Antimicrobial Activity of Extract

삼백초(Saururus chinensis Baill) 추출물의 항균활성

Abstract

In order to develop natural food preservatives, the ethanol and water extracts of the Saururus chinensis Baill were prepared. Antimicrobial activity was examined against 10 kinds of harmful microorganisms. The ethanol and water extracts showed the most active antimicrobial activity against B. subtilis and E. coli. The ethanol extract showed stronger antimicrobial activity than that of the water extract. However, the extracts did not show any antimicrobial activity against lactic acid bacteria and yeast. The minimum inhibitory concentrations (MIC) of ethanol extracts against B. subtilis and E. coli were 5 to 10 mg/mL, respectively. Antimicrobial activity of the ethanol extract was not destroyed at 40-12$0^{\circ}C$ and pH 3∼11. The ethanol extract was fractionated in the order of hexane, diethyl ether, ethyl acetate, and water fractions. The highest antimicrobial activity was found in the diethyl ether fraction.

삼백초 잎과 줄기의 에탄올 추출물과 그의 분획물을 일반세균, 젖산균 및 효모 총 10 균주에 대한 항균활성을 검색하고, 항균활성 물질의 미생물에 대한 최소저해농도와 증식에 미치는 영향 및 열과 pH에 대한 안정성, 용매분획 후 각 분획별 항균활성을 조사한 결과는 다음과 같다. 삼백초 에탄올추출물과 물추출물에서 항균활성 이 가장 높게 나타난 것은 그람양성균인 B. subtilis와 그람음성균인 E. coli이었으며, 에탄올추출물의 경우에는 물추출물보다 항균활성은 모든 균주에서 대부분 더 높게 나타났다. 삼백초 에탄올추출물에 대한 그람양성균과 그람음성균의 최소저해농도는 5∼10 mg/mL으로 나타났으며, 젖산균과 효모는 20 mg/mL 이하의 농도에서는 항균활성이 나타나지 않았다. 에탄올추출물이 미생물 증식에 미치는 영향은 B. subtilis와 E. coli이 경우 5mg/mL 이상의 농도에서 균증식이 현저하게 억제되었다. 젖산균과 효모는 에탄올 추출물의 농도가 증가함에 따라 항균활성이 증가하였으나,1.5 mg/mL 이하의 저농도에서는 균증식이 8시간까지 억제되다가 그 후 시간이 경과함에 따라 현저하게 균증식이 증가하였다. 에탄올 추출물이 대수 증식기에 미치는 영향은 5. aureus, E. coil의 경우 대조구에 비하여 균증식이 억제되었으며 2.5 mg/mL 이상의 농도에서는 농도에 따른 차이는 거의 나타나지 않았다. 항균활성 물질의 열 및 pH 안정성은 40∼ 12$0^{\circ}C$ 및 pH 3∼11까지 모든 균주의 생육환의 크기가 대조구와 비슷하게 나타나 열과 pH에 안정한 것으로 확인되었다. 에탄올추출물을 용매 분획한 결과 에테르 분획물의 항균활성이 가장 높았고, 그람양성균과 그람음성균의 항균활성은 젖산균과 효모보다 더 높게 나타났다.
