Effects of Edge Activator on the Droplet Size and Skin Permeation of Hydrated Liquid Crystalline Vesicles (Edge Activator가 수화 액정형 베시클의 입자크기와 피부 침투에 미치는 영향)

  • Hydrated liquid crystalline vesicles incorporating a edge activator, which confers flexibility to the vesicle membranes, were prepared and niacinamide was encapsulated in them. The formation of liquid crystalline phases and their thermal phase transitions were investigated by polarized optical microscopy and differential scanning calorimetry (DSC), respectively. Droplet sizes of the vesicles were reduced to several tens of nanometers by incorporating edge activators, such as sodium deoxycholate, lysolecithin, or polysorbate 80. The amount of niacinamide permeated into a pig skin increased greatly using the hydrated liquid crystalline vesicles compared to the case where niacinamide was applied in an aqueous solution state. The vesicles incorporating 10% sodium deoxycholate increased the amount of niacinamide permeated nearly four times. These results suggest that edge activators are effective in improving the skin permeability of vesicles.

  • For transdermal delivery of ceramides, various liposomes formulations were studied and evaluated. Sodium deoxycholate (SDC), Tween 20 and Span 85 were used as edge activators. The skin permeation of ceramides was performed using a Franz cell apparatus with hairless mouse skin. Among edge activators, SDC showed the higher values of deformability index and skin permeation than did others. For optimization of formulations, we varied the ratios of lipids to edge activators and the compositions between phosphatidylcholine (PC) and ceramides. The optimal ratio of lipid to SDC was observed to be 6:1 (w:w) and that of PC and ceramide was 1:1. Our results suggest that the skin permeation of ceramides could be enhanced by optimized deformable formulations of liposomes containing SDC as a major edge activator.

  • Cationic lipids have been used as one of the major components for making most promising non-viral gene delivery systems. whereas sodium cholate. an edge activator has been used as a surfactant in making ultradeformable and ultraflexible liposomes called Transfersomes. Using both a cationic lipid, DOTAP and sodium cholate. a newly formulated ultradeformable cationic liposome has been prepared. The average particle size of this formulation was approximately 80nm.   • To increase skin permeability of LMWH (Low Molecular Weight Heparin), ultradeformable liposomes were developed. Ultradeformable liposomes were developed by Egg phosphatidylcholine (Egg-PC) and edge activator. Entrapment efficiency, vesicle size and zeta potential of vesicles were determined and characterized for deformability and stability. Transepidermal permeation of LMWH was compared to saturated aqueous control in vitro. The steady-state flux and its maximum time were calculated from the flux curves. (omitted)

Hippo-YAP/TAZ signaling in angiogenesis

  • Angiogenesis is a complex, multistep process involving dynamic changes in endothelial cell (EC) shapes and behaviors, especially in specialized cell types such as tip cells (with active filopodial extensions), stalk cells (with less motility) and phalanx cells (with stable junction connections). The Hippo-Yes-associated protein (YAP)/ transcription activator with PDZ binding motif (TAZ) signaling plays a critical role in development, regeneration and organ size by regulating cell-cell contact and actin cytoskeleton dynamics. Recently, with the finding that YAP is expressed in the front edge of the developing retinal vessels, Hippo-YAP/TAZ signaling has emerged as a new pathway for blood vessel development. Intriguingly, the LATS1/2-mediated angiomotin (AMOT) family and YAP/TAZ activities contribute to EC shapes and behaviors by spatiotemporally modulating actin cytoskeleton dynamics and EC junction stability. Herein, we summarize the recent understanding of the role of Hippo-YAP/TAZ signaling in the processes of EC sprouting and junction maturation in angiogenesis.

  • The photoluminescence (PL) properties are studied for a new phosphor $BaZr(BO_{3})_{2}$ doped with $Eu^{3+}$ activator ion. The excitation spectrum shows strong absorption in the vacuum ultraviolet (VUV) region with an absorption band edge at 200 nm. The PL spectrum shows the strongest emission at 615 nm corresponding to the electric dipole $^{5}D_{0}\;{\rightarrow}\;^{7}F_{2}$ transition of $Eu^{3+}$, which results in a good color purity.

