Evaluation of antioxidant properties and oxidative stability of oregano seed solvent fraction

추출용매에 따른 오레가노 종자 분획물의 항산화 및 유지산화안정성 평가

Abstract

The in vitro antioxidant activity of oregano seed fractions, fractionizing with 80% ethanol, n-hexane, ethyl acetate, n-butanol, and water, was evaluated, and their effects on edible oils were determined in corn oil at 180℃. The ethyl acetate fraction had the highest radical-scavenging activity. The ferric reducing antioxidant power activity and total phenol content of the ethyl acetate fraction were determined as 6,130 µmol ascorbic acid equivalents/g extract and 770 µmol tannic acid equivalents/g extract, respectively, which were significantly higher than those of the other fractions (p<0.05). Primary and secondary oxidation products in corn oil added with the ethyl acetate fraction of oregano seed significantly decreased by 1.5 and 1.26 times, respectively, compared with those in the control groups. The major volatile ingredients in the ethyl acetate fraction of oregano seeds were determined to be carvacrol, thymoquinone, and 3-cyclopentylcyl-cyclopentan-1-one. Ethyl acetate is a suitable solvent for extracting antioxidant compounds from oregano seeds and can be used as a natural antioxidant.

본 연구는 항산화 활성이 우수한 것으로 알려진 오레가노 종자를 80% 에탄올, 헥세인, 에틸 아세테이트, 부탄올, 증류수로 분획하여 시험관 내 항산화 활성을 평가하고 180℃로 산화된 옥수수 기름에 적용하여 유지 산화에 미치는 영향을 확인하였다. DPPH 라디칼 소거 활성은 오레가노 종자 에틸 아세테이트 분획물의 라디칼 소거 활성이 92.3%으로 분획물 중 가장 높게 나타났으며, ABTS 양이온 라디칼 소거 활성 측정 결과 오레가노 종자 에틸 아세테이트 분획물의 ABTS 양이온 라디칼 소거 활성이 53.9%로 분획물 중 가장 높게 나타났다. FRAP 환원력은 오레가노 종자 에틸 아세테이트 분획물이 6,130 µmol ascorbic acid equivalent/g extract로 분획물 중 가장 높게 나타났으며, 총 페놀함량은 FRAP 환원력의 결과와 마찬가지로 오레가노 종자 에틸아세테이트 분획물이 770 µmol tannic acid equivalent/g extract로 분획물 중 가장 높은 결과를 나타내었다. 오레가노 종자 분획물간 헤드스페이스 산소 함량은 80분 이상 산화가 진행되면서 오레가노 종자 헥세인, 에틸 아세테이트, 80% 에탄올 분획물을 첨가한 시료군이 대조군에 비해 각각 1.25, 1.05, 1.02배 많은 산소함량 차이를 보였다. 유지의 일차 및 이차 산화생성물을 평가한 CDA, ρ-AV의 실험결과 CDA가는 오레가노 종자 에틸 아세테이트 분획물을 첨가하였을 때 대조군에 비해 conjugated dienes 생성이 1.5배 유의적으로 감소한 것을 관찰하였다. ρ-AV의 결과 또한 CDA가와 마찬가지로 오레가노 종자 에틸 아세테이트 분획물을 첨가한 유지 시료의 이차 산화물 양이 대조군에 비해 1.26배 유의적으로 감소된 것이 관찰되었다. 이와 더불어 오레가노 종자에틸 아세테이트 분획물의 GC 분석 결과 주요 휘발성 성분은 카바크롤, 타이모퀴논 등으로 나타났다. 이러한 오레가노 종자의 항산화 활성은 에틸 아세테이트를 사용한 분획물에서 크게 나타났으며, 이는 오레가노 종자가 가지고 있는 향기 성분 속 항산화물질이 실험에서 사용한 다른 용매들 보다 에틸 아세테이트 속에 다량 용출되어 높은 항산화 활성을 나타낸 것으로 사료되며, 오레가노 종자 에틸 아세테이트 분획물 속에 포함된 테르펜 및 폴리페놀 등의 물질로부터 기인하는 것으로 생각된다. 따라서 본 연구 결과, 향후 오레가노 종자를 천연항산화제로 활용하기 위하여 에틸 아세테이트 분획물의 활용이 유용할 것으로 판단된다.
