Antioxidative Activity and Physiological Function of the Angelica dahurica Roots

Abstract

We analyzed the contents of polyphenol compounds, the antioxidant activity and the physiological activity to investigate the functional effects of extracts from Angelica dahurica by the reflux water extraction (RW), reflux ethanol extraction (RE) and pressure heating water extraction (PW). The content of phenolic compounds of PW was the highest at 156.30 mg/g, and those of RW and RE were 31.69 mg/g and 26.34 mg/g, respectively. The electron donating ability (EDA) were in the range of 30.56% $\sim$ 52.74% and superoxide dismutase (SOD) like activity were 10.96% $\sim$ 23.24% at 1,000 ${\mu}g/mL$. The nitrite scavenging ability of PW at pH 1.2 was 61.47%, higher than those of RW (16.81%) and RE (17.78%). The xanthine oxidase inhibitory were 90.91% and tyrosinase inhibitory rate of RE was the highest (51.71%) at the concentration of 5,000 ${\mu}g/mL$. All extracts were increased with increments of the extract concentrations.

한방에서 생약재로 사용되는 백지를 새로운 기능성 소재로 활용하기 위한 연구의 일환으로 환류 추출방법 가압 열수 추출방법으로 물과 에탄올을 용매로 추출하여 각 추출물에 대한 폴리페놀, 전자공여능, SOD 유사활성능, 아질산염 소거능, xanthine oxidase 및 tyrosinase 저해 활성을 측정하여 백지의 생리활성에 대하여 분석하였다. 백지 추출물의 폴리페놀 함량은 25.34$\sim$156.30 mg/g으로 가압 열수 추출물인 PW가 환류 추출물보다 5배 이상 많이 함유된 것으로 분석되었다. 전자공여능은 1,000${\mu}g/mL$ 농도에서 30.56%$\sim$52.74%로 환류 물 추출물인 RW가 가장 높았으며, SOD 유사활성능은 10.96%$\sim$23.24%로 RE가 PW보다 2배 이상 높은 활성을 나타내었다. 아질산염 소거능은 pH 1.2의 1,000 ${\mu}g/mL$의 농도에서 16.81%$\sim$61.47%로 PW에서 가장 높은 소거효과를 나타내었으며 pH 3.0에서도 54.09%로 RW와 RE보다도 3배 이상의 높은 아질산염 소거능을 나타내었다. Xanthine oxidase 저해 활성은 5,000 ${\mu}g/mL$농도에서 85.92%$\sim$90.91%로 RW가 가장 높았으며, RE도 유사한 저해활성을 나타내었다. Tyrosinase 저해 활성은 18.74%$\sim$51.71%로 RE에서 가장 높았다. 에탄올을 용매로 이용한 RE는 SOD유사활성능과 tyrosinase저해 활성이 다른 추출물에 비해 높은 것으로 나타났으며, 가압 열수 추출물인 PW는 고온과 고압으로 가장 많은 폴리페놀이 추출되었으며, 아질산염 소거능도 매우 우수한 것으로 분석되었다. 그러므로 백지는 다량의 폴리페놀과 우수한 생리활성을 나타내므로 천연 항산화제 기능성 제품을 위한 원료로서 유용한 한방 생약자원으로 생각된다.
