Hypoglycemic Effects of Extract Mixture of Red Ginseng and Steamed RehManiae Radix on Streptozotocin-induced Diabetic Rats (홍삼 및 숙지황의 혼합추출물의 고혈당강하작용)

  • The effects of extract mixture of red ginseng and steamed Rehmaniae radix prepared for antidiabetic activity was examined in streptozotocin-Induced diabetic rats. The increased blood glucosec level in the streptozotocin-induced diabetic rats was significantly decreased by the treatment with the mixture (800, 1600 mg/kg, p.o.). However, neither red ginseng extract nor steamed Rehmaniae radix extract alone showed significant hypoglycemic effects. The mixture prevented a weight loss in streptozotocin-induced diabetic rats. These results suggest that the mixture has the relieving action against streptozotocin-induced hyperglycemia.

  • The effectsof garlic vinegar supplementation on body weight, blood blucose, and plasma triglyceride and cholesterol were investigated in streptozotocin -induced diabetic rats fed high-cholesterol(1%) diet for 4 wk. The garlic vinegar was made by fermenting 20% frsh garlic juice. There was no effect of garlic vinegar on body weight, plasma glucose or triglyceride concentrations in the diabetic rats. Plasma total -cholesterol concentrations were unaffected by garlic vinegarr supplementation. However, plasma LDL-cholesterol concentrations and atherogenic index were significantly lower in the diabetic rats supplemented with garlic vinegar diet than tin the cotnrol rats. The diabetic-rats supplemented with garlic vinegar not only had increased HDL-cholesterol levels but had decreased LDL-cholesterol. This alteration in the HDL/LDL-cholesterol ratio in the diabetic rats may decrease the risk of atheroscelrosis. Therefore, the over-all effect of garlic vinegar supplement may contribute to the antiatherogenic role in streptozotocin-induced diabetic rats.

  • This study evaluated effects of Yanggyuksanhwa-tang(YST) on diabetic rats. Diabetic condition in rats was induced by streptozotocin injection. Then control effect of YST was observed with changes of serum glucose, insulin, and triglyceride levels and hepatic glucokinse activity. YST treatment was resulted significant decrease of high serum glucose level of diabetic rats induced by streptozotocin at 5th day of YST treatment. YST treatment was resulted increase of low serum insulin level of diabetic rats induced by streptozotocin, but it was not significant statistically. YST treatment was resulted significant decrease of high serum triglyceride level of diabetic rats induced by streptozotocin at 3rd and 5th of YST treatment. YST treatment was resulted significant increase of low hepatic glucokinase activity of diabetic rats induced by streptozotocin at 5th day of YST treatment. These results suggest that YST has effects on serum glucose control against cerebral inchemic damage under diabetic condition.

Anti-diabetic Effect of Wen-Pi-Tang-Hab-Wu-Ling-San Extract in Streptozotocin-induced Diabetic Rats (Streptozotocin으로 유도한 당뇨병 쥐에서 $WHW^{(R)}$의 항당뇨 효과에 대한 연구)

  • Objectives : This study aimed to evaluate the anti-diabetic effect of Wen-Pi-Tang-Hab-Wu-Ling-San (WHW) extract in streptozotocin(STZ)-induced type-1 diabetic rats. Methods : Experimental diabetes were induced by intraperitoneal injection of streptozotocin (60 mg/kg). Two groups of STZ-induced diabetic rats were given the following treatments for 2 weeks by oral Administrations : (1) WHW 10 mg/kg, (2) WHW 100 mg/kg. In addition, vehicle-treated diabetic and nondiabetic controls were used in the experiment. The effects of WHW extract on STZ-induced diabetes were observed by measuring the changes of body weights and the levels of fasting blood glucose, insulin, urea nitrogen (BUN) and creatinine level in sera of rats, respectively. Results : In comparison control group, WHW-treated groups (100 mg/kg) were significantly decreased fasting blood glucose levels and increased serum insulin levels in STZ-induced diabetic rats. Moreover, WHW-treated groups (100 mg/kg) were reduced s-creatinie levels in STZ-induced diabetic rats. In addition, the changes related to diabetic nephropathy with body weight were significantly lower in WHW extract-dosing groups than in the diabetic control. Conclusions : The study thus showed that WHW extract enhanced the anti-diabetic effect in STZ-induced diabetic rats by improving the hypoglycemia. It also increased pancreatic insulin content in these rats.

Impaired Endothelium-Dependent Relaxation is Mediated by Reduced Production of Nitric Oxide in the Streptozotocin-Induced Diabetic Rats

  • To evaluate the involvement of nitric oxide production on the endothelium-dependent relaxation in diabetes, we have measured vascular and endothelial function and nitric oxide concentration, and the expression level of endothelial nitric oxide synthase in the streptozotocin-induced diabetic rats. Diabetic rats were induced by the injection of streptozotocin (50 mg/kg i.v.) in the Sprague-Dawley rats. Vasoconstrictor responses to norepinephrine (NE) showed that maximal contraction to norepinephrine $(10^{-5}\;M)$ was significantly enhanced in the aorta of diabetic rats. Endothelium-dependent relaxation induced by acetylcholine was markedly impaired in the aorta of diabetic rats, these responses were little improved by the pretreatment with indomethacin. However, endothelium-independent relaxation induced by nitroprusside was not altered in the diabetic rats. Plasma nitrite and nitrate $(NO_2/_3)$ levels in diabetic rats were significantly lower than in non-diabetic rats. Western blot analysis using a monoclonal antibody against endothelial cell nitric oxide synthase (eNOS) revealed that the protein level was lower in the aorta of diabetic rats than in non-diabetic rats. These data indicate that nitric oxide formation and eNOS expression is reduced in diabetes, and this would, in part, account for the impaired endothelium-dependent relaxation in the aorta of streptozotocin-induced diabetic rats.

