Effects of Gamcho on Cytokine Production in Asthma Model Mouse (감초가 천식모델 생쥐의 혈청내 Cytokine에 미치는 효과)

  • This experiment was designed to investigate the effect of Gamcho(Glycyrrhiza uralensis Fisch, GLU) on Antiallergy in asthma. We measured IL-4, IL-5, UL-13, IgE in bronchoalveolar lavage fluid of ovalbumin induced asthmatic mouse and observed murine lung tissue. The results were obtained as follows: 1. GLU decreased the proliferation of IL-4, IL-5, IL-13, IgE significantly. According to the above results, it is suggested that GLU extract might be useful applied for prevention and treatment of allergic asthma.

Suppressive Effects of Hesperidin on Th2-associated Cytokines Expression in RBL-2H3 Cells (RBL-2H3 세포에서 Hesperidin의 Th2 사이토카인 발현 억제 효과)

  • Hesperidin (HES), a flavonone glycoside isolated from the citrus fruits such as lemons and oranges, has been reported to have many biological properties including antiinflammatory, antioxidant, and antiallergy activities. In this study, we focused on the action of HES modulating Th2-associated cytokines such as IL-4 and IL-13 expression in PMA/ionomycin (PI)-stimulated rat basophilic leukemia (RBL-2H3) cells. The production of IL-4 and IL-13 was quantified by ELISA and the mRNA expression was detected by using RT-PCR assay. In addition, western blot analysis was performed to determine the transcription factors involved in the cytokine expression. We found that HES significantly decreased PI-induced IL-4 and IL-13 productions and also decreased the level of mRNA in a dose-dependent manner. Furthermore, western blot analysis of the transcription factors implied that HES down-regulated the protein level of c-Jun and c-Fos, which are the activating protein 1 (AP-1) family and nuclear factor-kappaB (NF-${\kappa}B$) characterized as a transcription factors related to the Th2-associated cytokine expression. Taken together, our data showed that the action of HES responsible for antiallergy activities is based on suppression of Th2-associated cytokines through inhibition of AP-1 and NF-${\kappa}B$ transcription factors.

  • This study was conducted to investigate antiallergy of the polyphenol fractions isolated from persimmon leaves (Diospyros kaki folum). The result of the passive cutaneous anaphylaxis (PCA) reaction of expriment on anti-allergy, the allergy remarkably decreased in polyphenol fraction I, II and III groups in comparison with the control group. After antigen challenge, histamine content of blood of the polyphenol groups was lowered to near the normal group.

Effect of Mycelia Extracts from Lentinus edodes Mushroom-Cultured Lonicera japonica Thunberg on Anticancer and Antiallergy Activities (인동초로 배양한 표고버섯 균사체 추출물의 항암 및 알레르기 억제효과 검증)

  • This study was conducted to investigate the effect of mycelia of Lentinus edoes mushroom-cultured Lonicera japonica Thunberg (LLJ) on proliferation of the cancer cell lines (Hep3B, MCF-7 and HeLa), sarcoma 180 (S-180) and antiallergy. In an anti-cancer test using Hep3B (hepatic cancer cell), MCF-7 (breast cancer cell) and HeLa (uterine cancer cell), LLJ extract showed higher antiproliferating effect than that of LJ (Lonicera japonica Thunberg) extract. In an anti-cancer testing using Hep3B cells, LLJ extract showed growth-inhibitory effect of $85.60{\pm}4.66%$ at 3mg/rnL. In an anti-cancer testing using MCF-7 cells, LLJ and LJ extracts showed high antiproliferating effect. LLJ showed the tumor suppressive effect in mice injected with S-180 cells. The growth-inhibitory rates against tumor cells were 61% for LLJ, 37% for LJ. LLJ inhibited histamine release from rat peritoneal mast cells activated by compound 48/80. These results suggest that Lentinus edodes mushroom-cultured herb has an antiproliferating effect against cancer cell lines (Hep3B, MCF-7 and HeLa) and S-180 tumor, and will be beneficial in the treatment of allergic reaction.

