Gene Expression Analysis Using cDNA Microarray Assay by Cervi Pantotricuhum Cornu Herbal Acupuncture (녹용약침액(鹿茸藥鍼液)의 DNA chip을 이용(利用)한 유전자(遺傳子) 발현(發顯) 분석(分析))

  • Objective : Bone homeostasis is maintained by balance of bone formation and resorption. Therefore, bone related diseases arose by disturbance of this balance between osteoblast and osteoclast activities. To develop a successful screening system the therapeutic components based on oriental medicine is essential to set up systematic approach for that purpose. The purpose of this study is to the know the gene expression using cDNA microarray assay. Methods : Cervi Pantotricuhum Cornu Herbal-acupuncture extract was prepared by boiling. human osteosarcoma cells(HOS) were treated with Cervi Pantotricuhum Cornu Herbal-acupuncture solution. Then mRNA was extracted and cDNA microarray assay was performed. Results : Human osteosarcoma cells(HOS) treated with Cervi Pantotricuhum Cornu Herbal-acupuncture solution($500{\mu}g/m{\ell}$) showed that thioredoxin, TAFII31 and two novel genes were increased. However many genes decreased their expression by Cervi Pantotricuhum Cornu Herbal-acupuncture. Conclusions : This type of approach will give a good chance to explore the favorable effects of Cervi Pantotricuhum Cornu Herbal-acupuncture. Further study is needed for investigating the effect of Cervi Pantotricuhum Cornu Herbal-acupuncture.

A Study on Effects of the Cervi Pantotricuhum Cornu Herb-acupuncture on the Osteoporosis Induced by Ovariectomy in Rats (녹용(鹿茸) 약침(藥鍼)이 난소적출(卵巢摘出)로 유발(誘發)된 백서(白鼠)의 골다공증(骨多孔症)에 미치는 영향(影響))

  • In order to investigate the effect of the Cervi Pantotricuhum Cornu Herb-acupuncture treated on Shinsu($BL_{23}$) and Taejo($BL_{11}$) loci on the estrogen-deficiency osteoporosis induced by ovariectomy in rats, serum osteocalcin estradiol progesterone follicular stimulating hormone(FSH) calcium(Ca) and phosphorous(P) levels were monitored. The results were as follows : 1. Serum osteocalcin levels were significantly (p<0.01, p<0.05) increased in the Cervi Pantotricuhum Cornu Herb-acupuncture treated on Shinsu and Taejo group compared with control group. 2. Serum estradiol levels were increased In the Cervi Pantotricuhum Cornu Herb-acupuncture treated on Shinsu and Taejo group compared with control group, but Taejo group did not show significant value. 3. Serum progesterone levels were significantly(p<0.01) increased in the Cervi Pantotricuhum Cornu Herb-acupuncture treated on Shinsu and Taejo group compared with control group. 4. Serum FSH levels were significantly(p<0.01, p<0.05) decreased in the Cervi Pantotricuhum Cornu Herb-acupuncture treated on Shinsu and Taejo group compared with control group. 5. Serum Ca levels were significantly(p<0.01) increased in the Cervi Pantotricuhum Cornu Herb-acupuncture treated on Shinsu group compared with control group, but Taejo group did not show significant value. 6. Serum P levels were significantly(p<0.01, p<0.05) decreased in the Cervi Pantotricuhum Cornu Herb-acupuncture treated on Shinsu and Taejo group compared with control group. According to these results, the Cervi Pantotricuhum Cornu Herb-acupuncture treated on Shinsu and Taejo loci have favorable effect on treatment and prevention of osteoporosis induced by ovariectomy in rats.

Healing of Bony Defects by Cervi Pantotricuhum Cornu Herbal Acupuncture (녹용약침(鹿茸藥鍼)이 골형성(骨形成)에 미치는 영향(影響))

Methods : This study was perforated Sprague-Dawley rat femur for bony defects(${\phi}5mm$) by the fissure bur. And experimetal group were treated with Cervi Pantotricuhum Cornu injection at both $Sh{\grave{e}}nsh{\tilde{u}}$(BL23) & $D{\grave{a}}zh{\grave{u}}(BL11)(0.2m{\ell})$. This was evaluated by radiography and histological analysis with in situ hybridization. Results : Cervi Pantotricuhum Cornu Herbal Acupuncture has weak effect on bony defect healing and this was evaluated by X-ray taking and histological analysis with in situ hybridization. Osteocalcin gene expression was not changed by Cervi Pantotricuhum Cornu Herbal Acupuncture in bony defects animal model. Conclusion : Taken together this study show that the Cervi Pantotricuhum Cornu herbal-acupuncture has a weak effect healing of bony defects, this type of approach might give a good chance to explore the favorable effects of Cervi Pantotricuhum Cornu herbal-acupuncture on bone tissue.

  • Object : The tinnitus is a very common otorhinolaryngologic disease. But, we don't know the exact cause and the healing method of that. Treatment : We have three cases report of the patients who have tinnitus. Their tinnitus is diagnosed weakness of the kidney because of chronic symptoms, the Cheek pulse weakening, the whole body weakness, fatigue. So we treat them with herbal medicine and Cervi Pantotricuhum Cornu herbal-acupunture. Result : In the result, their symptoms is reduced. So we report these cases for the improvement of tinnitus treatment efficacy and making the best use of Cervi Pantotricuhum Cornu herbal-acupunture.

Molecular Identification of Deer Antlers using Nucleotide Sequences of Mitochondrial Displacement Loop Region (미토콘드리아 displacement loop 영역의 염기서열을 이용한 녹용의 원산지 동정)

  • It is reported that about 80% of deer antlers (Cervi Pantotricuhum Cornu) produced in the world are consumed in Korea. Fraudulent replacement or mislabeling of costly deer antlers with cheaper ones, however, is one of the most common problems in the Korean deer antler market. Therefore, there is a continuous need for the development of genetic markers to discriminate between genuine and fraudulent deer antlers. This study was performed to develop a method for the identification and authentication of deer antlers using nucleotide sequence analysis against displacement loop of mitochondrial genome among four deer antlers, Cervus eleaphus sibericus, Cervus eleaphus bactrianus, Cervus eleaphus Canadensis, and Cervus eleaphus, originated from Russia, China, North America and New Zealand, respectively. As a result, multiple-alignment of mitochondrial displacement (D) loop region in 1.2 kb showed that, among the four deer antlers, a deleted sequence of about 70 bps was only found in Cervus elaphus bactrianus from China. Finally, Cervus elaphus bactrianus among nine samples of deer antlers were successfully identified by PCR using primer amplifying deleted D-loop. Cervus elaphus bactrianus was also confirmed from cloning the PCR products and their nucleotide sequence analyses were confirmed. However, no marker to identify Cervus eleaphus sibericus, Cervus eleaphus canadensis and Cervus eleaphus were found in the nucleotide sequences of mitochondrial D-loop. Our results suggest that PCR for deleted D-loop region of mitochondrial DNA are useful for identification and authentication of deer antlers of Cervus elaphus bactrianus originating from China.
