resulted in a significantly increase of 16.5% and 67.0%, respectively, compared with control group. These results suggest that anti-stress rose fruit (ASRF) drink also can effectively ridded the sociopsychological stress.
Growth Characteristics and Change of Inorganic Element in the Leaf of Perilla by Replanting

This study was conducted to elucidate the growth characteristics and change of inorganic element in the leaf of perilla by replanting. The replanting injury in the height of perilla appeared from the 2nd and 3rd year after replanting, and the sickness of soil occurred from the 4th year. Number of node of perilla by replanting was significantly affected to the middle stage of growth, but was similar at the latter stage of growth. Weight of a leaf was the highest at the 1st and 2nd year, and decreased with the replanting. Also, the rate of dry leaf decreased with the replanting. In the content of inorganic element of the perilla leaf by replanting, N in the leaf increased with the replanting, and K and S decreased. However the content of Ca, Cu and Mn did not affected to replanting.
Effect of Fertilizer Level of Organic Compost on Leaf Characteristics, Leaf Number and Replanting in Perilla frutescens

This study was conducted to elucidate the leaf characteristics, leaf number and fertilizing level in perilla, Perilla frutescens, which was cultivated using five types organic compost with three level. Number of the harvested leaves was effective in Heulgnara with 800 kg/10 and Poongjag with 1200 kg/10 at 70 days and organic compost were the highest in Sarang followed by Heulgnara and Poongjag in order. Number of leaf harvested during the fifth times was the highest Poongjag followed by Heulgnara and Sarang in order. Leaf length and leaf width were the highest in Poongjag with 1200 kg/10 a at 70 days. Simple method for calculation of fertilizer level was N 6.09 kg and slaked lime 36.56 kg in Heulgnara 100 kg per 10 a. Recommendation level of chemical fertilizer in Heulgnara due to fertilizer method was N 275.10 kg, P 4.3 kg and K 10 kg per 10 a.
