Synthesis of Allylthiopyridazine Derivatives and their Protective Effects of W-C Irradiation (알릴티오피리다진 유도체 합성 및 UV-C조사에 대한 방어효과)

  • Four 3-alkoxy-6-allylthiopyridazines and 3-chloro-6-allylthiopyridazine were synthesized and their protective effects against oxidative stress and UV-C irradiation were tested. 3-Methoxy-6-allylthiopyridazine and 3-ethoxy-6-allylthiopyridazine did not show protective effect on the oxidative stress but showed the strongest protective effect on UV-C irradiation among the tested compounds. Especially 500 $\mu\textrm{g}$/$m\ell$ of the two compounds was the most effective concentration.

  • Lightning surge damages of low voltage equipments in building are increasing due to increase in electrical and communication networks in the information-oriented society. And electronic equipments contained electrical circuits with semiconductor are very weak against lightning surge. The surge protective devices for electronic circuit in electronic equipments and AC power lines are becoming more widely. To achieve effective method of surge protection, there are needs for correlation between lightning surge protective effect of electronic equipment and installation method of surge protective device. This paper describes as a result of experiments for correlation between lightning surge protective effect and installation method of surge protective device.

  • Electronic equipments made from electrical circuits with small-sized semiconductor are very weak against lightning surge. So, electronic equipments is protected by SPD(surge protective devices) such as Zno varistor. The SPD protect electronic circuit in electronic equipments and AC power lines from the lightning surge. Therefore to achieve effective method of surge protection, there are needs for correlation between lightning surge protective effect and installation method of surge protective device. Therefore this paper describes as a result of experiments for correlation between lightning surge protective effect and installation method of surge protective device.

Protective Effect of Yangguksanwha-tang Metabolized by Liver Homogenate on Hypoxia-reperfusion Induced PC12 Cell Damage (간효소에 의해 대사된 양격산화탕의 저산소/재관류로부터 PC12 세포 보호효과)

  • The protective effect of Yangguksanwha-tang (YST) against hypoxia-reperfusion insult was investigated in PC12 cells. To elucidate the mechanism of the protective effect of YST, cell viability, the changes in activities of superoxide dismutase, glutathione peroxidase, catalase, caspase 3 and the production of malondialdehyde were observed after treating PC12 cells with YST which was metabolized by rat liver homogenate. Pretreatment of YST with liver homogenate appeared to increase its protective effect against hypoxia-reperfusion insult. The result showed that YST had the highest protective effect against hypoxia/reperfusion at the dose of $2\;{\mu}g/ml$ in PC12 cells, probably by recovering the redox enzyme activities and MDA to control level.

  • This research was performed to investigate the protective effect of Chungpesagan-tang (CST) from hypoxia/reperfusion induced-PC12 cell damage. To elucidate the mechanism of the protective effect of CST, cell viability, changes in activities of superoxide dismutase, glutathione peroxidase, catalase, caspase 3 and the production of malondialdehyde were observed after treating PC12 cells with CST which was metabolized by rat liver homogenate. Protective Effect of Bupleuri Radix on Hypoxia Reperfusion Induced by PC12 Cell Damage and Global Ischemia in Gerbil (PC12 손상 세포 및 전뇌허혈 유발 Gerbil에 대한 시호 세포보호효과)

  • Objects: This research was conducted to investigate the protective effect of Bupleuri Radix against ischemic damage using PC12 cells and global ischemia in gerbils, Methods: To observe the protective effect of Bupleuri Radixon ischemic damage, viability and changes in activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase and production of malondialdehyde (MDA) were observed after treating PC12 cells with Bupleuri Radix during ischemic damage. Gerbils were divided into three groups: a normal group, a 5-minute two-vessel occlusion (2VO) group and a Bupleun Radix administered group after 2VO. The CCAs were occluded by microclip for 5 minutes, Bupleuri Radix was administered orally for 7 days after 2VO. Histological analysis was performed on the 7th day. For histological analysis, the brain tissue was stained with 1 % of cresyl violet solution. Results: 1. Bupleuri Radix has a protective effect against ischemia in the CA1 area of the gerbil's hippocampus 7 days after 5-minute occlusion. 2. In the hypoxia/reperfusion model using PC12 cells, the Bupleuri Radix has a protective effect against ischemia in the dose of 0.2{\;}\mu\textrm{g}/ml,2{\;}\mu\textrm{g}/ml{\;}and{\;} 20{\;}\mu\textrm{g}/ml$. 3. Bupleuri Radix increased the activities of glutathione peroxidase and catalase. 4. The increased activity of superoxidedismutase (SOD) by ischemic damage might have been induced as an act of self-protection. This study suggests that Bupleuri Radix has some neuroprotective effect against neuronal damage following cerebral ischemia in vivo with a widely used experimental model of cerebral ischemia in Mongolian gerbils. Bupleuri Radix also has protective effect on a hypoxia/reperfusion cell culture model using PC12 cells. Conclusions: Bupleuri Radix has protective effect against ischemic brain damage during the early stages of ischemia.

To elucidate the mechanism of the protective effect of CPS on ischemic insult, cell viability and changes in activities of Superoxide dismutase, Glutathione Peroxidase, Catalase, Caspase 3 and the production of Malondialdehyde were observed after treating PC12 cells with CPS which was metabolized by rat liver homogenate. Pretreatment of CPS with liver homogenate increased its protective effect against ischemic insult by reducing the harmful effect of CPS itself. (omitted)
