Functional Properties of 50% Methanol Extracts from Different Parts of Acanthopanax sessiliflorus

오가피 부위별 50% 메탄올 추출액의 기능적 특성

Abstract

This study was carried out in order to investigate the functional properties of 50% methanol extracts from four parts (root, stem, leaf and fruit) of Acanthopanax sessiliflorus by means of measuring the contents of eleutheroside B, E, and total polyphenols as well as determining electron donating ability (EDA), nitrite scavenging ability (NSA), and anticancer activity. The highest contents of eleutheroside B and E were found in the fruit (538.99 ${\mu}g/g$) and the stem (556.00 ${\mu}g/g$). The root extract demonstrated the highest polyphenol content (2.97 mg/g). EDA of the stem and root extracts were 90.21% and 85.71%. All of the extracts showed 81.5-93.0% of NSA at pH 1.2. In addition all extracts indicated no cytotoxicity to normal cell line (DC2.4). The root extract had a 23% inhibitory effect against the stomach cancer cell line (SNU-719). These results revealed that 50% methanol extracts from A. sessiliflorus can be used as a potential resource of nutraceuticals.

오가피는 다양한 생리활성을 가진 약재로 사용되어왔다. 본 연구에서는 오가피 부위별 50% (v/v) 메탄올 추출액의 기능적 특징을 규명하기 위해 생리활성 물질(eleutheroside B, eleutheroside E)의 함량, 추출부위별 총 폴리페놀 함량, 항산화활성, 아질산염소거능 및 항암활성을 알아보았다. 오가피의 생리활성 표준물질인 eleutheroside E는 줄기(556.00 ${\mu}g/g$)>뿌리(430.22 ${\mu}g/g$)>열매 (32.70 ${\mu}g/g$) 순으로 함유되어 있었고 eleutheroside B는 열매 (538.99 ${\mu}g/g$)>줄기(423.60 ${\mu}g/g$)>뿌리(352.30 ${\mu}g/g$) 순으로 많이 함유되어 있었다. 총 폴리페놀은 뿌리(2.97 mg/g) 부위에 가장 많이 함유되어 있었고 잎, 줄기에 각각 0.95, 0.77 mg/g 함유되어 있었다. 전자공여능은 줄기 추출물에서 90.21%로 대조구인 1% (w/v) L-ascorbic acid의 전자공여능(91.75%)과 유사하게 높은 활성을 보였고 뿌리 추출물에서도 85.71%로 높은 활성을 보였다. 발암 전단계를 촉진하며 산성 환경에서 식품 중에 nitrosoamine을 생성하는 아질산염에 대한 오가피의 제거 효과는 추출부위와 관계없이 pH 1.2에서 81.5-93.0%로 높게 나타났다. pH가 높아짐에 따라 아질산염 소거능은 대개 소실되었으나 잎 추출액의 경우 pH 6.0에서도 62.9%의 아질산염 소거능을 유지하고 있었다. 각 부위별 추출액은 모두 정상세포인 DC2.4의 생육 저해 효과를 보이지 않아 세포독성은 없는 것으로 판단되었다. 뿌리 추출물에서는 위암세포주인 SNU-719에 대해 약 23%의 저해 효과가 발견되었다. 따라서 오가피 부위별 50% (v/v) 메탄올 추출액은 높은 항산화 활성과 아질산염 제거효과 등 다양한 기능성을 가진 식품소재로 활용이 가능 할 것으로 판단되었다.