The Inhibitory Effects of SoCheongRyongTang(SCRT) on Cytokine Production and Secretion of IgE in Highly Purified Mouse B cells (생쥐의 B 세포에서 IgE의 분비와 Cytokine 생산에 대한 소청룡탕(小靑龍湯)의 효과(效果))

  • Objective : We aimed to identify the inhibitory effects of SocheongRyoungTang on Cytokine Production and Secretion of IgE in Highly Purified Mouse B cells. This experiment was designed to investigate the effect of SoCheongRyongTang(SCRT) on Antiallergy. Materials and Methods : We measured the cytotoxic activity for cytokines transcript expression, production of $IL-1{\beta},\;IL-4,\;IL-6,\;IL-10,\;IFN-{\gamma},\;TNF-{\alpha},\;TGF-{\beta}$ proliferation of B cell in anti-CD40mAb plus rIL-4 plus HRF stimulated murine splenic B cells and histamine in anti-CD40mAb plus rIL-4 plus HRF stimulated mast cells. Results : 1. SCRT increased the gene synthesis of $IFN-{\gamma}(m-RNA)$, the appearance of IL-10, $IFN-{\gamma}$. 2. SCRT decreased the gene synthesis of $IL-l{\beta},\;IL-4,\;TGF-{\beta}(m-RNA)$ and the appearance of $IL-l{\beta},\;IL-4,\;TGF-{\beta},\;IgE$ significantly. Conclusion : SCRT decreased the proliferation of B cell significantly. According to the above results, it is suggested that SCRT extract might be usefully applied for prevention and treatment of Allergic disease.

A Review of Experimental Study for Herbal Medicines of Anti-allergic Effects (항알레르기 효능을 가진 한약물의 실험 연구 결과에 대한 고찰)

  • Objective : The purpose of this study is to survey the experimental studies for Herbal Medicines of Anti-allergic Effects by reviewing oriental medicine journals. Methods : We collected 48 research papers on Oriental herbal medicines (prescriptions, herbs and aqua-acupunctures) of Anti-allergic effects in the internet site OASIS using the keyword 'Antiallergy'. Then we analyzed them according to published journals, years, and research methods, etc. Results : 48 papers in relation to Oriental herbal medicines of anti-allergic effects have been published in many kinds of journals and shows a tendency to increase every year. There are 23 cases of using herb simplexes, 21 cases of herb-combined prescription, and 4 cases of aqua-acupuncture. Conclusions : This result will provide useful information of field of Oriental herbal medicine for the development of anti-allergic related herb simplexes and herb combined prescriptions. In the future, we have to perform more studies such as clinical trial and research papers on combined medication of the oriental and the western medicines for anti-allergic effects.

  • Although Oriental medicines have long been used effectively in treating many diseases throughout the world, the pharmacological mechanisms of most Oriental medicines used have not been defined. As part of our continuing search for biologically active antiallergic drugs from natural sources, Oriental medicines were analyzed. Some Oriental medicines have been used against various allergic diseases for generations, and still occupies an important place in traditional medicine in Korea. It is also still unclear how Oriental medicine prevents allergic disease in experimental animal models. Some Korean folk medicines inhibited the mast cell-mediated allergic reaction. This review summarizes the effective folk medicine in experimental effect of allergic reaction. Potential antiallergic folk medicines include: Poncirus trifoliata; Siegesbeckia glabrescence; Solanum lyratum; Aquilaria agallocha; Ulmi radicis; Polygonum tinctorium; Hwanglyun-Haedok-Tang; Rehmannia glutinosa; Kum- Hwag-San; Syzygium aromaticm; Spirulina platensis; Sosiho-Tang; Sinomenium acutum; Schizonepta tenuifolia; Shini-San; Magnoliae flos; Sochungryoung-Tang; Oryza sativa; Cryptotympana atrata; Salviae radix; Rosa davurica; Asiasari radix; Chung-Dae-San; and Cichorium intybus. Understanding the mechanisms of action for these Oriental medicines can permit drug development and laying of the ground-work for evaluating potential synergistic effects by addition and subtraction of prescriptions.

This experiment was designed to investigate the antiallergic and antiinflamatary the effects of Glycyrrhiza uralensis Fisch on asthma. Materials and Methods : We measured the eosinophil, IL-4, IL-5, IL-13, $IFN-{\gamma}$, CD4, CDS, CD69, CCR3, CD11b, Gr-1 in bronchoalveolar lavage fluid of ovalbumin induced asthmatic mice. Results : Glycyrrhiza uralensis Fisch increased the proliferation of eosinophils, IL-4, IL-5, IL-13, IgE, granulocyte, CCR3, CD4, IgE, CD69. Glycyrrhiza uralensis Fisch increased the proliferation of $IFN-{\gamma}$. Conclusion : Results suggest that Glycyrrhiza uralensis Fisch extract is useful in treatment and prevention allergic asthma.