  • In this study, the elastic liposome consisted of egg phospholipids and edge activator ($Tego^{(R)}$ care 450) was prepared in order to supplement the defect of the conventional liposome. We prepared elastic liposome containing quercetin, known as natural antioxidant, and evaluated the vesicles size, elasticity, loading efficiency, stability, and in vitro skin permeation. The mean diameter of quercetin loaded elastic liposome formulations ranged between 208.2~303.4 nm and loading efficiency was observed 64.1~87.5%. The highest loading efficiency (87.5%) and deformability (28.3) were observed at the optimal ratio of 90 : 10 (egg phospholipids : $Tego^{(R)}$ care 450) among 0.1% quercetin loaded elastic liposome formulations. The elastic liposome formulation was selected for further transdermal permeation study. The elastic liposome ($129.9{\mu}g/cm^2$) exhibited more skin permeability than general liposome ($114.8{\mu}g/cm^2$) and 1,3-butylene glycol ($75.1{\mu}g/cm^2$) solution. This results suggest that the elastic liposome formulation using $Tego^{(R)}$ care 450 as a major edge activator could be useful for the delivery of active ingredient through the skin transdermal.

Growth and Scintillation Characteristics of CsI(Br) Single Crystals (CsI(Br) 단결정의 육성과 섬광특성)

  • CsI(Br) single crystals doped with 1, 3, 5 or 10 mole% $Br^-$ ions, as an activator, were grown by Czochralski method. The lattice structure of grown CsI(Br) single crystal was bcc and its lattice constant was $4.568\;{\AA}$. The absorption edge of the CsI(Br) single crystals was observed at 243 nm. The spectral range of the luminescence excited by 243 nm of wavelength was $300{\sim}600\;nm$, and its peak emission appeared at 440 nm. The luminescence intensity was maximum when CsI(Br) was doped with 3 mole % $Br^-$ ions. The energy resolutions of the CsI(Br) scintillator doped with 3 mole % $Br^-$ ions were 15.0% for $^{137}Cs$(662 keV), 13.1% for $^{54}Mn$(835 keV), and 18.0% and 6.3% for $^{22}Na$(511 keV and 1275 keV), respectively. The decay curves had fast and slow components, and the fast component was about 41 ns independent on the concentration of the $Br^-$ ions. The time resolution of CsI(Br) scintillators decreased with increasing of the concentration of $Br^-$ ions.

  • CsI(Li) single crystals doped with 0.02, 0.1, 0.2 and 0.3 mole% lithium as an activator were grown by Czochralski method. The lattice structure of grown CsI(Li) single crystal was bcc, its lattice constant was $4.568\;{\AA}$. The absorption edge of CsI(Li) single crystal was 245 nm, and the spectral range of luminescence was $300{\sim}600\;nm$, its maximum luminescence intensity appeared at 425 nm. The energy resolutions of CsI(Li) single crystal doped with 0.2 mole% lithium were 14.5% for $^{137}Cs$(662 keV), 11.4% for $^{54}Mn$(835 keV) and 17.7% and 7.9% for $^{22}Na$(511 keV and 1275 keV), respectively. The relation formula of $\gamma$-ray energy versus energy resolution was ln (FWHM%) = -0.893lnE + 8.456 and energy calibration formula was ${\log}E_r=1.455\;{\log}(ch.)-1.277$. The phosphorescence decay time of CsI(Li) crystal doped with 0.2 mole% lithium was 0.51 s at room temperature, and its time resolution measured by CFT(constant-fraction timing method) was 9.0 ns.

Elastic Liposome Formulation for Transdermal Delivery of Rutin (루틴의 피부 흡수 증진을 위한 탄성 리포좀 제형 연구)

  • In this study, we prepared elastic liposome containing rutin, known as antioxidants, and evaluated the physical characterization and enhanced skin permeation effect. The elastic liposome was prepared using the different ratios of egg phospholipids and $Tego^{(R)}$ care 450. The mean diameter of rutin loaded elastic liposomes formulations ranged between 205.7 ~ 298.0 nm and deforability 20.9 ~ 42.5, The loading efficiency was observed to be 52.0 ~ 71.0 %. The highest loading efficiency (71.0 %) and deformability (42.5) were observed at the optimal ratio of 85 : 15 (egg phospholipids : $Tego^{(R)}$ care 450) in the 0.1 % rutin loaded elastic liposome formulations. The elastic liposome formulation was selected for further transdermal permeation study. The elastic liposome(129.9 ${\mu}g/cm^2$) exhibited a significantly higher skin permeation compared with general liposome (98.0 ${\mu}g/cm^2$) and 1,3-butylene glycol (76.3 ${\mu}g/cm^2$) solution. These results suggest that the elastic liposome formulation using $Tego^{(R)}$ care 450 as a major edge activator could be useful for the delivery of active ingredient through the skin barrier.