  • In order to study the effects of Chungpyesagantang(淸肺瀉肝湯) on the diabetic rats induced by streptozotocin, during 15 days rats were administered Chungpyesagantang extract after streptozotocin injection (50mg/kg) (sample group). On 2nd, 9th, and 15th day, I investigated the levels of body weight, serum glucose, serum total cholestrol, serum triglyceride. The last day, I killed rats and investigated hepatic lipid peroxidase. The results were obtained as follows: 1. Body weight of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 15th day, increased effectively. (p〈0.05 respectively) 2. Glucose levels in serum of the diabetic rats induced by streptozotoc in and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.05 respectively) 3. Total cholesterol levels in serum of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.05 respectively ) 4. Triglyceride levels in serum of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.05 respectively) 5. On hepatic lipid peroxidethe of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.01 respectively)

  • Hypoglycemic efficacy of natural functional mixture(FM) and level of the diabete related hormones in streptozotocin(STZ)-induced diabetic rats were investigated. Male Sprague-Dawley rats were divided into three groups (normal, diabetic fed diets with/without FM). Supplement of FM did not affect the body weight and feed intake of STZ-induced diabetic rats. The increase in the weight of liver of STZ-induced diabetic rats was weakened by supplement of FM, whereas the weight of kidney and heart was not affected. Blood glucose level was slightly, and glucose tolerance of post-feeding was significantly improved by functional mixture. The mixture significantly reduced the elevated HbA1C level of diabetic rats by 15%, and it increased the level of insulin and C-peptide in blood and decreased glucagon level. Therefore, we conclude that FM in this study has a potency of prevention and treatment of diabetes mellitus.

Antidiabetic and Antioxidant Effects of Chunggugjang Powder in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발된 당뇨쥐에서 청국장 분말의 항당뇨 및 항산화 효과)

  • The purpose of this study was to investigate the effects of dietary chungguajang powder on blood glucose level and hepatic activities of antioxidant enzymes in normal and streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male ratt ($200{\sim}220g$) of six groups including normal group fed normal diet (N), diabetic group fed normal diet (C), diabetic groups fed chunggugjang powder diet (DC-1%, DC-5%, DC-10%, DC-20%) were used for the experiments. Diabetes was induced by single intraperitoneal injection of streptozotocin as a dose of 70 mg/kg body weight. After 3 weeks the animals were sacrificed and hepatic activities of antioxidant enzymes, serum level of glucose and organ weight were evaluated. Food and water intakes were higher in diabetic groups than normal group. Body weight gain and food efficiency ratio were lower in diabetic groups. However, they were higher in chunggugjang diet groups (DC) than normal diet group (C). The serum glucose levels were significantly lower (p<0.05) in the diabetic groups fed chunggugjang diet (DC-10%, DC-20%) than diabetic group fed normal diet (C). Hepatic activities of superoxide dismutase, catalase, glutathione peroxidase and glutathione S-transferase were lower in diabetic groups than normal group and they were significantly lower in diabetic groups fed chunggugjang diet (DC-20%) compared to diabetic rats fed normal diet (p<0.05). In conclusion, these results indicated that chunggugjang powder would have antidiabetic and antioxidant effects in STZ-induced diabetic rats.

Beneficial effects of Phellodendri Cortex extract on hyperglycemia and diabetic nephropathy in streptozotocin-induced diabetic rats

  • This study investigated the effect of Phellodendri Cortex extract on hyperglycemia and diabetic nephropathy in streptozotocin-induced diabetic rats. Male Sprague-Dawley rats were divided into normal control (NC), diabetic control (DC), and diabetic treatment with Phellodendri Cortex extract (DP). Over a 4-week experimental period, Phellodendri Cortex extract was administered orally at 379 mg/kg BW/day. The final fasting serum glucose level, urine total protein level, and relative left kidney weight in the DP group were significantly lower than the DC group. Renal XO and SOD activities in the DP group were significantly lower than the DC group and renal CAT activity in the DP group was significantly higher than the DC group. Tubular epithelial change was reduced in the DP group compared to the DC group. These results indicated that Phellodendri Cortex can reduce glucose level and prevent or retard the development of diabetic nephropathy in streptozotocin-induced diabetic rats.

Effects of Polygonatum odoratum on In vivo Insulin Activity in Streptozotocin-Induced Diabetic Rats (둥글레 섭취가 Streptozotocin 유발 당뇨병 쥐의 In vivo 인슐린 작용에 미치는 영향)

  • This study investigated the in vivo insulin function of Polygonatum odoratum in normal and diabetic male Sprague-Dawley rats. Diabetes mellitus was induced by an i.p. injection of streptozotocin. Normal and diabetic rats were assigned to the diet groups of the control basal diet and Polygonatum odoratum diet. The animals were fed the diet and water ad libitum for 15 days. Initial and final body weights, total food intake and serum glucose and insulin levels were measured. An insulin suppression test was performed to elucidate the insulin function in the peripheral tissues. The results showed that the final serum glucose levels significantly decreased in the diabetic rats on the Polygonatum odoratum diet compared with the diabetic rats on the control diet. The final serum insulin levels were increased in the diabetic rats on the Polygonatum odoratum diet compared with the diabetic rats on the control diet. The in vivo function of the insulin increased in the diabetic rats on the Polygonatum odoratum compared with the diabetic rats on the control diet. These data indicate that Polygonatum odoratum may be beneficial in improving the in vivo insulin function in streptozotocin-induced diabetic rats.
