In Vitro Hepatoprotective Effects of Fermented Curcuma longa L. by Aspergillus oryzae against Alcohol-Induced Oxidative Stress

알코올성 산화적 손상에 대한 발효울금의 간세포 보호 효과

Abstract

Protective effects of fermented Curcuma longa L. (CL) against alcoholic liver damage were investigated in HepG2/2E1 cells. Fermented CL was extracted by cold water (FCC), hot water, 80% ethanol, and methanol. Of the four extracts, the strongest hepatoprotective effect against ethanol-induced oxidative stress was observed in FCC. Pretreatment with FCC also reduced intracellular reactive oxygen species formation compared to ethanol-alone treated cells. FCC also enhanced catalase, glutathione-S-transferase, glutathione reductase, glutathione peroxidase, and superoxide dismutase, and non-enzymatic antioxidative activities such as glutathione compared to alcohol-treated HepG2/2E1 cells. Our findings suggest that FCC might be considered as a useful agent in the prevention of liver damage induced by oxidative stress by increasing the antioxidant defense mechanism.

본 연구는 발효울금 추출물에서 알코올로 유발된 산화적 스트레스에 대한 간세포 손상에 대한 보호 효과를 평가하기 위해 발효울금을 열수 추출, 냉수 추출, 80% 에탄올 추출 및 메탄올 추출하여 HepG2/2E1 세포에서 알코올과 추출물을 함께 처리 후 세포생존율을 평가하고 세포생존율이 가장 높은 냉수 추출물에서 reactive oxygen species(ROS) 함량, 항산화 지표와 malondialdehyde(MDA) 함량을 측정하였다. 알코올에 대한 간 보호 효과에서는 알코올만 처리한 군의 세포생존율이 72.8%로 감소하였으나 발효울금 냉수 추출물을 함께 처리 시에 세포생존율이 86.1%로 유의적으로 증가하며 간 보호 효과를 확인하였다. 또한, 발효울금 냉수 추출물에서 세포 내 ROS 생성 억제능은 에탄올만 처리한 군에서 무처리군에 비해 유의적으로 ROS 생성이 증가하였으나 발효울금 냉수 추출물 $100{\mu}g/mL$, $250{\mu}g/mL$, $500{\mu}g/mL$ 농도 처리군 모두 알코올만 처리한 군과 비교 시 유의적인 차이를 보이며 ROS level을 감소시켰다. 항산화 지표에서 효소적 항산화인 catalase, glutathione-S-transferase, superoxide dismutase, glutathione peroxidase 및 glutathione reductase는 알코올만 처리한 군에서 무처리군보다 감소하였으나 발효울금 냉수 추출물 $500{\mu}g/mL$ 농도 처리군에서는 알코올만 처리한 군에 비해 유의적인 차이를 보이며 항산화 효소의 감소를 억제하였다. GSH 함량은 에탄올만 처리한 군에서 무처리군보다 유의적으로 함량이 감소하였으나 발효울금 냉수 추출물 $250{\mu}g/mL$, $500{\mu}g/mL$ 농도 처리군 모두 에탄올만 처리한 군에 비해 GSH 함량의 감소를 억제하였다. MDA 함량은 알코올만 처리한 군에서 무처리군에 비해 유의적으로 증가하였으나 발효울금 냉수 추출물 고농도 처리군($500{\mu}g/mL$)에서는 알코올만 처리한 군에 비해 유의적인 차이를 보이며 MDA 함량을 감소시켰다. 이상의 결과로부터 발효울금 추출물 중 냉수 추출물에서 알코올성 간 손상에 대한 간 보호 효과가 높게 나타나는 것을 확인하였다. 결과적으로 본 연구를 통해 발효울금을 냉수 추출하는 것은 발효울금의 알코올성 간 손상에 대한 간 보호 효과를 증대시키고 ROS 생성을 억제, 항산화 효소와 GSH 함량의 감소 억제 및 MDA 함량의 증가 억제 효과를 높이는 것으로 확인되었다. 이에 발효울금을 냉수로 처리하여 추출하는 것이 간 보호 효과를 증대시킬 수 있는 효과적인 방법이며, 이러한 방법을 통해 얻은 발효울금 냉수 추출물은 알코올로 유발된 산화적 스트레스에 대한 간세포 손상에 대한 보호 효과가 있는 기능성 소재로 활용될 수 있을 것으로 생각한다.
